Abstract:As a peroxidase mimic, cupric oxide nanoparticles were found to enhance the chemiluminescence (CL) of luminol-H(2)O(2) system up to 400 folds. The CL spectra and radical scavengers were conducted to investigate the possible CL enhancement mechanism. It was suggested that the enhanced CL could be attributed to the peroxidase-like activity of CuO nanoparticles, which effectively catalyzed the decomposition of hydrogen peroxide into hydroxyl radicals. The effects of the reactant concentrations and some organic compounds were also investigated. The proposed method could be used as a sensitive detection tool for hydrogen peroxide and glucose.

Abstract:Inspired by natural hemin structure, the fabrication of metal organic framework (MOF) nanomaterials constructed by metal ions and iron porphyrin for peroxidase mimicking has attracted extensive attention recently due to their high concentration of active sites and great channeling for substrate diffusion. Herein, by using benzoic acid as capping agent, we fabricated uniform PCN-224 nanoparticles (NPs), which were composed of Zr6 cluster and tetrakis(4-carboxyphenyl)porphyrin (H2TCPP). Then Fe(Ⅲ) ions were incorporated into the center of H2TCPP producing iron porphyrin unit similar to hemin serving as catalytic active site. The procedure of iron incorporation didn't change the size and morphology of PCN-224 NPs. The newly formed Fe@PCN-224 NPs possessed peroxidase-like activity with much lower Km values and higher Kcat values than most of peroxidase-like nanozymes as well as natural horseradish peroxidase (HRP), indicating their enhanced catalytic activity. Taking advantage of the excellent peroxidase-like activity of Fe@PCN-224 NPs, we successfully developed a colorimetric method for simple and sensitive detection of hydrogen peroxide and glucose in combination with glucose oxidase.

Abstract:Using a heterogeneous catalyst, Co(II)-ethanolamine complex sorbed on Dowex-50W resin, the chemiluminescence (CL) of luminol in unbuffered or weakly acidic solution was studied in the presence of H2O2. The maximum luminol CL wavelength at pH 5.7 was 448 nm, 23 nm longer than that in a basic solution (pH 10.5). Three different ligands, mono-, di-, and triethanolamine, and six transition metal ions, Co(II), Cu(II), Ni(II), Mn-(II), Fe(II), and Fe(III) were compared by CL measurements. The CL intensity decreased in the order mono- > di- > triethanolamine and Co(II) > Cu(II) > Ni(II) > Fe-(III) > Mn(II) > Fe(II). This heterogeneous CL system was developed as H2O2 and glucose flow-through sensors. Detection limits (S/N = 3) of H2O2 and glucose using Dowex-50W-X4-Co(II)-monoethanolamine as catalyst are 1 x 10(-7) M and 1 x 10(-6) M, respectively. On the basis of the studies of the CL, fluorescence, UV-vis and ESCA spectra and the effect of dissolved oxygen in luminol solution, a mechanism for CL emission in unbuffered solution was considered as the formation of a superoxide radical ion during the decomposition of H2O2 catalyzed by the Co(II)-ethanolamine immobilized resin. Then the superoxide radical ion acted on luminol and the CL was emitted. The applications of the proposed method to determine H2O2 in rainwater without any special pretreatment and glucose in human urine and orange juice samples give satisfactory results.

Abstract:It was found that chloroauric acid (HAuCl(4)) could be directly reduced by the luminescent reagent luminol in aqueous solution to form gold nanoparticles (AuNPs), the size of which depended on the amount of luminol. The morphology and surface state of as-prepared AuNPs were characterized by transmission electron microscopy, UV/visible spectroscopy, X-ray photoelectron spectroscopy, FTIR spectroscopy, and thermogravimetric analysis. All results indicated that residual luminol and its oxidation product 3-aminophthalate coexisted on the surface of AuNPs through the weak covalent interaction between gold and nitrogen atoms in their amino groups. Subsequently, a luminol-capped AuNP-modified electrode was fabricated by the immobilization of AuNPs on a gold electrode by virtue of cysteine molecules and then immersion in a luminol solution. The modified electrode was characterized by cyclic voltammetry, electrochemical impedance spectroscopy, and scanning electron microscopy. The as-prepared modified electrode exhibited an electrochemiluminescence (ECL) response in alkaline aqueous solution under a double-step potential. H2O2 was found to enhance the ECL. On this basis, an ECL sensor for the detection of H2O2 was developed. The method is simple, fast, and reagent free. It is applicable to the determination of H2O2 in the range of 3x10(-7)-1x10(-3) mol L(-1) with a detection limit of 1x10(-7) mol L(-1) (S/N=3).

Abstract:In this paper, a novel fluorescent detection method for glucose and lactic acid was developed based on fluorescent iron nanoclusters (Fe NCs). The Fe NCs prepared using hemin as the main raw material exhibited excellent water solubility, bright red fluorescence, and super sensitive response to hydrogen peroxide (H2O2). This paper demonstrates that Fe NCs exhibit excellent peroxide-like activity, catalyzing H2O2 to produce hydroxyl radicals (•OH) that can quench the red fluorescence of Fe NCs. In this paper, a new type of glucose sensor was established by combining Fe NCs with glucose oxidase (GluOx). With the increase in glucose content, the fluorescence of Fe NCs decreases correspondingly, and the glucose content can be detected in the scope of 0–200 μmol·L−1 (μM). Similarly, the lactic acid sensor can also be established by combining Fe NCs with lactate oxidase (LacOx). With the increase in lactic acid concentration, the fluorescence of Fe NCs decreases correspondingly, and the lactic acid content can be detected in the range of 0–100 μM. Furthermore, Fe NCs were used in the preparation of gel test strip, which can be used to detect H2O2, glucose and lactic acid successfully by the changes of fluorescent intensity.

Abstract:Hollow nanocages of nickel‐cobalt layered bimetallic hydroxide with a hollow structure and defective states are employed as co‐reaction promoters in the luminol‐H2O electrochemiluminescence system and reacted by oxygen evolution reaction to generate reactive oxygen species, which enhances the electrochemiluminescence emission of luminol and achieves sensitive biosensing. Conventional luminol co‐reactant electrochemiluminescence (ECL) systems suffer from low stability and accuracy due to factors such as the ease of decomposition of hydrogen peroxide and inefficient generation of reactive oxygen species (ROS) from dissolved oxygen. Inspired by the luminol ECL mechanism mediated by oxygen evolution reaction (OER), the nickel‐cobalt layered double hydroxide (NiCo‐LDH) hollow nanocages with hollow structure and defect state are used as co‐reaction promoters to enhance the ECL emission from the luminol‐H2O system. Thanks to the hollow structure and defect state, NiCo‐LDH hollow nanocages show excellent OER catalytic activity, which can stabilize and efficiently produce ROS and enhance the ECL emission. Additionally, mechanistic exploration suggests that the ROS involved in the co‐reaction of the luminol‐H2O system are derived from the OER reaction process, and there is a positive correlation between ECL intensity and the OER catalytic activity of the co‐reaction promoter. The selection of catalysts with excellent OER catalytic activity is a key factor in improving ECL emission. Finally, a dual‐mode immunosensor is constructed for the detection and analysis of alpha‐fetoprotein (AFP) based on the promoting effect of NiCo‐LDH hollow nanocages on the luminol‐H2O ECL system.

Abstract:A glassy carbon electrode (GCE) modified with cobalt(II) meso-tetraphenylporphrine/multiwall-carbon nanotube (CoTPP/MWNT) was applied to investigate the electrochemiluminescent (ECL) behavior of luminol. The ECL intensity of luminol was found to be increased greatly on this modified electrode. The presence of cobalt(II) meso-tetraphenylporphrine (CoTPP) can catalyze the reduction of oxygen on the electrode surface to produce HOO−, which can increase the ECL intensity of luminol. Moreover, MWNT can provide the more effective area of the electrode, and can act as a promoter to enhance the electrochemical reaction. The proposed method enables a detection limit for luminol of 1.0×10−8mol/L in the neutral solution. Under the optimum condition, the enhanced ECL intensity of luminol by H2O2 had a linear relationship with the concentration of H2O2 in the range of 1.0×10−7 to 8.0×10−8mol/L with the detection limit of 5.0×10−9mol/L.

Abstract:In this study, Ag and N-doped carbon dots (Ag-NCDs) were used to enhance the ultraweak H2O2-Co2+ (a Fenton-like reaction) chemiluminescence (CL) reaction, in which Ag-NCDs were found to have the functions of catalyst, chelator, and luminophore. Under the co-catalysis of Co2+ and Ag-NCDs, H2O2 dissociation was was facilitated, resulting in increased generation of active •OH and •O2– radicals. Thus, the CL intensity of the Ag-NCDs-H2O2-Co2+ system was significantly higher than that of the H2O2-Co2+ system and the Ag-NCDs-H2O2 system. Co2+ catalyzes the reaction through complex formation with Ag-NCDs' surface amino groups, thereby accelerating H2O2 decomposition. The CL intensity of H2O2-Co2+ system increased with increasing Co2+ concentration in a range from 0.4 μM to 40.0 μM with a detection limit of 0.07 μM. Furthermore, we observed that HQ can effectively quench the Ag-NCDs-H2O2-Co2+ CL signal. This led to the application of the CL system for HQ detection, achieving quantitative measurements within the 0.2–20.0 μM concentration range with a detection limit of 0.04 μM. It also enabled sequential determination of Co2+ and HQ in egg yolk, vitamin B12, and water samples, offering simplicity, time-saving, and higher selectivity.

Abstract:A novel fluorescent hydrogen peroxide sensor was developed based on the peroxidase-like activity of cupric oxide nanoparticles. Cupric oxide nanoparticles effectively catalyzed the decomposition of hydrogen peroxide into hydroxyl radicals. Then terephthalic acid was oxidized by hydroxyl radical to form a highly fluorescent product. The linear range of hydrogen peroxide estimated to be 5.0 × 10(-6)-2.0 × 10(-4)M with a detection limit of 3.4 × 10(-7)M. Moreover, this detection system enabled the sensing of analytes which can enzymatically generate hydrogen peroxide. By coupling the oxidation of glucose or L-lactate catalyzed by their corresponding oxidase enzymes with terephthalic acid oxidation catalyzed by cupric oxide nanoparticles, sensitive assays of glucose and l-lactate with detection limits of 1.0 × 10(-6) and 4.5 × 10(-8)M were realized. The successful applications of this approach in human serum samples have also been demonstrated.

Abstract:Hydrogen peroxide (H2O2) and glutathione (GSH) have been reported to be associated with a variety of diseases. Therefore, the accurate and rapid determination of H2O2 and GSH was very important. The traditional assay was the colorimetric method, which suffered from the bottleneck of single-mode analysis. In this work, we designed a dual-mode fluorescence and colorimetric sensor for detecting H2O2 and GSH based on bifunctional Fe, Co-CDs, which possessed peroxidase (POD)-mimetic activity and fluorescence property. Concretely, the Fe, Co-CDs catalyzed oxidation of 3,3 ',5,5 '-tetramethylbenzidine (TMB) to a blue product (oxTMB) with the help of H2O2, which could be inhibited by GSH. Meanwhile, the fluorescence of Fe, Co-CDs weakened with increasing oxTMB, realizing the fluorescence analysis of H2O2 and GSH. In fluorescence and colorimetric sensors, the detection limit (LOD) of H2O2 could reach as low as 0.54 mu M and 0.81 mu M, as well as the LOD of GSH was 0.39 and 0.61 mu M, respectively. Besides, the smartphone-assisted hydrogel sensor was developed for on-site detection of GSH via color change. This study not only designed a dual-mode sensor for dual-target detection but also provided insights for further exploration of multifunctional nanozymes.
Molybdenum disulfide nanosheet induced reactive oxygen species for high-efficiency luminol chemiluminescence

Abstract:Background Luminol chemiluminescence (CL) sensing system remains an excellent candidate for application in bioanalysis due to its good water solubility. However, traditional luminol CL system usually requires the addition of oxidizing agents and sensitizers to obtain high efficiency for the improvement of detection sensitivity. Although numerous studies on the nanomaterial-enhanced luminol CL systems have been carried out, there is still great potential to develop inexpensive, readily available and easily handled catalysts to construct simple and effective CL platform for biomolecular sensing. Results Few-layered MoS 2 nanosheets (NS) prepared by sonication-assisted exfoliation of commercially available bulk MoS2 were found to significantly enhance the CL of luminol‒dissolved oxygen in the absence of additional oxidants. The mechanism study demonstrated that exfoliated MoS2 NS could catalyze the decomposition of dissolved oxygen by virtue of its exposed active sites on the surface, generating increased reactive oxygen intermediates, which then oxidize luminol and produce intense CL emission. The proposed high-efficiency luminol CL system was then employed for the extremely sensitive identification of dopamine based on the quenching of CL by dopamine. The limit of detection (LOD) for dopamine can be as low as 2.07 nM. Besides, it also works well in the actual urine sample with good recoveries (99.6–100.6 %), confirming the practicability of the method. Significance As a new type of CL catalyst, MoS2 NS developed in this work are easy to obtain, simple to prepare and can be produced in large quantities, which lays a foundation for extending applicability of MoS2 NS in the CL field, and provides a new idea for developing simple and highly sensitive CL sensing system.

Enhanced Solid-State Electrogenerated Chemiluminescence of Au/CdS Nanocomposite and Its Sensing to H2O2

Abstract:Au nanoparticles (AuNPs) are good quenchers once they closely contact with luminophore. Here we reported a simple approach to obtain enhanced electrogenerated chemiluminescence (ECL) behavior based on Au/CdS nanocomposite films by adjusting the amount of AuNPs in the nanocomposite. The maximum enhancement factor of about 4 was obtained at an indium tin oxide (ITO) electrode in the presence of co-reactant H2O2. The mechanism of this enhancement was discussed in detail. The strong ECL emission from Au/CdS nanocomposites film was exploited to determine H2O2. The resulting ECL biosensors showed a linear response to the concentration of H2O2 ranging from 1.0 × 10−8 to 6.6 × 10−4 mol L−1 with a detection limit of 5 nmol L−1 (S/N = 3) and good stability and reproducibility.

Abstract:An aminated Fe–Ni bimetallic metal–organic framework (Fe 3 Ni-MOF-NH 2 ) with both peroxidase-like activity and fluorescence properties was developed. Fe 3 Ni-MOF-NH 2 possessed the enhanced peroxidase-like activity through the enhanced electron transfer process and hydroxyl radical (·OH) generation. It was found that the amino group endowed the material with fluorescent property and the metal site Ni in Fe 3 Ni-MOF-NH 2 could also enhance the fluorescence emission intensity (Ex = 345 nm, Em = 452 nm). Based on the dual excellent performance of Fe 3 Ni-MOF-NH 2 , a novel sensitive fluorescence detection strategy for H 2 O 2 and glucose was designed and achieved. First, Fe 3 Ni-MOF-NH 2 converted H 2 O 2 to ·OH by exerting peroxidase-like activity, and ·OH converts catechol to o-benzoquinone. Then, the amino group in Fe 3 Ni-MOF-NH 2 connected to o-benzoquinone, which resulted in its fluorescence quenching. The detection limit of H 2 O 2 was as low as 5 nM. Combined with glucose oxidase which can oxidize glucose and produce H 2 O 2 the glucose could be indirectly determined with a detection limit of 40 nM. The method was applied to the detection of low-level glucose in human urine samples with good recoveries and reproducibilities.

Abstract:Abstract Hydrogen peroxide (H 2 O 2 ) is an important product generated in the body and related to many pathophysiological processes and glucose metabolism disorder can cause many fatal diseases in living bodies. Therefore, the sensing of H 2 O 2 and glucose is of great significance in disease diagnostics and treatment. Fluorescent carbon dots (CDs) are one new class of nanoprobes for H 2 O 2 and glucose. Nevertheless, the CD-based sensor is always based on its fluorescence response, which is influenced by the auto-fluorescent interference. Herein, efficient fluorescent CDs were synthesized by one-pot solvothermal method, and the CDs exhibit bright and persistent deep-red (DR) chemiluminescence (CL) in bis(2,4,6-trichlorophenyl) oxalate and H 2 O 2 solution with a CL quantum yield of (8.22 ± 0.30) × 10 −3 , which is amongst the highest values in ever reported nanomaterials for chemical analysis. Employing the CDs as CL nanoprobes, sensitive sensing for H 2 O 2 has been achieved with a detection limit of 11.7 μM, and further for glucose detection with a detection limit of 12.6 μM. The DR CL CDs is promising to be applied in blood glucose analysis or in vivo biosensor.

Direct placing of the resultant dispersion on a glassy carbon electrode (GCE) surface gives a very stable AuPs-containing film exhibiting remarkable catalytic performance toward both the oxidation and reduction of H2O2. This enzymeless H2O2 sensor shows a fast amperometric response time of less than 3s and the corresponding linear range and detection limit are estimated to be from 0.1mM to 50mM (r=0.999) and 4μM at a signal-to-noise ratio of 3, respectively.

Abstract:Chemiluminescence (CL) assay has been considered as a sensitive method for quantitative detection of glutathione (GSH), a significant regulatory metabolite in cells. However, most reported CL assays present flash-type CL emission with limited detection accuracy and repeatability. Herein, an intensive and long-lasting glow-type CL system was proposed based on a novel bimetallic Co-doped ceria mesoporous nanocomposite (), which was prepared by in-situ growth of sub-3 nm ultrasmall Co-ceria nanodots within the pore channels of mesoporous silica nanospheres (MSN). Attributing to the confinement effect of MSN and the superior peroxidase-like catalytic activity of Co-ceria nanodots, this nanocomposite could function as a nanozyme to catalyze luminol/H 2 O 2 CL reaction, producing a robust and long-lasting CL emission with a long plateau for over 1.2 h at physiological pH. Surprisingly, both the CL intensity and duration time were found to exist a strong dependence on GSH concentration. Accordingly, the proposed glow-type CL platform was further applied for GSH determination with a limit of detection down to 10 nM. This strategy was successfully utilized for quantitative evaluation of cellular GSH levels, indicating its broad prospects in clinical diagnosis.

Abstract:Local enrichment of free radicals at the electrode interface may open new opportunities for the development of electrochemiluminescence (ECL) applications. The sensing platform was constructed by assembling ECL-emitting luminol derived carbon dots (Lu CDs) onto the heterojunction Tungsten disulfide/Covalent organic frameworks (WS2@COF) for the first time, establishing a nanoconfinement-reactor with significantly heightened ECL intensity and stability compared to the Lu CDs-H2O2 system. This enhanced performance is credited to the COF domain's restricted pore environment, where WS2@COF exhibits a more negative adsorption energy for H2O2, effectively enriching H2O2 in the catalytic edge sites of WS2. Furthermore, the internal electric field at the WS2 and COF interface accelerates electron flow, boosting WS2's catalytic activity and achieving domain-limited catalytic enhancement of ECL. Self-designed DNA nanomachines combined with cascading molecular keypad locking mechanisms are integrated into the biosensors, effectively guaranteeing the accuracy of the sensing process while providing crucial safeguards for molecular diagnostics and information security applications. In essence, this innovative approach represents the first system to enhance local free radical concentrations by enriching co-reactants on the electrode surface through nanoconfinement catalysis, yielding heightened ECL luminescence intensity. The potential impact of this novel strategy and sensing mechanism on real-bioanalysis applications is promising.
