Abstract:Visible-light-induced radical decarboxylative functionalization of carboxylic acids and their derivatives has recently received considerable attention as a novel and efficient method to create CC and CX bonds. Generally, this visible-light-promoted decarboxylation process can smoothly occur under mild reaction conditions with a broad range of substrates and an excellent functional-group tolerance. The radical species formed from the decarboxylation step can participate in not only single photocatalytic transformations, but also dual-catalytic cross-coupling reactions by combining photoredox catalysis with other catalytic processes. Recent advances in this research area are discussed herein.

Abstract:Visible light promoted difunctionalization of alkynes is reviewed. The difunctionalization reaction is achieved by different reagents. Radicals such as carbon (sp3), carbon (sp2), and other heteroatom (P, S, N, Se, O, and halide) radicals initiated by visible light can undergo radical addition to a carbon-carbon triple bond. Upon further transformation, the desired difunctionalized products are obtained. Some organometallic complexes can be activated by visible light; the difunctionalization of alkynes is catalyzed by these species. Other reagents like 1,3-dipole precursors could also react with alkynes to give difunctionalization products; here, the 1,3-dipole derivatives are obtained by visible light photocatalysis. So far, the strategy has been succeeded in the formation of C–C bonds and C–X bonds. Several valuable chemical skeletons have been constructed under mild conditions. However, high regio- and stereoselectivities in some direct difunctionalization methodologies are yet to be achieved.

Abstract:A gentle and effective method for the photocatalytic dual functionalization of allenes with high regio- and stereoselectivity using a nonmetallic catalyst is described. Inexpensive and easily available sulfinates and TMSN3 were employed as sulfone and azido sources, respectively. The method is characterized by satisfactory substrate compatibility and tolerance toward functional groups. The straightforward initial mechanistic experiments suggested that the reaction could follow a radical pathway. The synthesis of vinylsulfone azide derivatives presented here offers a promising scaffold for the future development of vinyl sulfone-based drugs and functional bioorthogonal reagents.

Abstract:A general visible-light-induced photocatalyst-/additive-free strategy was developed for the construction of various nitrogen-heterocycles (42 examples, up to 97% yield) such as γ-lactams, δ-lactams, pyrrolidines, indolones, quinolinones, and fused polycyclic structures at room temperature. The prominent features of this protocol are mild and environmentally friendly conditions, broad substrate scope, and good functional group tolerance. Importantly, the reaction can be performed under natural sunlight, the most sustainable energy source imaginable. Furthermore, this strategy provides straightforward access to a range of derivatives through subsequent elaboration of the xanthate group.

Abstract:Herein, we describe a green and efficient photoredox catalytic C3-H alkoxycarbonylation between quinoxalin-2(1H)-ones or coumarins and readily available alkyloxalyl chlorides under ambient conditions. A series of quinoxaline-3-carbonyl and coumarin-3-carbonyl compounds are prepared through the radical addition of the in situ-generated alkoxycarbonyl radicals. Notably, this protocol features mild conditions, operational simplicity, and excellent functional group tolerability. More importantly, the carboxylated products can be readily derivatized into other important compounds that would be of great potential for the exploitation of pharmaceutically active compounds.

Abstract:We report a novel visible-light photoredox strategy for the efficient dehydroxylative alkylation of a wide array of α-hydroxy carboxylic acid derivatives using diaryl boron radical. The reaction features readily accessible starting materials, broad substrate scope with excellent functionality tolerance. Preliminary mechanistic studies reveal that the spin-center shift (SCS) process is responsible for the C-O bond activation, which is promoted by the diaryl boron radical generated from bench-stable and commercially available tetraphenyl borate (NaBPh4).

Abstract:3-Alkylated coumarins have many applications in medicinal chemistry, however, methods access to such structures are still limited. Herein, we report a site-selective photocatalytic deaminative alkylation of coumarins utilizing pyridinium-activated aliphatic primary amines as alkylation reagents. The protocol was highlighted by its mild reaction conditions, operational simplicity, and broad functional group compatibility. Moreover, this strategy enables late-stage modification of some pharmaceuticals and natural products, thus providing an appealing approach to valuable molecules in medicinal chemistry.

Abstract:Aryl radicals are important intermediates in organic synthesis. The generation of these reactive species via direct decarboxylation of inexpensive and readily available aromatic carboxylic acids is an attractive goal. However, such a process intrinsically exhibits high energy barriers to overcome, which in consequence usually require precious metal catalysis, stoichiometric oxidants and harsh conditions, suffering from limitations such as poor functional group tolerance and low atom economy. In recent years, visible‐light‐induced photochemical reactions have provided new approaches to address this challenge. Three major strategies have been introduced in this emerging field: 1) one‐pot in‐situ activation of benzoic acids to generate intermediates such as benzoyl hypobromites or hypoiodites; 2) the use of specialized photocatalysts like biphenyl/1,4‐dicyanobenzene to promote decarboxylation through photo‐induced electron transfer or charge transfer processes; 3) photo‐induced LMCT (Ligand‐to‐Metal Charge Transfer) strategy where copper or iron salts coordinate to the carboxylate anion and generate aromatic radicals upon visible light excitation. On the basis of these three strategies, this review will systematically summarize the development of visible‐light‐induced direct decarboxylative functionalization of aromatic carboxylic acids, focusing on the reaction mechanism and substrate scope, and discuss their prospects in organic synthesis.

Abstract:A photocatalytic acyl lactonization of unsaturated carboxylic acids using simple and inexpensive acyl chlorides has been developed for a modular synthesis of acyl lactones. Significantly, the simple protocol could allow an efficient construction of biologically important phthalide framework. The transformation could be extended to a wide variety of unsaturated carboxylic acids, including substituted 2‐vinyl benzoic acids and different types of alkenoic acids. Moreover, a series of aroyl, heteroaroyl chlorides could serve as coupling partners. Notably, functional groups including MeO, F, Cl and Br could survive. It is believed that acyl radicals generated from acyl chlorides under photoredox catalysis reacted with alkenes via atom‐transfer radical addition (ATRA) and triggered subsequent lactonization in the process. Preliminary mechanistic investigations revealed that the transformation probably proceed through a free radical pathway.

Abstract:A visible-light-induced radical-radical cross-coupling reaction between 1,3,4-oxadiazoles and hydroxamic acid derivatives has been realized under base- and metal-free conditions. The protocol was characterized by broad substrate scope, excellent functional group tolerance, and simple operation procedures. By using this protocol, a variety of biologically important 5-aryl-1,3,4-oxadiazole-2-methylamines were obtained in good yields with excellent chemoselectivity.

Abstract:Thermal C–C bond cleavage reactions allow the construction of structurally diverse molecular skeletons via predictable and efficient bond reorganizations. Visible light photoredox-catalyzed radical-mediated C–C bond cleavage reactions have recently emerged as a powerful alternative method for overcoming the thermodynamic and kinetic barrier of C–C bond cleavage in diverse molecular scaffolds. In recent years, a plethora of elegant and useful reactions have been invented, and the products are sometimes otherwise inaccessible by classic thermal reactions. Considering the great influence and synthetic potential of these reactions, we provide a summary of the state of art visible light-driven radical-mediated C–C bond cleavage/functionalization strategies with a specific emphasis on the working models. We hoped that this review will be useful for medicinal and synthetic organic chemists and will inspire further reaction development in this interesting area.This article has not yet been cited by other publications.

Abstract:Light-driven utilization of CO2 in organic synthesis is highly attractive because it mimics nature. However, such transformations are mainly limited to the incorporation of only a single CO2 molecule into organic compounds, far less than the number of CO2 molecules fixed in the product in photosynthesis. Here we report the visible-light photoredox-catalysed dicarboxylation of alkenes, allenes and (hetero)arenes with the incorporation of two CO2 molecules. This method realizes the formation of multiple C–C bonds with high chemo- and diastereoselectivities under mild conditions, which represents a simple, rapid and sustainable approach to valuable dicarboxylic acids. Moreover, this transition-metal-free protocol exhibits a low catalyst loading, good functional group tolerance, broad substrate scope, facile scalability and easy product derivatizations to give drug and material molecules. Mechanistic studies indicate a pathway by which a visible-light-induced two-electron reduction via sequential single electron transfer generates radical anions of such unsaturated substrates, broadening the repertoire of strategies.
