Abstract:Ochratoxin A (OTA) is a class of mycotoxin that are mainly produced by Aspergillus and Penicillium and widely found in plant origin food. OTA-contaminated foods can cause serious harm to animals and humans, while high stability of OTA makes it difficult to remove in conventional food processing. Thus, sensitive and rapid detection of OTA undoubtedly plays an important role in OTA prevention and control. In this paper, the conventional and novel methods of OTA at home and abroad are summarized and compared. The latest research progress and related applications of novel OTA electrochemical biosensors are mainly described with a new perspective. We innovatively divided the recognition element into single and combined recognition elements. Specifically, signal amplification technologies applied to the OTA electrochemical aptasensor are proposed. Furthermore, summary of the current limitations and future challenges in OTA analysis is included, which provide reference for the further research and applications.
CRISPR/Cas12a-Based Indirect Competitive Enzyme-Linked Immunosorbent Assay for Sensitive Detection of Ochratoxin A

Abstract:The high toxicity and widespread contamination of ochratoxin A (OTA) make it urgent to develop a sensitive method to detect trace OTA in complex food matrices. Herein, an indirect competitive enzyme-linked immunosorbent assay (icELISA)-based on the CRISPR/Cas12a system is described. DNA amplicons with multiple activation sequences of the CRISPR/Cas12a system were pre-prepared to improve detection sensitivity. In the absence of OTA, streptavidin-mediated biotinylated DNA amplicons were captured by the biotinylated secondary antibody on the microplate. The captured DNA amplicons activated the CRISPR/Cas12a system, which thereby effectively cleaved the reporter DNA, producing strong fluorescence. The presence of OTA led to a decrease in DNA amplicons on the microplate, resulting in a decrease in activated Cas12a and ultimately a drop in fluorescence intensity. OTA in food matrices at nanogram per milliliter levels can be detected. Therefore, the new method has great potential in monitoring OTA.

Abstract:We report herein a "turn off-on" fluorescent sensor based on ZnCdSe quantum dots (ZnCdSe QDs) and self-assembled zinc porphyrin for sensitive and rapid detection of ochratoxin A (OTA). Using environmentally friendly dodecyl dimethyl betaine as "soft template", zinc 5, 10, 15, 20-tetra(4-pyridyl)-21H-23H-porphine (ZnTPyP) was self-assembled into nanorods (SA-ZnTPyP) through a green process. The OTA detection process involves two steps: (1) the fluorescence of ZnCdSe QDs is significantly quenched by SA-ZnTPyP, and (2) the combination between SA-ZnTPyP and OTA releases ZnCdSe QDs from their quenchers such that the fluorescence is recovered. Under the optimized experimental conditions, the newly developed fluorescent sensor showed a linear response in concentration range 0.5-80 ng mL(-1), with a detection limit of 0.33 ng mL(-1). The excellent applicability of the system has been verified in milk and coffee samples, with relative recoveries between 98.33% and 103.70%. The proposed fluorescent sensor exhibits excellent selectivity and high sensitivity, and thus could be a promising tool for on-site determination of OTA in food safety control and environmental monitoring.
Development of a Magnetic Nanoparticles-Based Screen-Printed Electrodes (Mnps-Spes) Biosensor for the Quantification of Ochratoxin A in Cereal and Feed Samples

Abstract:A rapid and sensitive electrochemical biosensor based on magnetic nanoparticles and screen-printed electrodes (MNPs-SPEs sensor) was developed for the detection of ochratoxin A (OTA) in cereal and feed samples. Different types of magnetic nanoparticles-based ELISA (MNPs-ELISA) were optimized, and the signal detection, as well as sensitivity, was enhanced by the combined use of screen-printed electrodes (SPEs). Under the optimized conditions, the calibration curve of the MNPs-SPEs sensor was y = 0.3372x + 0.8324 (R2 = 0.9805). The linear range of detection and the detection limit were 0.01–0.82 ng/mL and 0.007 ng/mL, respectively. In addition, 50% inhibition (IC50) was detectable at 0.10 ng/mL. The limit of detection (LOD) of this MNPs-SPEs sensor in cereal and feed samples was 0.28 μg/kg. The recovery rates in spiked samples were between 78.7% and 113.5%, and the relative standard deviations (RSDs) were 3.6–9.8%, with the coefficient of variation lower than 15%. Parallel analysis of commercial samples (corn, wheat, and feedstuff) showed a good correlation between MNPs-SPEs sensor and liquid chromatography tandem mass spectrometry (LC/MS-MS). This new method provides a rapid, highly sensitive, and less time-consuming method to determine levels of ochratoxin A in cereal and feedstuff samples.

Abstract:Ochratoxin A (OTA) is a mycotoxin identified as a contaminant in grains and wine throughout the world, and convenient, rapid and sensitive detection methods for OTA have been a long-felt need for food safety monitoring. Herein, we presented a new competitive format based lateral flow strip fluorescent aptasensor for one-step determination of OTA in corn samples. Briefly, biotin-cDNA was immobilized on the surface of a nitrocellulose filter on the test line. Without OTA, Cy5-labeled aptamer combined with complementary strands formed a stable double helix. In the presence of OTA, however, the Cy5-aptamer/OTA complexes were generated, and therefore less free aptamer was captured in the test zone, leading to an obvious decrease in fluorescent signals on the test line. The test strip showed an excellent linear relationship in the range from 1 ng·mL-1 to 1000 ng·mL-1 with the LOD of 0.40 ng·mL-1, IC15 value of 3.46 ng·mL-1 and recoveries from 96.4% to 104.67% in spiked corn samples. Thus, the strip sensor developed in this study is an acceptable alternative for rapid detection of the OTA level in grain samples.

Abstract:The mycotoxin ochratoxin A (OTA) is a secondary metabolite derived from multiple Aspergillus and Penicillium strains. The development of a rapid, sensitive, and simple method for OTA detection is important to ensure food biosafety and safeguard public health. In this study, we designed a highly specific and sensitive assay for the detection of OTA using copper monosulfide (CuS) nanoparticles conjugated to an anti-OTA antibody (CuS-Ab NPs) and a fluorescent probe for Cu2+. When OTA is present in the solution, the OTA antigen, bound to the microplate, is competed off by the soluble OTA for binding to CuS-Ab NPs. After washing, the CuS-Ab NPs and bound OTA are removed. Subsequently, HCl is added to dissolve the CuS-Ab NPs bound to the OTA antigen, releasing Cu2+ and activating the Cu2+ fluorescent probe. Thus, the resultant fluorescence emission is inversely proportional to the OTA content in the solution. Under optimal conditions, this method detected 0.1–100 ng mL−1 OTA with a limit of detection of 0.01 ng mL−1. The assay was tested using corn, soybean, and coffee samples, with recoveries ranging from 94% to 110%. This strategy provides a new approach for the detection of mycotoxins and other small-molecule analytes with broad application potential in food safety and quality control.

Abstract:With the widespread contamination of ochratoxin A (OTA), it is of significant importance for detecting OTA in foods and traditional Chinese medicine (TCM). In this study, a novel label-free fluorescent aptasensor utilizing the interaction between OTA-triggered antiparallel G-quadruplex and (N-methyl-4-pyridy) porphyrin (TMPyP) for the rapid and sensitive determination of OTA was established. The fluorescence of CdTe quantum dots (QDs) could be quenched by TMPyP. In the presence of analyte (OTA), the aptamer could recognize OTA and transform from a random coil to the antiparallel G-quadruplex. The interaction between G-quadruplex and TMPyP could release CdTe QDs from TMPyP, and thus recover the fluorescence of CdTe QDs. Under optimized conditions, the detection limit of the designed aptasensor was 0.16 ng mL-1, with a linear range of 0.2 to 20 ng mL-1. Furthermore, this aptasensor showed high selectivity toward OTA against other structural analogs and other mycotoxins, and was successfully applied in Astragalus membranaceus samples. The presented aptasensor for OTA detection could be a promising tool for the field monitoring of food and TCM.

Abstract:Ochratoxin A(OTA), a highly toxic mycotoxin commonly found in food, poses a serious threat to health even at low concentrations. Developing a sensitive, accurate, simple, and cost-effective detection method is of great significance for food safety and quality control. Herein, a triple cascade amplification strategy was used to construct the colorimetric assay for the detection of OTA, where the amplification process consists of an entropy-driven DNA circuit (EDC), a catalytic hairpin assembly (CHA), and Mg2+-assisted DNAzyme catalysis (MNAzyme). Through the specific binding of ochratoxin A (OTA) and its aptamer, an initiator strand is released to initiate upstream EDC and then produce a new trigger unit that motivates downstream CHA to generate MNAzyme, which further cleaves the substrate strand to induce the formation of G-quadruplex/hemin DNAzyme as a signal readout. The aptasensor was shown to detect OTA, with a low detection limit of 8.7 fM and good selectivity. The developed method could be used as a highly colorimetric aptasensor for the detection of OTA in spiked rice samples.
Dual Flow Immunochromatographic Assay for Rapid and Simultaneous Quantitative Detection of Ochratoxin A and Zearalenone in Corn, Wheat, and Feed Samples

Abstract:A one-step dual flow immunochromatographic assay (DICGA), based on a competitive format, was developed for simultaneous quantification of ochratoxin A (OTA) and zearalenone (ZEN) in corn, wheat, and feed samples. The limit of detection for OTA was 0.32 ng/ml with a detection range of 0.53‒12.16 ng/ml, while for ZEN it was 0.58 ng/ml with a detection range of 1.06‒39.72 ng/ml. The recovery rates in corn, wheat, and feed samples ranged from 77.3% to 106.3% with the coefficient of variation lower than 15%. Naturally contaminated corn, wheat, and feed samples were analyzed using both DICGA and liquid chromatography-tandem mass spectrometry (LC-MS/MS) and the correlation between the two methods was evaluated using a regression analysis. The DICGA method shows great potential for simple, rapid, sensitive, and cost-effective quantitative detection of OTA and ZEN in food safety control.

Abstract:Ochratoxin A (OTA) is one of the most prevalent and toxic mycotoxins. Ultrasensitive and convenient detection of OTA is urgent demanded for public health. In this work, a dual-readout immunoassay was established for the detection of OTA based on Ce4+ oxidizing 3,3′,5,5′-tetramethylbenzidine (TMB) and Ce3+ inducing aggregation induced emission (AIE) of Au nanoclusters (AuNCs). Under alkaline phosphatase (ALP), the ascorbic acid 2-phosphate (AA2P) can form ascorbic acid (AA) by dephosphorylation. The AA can reduce Ce4+ to generate Ce3+, which induced the AIE of AuNCs to enhance the fluorescence intensity of AuNCs. Meanwhile, unreacted Ce4+ oxidized TMB to form blue oxTMB. Thus, a dual-readout immunoassay was developed based on AIE of AuNCs and TMB as substrate. The limits of detection (LODs) were as low as 0.62 ng/mL for fluorescent assay and 0.81 ng/mL for colorimetric assay. The recoveries of OTA from corn were 94.4%–107.7% for the fluorescent mode and 93.7%–106.9% for the colorimetric mode. The results verified that the cerium ions triggered dual-readout immunoassay was reliable to sensitive detect OTA in corn samples.

Abstract:In this work, a quantum dots (QDs) based label-free electrochemiluminescence (ECL) aptasensor was developed for the specific and sensitive detection of ochratoxin A (OTA). Chitosan (CHI) could immobilize abundant QDs on the surface of an Au electrode as the luminescent nanomaterials. Glutaraldehyde was used as the crosslinking agent for coupling a large number of OTA aptamers. Thanks to the excellent stability, good biocompatibility, and strong ECL intensity of QDs, as well as the quick reactions of the generated SO4•− in the electrolyte, strong ECL signals were measured. Because of the specific recognition of aptamer toward OTA, the reduced ECL signals caused by OTA in the samples were recorded for quantify the content of OTA. After optimizing a series of crucial conditions, the ECL aptasensor displayed superior sensitivity for OTA with a detection limit of 0.89 ng/mL and a wide linear concentration range of 1–100 ng/mL. The practicability and viability were verified through the rapid and facile analysis of OTA in real Lily and Rhubarb samples with recovery rates (n = 3) of 98.1–105.6% and 97.3–101.5%, respectively. The newly-developed QDs-based ECL aptasensor provided a new universal analytical tool for more mycotoxins in safety assessment of foods and feeds, environmental monitoring, and clinical diagnostics.

An Extremely Highly Sensitive ELISA in pg mL−1 Level Based on a Newly Produced Monoclonal Antibody for the Detection of Ochratoxin A in Food Samples

Abstract:In this study, an extremely highly sensitive enzyme-linked immunosorbent assay (ELISA) based on a newly produced monoclonal antibody (mAb) for the detection of ochratoxin A (OTA) in food samples was developed. OTA-Bovine serum albumin (BSA) conjugate was prepared and used as the immunogen for the production of the mAb. Among four hybridoma clones (8B10, 5C2, 9B7, and 5E11), the antibody from 8B10 displayed the highest affinity recognition for OTA. Based on the mAb (8B10), the IC50 and LOD of the ELISA for OTA were 34.8 pg mL−1 and 1.5 pg mL−1, respectively, which was 1.53~147 times lower than those in published ELISAs, indicating the ultra-sensitivity of our assay. There was no cross-reactivity of the mAb with the other four mycotoxins (AFB1, ZEN, DON, and T-2). Due to the high similarity in molecular structures among OTA, ochratoxin B (OTB), and ochratoxin C (OTC), the CR values of the mAb with OTB and OTC were 96.67% and 22.02%, respectively. Taking this advantage, the ELISA may be able to evaluate total ochratoxin levels in food samples. The recoveries of the ELISA for OTA in spiked samples (corn, wheat, and feed) were 96.5–110.8%, 89.5–94.4%, and 91.8–113.3%; and the RSDs were 5.2–13.6%, 8.2–13.0%, and 7.7–13.7% (n = 3), respectively. The spiked food samples (corn) were measured by ELISA and HPLC-FLD simultaneously. A good correlation between ELISA (x) and HPLC-FLD (y) with the linear regression equation of y = 0.918x − 0.034 (R2 = 0.985, n = 5) was obtained. These results demonstrated that the newly produced mAb-based ELISA was a feasible and ultra-sensitive analytical method for the detection of OTA in food samples.

Identification of a high-affinity monoclonal antibody against ochratoxin A and its application in enzyme-linked immunosorbent assay

Abstract:Ochratoxin A (OTA) is one of the most commonly occurring mycotoxins produced by some species of Aspergillus and can contaminate cereal and cereal products. A high-affinity anti-OTA monoclonal antibody (mAb) was generated from a hybridoma cell line 2D8 using splenocytes from a BALB/c mouse immunized with synthesized OTA-bovine serum albumin conjugate. The mAb 2D8 is specific with high affinity (3.75 × 10(9) L/M). An indirect competitive ELISA (ic-ELISA) was then developed using this mAb for quantitative determination of OTA in corn and feed samples. Using the optimized conditions, there was good linearity between OTA concentration and competitive inhibition (y = -0.6076x + 0.2441, R(2) = 0.9923) with the working range from 2.4 to 23.6 μg/kg, IC50 at 7.6 μg/kg and lower limit of detection at 1.4 μg/kg. The recovery rates in spiked samples were 91.2-110.3%. Of the 56 corn and feed samples, this ic-ELISA and a commercial kit both found the same 13 samples positive for OTA with good linear correlation between the two methods in OTA quantification (R(2) = 0.9706). We conclude that this ic-ELISA can be used for rapid and quantitative screening of corn and feed samples for the presence of OTA.
Electrochemiluminescence immunoassay system based on PCN-224-Mn and gold-platinum bimetallic nanoflowers for sensitive detection of ochratoxin A

Abstract:In this work, a novel Electrochemiluminescence Immunosensor was constructed using PCN-224-Mn and gold-platinum nanoflowers (AuPt NFs) for the ultrasensitive detection of ochratoxin A (OTA). PCN-224 modified with Mn (II) was synthesized as a probe material. The interaction efficiency of PCN-224 with S2O82- was also greatly improved. AuPt NFs were used as the substrate material for the electrodes. It has favorable biocompatibility, large specific surface area and can bind more antigen. Also greatly increased the electroactive surface area and conductivity of the electrode. OTA was detected using a competitive immunoassay strategy, in which OTA in the sample competes with the encapsulated antigen for a finite number of antibodies. ECLIA for the detection of OTA was designed to be highly sensitive, with a linear range from 0.0002 ng mL-1 to 1000 ng mL-1 and a LOD as low as 0.067 pg mL-1. In addition, it was evident from the electrochemical analyses that PCN-224-Mn had a stronger and more stable ECL signal compared to the plain PCN-224. The successful preparation of specific, sensitive and reproducible ECL immunosensors confirms the great promise for the detection of OTA or other small molecule mycotoxins.

Abstract:A polyvinylidene fluoride membrane-based dot immunoassay using nanobody (Nb) for rapid, qualitative, and visual detection of ochratoxin A (OTA) in cereals was developed. On the basis of optimal assay conditions, the cut-off level of this method assessed visually was 5 μg/kg for OTA, and the final results were obtained within 20 min. This method was simple with no time-consuming cleanup procedure. Good accuracy and reproducibility were obtained in recovery experiments. Results of the Nb-based dot ELISA are in agreement with the ELISA kit, except for samples that were negative for OTA presence assessed through Nb-based dot ELISA but found positive through the ELISA kit. Abstract:A highly selective and sensitive "on-off-on" electrochemiluminescence (ECL) aptasensor based on a self-enhanced luminophore was developed for the detection of ochratoxin A (OTA). Specifically, polyethyleneimine functionalized multi-walled carbon nanotubes decorated with gold nanoparticles (AuNPs-PEI-MWCNTs) were used as the electrode matrix to accelerate electron transfer and provide a favorable microenvironment for self-enhanced luminophore loading and ECL signal enhancement. In addition, black phosphorus quantum dots (BPQDs) were used as co-reactants of the ECL reagent tris (2,2'-bipyridyl) ruthenium(II) (Ru(bpy)32+) in ECL experiments, and the reaction mechanism was investigated. The self-enhanced luminophore Ru@SiO2-BPQDs was obtained by encapsulating Ru(bpy)32+ in silica (SiO2) nanoparticles and then combining it with BPQDs through electrostatic interaction. In conventional ECL systems, the emitter and its co-reactants reacted via the inter-nanoparticle pathway, leading to long distance electron transfer. However, the electron transfer distance in the self-enhanced luminophore was significantly shortened due to the intra-nanoparticle electron transfer pathway because BPQDs and oxidized Ru(bpy)32+ were bound within one nanoparticle, thereby improving ECL efficiency to achieve the first "switch-on" state. Then, the luminophore was quenched using ferrocenes (Fc) modified on an aptamer to achieve the "switch-off" state. Finally, OTA was specifically identified by the adapter, causing Fc to be released from the sensor interface, restoring the ECL intensity to achieve the second "switch-on" state. Under optimal conditions, the aptasensor exhibited good sensitivity, stability, and reproducibility, with a linear detection range from 0.1 to 320 ng/mL and a detection limit of 0.03 ng/mL. The novel ECL aptasensor provided a common analytical tool for the detection of mycotoxins and other small molecules.

Abstract:In this study, a novel aptasensor based on Aptamer/NH2 Janus particles is developed for the detection of Ochratoxin A(OTA). By coating gold on the hemispherical surface of the aminated polystyrene particles, Ochratoxin A aptamer is immobilized on the surface of the gold layer for selective identification and the other hemispherical able to bind to Glassy carbon electrode via peptide bond. Under optimum conditions, the sensor exhibited a wide dynamic range of OTA concentration from 1 x 10(-5) nM to 10 nM, and the detection limit is 3.3 x 10(-3) pM on condition of acceptable stability and reproducibility. The sensors were showed excellent performance in the detection of OTA in red wine sample with recoveries between 95.7% and 100.18%, which studied by the standard addition spiking technique. This work provides a new idea and method for preparing immune electrochemical sensors and is expected to be used for the OTA detection in red wine sample analysis.
Dramatically Enhancing the Sensitivity of Immunoassay for Ochratoxin A Detection by Cascade-Amplifying Enzyme Loading

Abstract:Enzyme-linked immunosorbent assay (ELISA) is widely used in the routine screening of mycotoxin contamination in various agricultural and food products. Herein, a cascade-amplifying system was introduced to dramatically promote the sensitivity of an immunoassay for ochratoxin A (OTA) detection. Specifically, a biotinylated M13 bacteriophage was introduced as a biofunctional competing antigen, in which a seven-peptide OTA mimotope fused on the p3 protein of M13 was used to specifically recognize an anti-OTA monoclonal antibody, and the biotin molecules modified on capsid p8 proteins were used in loading numerous streptavidin-labeled polymeric horseradish peroxidases (HRPs). Owing to the abundance of biotinylated p8 proteins in M13 and the high molar ratio between HRP and streptavidin in streptavidin-polyHRP, the loading amount of HRP enzymes on the M13 bacteriophage were greatly boosted. Hence, the proposed method exhibited high sensitivity, with a limit of detection of 2.0 pg/mL for OTA detection, which was 250-fold lower than that of conventional ELISA. In addition, the proposed method showed a slight cross-reaction of 2.3% to OTB, a negligible cross-reaction for other common mycotoxins, and an acceptable accuracy for OTA quantitative detection in real corn samples. The practicability of the method was further confirmed with a traditional HRP-based ELISA method. In conclusion, the biotinylated bacteriophage and polyHRP structure showed potential as a cascade-amplifying enzyme loading system for ultra-trace OTA detemination, and its application can be extended to the detection of other analytes by altering specific mimic peptide sequences.

Abstract:Ochratoxins are the major components of mycotoxin that exist extensively in plant origin food. In the ochratoxins group, ochratoxin A (OTA) as the most toxic compound, which possesses a high chemical and thermal stability in foodstuff and would do harm to human beings. Hence, it is imperative to develop a simple, rapid, sensitive, robust, and accurate way to determine OTA. Nanomaterial-based biosensors seem to be ideal analysis tools for identifying OTA on account of their tremendous merits, such as high sensitivity, excellent selectivity, simplicity of operation, and rapidity. This review focuses on the application and development of nanomaterial-enabled biosensors in OTA detection in the past years (2007–2018). Three dominant signal transduction mechanisms, i.e. optical, electrochemical, and piezoelectric, are discussed in details. Moreover, the major challenges and perspectives for future developments of OTA biosensors are also contained to provide an overview for the forthcoming research orientation.
