Abstract:The synthesis of primary amines via the reductive amination of alcohols involves a hydrogen-borrowing or hydrogen-transfer mechanism, which consists of three main steps: alcohol hydroxyl dehydrogenation, carbonyl imidization, and imine hydrogenation. Heterogeneous catalysts are widely used for this reaction because of their high performance and amenability to separation and reuse. However, the efficiency of reductive amination is limited by the dehydrogenation step, which is severely affected by the competitive adsorption of NH3. We hope to improve the efficiency of reductive amination by increasing dehydrogenation efficiency. Therefore, in this overview, we introduce the research progress of alcohol reductive amination reaction catalyzed by heterogeneous metal catalysts, focusing on methods of enhancing dehydrogenation efficiency by screening the metal component and the acidity/alkalinity of the support. Finally, we propose some new strategies for the preparation of catalysts from the perspective of overcoming the competitive adsorption of NH3 and speculate on the design and synthesis of novel catalysts with high performance in the future.

Abstract:The functionalization of remote unactivated C(sp3)-H and the reaction selectivity are among the core pursuits for transition-metal catalytic system development. Herein, we report Pd-catalyzed γ-C(sp3)-H-selective alkylation and alkenylation with removable 7-azaindole as a directing group. Acid and base were found to be the decisive regulators for the selective alkylation and alkenylation, respectively, on the same single substrate under otherwise the same reaction conditions. Various acrylates were compatible for the formation of C(sp3)-C(sp3) and C(sp3)-C(sp2) bonds. The alkenylation protocol could be further extended to acrylates with natural product units and α,β-unsaturated ketones. The preliminary synthetic manipulation of the alkylation and alkenylation products demonstrates the potential of this strategy for structurally diverse aliphatic chain extension and functionalization. Mechanistic experimental studies showed that the acidic and basic catalytic transformations shared the same six-membered dimer palladacycle.

Abstract:Amines are common scaffolds in bioactive molecules as well as building blocks for chemical synthesis. Despite significant advances in palladium-catalyzed C(sp3)−H functionalization of amines over the past decades, it remains challenging to perform directed C−H metalation with native amine groups rather than amine-derived or transient directing groups (DGs). Recently, our group developed the Pd(II)/sulfoxide-2-hydroxypyridine (SOHP) catalytic system, in which SOHP ligands play a pivotal role as a crucial functional module, enabling regioselective C(sp2)−H and enantioselective C(sp3)−H functionalization reactions. In this work, we demonstrate that this chemistry provides an ideal solution for native primary amine-directed γ-C(sp3)-H arylation. Primary amines of varying degrees of complexity, encompassing amino acid esters and aminol silyl ethers, were found to be compatible with the established methodology. Additionally, we achieved a preliminary implementation of the asymmetric variant of this γ-C−H arylation reaction by employing a chiral SOHP ligand. Moreover, the range of applicable substrates could be extended to pyridine, oxime ether, and pyridine-N-oxide.

Abstract:Remarkable progress has been made on chelation-assisted α and β C–H functionalization of aryl alkenes; however, there is no report on C–H allylation reactions. This work focuses on the first α and β C–H allylation of aryl alkenes using allyl carbonates to produce linear and branched 1,4-dienes enabled by chelation-assistance of pyridine-2-carboxamide, simply performed with Pd(OAc) 2 /AcOH in ethanol. This operationally simple protocol exhibited wide functionality tolerance and broad substrate scope and enabled successful gram-scaled preparation.

Abstract:An efficient palladium-catalyzed ortho-alkoxylation of N-benzoyl α-amino acid derivatives at room temperature has been explored. This novel transformation, using amino acids as directing groups, Pd(OAc)2 as catalyst, alcohols as the alkoxylation reagents, and PhI(OAc)2 as the oxidant, showed wide generality, good functional tolerance, and high monoselectivity and regioselectivity.

Abstract:The amino group is one of the most prevalent functional groups present in a wide range of organic molecules, which has been widely applied in various organic synthesis and pharmaceuticals. Alkyl amines could be converted in a single step into Katritzky salts which could serve as alkyl radical precursors to construct C‐C bonds and C‐X bonds via C(sp 3 )‐N cleavage. This deaminative functionalization could be carried out under transition‐metal catalysis, photocatalysis, carbene catalysis or base. In this paper, recent developments in the deaminative functionalization of alkyl amines were covered. This review was categorized into four parts according to the nature of the bond formation. Some representative examples and corresponding reaction mechanisms were also summarized.

Abstract:Amines are prominent in natural products, pharmaceutical agents and agrochemicals. Moreover, they are synthetically valuable building blocks for the construction of complex organic molecules and functional materials. However, amines, especially aliphatic and aromatic amines with free N–H bonds, tend to coordinate with transition metals and deactivate the catalyst, posing a tremendous challenge to applying Lewis basic amines in the amination of olefins. Here we present an example of oxidative amination of simple olefins with various Lewis basic amines. The combination of a palladium catalyst, 2,6-dimethyl-1,4-benzoquinone and a phosphorous ligand leads to the efficient synthesis of alkyl and aryl allylamines. A series of allylamines were obtained with good yields and excellent regio- and stereoselectivities. Intramolecular amination to synthesize tetrahydropyrrole and piperidine derivatives was also realized. Mechanistic investigations reveal that the reaction undergoes allylic C( sp 3 )–H activation and subsequent functionalization.

Abstract:A palladium-catalyzed monoarylation of β-methyl C(sp(3))-H of an alanine derivative with aryl iodides using an 8-aminoquinoline auxiliary is described. The reaction is highly efficient, scalable and compatible with a variety of functional groups with complete retention of chirality, providing a general and practical access to various β-aryl-α-amino acids. The synthetic potential of this protocol is further demonstrated in the sequential synthesis of diverse β-branched α-amino acids.

Abstract:The utilization of readily available amino acids, which is not only an oxygen nucleophile but also a nitrogen nucleophile, in palladium-catalyzed allylic substitution is realized under mild conditions. The chemoselectivity and multiple allylation are controlled by adjusting the reaction conditions. This represents the first example of this convenient access to valuable N,O-diallylated amino acids. Under the title conditions, a range of amino acids (α-, β-, γ-) and dipeptides can be readily converted in to the corresponding allylic products with excellent yields (67 examples, up to 99% yield) as well as good functional group tolerance.

Abstract:We report an arene-containing chiral primary amine as a dual aminocatalyst and ligand: the π-coordinating aminocatalyst/palladium synergistic catalysis for asymmetric allylic alkylation of α-branched β-ketocarbonyls. The use of arene-containing chiral primary amine catalyst led to not only enhanced reaction rate but also reversed chiral induction compared with its sterically bulky derivative. Both enantiomers of the allylic adducts bearing acyclic all-carbon quaternary stereocenters could be obtained from the same configured chiral aminocatalysts with high efficiency and excellent regio-, stereo-, and enantioselectivity. Mechanistic studies revealed a distinctive Pd-arene π-coordination mode for effective catalysis. The π-coordinating chiral primary amine catalyst could be successfully applied in the asymmetric allylation reactions of vinylethylene carbonates, vinyl epoxides, or simple allylic alcohols.

Abstract:A range of primary allylic amines were resolved with selectivity factors of up to 491 through [Pd(allyl)Cl](2)/(S)-BINAP-catalyzed and mesitylsulfonyl hydrazide-accelerated asymmetric allylic alkylation of malononitriles involving enantioselective C-N bond cleavage under aerobic conditions. Moreover, the reaction proved useful for the asymmetric synthesis of a-branched allyl-substituted malononitriles with high enantiopurity.

Abstract:AbstractDespite much progress has been made in the asymmetric α‐arylation reactions of cyclic ketones, lactones and lactams, the enantioselective α‐arylation of acyclic carbonyl compounds lagged much behind due to the in situ generated Z/E‐enolate intermediates leading to opposite enantiomers. Especially, the asymmetric α‐arylation of acyclic aldehydes is a long‐standing challenge, because of the highest activity among carbonyl compounds leading to various competitive side reactions. Herein we reported an efficient Pd‐catalyzed asymmetric intermolecular α‐arylation reaction of α‐alkyl‐α‐aryl disubstituted aldehydes with aryl bromides, which provides a rapid access to chiral aldehydes bearing an α‐all‐carbon quaternary stereocenter in moderate to good yields with good er in most cases. In addition, a pair of enantiomers could be easily prepared with the use of the same ligand by exchanging the aryl groups of aldehyde and aryl bromide.

Abstract:A palladium-catalyzed alkylation of primary and secondary C(sp(3))-H bonds with alkyl iodides and/or bromides for the synthesis of optically active unnatural alpha-amino acids (alpha-AAs) is described. This process is scalable and tolerates a variety of functional groups with complete retention of chirality, providing an efficient new strategy for the synthesis of various unnatural alpha-amino acid derivatives.
