Phylogenetic Analysis of Endophytic and Pathogenic Pestalotiopsis Species from Myrica Rubra Based On ITS, Β-Tubulin, EF1-Α, And GPDH Gene Sequences

Abstract:Background: Myrica rubra is a fruit tree native to the subtropical regions of China. Recently, a twig blight disease what caused by Pestalotiopsis occurred on the fruit tree and caused destructive damage of plantings in the Zhejiang, Guizhou, Sichuan, Yunnan and Fujian provinces in recent years. However, there is little morbidity in Guangxi. Endophytic fungi are referred those that live in plants for a certain period of time, but do not cause obvious symptoms of disease on host plant tissue .There are few reports about the relationship between endophytic and pathogenic Pestalotiopsis. Materials and methods: Isolates of endophytic and pathogenic Pestalotiopsis from Myrica rubra from 8 regions/townships in Zhejiang Province and Guangxi Province, China were collected and morphologically identified. Isolates from undomesticated plants were all endophytic. Both endophytic and pathogenic strains were isolated from different environments on same domesticated varieties. Regions/township appeared to be a determining factor on the states of endophytic/pathogenic. The relationship between endophytic and pathogenic Pestalotiopsis species was investigated based on molecular phylogenetic analysis of rDNA ITS, β-tubulin, EF1-α, and GPDH gene sequences. Results:It was found that Pestalotiopsis isolates of the same species, regardless of if they were endophytic or pathogenic, clustered together in all four phylogenetic trees. On M. rubra , Pestalotiopsis versicolor and Pestalotiopsis microspora were all endophytic and pathogenic on trunks, stems, twigs and leaves; Pestalotiopsis theae was only endophytic on twigs and leaves; the results revealed that some Pestalotiopsis species could have endophytic and pathogenic states on different host varieties of the same species or parts of a plant. and we hypothesize that a Pestalotiopsis species could be endophytic,  pathogenic or both states and a species could be transformed from one state into another state under certain environmental conditions. Our results also revealed that Pestalotiopsis species are not generally specific to tissue of plants. Conclusion:In this study, environment (geographic location) other than genetic variation of Pestalotiopsis was a main factor determining the endophytic or pathogenic state of a Pestalotiopsis species in Chinese bayberry, which supports the endophytic continuum hypothesis.
