Abstract:Polybrominated diphenyl ethers (PBDEs) have been shown to have a disruptive effect on the thyroid hormone system, and one possible mechanism is the direct binding of their hydroxylated metabolites (HO-PBDEs) to thyroid hormone receptors (TRs). However, the experimental data on the thyroid hormone activity of HO-PBDEs are limited, and the molecular interaction mechanism remains unclear, impeding the ecological risk assessment for these widespread contaminants. In the present research, a quantum chemical approach was developed to predict the thyroid hormone activity of HO-PBDEs using the electronic structure parameters of neutral molecules. The ab initio HF/6-31G** algorithm was employed to optimize the molecular geometry and to calculate local molecular parameters regarding effective energy and electron transfer amount. The mechanistic analysis shows that the ability of the hydroxyl oxygen and hydrogen atom to donate or accept additional electron charges is an important property affecting the chemical activity of the thyroid hormone. The derived regression model was shown to have a good statistical performance and could be used to predict the thyroid hormone activity of other HO-PBDE congeners for which experimental measurements are not possible or are restricted. Therefore, the model has the potential to be a useful tool in the application of integrated testing strategies.

Abstract:Endocrine-disrupting chemicals have attracted great concern. As major metabolites of polychlorinated biphenyls (PCBs), hydroxylated polychlorinated biphenyls (HO-PCBs) may disrupt estrogen hormone status because of their structural similarity to estrogen endogenous compounds. However, interactions between HO-PCBs and estrogen receptors (ERs) are not fully understood. In the present work, a molecular modeling study combining molecular docking, molecular dynamics simulations, and binding free energy calculations was performed to characterize the interactions of three HO-PCBs (4′-HO-PCB50, 2′-HO-PCB65, and 4′-HO-PCB69) having much different estrogenic activities with ERβ. Docking results showed that binding between ligands and ERβ was stabilized by hydrogen bond and hydrophobic interactions. The binding free energies of three ligands with ERβ were calculated, and further binding free energy decomposition analysis indicated that the dominating driving force of the binding between the ligands and ERβ was the van der Waals interaction. Some key residues, such as Leu298, Phe356, Gly472, His475, and Leu476, played important roles in ligand–receptor interactions by forming hydrophobic and hydrogen bond interactions with ligands. The results may be beneficial to increase understanding of the interactions between HO-PCBs and ERβ.
Binding modes of environmental endocrine disruptors to human serum albumin: insights from STD-NMR, ITC, spectroscopic and molecular docking studies.

Abstract:Given that bisphenols have an endocrine-disrupting effect on human bodies, thoroughly exposing their potential effects at the molecular level is important. Saturation transfer difference (STD) NMR-based binding studies were performed to investigate the binding potential of two bisphenol representatives, namely, bisphenol B (BPB) and bisphenol E (BPE), toward human serum albumin (HSA). The relative STD (%) suggested that BPB and BPE show similar binding modes and orientations, in which the phenolic rings were spatially close to HSA binding site. ITC analysis results showed that BPB and BPE were bound to HSA with moderately strong binding affinity through electrostatic interactions and hydrogen bonds. The order of binding affinity of HSA for two test bisphenols is as follows: BPE > BPB. The results of fluorescence competitive experiments using 5-dimethylaminonaphthalene-1-sulfonamide and dansylsarcosine as competitors, combined with molecular docking indicated that both bisphenols are prone to attach to the binding site II in HSA. Spectroscopic results (FT-IR, CD, synchronous and 3D fluorescence spectra) showed that BPB/BPE induces different degrees of microenvironmental and conformational changes to HSA.

Abstract:Bisphenol F (BPF) as an endocrine disrupting compounds (EDCs) pollutant in the environment poses a great threat to human health. To evaluate the toxicity of BPF at the protein level, the effects of BPF on human serum albumin (HSA) were investigated at three temperatures 283, 298, and 308 K by multiple spectroscopic techniques. The experimental results showed that BPF effectively quenched the intrinsic fluorescence of HSA via static quenching. The number of binding sites, the binding constant, the thermodynamic parameters and the binding subdomain were measured, and indicated that BPF could spontaneously bind with HSA on subdomain IIA through H-bond and van der Waals interactions. Furthermore, the conformation of HSA was demonstrably changed in the presence of BPF. The work provides accurate and full basic data for clarifying the binding mechanisms of BPF with HSA in vivo and is helpful for understanding its effect on protein function during its transportation and distribution in blood.

Abstract:Polybrominated diphenyl ethers (PBDEs) are becoming a public health concern because of their potential toxicity, from endocrine disruption system to neurodevelopmental impairments. Nonetheless, information on their levels in human blood is scarce. In this study, human serum samples collected in Shanghai, China, were analyzed for the concentrations of PBDEs and their hydroxylated metabolites (OH-PBDEs). Eight PBDE congeners and six OH-PBDE congeners were quantified in serum samples by gas chromatography with mass spectrometry (GC-MS) and high-performance liquid chromatography with tandem mass spectrometry (HPLC-MS/MS). As a result, total PBDE concentration ranged from 0.280 to 12.330 ng g−1 on a lipid weight basis lw (median: 1.100 ng g−1 lw) and the total OH-PBDE level ranged from 0.045 to 0.363 ng g−1 (lw) (median: 0.187 ng g−1 lw). Among them, BDE-47 and 6-OH-BDE-47 were the predominant PBDEs and OH-PBDEs, respectively. In addition, based on the results of the Bartelett X2 test, BDE-47 significantly (p < 0.05) correlated with BDE-28, BDE-100, BDE-85, and BDE-154, whereas 3′-OH-BDE-7 significantly (p < 0.01) correlated with 3-OH-BDE-47, 2-OH-BDE-68, and 6′-OH-BDE-99. Among all donors, no significant association between age and PBDEs (or OH-PBDEs) was found. Further research on the exposure routes in the environment and metabolic processing of PBDEs in human blood is necessary.

Abstract:Hydroxylated (OH-) and methoxylated (MeO-) polybrominated diphenyl ethers (PBDE) are suspected endocrine disruptors. Little is known about the accumulation or sources of these chemicals in tissues of humans, particularly those residing in Hong Kong, which is one of the most densely populated cities in the world. Seven MeO-BDEs, fifteen OH-BDEs and three bromophenols (BRPs) were analyzed in blood plasma of 116 humans that had been collected by the Hong Kong Red Cross. Total concentrations of MeO-BDEs, OH-BDEs and BRPs ranged from 3.8 × 102 to 52 × 103 pg g− 1 lipid (median 4.5 × 103 pg g− 1), 5.3 to 4.9 × 102 pg g− 1 lipid (81 pg g− 1) and ND to 1.1 × 102 pg g− 1 lipid (3.7 pg g− 1), respectively. 3-MeO-BDE-47, 6-OH-BDE-47 and 2, 4, 5-TBP were the predominant MeO-BDEs, OH-BDEs and BRPs, respectively. These results are consistent with accumulation of MeO-BDEs, OH-BDEs and BRPs in human plasma being primarily from natural products and inter-conversion of natural products. Coefficients of determination for some pairs of congeners such as 3-OH-BDE-100 and 6-OH-BDE-47, 6-OH-BDE-85 and 5′-OH-BDE-99, and 2, 4-DBP and 6-OH-BDE-85, were near 1.0, which is consistent with them having common sources. Patterns of relative concentrations of the target analytes were similar in the diet, particularly fish, as in blood plasma of humans, which suggests that the diet and particularly seafood might be a source of these compounds and PBDEs. Furthermore, biotransformation of natural chemicals such as OH-BDEs to BRPs might be the primary route of their elimination from humans.

Abstract:Polybrominated diphenyl ethers (PBDEs), a specific group of brominated flame retardants (BFRs), are widely used in household and industrial products, including electronics and household furnishings. PBDEs and their metabolites have recently been reported to be present in various environmental and biotic samples. PBDEs have reproductive and developmental toxicity, immunity toxicity, neurotoxicity and endocrine disruption toxicity. This paper summarizes the endocrine disrupting activity of PBDEs and their metabolites, mainly focusing on disrupting thyroid hormone, estrogen hormone, androgen hormone and various enzyme activities (e.g. aromatase CYP19, CYP17 enzyme and estradiol sulfotransferase E2SULT, which affect the transformation and metabolism of sex hormone), and analyzes the structure feature of PBDE compounds corresponding to different endpoints. It is found that PBDEs metabolites exhibit higher endocrine disrupting activity than PBDEs parent compounds do, especially HO -PBDEs induce serious endocrine disrupting interaction. The endocrine disrupting activities of PBDEs need metabolism in vitro. In order to evaluate environmental and human health risks from PBDEs exposure, their toxicity action mechanisms and quantitative structure-activity relationship (QSAR) should be investigated further.

Abstract:Seven polybrominated diphenyl ethers (PBDEs) and eleven hydroxylated PBDEs (OH-PBDEs) were selected to investigate their contamination status in 32 female serum samples from Dalian, China. These studied PBDEs and OH-PBDEs were found in most of the serum samples with the concentrations of ΣPBDEs and ΣOH-PBDEs ranging from 1.07 to 38.7ng/g lipid (median: 5.56ng/g lipid) and from 0.011 to 0.436ng/g lipid (median: 0.069ng/g lipid), respectively. More impressively, BDE-47 and OH-BDE-47 were the predominant congeners in female serum, accounting for 52% of ΣPBDEs and 46% of ΣOH-PBDEs, respectively. Correspondence analysis indicated that significant (p<0.05) positive correlations were observed between BDE-47 and its metabolites such as 6-OH-BDE-47, 3-OH-BDE-47 and 4'-OH-BDE-49. In addition, the positive significant (p<0.05) correlation was shown between ∑PBDEs and lipid contents but between concentrations of ∑PBDEs and ages or BMIs, as well as for ∑OH-PBDEs.

Abstract:RATIONALE: Bisphenol A and its alternatives are widely used in common consumer products and are known as environmental endocrine disrupting chemicals. Five bisphenol analogues, namely, tetrabromobisphenol A (TBBPA), tetrachlorobisphenol A (TCBPA), 4,4'-sulfonyldiphenol (BPS), bisphenol A (BPA) and 4,4'-hexafluoroisopropylidenediphenol (BPAF), were selected to study their interactions with serum albumins, aiming at a better understanding of the toxicological mechanisms of bisphenol compounds. METHODS: The interactions between human and bovine serum albumins (HSA and BSA) with these five compounds were investigated by electrospray ionization mass spectrometry (ESI-MS). Fluorescence spectroscopy and molecular docking confirmed the ESI-MS observations and provided complementary information with respect to thermodynamic properties and binding modes. RESULTS: TBBPA showed the highest binding ability with HSA and BSA, followed by TBBPA and BPS, whereas BPA and BPAF exhibited little or no binding with these serum albumins. The calculated thermodynamic parameters suggested that hydrogen bonds and electrostatic forces played important roles for these interactions. Binding energies of TBBPA, TCBPA, and BPS calculated by molecular docking were -35.18, -34.39, and -25.89 kJ mol(-1), respectively, in good agreement with ESI-MS measurements. CONCLUSIONS: The results of this study showed that halogenated substituents on the phenolic rings of bisphenol could enhance the binding ability with serum albumins. This work could provide useful information for further research on the relationship between molecular structures and toxicities of bisphenols. Copyright (C) 2016 John Wiley & Sons, Ltd.

Abstract:Polybrominated diphenyl ethers (PBDEs),as additive brominated flame retardants,are widely used in a variety of industrial and commercial products.PBDEs are persistent organic pollutants with environmental endocrine disruption effect,neurotoxicity,and reproductive toxicity.PBDEs can be metabolized into hydroxylated PBDEs (OH-PBDEs) and methylated into methoxylated PBDEs (MeO-PBDEs) in vivo.Recently,PBDEs and their metabolites (OH-PBDEs,MeO-PBDEs) and bromophenols have raised social concerns due to the detection of above-mentioned chemicals in environment and organisms.In this review,the exposure levels and the methods of pretreatment and detection of PBDEs and their metabolites in environment and human body were summarized,aiming to provide methodological references for the exposure assessment and toxicological research of PBDEs.

Abstract:In this study, the interaction of the endocrine disruptor bisphenol A (BPA) and human serum albumin (HSA) was investigated by molecular modelling, fluorescence, ultraviolet–visible spectroscopy (UV–vis), Fourier transform infrared spectroscopy (FT-IR) and circular dichroism spectroscopy (CD). The association constants between BPA and HSA were determined using the Scatchard equation. The thermodynamic parameters of the binding reaction (ΔG0, ΔH0 and ΔS0) were measured, and they indicated the presence of hydrophobic forces in the BPA–HSA interaction, which agreed well with the results from molecular modelling. The alterations of protein secondary structure in the presence of BPA were confirmed by UV–vis, FT-IR and CD spectroscopy. Lastly, the average binding distance, r, between BPA and HSA was evaluated and found to be 1.82nm according to Förster’s theory of non-radiation energy transfer.

Abstract:This work has evaluated the interactions of HSA and typical PBDEs (BDE47, BDE99, BDE100, BDE153 and BDE209) at molecular level by modelling. Apart from the BDE209, PBDEs with higher molecular weight show higher binding energy with the residues of HSA. The BDE209 without H atoms has the lowest binding energy (-ΔGbinding: 4.30calmol-1) than other PBDEs (-ΔGbinding: 7.93-8.42calmol-1). The BDE99 shows a higher binding energy than its isomer (BDE100). On the other hand, the lgKow-depth plotting figure shows that a higher Kow value (hydrophobicity) of PBDEs is accompanied by a deeper binding site within the central channel of HSA. This work may provide a theoretical method to assess the transport and distribution of PBDEs in human body.

Abstract:BACKGROUND: Previous studies have shown that polybrominated diphenyl ethers (PBDEs) behave as weak estrogens in animal and cell culture bioassays. In vivo metabolites of PBDEs are suspected to cause these effects. OBJECTIVES: To identify candidate metabolites, mouse plasma samples were collected after continuous oral and subcutaneous exposure to DE-71, a widely used commercial pentabromodiphenyl ether product, for 34 days. METHODS: Samples were extracted, separated into neutral and phenolic fractions, and analyzed by gas chromatographic mass spectrometry. RESULTS: In the plasma samples of orally treated animals, 2,2',4,4',5,5'-hexabromodiphenyl ether (BDE-153) represented 52% of total measurable PBDEs, whereas it represented only 4.3% in the DE-71 mixture. This suggested that BDE-153 was more persistent than other congeners in mice. Several metabolites were detected and quantitated: 2,4-dibromophenol, 2,4,5-tribromophenol, and six hydroxylated PBDEs. The presence of the two phenols suggested cleavage of the ether bond of 2,2',4,4'-tetrabromodiphenyl ether (BDE-47) and 2,2',4,4',5-pentabromodipheny ether (BDE-99), respectively. The hydroxylated (HO)-PBDEs might come from hydroxylation or debromination/hydroxylation. Among the quantitated hydroxylated metabolites, the most abundant was 4-HO-2,2',3,4'-tetra-BDE, which suggested that there was a bromine shift during the hydroxylation process. para-HO-PBDEs have been proposed to behave as endocrine disruptors. CONCLUSIONS: There seem to be three metabolic pathways: cleavage of the diphenyl ether bond, hydroxylation, and debromination/hydroxylation. The cleavage of the diphenyl ether bond formed bromophenols, and the other two pathways formed hydroxylated PBDEs, of which,para-HO-PBDEs are most likely formed from BDE-47. These metabolites may be the most thyroxine-like and/or estrogen-like congeners among the HO-PBDEs.

Abstract:The noncovalent interactions of 24 diversely structured hydroxylated polybrominated diphenyl ethers (OH-PBDEs), ranging from hydroxylated monobromodiphenyl ether (OH-monoBDE) to hydroxylated octabromodiphenyl ether (OH-octaBDE), with bovine serum albumin (BSA) have been examined by employing an electrospray ionization source fitted on a quadrupole time-of-flight hybrid mass spectrometer equipped with a traveling wave ion mobility cell. The mass spectrometric parameters were finely optimized to favor the observation of noncovalent complexes. The experimental data confirm that due to the close structural resemblance with thyroid hormone thyroxine, some OH-PBDEs have been shown to conjugate with BSA. It is found that BSA preferentially interacts with one OH-octaBDE (4'-OH-BDE-201) and two hydroxylated heptabromodiphenyl ether (OH-heptaBDE) isomers (4-OH-BDE-187 and 6-OH-BDE-180) with 1:1 and 2:1 binding stoichiometries. The state of bound OH-PBDEs is inferred to be as both neutral and anionic species. The dissociation constants of corresponding noncovalent complexes are calculated by titration curve data fitting. Investigation into various OH-PBDE analogs demonstrates that both the degree of bromination and the positions of hydroxylation and bromine moieties may exert influence on the binding of OH-PBDEs to BSA. The possible conformational changes in BSA induced upon the binding of OH-PBDEs were explored by use of ion mobility-mass spectrometry (IM-MS) to understand the details of the interactions in structural aspects. Abstract:The interactions between eight polybrominated diphenyl ethers (PBDEs) and human serum albumin (HSA) were studied by using surface plasmon resonance (SPR) and molecular docking technologies. The results of SPR kinetic analysis showed that both the number and substitution position of bromine on PBDEs had regular influences on the interaction. Bromine atoms mainly affected the dissociation rate of interaction, causing the affinity of PBDE and HSA to increase with the number of bromine atoms. The position of bromine on PBDEs mainly affected the association rate of interaction, resulting in the high affinity of m-substituted PBDE and HSA than that of o-substituted PBDE in isomers. The results of molecular docking indicated although all PBDEs were mainly bound to site I of HSA, the difference of amino acid residues around the binding site affected the binding ability. The van der Waals force and hydrogen bond force had greater contribution to binding free energy than electrostatic force. The experimental datum of SPR were in good agreement with the results of computer simulation.
Measurement of Polybrominated Diphenyl Ethers and Metabolites in Mouse Plasma after Exposure to a Commercial Pentabromodiphenyl Ether Mixture

Abstract:Background Previous studies have shown that polybrominated diphenyl ethers (PBDEs) behave as weak estrogens in animal and cell culture bioassays. In vivo metabolites of PBDEs are suspected to cause these effects. Objectives To identify candidate metabolites, mouse plasma samples were collected after continuous oral and subcutaneous exposure to DE-71, a widely used commercial pentabromodiphenyl ether product, for 34 days. Methods Samples were extracted, separated into neutral and phenolic fractions, and analyzed by gas chromatographic mass spectrometry. Results In the plasma samples of orally treated animals, 2,2′,4,4′,5,5′-hexabromodiphenyl ether (BDE-153) represented 52% of total measurable PBDEs, whereas it represented only 4.3% in the DE-71 mixture. This suggested that BDE-153 was more persistent than other congeners in mice. Several metabolites were detected and quantitated: 2,4-dibromophenol, 2,4,5-tribromophenol, and six hydroxylated PBDEs. The presence of the two phenols suggested cleavage of the ether bond of 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47) and 2,2′,4,4′,5-pentabromodiphenyl ether (BDE-99), respectively. The hydroxylated (HO)-PBDEs might come from hydroxylation or debromination/hydroxylation. Among the quantitated hydroxylated metabolites, the most abundant was 4-HO-2,2′,3,4′-tetra-BDE, which suggested that there was a bromine shift during the hydroxylation process. para-HO-PBDEs have been proposed to behave as endocrine disruptors. Conclusions There seem to be three metabolic pathways: cleavage of the diphenyl ether bond, hydroxylation, and debromination/hydroxylation. The cleavage of the diphenyl ether bond formed bromophenols, and the other two pathways formed hydroxylated PBDEs, of which para-HO-PBDEs are most likely formed from BDE-47. These metabolites may be the most thyroxine-like and/or estrogen-like congeners among the HO-PBDEs.

Abstract:Hydroxylated polybrominated diphenyl ethers (OH-PBDEs) is a kind of emerging persistent organic pollutants that may interfere with regulating function of human thyroid hormone;however,the molecular mechanism behind remains unknown.In the present study,molecular docking was performed to explore the possible inhibition activity of OH-PBDEs to thyroxine-binding globulin (TBG) and transthyretin (TTR).Hydrogen bonding interaction was found to enhance the binding affinity of OH-PBDEs to these proteins through critical hydrogen bond amino acids of Lys270 (TBG),Leu110 (TTR) and Ser117 (TTR).Based on stimulated binding modes,the 3D-QSAR model on 14 OH-PBDEs,triiodothyronine (T3) and tyroxine (T4) was developed to predict binding affinities of OH-PBDEs to TBG and TTR.The optimal models from comparative molecular similarity index analysis (CoMSIA)were developed with r2 value of 0.966 (TBG) and 0.961 (TTR),and q2 value of 0.560 (TBG) and 0.525 (TTR).Electrostatic and hydrophobic field of OH-PBDEs account for 72％ (TBG) and 68％ (TTR) of binding affinities,respectively.These findings provide new insight into OH-PBDEs-TBG/TTR interactions,and will be conducive to evaluating damage of this type of chemicals to human health.
In Vitro Metabolism of Hydroxyl Polybrominated Diphenyl Ethers (Oh-Pbdes) and Effects on Estrogen Metabolism in Mouse Liver Microsomes

Abstract:Hydroxylation polybrominated diphenyl ethers (OH-PBDEs) are a class of phenolic compounds that have been found to be an endocrine disruptor, and the in vitro metabolism of OH-PBDEs and effect on metabolism of synthetic estrogen were investigated using mouse liver microsome. The results showed that all of the four OH-PBDEs can be metabolized, and the metabolic rate followed the order: 6-OH-BDE-99＞6'-OH-BDE-99＞6-OH-BDE-137＞5'-OH-BDE-99, which indicated OH-PBDEs were more readily bio-transformed when the hydroxyl functional group (-OH) was in the ortho position with the ether bond and the Br atom. The four OH-PBDEs showed different effects on the metabolism of natural estrogens, including estrone (E1), 17β-estradiol (E2), estriol (E3). As a whole, the inhibition on metabolic rate of E1 and E2 gradually increased with the increasing concentrations of OHPBDE, but showed a promotion effect on metabolism of E3 and no significant effect on metabolism of synthetic estrogen 17α-ethynylestradiol (EE2). The quantification of the metabolite 2-hydroxyestradiol (2-OH-E2) showed that6-OH-BDE-99 promoted the 2-OH-E2 production, while 2-OH-E2 production was inhibited by the other three OHPBDEs as the experimental concentration increased, and these results revealed that it may be related to the different metabolic mechanisms mediated by enzymes.
Study on the Biomolecular Competitive Mechanism of Polybrominated Diphenyl Ethers and Their Derivatives on Thyroid Hormones

Abstract:Polybrominated diphenyl ethers (PBDEs) are widely used brominated flame retardants. PBDEs and their derivatives, hydroxylated PBDEs (OH-PBDEs), can bind to hormone receptors and impact hormone secretion, transportation, and metabolism, leading to endocrine disruption and the development of various diseases. They have particularly strong interference effects on thyroid hormones. This study used decabromodiphenyl ether (BDE-209); 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47); and 6-OH-BDE-47 as representative compounds of PBDEs and their derivatives, OH-PBDEs. A fluorescence probe, fluorescein-isothiocyanate-L-thyroxine (FITC-T4, F-T4), specific for binding to transthyretin (TTR), a thyroid transport protein, was prepared. The binding capacity of PBDEs and their derivatives, OH-PBDEs, to TTR was quantitatively measured using fluorescence spectroscopy. The principle of quenching the fluorescence intensity of F-T4 after binding to TTR was used to analyze the competitive interaction between the probe and BDE-209, BDE-47, and 6-OH-BDE-47, thereby evaluating the toxic effects of PBDEs and their derivatives on the thyroid system. Additionally, AutoDock molecular docking software (1.5.6) was used to further analyze the interference mechanism of OH-PBDEs on T4. The results of the study are as follows: (1) Different types of PBDEs and OH-PBDEs exhibit varying degrees of interference with T4. Both the degree of bromination and hydroxylation affect their ability to competitively bind to TTR. Higher bromination and hydroxylation degrees result in stronger competitive substitution. (2) The competitive substitution ability of the same disruptor varies at different concentrations. Higher concentrations lead to stronger substitution ability, but there is a threshold beyond which the substitution ability no longer increases. (3) When OH-PBDEs have four or more bromine atoms and exhibit the most structural similarity to T4, their binding affinity to TTR is stronger than that of T4.
