Abstract:Chemical investigation of a deep sea sediment derived fungus Eutypella sp. MCCC 3A00281 resulted in the isolation of six new thiodiketopiperazine alkaloids, namely eutypellazines N-S (1-6). Their structures were elucidated on the basis of the extensive NMR and mass spectroscopic analysis, including the ECD data for the determination of absolute configuration. The structures of eutypellazines N-P (1-3) were characteristic of unique spirocyclic skeletons, while eutypellazines N-O bearing a spirocyclic tetrahydrobenzothiophene motif were found from wide type fungus for the first time. The biogenetic generation of the spirocyclic skeletons was postulated. Compounds 3-5 exhibited inhibitory effects against vancomycin-resistant enterococci (VRE), suggesting that they may be the potential inhibitors toward drug resistant pathogenic bacteria after the structural modification. Abstract:Cultivation of a deep-sea-derived fungus Eutypella sp. MCCC 3A00281 by a chemical epigenetic manipulation using suberohydroxamic acid (SBHA), a histone deacetylase inhibitor (HDI), resulted in the significant change of the metabolite profile. Chromatographic application of the extended metabolites led to the isolation of a total of 30 eremophilane-type sesquiterpenoids, of which 26 were identified as new entities namely eutyperemophilanes A-Z (1–26). Their structures were established by extensive analyses of the 1D and 2D NMR, and HRESIMS data, as well as the modified Mosher's method, X-ray crystallography, and ECD data for the assignment of their absolute configurations. Most eremophilanes from the fungal strain showed a trans-fusion of rings A and B, uncommonly found from nature. All compounds were evaluated for the inhibitory effects toward the nitric oxide (NO) production that was induced by lipopolysaccharide (LPS) in RAW 264.7 macrophage cells, while compounds 9 and 10 exerted significant inhibition.

Abstract:Five new diketopiperazine alkaloids, brevicompanines D-H (3-7), together with two known analogs, allobrevicompanine B (1) and fructigenine B (2), were isolated from a deep ocean sediment derived fungus Penicillium sp. Their structures were established by spectroscopic methods including 2D NMR and chiral HPLC analysis. Compounds 4 and 7 inhibited lipopolysaccharide (LPS)-induced nitric oxide production in BV2 microglial cells.

Abstract:Five new pyridone alkaloids Citridones H-L (1-5) along with one new natural product ent-Citridone A (6) were isolated from the marine-derived fungus Penicillium sp. XZD3-3, which came from the gut of a marine shrimp. The structures and absolute configurations of these compounds were elucidated on the basis of NMR, HRESIMS, UV, modified Mosher's method, single-crystal X-ray diffraction, and ECD calculation. Compounds 1 and 3 contained unprecedented polycyclic systems of phenyl-pyranopyridone and phenyl-spiro[cyclopentene-furopyridone], respectively. Compound 2 showed moderate inhibition activity against nitric oxide production with IC50 value of 52.5 mu M. All tested compounds were inactive in neither antibacterial nor cytotoxicity assays. Abstract:Eight new 12,8-eudesmanolide sesquiterpenes, eutypellaolides A–H ( 1 – 8 ), and two new eudesmane-type sesquiterpenes, eutypellaolides I–J ( 9 – 10 ), along with four known 12,8-eudesmanolide compounds 11 – 14 , were isolated from the culture extract of the polar fungus Eutypella sp. D-1 by one strain many compounds (OSMAC) approach. The structures of these compounds were determined through comprehensive spectroscopic data and experimental and calculated ECD analysis. Antibacterial, immunosuppressive, and PTP1B inhibition activities of these compounds were evaluated. Compounds 1 and 11 exhibited strong inhibitory activities against Bacillus subtilis and Staphylococcus aureus , with each showing an MIC value of 2 μg/mL. Compound 9 displayed weak immunosuppressive activity against ConA-induced T-cell proliferation with an inhibitory rate of 61.7% at a concentration of 19.8 μM. Compounds 5 , 11 , and 14 exhibited weak PTP1B inhibition activities with IC 50 values of 44.8, 43.2, and 49.5 μM, respectively.

Abstract:Three new polyketides, eutyketides A and B (1 and 2) and cytosporin X (3), along with four known compounds (4−7), were obtained from the marine-derived fungus Eutypella scoparia. The planar structures of 1 and 2 were elucidated by extensive HRMS and 1D and 2D NMR analyses. Their relative configurations of C-13 and C-14 were determined with chemical conversions by introducing an acetonylidene group. The absolute configurations of 1−3 were determined by comparing their experimental electronic circular dichroism (ECD) data with their computed ECD results. All of the isolated compounds were tested for their anti-inflammatory activities on lipopolysaccharide-induced nitric oxide production in RAW 264.7 macrophages. Compounds 5 and 6 showed stronger anti-inflammatory activities than the other compounds, with the inhibition of 49.0% and 54.9% at a concentration of 50.0 μg/mL, respectively.

Abstract:Four new alkaloids, including two new meleagrin analogs, meleagrin D ( 1 ) and E ( 2 ), and two new diketopiperazines, roquefortine H ( 3 ) and I ( 4 ), were isolated from a deep ocean sediment-derived fungus Penicillium sp. Meleagrin D ( 1 ) and E ( 2 ) possess unprecedented acetate–mevalonate-derived side chains on the imidazole moiety. These new meleagrins showed weak cytotoxicity against the A-549 cell line, whereas meleagrin B ( 5 ) and meleagrin ( 6 ), which were isolated previously from the same strain, induced HL-60 cell apoptosis or arrested the cell cycle through G 2 /M phase, respectively. The results indicate that the distinct substitutions on the imidazole ring significantly influence the cytotoxicity of the meleagrin alkaloids.

Abstract:Marine algae represent a prolific source of filamentous fungi for bioprospecting. In continuation of our search for new anticancer leads from fungi derived from the brown alga Fucus vesiculosus, an endophytic Pyrenochaetopsis sp. FVE-001 was selected for an in-depth chemical analysis. The crude fungal extract inhibited several cancer cell lines in vitro, and the highest anticancer activity was tracked to its CHCl3–soluble portion. A bioactivity-based molecular networking approach was applied to C18-SPE fractions of the CHCl3 subextract to predict the bioactivity scores of metabolites in the fractions and to aid targeted purification of anticancer metabolites. This approach led to a rapid isolation of three new decalinoylspirotetramic acid derivatives, pyrenosetins A–C (1–3) and the known decalin tetramic acid phomasetin (4). The structures of the compounds were elucidated by extensive NMR, HR-ESIMS, FT-IR spectroscopy, [α]D and Mosher’s ester method. Compounds 1 and 2 showed high anticancer activity against malignant melanoma cell line A-375 (IC50 values 2.8 and 6.3 μM, respectively), in line with the bioactivity predictions. This is the first study focusing on secondary metabolites of a marine-derived Pyrenochaetopsis sp. and the second investigation performed on the member of the genus Pyrenochaetopsis.

Abstract:Seven new alkaloids including tryptoquivaline K (1) and fumiquinazolines K-P (2-7), bearing a rare 1-aminocyclopropane- 1-carboxylic acid residue, together with six known compounds (8-13), were isolated from the fungus Aspergillus sp. obtained from the Mediterranean sponge Tethya aurantium. The structures of the new compounds were determined by extensive analysis by 1D and 2D NMR spectroscopy and mass spectrometry. The absolute configurations of tryptoquivaline K (1) and fumiquinazolines K and L (2, 3) were determined by TDDFT ECD calculations of their solution conformers, and the configurational assignment of the related fu-miquinazolines M-P (4-7) was achieved by comparison of their ECD spectra with those of compounds 2 and 3. All compounds were evaluated for their cytotoxic activity by the MTT method. Compound 8 exhibited pronounced cytotoxicity against the mouse lymphoma cell line (L5178Y) with an IC50 value of 3.6 M, but only moderate activity was observed against human ovarian cancer (A2780) and human Philadelphia chromosome-positive chronic myelogenous leukemia (K562) cell lines with IC50 values of 18.5 and 15.0 mu M, respectively.

Abstract:Two rare 6(D)/5(E) ring‐fused meroterpenoids, eupeniacetal A (1) and eupeniacetal B (2), and three new meroterpenoids, 1‐methoxy‐hydropreaustinoid A1 (3), hydroberkeleyone B (4), and 22‐deoxy‐10‐oxominiolutelide B (5), as well as five known meroterpenoids, 22‐deoxyminiolutelide B (6), preaustinoid A1 (7), berkeleyone C (8), berkeleyacetal A (9), and preaustinoid A3 (10), have been isolated, through the aid of LC–MS, from the sponge‐derived fungus Eupenicillium sp. 6A‐9. The structures of these meroterpenoids were elucidated by detailed spectroscopic analysis, comparison with literature data, and single‐crystal X‐ray diffraction. The absolute configurations were assigned by means of TDDFT/ECD and [α]D calculations as well as by X‐ray diffraction analysis. These compounds showed inhibitory activity towards TNF‐α production in the induced THP‐1 cell line but were inactive against normal tumor cell lines.

Abstract:Three new tetramic acid derivatives, tolypocladenols A1, A2, and B (1-3), a new pyridone alkaloid, tolypyridone A (4), and a new coumarin derivative, 3,8-dihydroxy-4-(4-hydroxyphenyl)-6-methylcoumarin (5), together with four known compounds (6-9) were isolated from the endolichenic fungus Tolypocladium cylindrosporum, which inhabits the lichen Lethariella zahlbruckneri. Structures of these compounds were determined by comprehensive analysis of spectroscopic data and single-crystal X-ray diffraction determination. Bioassay of the isolated compounds found that pyridoxatin (7) was cytotoxic to human cancer cells by induction of G0/G1 cell cycle arrest and apoptosis.

Abstract:Two new alkaloids tryptoquivaline Y (1) and pseurotin I (2), together with eight known compounds (3–10), were purified from a fungal strain Aspergillus felis FM324, which was isolated from a Hawaiian beach soil sample. The absolute configuration and physicochemical data of tryptoquivaline Z (3) were reported for the first time here in this paper. Compound 1 is an uncommon tryptoquivaline analog containing a 3- O -isobutanoyl group. The structures of the new compounds 1–2 and known compound 3 were elucidated through HRESIMS, NMR spectroscopy and ECD analysis. All the compounds were evaluated for their antiproliferative, antibacterial and NF-κB inhibitory activities. Compound 4 showed weak antibacterial activity against Staphylococcus aureus , methicillin resistant Staphylococcus aureus and Bacillus subtilis with the same MIC value of 59.2 μM. Compounds 3 and 2 inhibited NF-κB with IC 50 values of 26.7 and 30.9 μM, respectively.

Abstract:The fungi Eutypella could metabolize a myriad of natural products with unique structures and diverse bioactivities, which were deemed as key sources for lead compounds in drug discovery. Since the first research on the genus Eutypella in 2009, a myriad of secondary metabolites including terpenoids, alkaloids, and polyketides have been discovered in this genus, and most of them exhibited significant pharmacological activities. However, there are no systematic reviews that reported about the structures and bioactivities of Eutypella up to now. In this review, a total of 153 secondary metabolites and 42 references have been systematically summarized, and we found that the terpenoids (68.09%) and alkaloids (19.15%) were the new main components of this fungi, and the primary antiproliferative activity (64.89%) was mainly derived from the terpenoids and alkaloids. Thus, this review about the chemical diversity and biological activities of the metabolites from the fungus Eutypella provided a new perspective for the development of new drugs for pharmacologists.

Abstract:Three new polyketides, a griseofulvin derivative 1 , a hydroanthraquinone derivative 8 and a pyranolactone derivative 10 , together with eight known compounds ( 2 – 7 , 9 and 11 ), were isolated from the marine-derived fungus Nigrospora sp. MG36–1. The structures of the three new compounds were unambiguously determined by nuclear magnetic resonance (NMR), mass spectrometry, 13 C NMR calculation in combination with DP4+ and ECD calculations. The antitumor, antibacterial and antifungal activities of the compounds 1 – 9 were evaluated in vitro. Compound 1 showed antibacterial activity against Acinetobacter baumannii with MIC 42.5 μg/mL. Compounds 1 and 8 exhibited antifungal activity against Candida albicans with MICs 21.5 μg/mL and 17.5 μg/mL, respectively.

Abstract:The novel diketopiperazine alkaloid toxicocladine A ( 1 ), two new natural products cyclo(L-Phe-ΔAla) ( 2 ) and cyclo(L-Tyr-ΔAla) ( 3 ), and three known compounds protocatechuic acid ( 4 ), protocatechuic acid methyl ester ( 5 ) and ergosterol peroxide ( 6 ), were isolated from the fungus Toxicocladosporium sp. CYH-18 collected from the Haima cold seep. The structures and absolute configurations of the compounds were determined via NMR, MS, optical rotation, and X-ray diffraction analysis and by comparison with the literature. Structurally, compound 1 was a diketopiperazine with an unprecedented methylene sulfide bridge between C-5 and C-8, while compounds 2 and 3 were diketopiperazines with terminal olefins. Compounds 4 and 5 showed antioxidant activity. Compound 6 displayed antibacterial activity against Bacillus subtilis and Exiguobacterium profundum .

Chlorinated Metabolites with Antibacterial Activities from a Deep-Sea-derived Spiromastix Fungus

Abstract:Chromatographic separation of the solid cultures of a deep-sea-derived Spiromastix fungus (MCCC 3A00308) resulted in the isolation of eight compounds. Their structures were identified on the basis of the spectroscopic data. Compounds 1-8 are classified as depsidone-type (1-4), isocoumarin-type (5 and 6), and benzothiazole-type (7 and 8), of which 1-7 are new compounds and 1-3 along with 5 and 6 are chlorinated. Compound 3 is characterized by trichlorination and shows potent activities against Gram-positive pathogenic bacteria including Staphylococcus aureus ATCC 25923, Bacillus thuringiensis ATCC 10792, and Bacillus subtilis CMCC 63501, with minimum inhibitory concentration (MIC) values ranging from 0.5 to 1.0 μg mL-1. This study extends the chemical diversity of chlorinated natural products from marine-derived fungi and provides a promising lead for the development of antibacterial agents.
