Abstract:Chitosan, the cationic polysaccharide derived from the natural polysaccharide chitin, has been studied as a biomaterial for more than two decades. As a polycationic polymer with favorable properties, it has been widely used to form polyelectrolyte complexes with polyanions for various applications in drug delivery fields. In recent years, a growing number of studies have been focused on the preparation of polyelectrolyte complexes based on chitosan and its water soluble derivatives. They have been considered well-suited as biomaterials for a number of vital drug carriers with targeted/controlled release profiles, e.g., films, capsules, microcapsules. In this work, an overview highlights not only the favorable properties of chitosan and its water soluble derivatives but also the good performance of the polyelectrolyte complexes produced based on chitosan. Their various types of applications as drug carriers are reviewed in detail.

Abstract:A novel type of melamine formaldehyde microcapsule with a desirable barrier has been used to encapsulate water soluble ingredients, including potassium chloride (KCl) and allura red (dye) as models of an inorganic salt and organic molecule, respectively, via a facile method, and it has shown a sustained release of KCl and allura red for 12 h and 10 days in aqueous environment, respectively.

Abstract:The current study demonstrates a desired drug delivery-controlled system with a wide range of release parameters. It illustrates the feasibility of carriers in the systems for extended and short drug release. Herein, mesoporous silica nanoparticles (MSNs) coated with chitosan (Cs) grafted polylactic acid (PLA) composite were fabricated by a sol-gel method in the form of micrometric porous particles (∼270 nm) with ζ -potential of −17.7 ± 1.7 mV and a specific surface area and total pore volume of 122700 cm 2 /g and 0.057 cm 3 /g, respectively. The prepared template of polymer-silica composite pores proved to be essential in the incorporation of Doxorubicin (Dox) drug molecules with adsorption capacity of 83.33 mg/g and encapsulation efficiency of 88 %. Dox was incorporated via the solvent diffusion method into the polymer-silica composites at suitable pH and concentration, resulting in solid drug dispersions. The silica-Cs-g-PLA composites showed greater effectiveness in the delayed release of the Dox with complete release after 10 days as compared to pristine MSNs and silica-Cs, which were completed after 12 h and 24 h, respectively. The drug release profiles were studied at different pH (2, 7, and 8), with maximum results obtained at pH 2. Both pristine MSNs and silica-Cs suffered from the burst effect, which was overcome by the grafting of PLA. The functionalized silica-Cs with PLA shows a significant change in the drug release properties from a sustained release process for pristine MSNs and silica-Cs to an enhanced delayed process for the silica-Cs-g-PLA composite, which makes this composite attractive for oral multi-particulate formulations of modified release.

Abstract:Functionalized PS/SiO2 composite nanoparticles bearing sulfonic groups on the surface were successfully synthesized via emulsion copolymerization using a polymerizable emulsifier a olefin solfonate (AOS). As demonstrated by transmission electron microscopy and atomic force microscopy, well-defined core-shell PS/SiO2 composite nanoparticles with a diameter of 50 nm were obtained. Sulfonic groups introduced onto the surface of the composite nanoparticles were quantified by FTIR, and can be controlled to some extent via a two-stage procedure.

Abstract:pH-responsive hydrogel antibacterial microspheres were prepared by complex coacervation method and used as drug carriers to load the antibiotic substitute potassium diformate (KDF). The results showed that the three-dimensional network structure of the composite hydrogel was formed among the polysaccharide components, and Zeolite A loaded with KDF was uniformly dispersed in it. The results of drug release behavior and kinetics in vitro showed that the drug release rate was higher and the release mechanism followed the Ritger-Peppas model, which indicated that the antibacterial microspheres had a good pH response. In vitro , the carrier showed broad-spectrum antibacterial activity, good biocompatibility, and degradability. Graphical Sodium carboxymethylcellulose and pectin synergistic by hydrogen bonding, potassium dicarboxylate-loaded Zeolite A is filled in a three-dimensional hydrogel network, and chitosan quaternary ammonium salts coat the outer layer.

Abstract:Chitosan based dual microspheres are expected to achieve multiple functions in drug delivery area. Herein, the dual microspheres based on thiolated chitosan (CS-TBA)/CaCO3 were fabricated by ionic gelation method. The obtained dual microspheres combine the benefits of CS-TBA for improved water solubility and CaCO3 for mesoporous structure. The microspheres display a sphere morphology with a diameter of 400–500 μm, in which CaCO3 particles with mesoporous structure are encapsulated. In vitro dual drug release shows a controlled manner for different drugs from CS-TBA/CaCO3 microspheres at different pH. The dual CS-TBA/CaCO3 microspheres can serve as a dual-drug delivery system and have potential applications in tissue engineering and regenerative medicine.

Abstract:Finding novel systems for controlled delivery of drugs is still a stringent need. Fabrication of drug delivery systems (DDS) having a cross-linked low molar mass synthetic polycation such as poly(ethylene imine) (PEI) entrapped into the pores of a mesoporous silica followed by coating the silica/PEI composite microspheres with a pH responsive layer-by-layer (LbL) film is presented for the first time in this work. The LbL film, which behaves as drug release lingering component of the composite, was constructed with carboxymethyl cellulose (CMC) and chitosan (CS) as building blocks. Silica/PEI/(CMC/CS)n composites were fully characterized by Fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), scanning electron microscopy (SEM) and EDX spectra, and by the nitrogen sorption/desorption isotherms (BET analysis). The dynamic of LbL film construction was followed by potentiometric and polyelectrolytic titrations. Diclofenac sodium (DS) was used as a model drug to demonstrate the potential of the as prepared composite as novel system for controlling the release of drugs. Investigation of the release kinetics evidenced the influence of the LbL film on the mechanism of DS release. Taking into account the values of nr in the Korsmeyer-Peppas equation, pseudo-Fickian release, with nr < 0.45, was found for silica and silica/PEI composites, while anomalous release, with 0.59 < nr < 0.85, was found for the silica/PEI composite microspheres coated with (CMC/CS)9 films (diffusion and chain relaxation). Silica/PEI/(CMC/CS)n composite microspheres fabricated in this work have great potential in the sustained delivery of drugs to reduce the fluctuations in the drug level in blood.Graphic abstract

Abstract:Polysaccharide/silica hybrid microcapsules were prepared using ionic gelation followed by spray-drying. Chitosan and alginate were used as biopolymer matrices, and in situ prepared silica was used as a structuring additive. The prepared microparticles were used in two very different applications: the encapsulation of hydrophilic molecules, and as a support for palladium nanoparticles used as catalysts for a model organic reaction, namely the reduction of p-nitrophenol by sodium borhydride. In the first application, erioglaucine disodium salt, taken as a model hydrophilic substance, was encapsulated in situ during the preparation of the microparticles. The results indicate that the presence of silica nanostructures, integrated within the polymer matrix, affect the morphology and the stability of the particles, retarding the release of the encapsulated substance. In the second application, chloropalladate was complexed on the surface of chitosan microparticles, and palladium(II) was subsequently reduced to palladium(0) to obtain heterogeneous catalysts with an excellent performance.

Abstract:A novel nanocontainer, which could regulate the release of payloads, has been successfully fabricated by attaching zwitterionic sulfobetaine copolymer onto the mesoporous silica nanoparticles (MSNs). RAFT polymerization is employed to prepare the hybrid poly(2-(dimethylamino)ethyl methacrylate)-coated MSNs (MSN-PDMAEMA). Subsequently, the tertiary amine groups in PDMAEMA are quaternized with 1,3-propanesultone to get poly(DMAEMA-co-3-dimethyl(methacryloyloxyethyl)ammonium propanesulfonate)-coated MSNs [MSN-Poly(DMAEMA-co-DMAPS)]. The zwitterionic PDMAPS component endows the nanocarrier with biocompatibility, and the PDMAEMA component makes the copolymer shell temperature-responsive. Controlled release of loaded rhodamine B has been achieved in the saline solutions.

Abstract:Novel hollow mesoporous silica spheres (HMSs) with uniform size and morphology have been successfully synthesized in a facile route using poly(vinylpyrrolidone) (PVP) and cetyltrimethylammonium bromide (CTAB) as co-templates at room temperature. XRD, N2 adsorption–desorption analysis, FE-SEM, and TEM are used for the characterization of the structure. The hexagonally ordered pore channels are formed on the shell. Ibuprofen (IBU) as a model drug is used to examine the storage capacity and the release behaviour of drug molecules. The storage capacity of hollow mesoporous silica spheres can reach 1133 ± 52.4 mg g−1 for ibuprofen, which is three times higher than that of conventional mesoporous materials previously reported (337 mg g−1), and the storage capacity is adjustable in a wide range. Furthermore, this system also shows a sustained-release behaviour. The release rates from the IBU-HMSs system (IBU stored in HMSs) increase with the decrease of pH values and the release process follows a Fick's law.

Abstract:pH-responsive polymer shell chitosan/poly (methacrylic acid) (CS-PMAA) was coated on mesoporous silica nanoparticles (MSN) through the facile in situ polymerization method. The resultant composite microspheres showed a flexible control over shell thickness, surface charges and hydrodynamic size by adjusting the feeding amount of MSN and the molar ratio of [–NH2]/MAA. The MSN/CS-PMAA composite microspheres were stable in the pH range of 5–8 as well as in the physiological saline (0.15M NaCl). Doxorubicin hydrochloride (DOX) was applied as a model drug to investigate the drug storage and release behavior. The results demonstrated that DOX could be effectively loaded into the composite microspheres. The cumulative release of DOX-loaded composite microspheres was pH dependent and the release rate was much faster at low pH (5.5) than that of pH 7.4. The cytotoxicity test by MTT assay showed that the blank carrier MSN/CS-PMAA microspheres were suitable as drug carriers. The cellular uptake of composite microspheres was investigated by confocal laser scanning microscopy (CLSM), which indicated that MSN/CS-PMAA could deliver the drugs into HeLa cell. The above results imply that the composite microspheres are a promising drug delivery system for cancer therapy.
Preparation of Ethyl Cellulose Microspheres for Sustained Release of Sodium Bicarbonate.

Abstract:Sustained release of thermal-instable and water-soluble drugs with low molecule weight is a challenge. In this study, sodium bicarbonate was encapsulated in ethyl cellulose microspheres by a novel solid-in-oil-in-oil (S/O/O) emulsification method using acetonitrile/soybean oil as new solvent pairs. Properties of the microspheres such as size, recovery rate, morphology, drug content, and drug release behavior were evaluated to investigate the suitable preparation techniques. In the case of that the ratio of the internal and external oil phase was 1: 9, Tween 80 as a stabilizer resulted in the highest drug content (2.68%) and a good spherical shape of microspheres. After the ratio increased to 1: 4, the microspheres using Tween 80 as the stabilizer also had high drug content (1.96%) and exhibited a sustained release behavior, with 70% of drug released within 12 h and a sustained release of more than 40 h. Otherwise, different emulsification temperatures at which acetonitrile was evaporated could influence the drug release behaviour of microspheres obtained. This novel method is a potential and effective method to achieve the encapsulation and the sustained release of thermal-instable and water-soluble drugs with low molecule weight.
Coating of Sodium Percarbonate Particles Using Water Soluble Materials in a Fluidised Bed to Achieve Delayed Release in Aqueous Environment

Abstract:Three coating materials, namely sodium sulphate, 1.6R and 2.35R sodium silicate, were respectively used to coat sodium percarbonate (SPC) particles in a fluidised bed coater to achieve its delayed release in aqueous environment. The size of SPC particles was measured using image analysis. The thickness and porosity of the shell materials were analysed using scanning electron microscopy (SEM) and helium pycnometry respectively. The rates of SPC release from uncoated and the coated particles were measured using an iodide molybdate titration method coupled with UV-vis spectrometry. The results indicate that sodium sulphate coating with an average thickness of 53 ± 9 μm only reduced the release rate of SPC as no delayed release was observed. In contrast, sodium silicate coating generated a significant delayed release. 1.6R sodium silicate coating with a thickness of 109 ± 8 μm delayed the release of SPC by approximate 60 s under a static condition. At the same condition, 2.35R sodium silicate coating with a thickness of 71 ± 10 μm delayed the release by approximately 7 min. When the coated SPC particles immersed in water were shaken using an orbital shaker at 150 rpm, the delayed time was reduced by 50% in comparison with the static condition. The 1.6R sodium silicate shell in solid phase transformed to gel-like structure during dissolution and the hydrodynamic forces generated in the shaker accelerated its dissolution. However, there was no significant change of 2.35R sodium silicate shell when the capsules were immersed in water under the static condition, and they broke into pieces in the shaker. For both 1.6R and 2.35R sodium silicate, the further increase in shell thickness increased their shell porosity, which facilitated the water penetration and thus resulted in no significant benefit to additional delay. Moreover, the thermal stability of SPC after coating was slightly improved and the flowability did not change significantly. This study demonstrates that a significant delay in release of SPC can be achieved using 2.35R sodium silicate as a coating material.
Design of microencapsulated chitosan microspheres for colonic drug delivery

Abstract:Among the different approaches to achieve colon-selective drug delivery, the use of polymers, specifically biodegraded by colonic bacteria, holds great promise. In this work a new system which combines specific biodegradability and pH-dependent release is presented. The system consists of chitosan (CS) microcores entrapped within acrylic microspheres. Sodium diclofenac (SD), used as a model drug, was efficiently entrapped within CS microcores using spray-drying and then microencapsulated into Eudragit L-100 and Eudragit S-100 using an oil-in-oil solvent evaporation method. The size of the CS microcores was small (1.8-2.9 microns) and they were encapsulated within Eudragit microspheres (size between 152 and 233 microns) forming a multireservoir system. Even though CS dissolves very fast in acidic media, at pH 7.4, SD release from CS microcores was delayed, the release rate being adjustable (50% dissolved within 30-120 min) by changing the CS molecular weight (MW) or the type of CS salt. Furthermore, by coating the CS microcores with Eudragit, perfect pH-dependent release profiles were attained. No release was observed at acidic pHs, however, when reaching the Eudragit pH solubility, a continuous release for a variable time (8-12 h) was achieved. A combined mechanism of release is proposed, which considers the dissolution of the Eudragit coating, the swelling of the CS microcores and the dissolution of SD and its further diffusion through the CS gel cores. In addition, infrared (IR) spectra revealed that there was an ionic interaction between the amine groups of CS and the carboxyl groups of Eudragit, which provided the system with a new element for controlling the release. In conclusion, this work presents new approaches for the modification of CS as well as a new system with a great potential for colonic drug delivery.

Abstract:In present study, protein loaded poly (lactide-co-glycolide)/chitosan microspheres (PLGA/CS MSs) with spheres-in-sphere structure were prepared in order to weaken the burst release of protein from PLGA microspheres (PLGA MSs) and to buffer acidic micro-milieu. The PLGA MSs and PLGA/CS MSs were characterized in terms of their size distribution, morphology, drug-loading rate, zeta potential and physical-chemical properties. The incubation experiments of PLGA MSs and PLGA/CS MSs; were manipulated in PBS solution at pH 7.4, 37 degrees C to monitor the release of BSA and the vehicles degradation. The release kinetic of BSA was illuminated mainly based on the degradation processes of the matrices. External CS crusts were proved to strikingly improve the release kinetic of the model protein by reducing initial burst release and extending continuous release while acting as a diffusion barrier. Moreover, using PLGA/CS MSs could avoid the decrease of pH value resulted from the acidic products of PLGA MSs because of the effective buffer action of the basic groups in CS. The results demonstrated that the spheres-in-sphere structure is an effective way to control the initial burst release of protein and to overcome the acidic problem of protein-loading PLGA MSs. Thermo and Ph Dual Responsive, Polymer Shell Coated, Magnetic Mesoporous Silica Nanoparticles for Controlled Drug Release

Abstract:In this paper, a kind of core-shell composite microsphere was prepared based on poly(N-isopropylacrylamide-co-methacrylic acid) (P(NIPAM-co-MAA)) coated magnetic mesoporous silica nanoparticles (M-MSN) via precipitation polymerization. The composite microsphere presented a thermo/pH-coupling sensitivity and the volume phase transition could be precisely regulated by the molar ratio of MAA to NIPAM or the concentration of NaCl. At physiological conditions (37 degrees C, 0.15 M NaCl), the P(NIPAM-co-MAA) shell underwent a distinct transition from a swollen state in pH 7.4 to a collapsed state in pH 5.0, so that the polymer shell was active in moderating the diffusion of embedded drugs in-and-out of the pore channels of MSN. Doxorubicin hydrochloride (DOX) was applied as a model drug and the behaviors of drug storage/release were investigated. The drug loaded behavior was pH-dependent, and the composite microsphere had a drug embed efficiency of about 91.3% under alkaline conditions. The cumulative in vitro release of the DOX-loaded composite microsphere showed a low level of leakage below the volume phase transition temperature (VPTT) and was significantly enhanced above its VPTT, exhibiting an apparent thermo/pH-response controlled drug release. The cytotoxicity assay of a blank carrier to normal cells indicated that the composite microspheres were suitable as drug carriers, while the DOX-loaded composite microspheres had a similar cytotoxicity to HeLa cells compared with free DOX. Therefore, the thermo/pH-sensitive composite microsphere could, in principle, be used for in vivo cancer therapy with a low premature drug release during blood circulation whilst having a rapid release upon reaching tumor tissues.

The inner part of solid spheres is more easily dissolved rather than the outer part, and the inner part of the solid spheres is dissolved in deionized water at room temperature to obtain the hollow mesoporous silica nanospheres (HMSS) with ordered mesochannels. A facile method for preparing HMSS by water dissoving at room temperature was developed. The performance of HMSS in drug loading and releasing was investigated. The results show that this method can synthesize the HMSS with a well-defined spherical morphology, uniform particle size (i.e., 460nm), high specific surface area (i.e., 1237m2/g) and ordered mesochannels (i.e., 2.7nm), and the superior performance in drug releasing. The loading content of CPT–11 is 14.50%, and the drug releasing amount in 108h at pH 7.2, 5.5 and 4.2can reach 28.49%, 39.22% and 77.72%, respectively.

Abstract:Mesoporous silica microspheres (MSMs) with a pore-size larger than 10nm and a large pore-volume have attracted considerable attention for their application in delivering poorly water-soluble drugs. Here we developed a simple method for large-scale synthesis of MSMs using sodium silicate as silica precursor. The novelty of this approach lies in the use of sucrose solution to achieve large size and volume of nanopores. The highest values of pore size and pore volume are 13.2 nm and 1.97 cm(3)/g, respectively. Importantly, the method is reliable and easily upscalable. The blank and drug-loaded MSMs were characterized by differential scanning calorimetry (DSC) and X-ray powder diffraction (XRPD). Ibuprofen and resveratrol were successfully loaded into the nanopores of MSMs in amorphous and nanocrystalline form and showed high drug-loadings and enhanced dissolution rates. This kind of MSMs appears to be a promising candidate as a new oral drug delivery vehicle providing a rapid drug release.
Preparation and evaluation of controllable drug delivery system: a pH and temperature responsive nanosphere based on chitosan copolymer/mesoporous silica

Abstract:A pH/temperature dual response drug release system (MSNs@CMCS-g-PNIPAM) was prepared in this study. Around the core of mesoporous silica nanoparticles (MSNs), the copolymer of carboxymethyl chitosan (CMCS) and poly (N-isopropylacrylamide) (PNIPAM) was shelled. The CMCS was used as a pH-sensitive functional component in copolymers, and the PNIPAM was used as a temperature-sensitive functional component. The copolymer modification on the surface of mesoporous silicon was achieved through hydrogen bonding. Several characterization techniques were utilized to characterize the structure of the carrier, which include FTIR, TGA, XRD, XPS, N 2 adsorption/desorption, and SEM. The adsorption results showed that the carrier had the best adsorption capacity for doxorubicin hydrochloride (DOX) at pH 7, with drug loading rate and encapsulation rate of 7.43% and 82.99%, respectively. In vitro release experiments demonstrated that the cumulative drug release rate was 4.63 times higher under dual stimulation of pH 5.5 and 45 °C than pH 7.4 and 37 °C, proving the carrier has pH and temperature responsiveness. In addition, the erythrocyte hemolysis test and CCK8 test proved the biocompatibility of MSNs@CMCS-g-PNIPAM. The results indicate that MSNs@CMCS-g-PNIPAM might be a strong candidate for drug carriers.
