Abstract:The paper presents a novel strategy to identify analytical markers of traditional Chinese medicine preparation (TCMP) rapidly via direct analysis in real time mass spectrometry (DART-MS). A commonly used TCMP, Danshen injection, was employed as a model. The optimal analysis conditions were achieved by measuring the contribution of various experimental parameters to the mass spectra. Salvianolic acids and saccharides were simultaneously determined within a single 1-min DART-MS run. Furthermore, spectra of Danshen injections supplied by five manufacturers were processed with principal component analysis (PCA). Obvious clustering was observed in the PCA score plot, and candidate markers were recognized from the contribution plots of PCA. The suitability of potential markers was then confirmed by contrasting with the results of traditional analysis methods. Using this strategy, fructose, glucose, sucrose, protocatechuic aldehyde and salvianolic acid A were rapidly identified as the markers of Danshen injections. The combination of DART-MS with PCA provides a reliable approach to the identification of analytical markers for quality control of TCMP.

Abstract:Tyrosinase is a key enzyme in melanin synthesis. Its inhibitor may be used to efficiently treat hyperpigmentation and widely applied in cosmetic products and food supplements. In the present study, a new assay based on ultrafiltration high-performance liquid chromatography coupled with diode array detector and mass spectrometry (HPLC-DAD-MS) was developed for the rapid screening and identification of ligands for tyrosinase. Experiments were carried out to select the optimal binding conditions, tyrosinase concentration, and incubation time. Non-specific binding to the denatured tyrosinase was also investigated. Twelve compounds with tyrosinase binding activity were found in mulberry leaf extracts. The identities of these compounds were characterised by HPLC-DAD-MS(n). Particularly, two compounds, namely, quercetin-3-O-(6-O-malonyl)-β-D-glucopyranoside and kaempferol-3-O-(6-O-malonyl)-β-D-glucopyranoside, were identified as new tyrosinase inhibitors. The screening results were verified by tyrosinase inhibition assays. Experimental results proved that the proposed method could rapidly screen tyrosinase inhibitors in complex mixtures.

Abstract:A new protocol is proposed for magnetic loading and sensitive electrochemical detection of phenol via the tyrosinase cross-linked mesoporous magnetic core/shell microspheres. The mesoporous magnetic microspheres, characterized by transmission electron microscopy, N2 adsorption/desorption isotherms, and magnetic curve displays high capacity for enzyme immobilization and strong magnetism to adhere to the magnetic electrode surface without any additional adhesive reagent. The biosensor exhibits a wide linear response to phenol ranging from 1.0×10−9 to 1.0×10−5M, a high sensitivity of 78μAmM−1, a low detection limit of 1nM, and a fast response rate (less than 5s). The proposed method is simple, rapid, inexpensive and convenient in electrode renewal, which is recommended as a promising experimental platform for wider applications in biosensing.
[Quantitative Analysis Method of Shengxuebao Mixture by HPLC-UV-MS Based on Quality by Design Concept].

Abstract:In this study, the HPLC-UV-MS method for the simultaneous determination of eight active ingredients of Shengxuebao Mixture were developed based on the concept of quality by design（QbD）with a stepwise optimization approach. After the analytical target profile（ATP）had been defined, albiflorin, paeoniflorin, 2, 3, 5, 4'-tetra-hydroxy-stilbene-2-O-β-D-glucopyranoside, specnuezhenide, ecliptasaponin D, emodin, calycosin-7-glucoside, and astragaloside Ⅳ were identified as the indicator components. The resolution and the signal-to-noise ratio of indicator components were then selected as critical method attributes (CMA) for the first step optimization. According to the results collected from fractional factorial design, critical method parameters (CMP) were determined with a multiple linear regression method, which included the amount of acid addition in the mobile phase, temperature, gradient, and wavelength. After that, the amount of acid addition and the wavelength were optimized to improve the resolution and the signal-to-noise ratio of the indicator components. The peak symmetry factors of specnuezhenide and emodin were then set as CMA for the second step optimization. The Box-Behnken designed experiments were conducted. The temperature and gradient were optimized after modelling. The design space were calculated and verified. The optimized analytical method was validated, and the results showed a good precision, accuracy and stability, which means that it can be used for the quantification of the indicator components in Shengxuebao Mixture.

Abstract:The inhibition of tyrosinase is considered to be a common therapeutic strategy for some hyperpigmentation disorders. Screening of tyrosinase inhibitors is of great significance to the treatment of pigmentation diseases. In this study, tyrosinase was covalently immobilized on magnetic multi‐walled carbon nanotubes for the first time, and the immobilized tyrosinase was applied for ligand fishing of tyrosinase inhibitors from complex medicinal plants. The immobilized tyrosinase was characterized by transmission electron microscopy, atomic force microscopy, Fourier‐transform infrared spectroscopy, vibrating sample magnetometry, and thermo‐gravimetric analyzer, which indicated that tyrosinase was immobilized onto magnetic multi‐walled carbon nanotubes. The immobilized tyrosinase showed better thermal stability and reusability than the free one. The ligand was fished out from Radix Paeoniae Alba and identified as 1,2,3,4,6‐pentagalloylglucose by ultra‐performance liquid chromatography‐quadrupole time‐of‐flight high‐resolution mass spectrometry. 1,2,3,4,6‐pentagalloylglucose was found to be a tyrosinase inhibitor with similar half maximal inhibitory concentration values of 57.13 ± 0.91 μM compared to kojic acid (41.96 ± 0.78 μM). This work not only established a new method for screening tyrosinase inhibitors but also holds considerable potential for exploring the new medicinal value of medicinal plants.

Abstract:A simple and rapid method for screening of tyrosinase (TYR) inhibitors present in traditional Chinese medicines (TCMs) was developed by combining ligand fishing and the fluorescent enzymatic assay based on dopamine-functionalized carbon quantum dots (CQDs-Dopa). Ligands of the enzyme present in the TCM extractions were firstly adsorbed on the enzyme-modified magnetic beads, and then the beads were magnetically separated and subjected directly to the CQDs-Dopa-based fluorescent assay. Finally, compounds were desorbed from the “active” beads and identified with ultra-performance liquid chromatography-triple quadrupole mass spectrometry. A known natural TYR inhibitor quercetin was selected to assess the feasibility and quantification performance of this method, and good linearity in the range of 0.01–0.16 mM (R2 = 0.992) with a low detection limit of 0.004 mM was obtained. This method was then applied to screen TYR inhibitors present in Scutellaria baicalensis and Sophora flavescens. Six TYR inhibitors including baicalin (1), baicalein (2), wogonin (3), oroxylin A (4), kurarinone (5), and sophoraflavanone G (6) were found, among which 1–4 were firstly discovered in this work. This is the first report on the in situ assessment of the target compounds obtained by ligand fishing in the form of a mixture, which exhibited the combined advantages of specific extraction ability of ligand fishing and the high sensitivity of CQDs-based fluorescent assay, showing great potential for fast screening of enzyme inhibitors from TCMs.Graphical abstract

Abstract:Background and Objective: Taohongsiwu decoction, a traditional Chinese medicine formula has been widely developed to treat thrombotic diseases for hundreds of years. It contains variations of multi-components, which pose a serious challenge to its quality control. This study aimed to develop a simple and reliable ultra high performance liquid chromatography coupled with diode array detector method to separate and detect the constituents of the decoction. Materials and Methods: A total of 15 compounds was detected and 6 compounds (including ferulic acid, hydroxysafflor yellow a, ligustilide, amygdalin, 5-hydroxymethyl furfuraldehyde and paeoniflorin) were tentatively identified, using high performance liquid chromatography coupled with diode array detector method. Results: The four components (Ferulic acid, hydroxysafflor yellow a, ligustilide, amygdalin) showed good regression (R>0.9996) within test ranges and the recovery method ranged from 101.73-106.44%. Ten batches of taohongsiwu decoction were analyzed. The similarity scores of common peaks from these samples ranged from 0.957-1.000, which indicating that samples were highly correlated. Conclusion: It is concluded that this extraction method was found to be sensitive and simple, so it was feasible for comprehensive quality control of THSWD.

Abstract:In this study, tyrosinase was immobilized on carboxyl functionalized silica‐coated magnetic nanoparticles for the first time to be used for fishing of tyrosinase's ligands present in complex plant extract. The immobilized tyrosinase was characterized by transmission electron microscopy, vibrating sample magnetometry, Fourier transform infrared spectroscopy, thermo‐gravimetric analyzer, and atomic force microscopy. The reusability and thermo‐stability of the immobilized tyrosinase were found significantly superior to its free counterpart. Two tyrosinase's ligands, i.e., caffeic acid (1) and rosmarinic acid (2), were fished out from extract of the traditional Chinese medicine Prunellae Spica by the immobilized tyrosinase. Compound 1 was found to be an activator of the enzyme with the half maximal effective concentration value of 0.27 ± 0.06 mM, while compound 2 was an inhibitor with the half maximal inhibitory concentration value of 0.14 ± 0.03 mM. Taking advantage of the convenience of magnetic separation and specific extraction ability of ligand fishing, the proposed method exhibited great potential for screening of bioactive compounds from complex matrices. Abstract:Tyrosinase-based TLC (thin layer chromatography) was developed for screening of anti-melanogenic drugs. In particular, this technique enables researchers to identify melanogenic inhibitor(s) in tested mixtures with the naked eye. In comparison with traditional colorimetric screening assays for tyrosinase inhibitor(s), not only is tyrosinase-based TLC a more cost-effective option (nearly one-tenth the enzyme cost of colorimetric methods) but also is a more sensitive detection approach for kojic acid (KA), a standard anti-melanogenic drug. The detection limit of tyrosinase-based TLC and colorimetric tyrosinase assay for KA was 0.0125 and 1.25 μg, respectively, demonstrating that the former was 100-fold more sensitive than the latter to determine the tyrosinase inhibitory rate of KA. Furthermore, the results of this method have demonstrated excellent precision by Gage Repeatability and Reproducibility (Gage R&R), with the variation of total Gage R&R being 28.24%. To verify the applicability of tyrosinase-based TLC, this platform was employed to screen melanogenic inhibitor(s) from Ganoderma formosanum extracts and two of all fractions (GFE-EA F4, F5) obtained showed depigmenting activity. It is noteworthy that these two fractions also exerted anti-melanogenesis activity on zebrafish, therefore verifying the credibility of tyrosinase-based TLC. In sum, this technique provides new insight into the discovery of novel melanogenic inhibitor(s).

Abstract:Using the commercially available and economical 6-hydroxycoumarin (6-HC) as the substrate, a dual-emission ratiometric fluorescence sensor was developed to detect tyrosinase (TYR) activity based on 3-aminophenyl boronic acid functionalized quantum dots (APBA-QDs). TYR can catalyze 6-HC, a monohydroxy compound, to form a fluorescence-enhancing o-hydroxy compound, 6,7-dihydroxycoumarin. Owing to the special covalent binding between the o-hydroxyl and boric acid groups, APBA-QDs react with 6,7-dihydroxycoumarin to form a five-membered ring ester dual-emission fluorescence probe for TYR. With an increase in TYR activity, the fluorescence at 675 nm originating from the QDs is gradually quenched, whereas that at 465 nm owing to 6,7-dihydroxycoumarin increases. Referencing the decreasing signal of the dual-emission probe at 675 nm to measure the increasing signal at 465 nm, a ratiometric fluorescence method was established to detect the TYR activity with high sensitivity and selectivity. Under the conditions optimized via response surface methodology, a linear range of 0-0.05 U/mL was obtained for the TYR activity. The detection limit was as low as 0.003 U/mL. This sensing strategy can also be adopted for the rapid screening of the TYR inhibitors.
Ultra-high performance liquid chromatography coupled with time-of-flight mass spectrometry screening and analysis of potential bioactive compounds from traditional chinese medicine Kai-Xin-San, using a multivariate data processing approach and the MetaboLynx tool

Abstract:Traditional Chinese Medicine (TCM) has been used in clinical practice for several thousand years, with an indispensable role in prevention and treatment of disease using multiple ingredients. Kai-Xin-San (KXS) is a TCM formula consisting of four herbs, Ginseng Radix, Polygalae Radix, Poria and Acori Tatarinowii Rhizoma, which has been used to treat Alzheimer's disease, depression and Parkinson's disease. However, the constituents absorbed into blood after oral administration of KXS remain unknown. Here, a sensitive and rapid method using ultra performance liquid chromatography/quadrupole time-of-flight mass spectrometry (UPLC/Q-TOF-MS/MS) combined with a multivariate data processing approach (Mdpa) and MetaboLynx software tool was used for analysis and identification of bioactive components and their metabolites in rat plasma following oral administration of KXS. A hyphenated MS/MS analyzer was used for determination of accurate fragment ions and mass spectrometric fragmentation mechanisms, and enhanced data acquisition. Metabolite elucidation was performed using UPLC/Q-TOF-MS/MS coupled with MetaboLynx tool. With the established method, a total of 49 peaks were tentatively characterized in vivo based on MS and MS/MS data, and comparison with available databases. Of the 49 compounds tentatively characterized in rat plasma, seven metabolites were detected and identified by comparing their fragmentation patterns using MetaboLynx software tool. On the basis of the chromatographic peak area, the glucuronidated conjugates were identified as major metabolites. This work demonstrated the potential of the UPLC/Q-TOF-MS/MS with Mdpa and MetaboLynx for rapid, simple, reliable and automated identification of metabolites of herbal medicine.

Abstract:Tyrosinase (TYR) is as a well-known polyphenol oxidase and important biomarker of melanocytic lesions. Thus, developing powerful methods to determine TYR activity is of great value in the early diagnosis of skin disease. Direct surface-enhanced Raman scattering (SERS) detection of biomolecules is usually affected by non-specific interference and complicate structure of the analytes. It is a challenge to develop Raman-active molecules with specific recognition to analytes in complex media. Here, we report a novel colorimetric and surface-enhanced Raman scattering (SERS) dual-readout assay for the determination of TYR using commercially available and economical 4-mercaptophenyl boronic acid (4-MPBA) as a Raman-active and recognition molecule. 4-MPBA provides a unique interactive boronic acid group to the diol group of TYR substrate and exhibits good SERS signal. Also, the introduction of magnetic beads could promptly improve the anti-interference ability of dual-mode sensor. The TYR-incubated tyramine-modified magnetic beads could obviously change the concentration of 4-MPBA-AuNPs in the presence of O<sub>2</sub> and ascorbic acid, where the ultraviolet visible (UV-vis) absorption and SERS intensity were directly related to the concentration of TYR added. The dual-mode sensor had a rapid response to TYR within 1 min under optimized conditions and had high selectivity for TYR with a limit of detection at 0.001 U/mL. In addition, the dual-mode strategy showed promising prospects in the determination of TYR activity in serum samples and could be used to screen TYR inhibitors.

Abstract:We developed an approach for screening bioactive compounds from botanical drug using multiple target-immobilized magnetic beads coupled with high performance liquid chromatography-mass spectrometry. This novel approach was called magnetic beads based multi-target affinity selection-mass spectrometry (MT-ASMS). It can enrich and identify different types of ligands from mixture extracts. Multiple targets (maltase, invertase, lipase) were immobilized on the magnetic beads by covalent linkage using 1-(3-dimethyl-aminopropyl)-3-ethyl-carbodiimide (EDC) and N-hydroxysuccinimide (NHS) as reaction reagents, respectively. The properties of enzyme conjugated magnetic beads were characterized using transmission electron microscopy, X-ray diffractometer and vibration sample magnetometer. Several factors including pH, ion strength, incubation time and temperature were optimized using three known ligands (caffeic acid, ferulic acid, and hesperidin). The established MT-ASMS approach was applied to screening for ligands from a Chinese medicine "Tang-Zhi-Qing", which was used to treat type II diabetes in China. Seven bound compounds were identified via liquid chromatography-mass spectrometry (LC/MS). Five active compounds including 2,3,4,6-tetra-O-galloyl-D-glucose, 1,2,3,4-tetra-O-galloyl-D-glucose, 1,2,3,4,6-penta-O-galloyl-d-glucose, quercetin-3-O-β-D-glucuronide and quercetin-3-O-β-D-glucoside were identified and their activities were validated by conventional inhibitory assay. Our findings suggested that the proposed approach is efficient in screening compounds with multiple activities from extracts of botanical drugs.

Abstract:Bao-Yuan Decoction (BYD), a widely used traditional Chinese medicine formula, is worth developing into modern dosage forms. To assess the quality of traditional decoction, the commonly used ultra-performance liquid chromatography coupled with diode array and evaporative light scattering detection (UPLC-DAD/ELSD) method was initially applied to develop the analytical methods for the qualitative fingerprints and simultaneous quantitation of multiple marker compounds in BYD. Based on 16 batches of BYD prepared from multiple batches of qualified crude herbs combined randomly, the characteristic fingerprints were generated, with 41 and 19 common peaks detected by DAD and ELSD, respectively. Furthermore, ginsenosides Re, Rg1 and Rb1, calycosin-7-glucoside, calycosin, liquiritin, isoliquiritin apioside, isoliquiritin, glycyrrhizic acid and cinnamic acid were qualified as marker compounds to represent the herbs composing the formula. The characteristic fingerprints and the content ranges of multiple batches of the decoction were obtained, thus providing guidance for the quality control of modern dosage forms. The combination of these qualitative and quantitative methods will be an effective operational measure by which to evaluate and control the quality of BYD from traditional decoction to modern dosage forms.

Abstract:A new method for screening tyrosinase inhibitors from traditional Chinese medicines (TCMs) was successfully developed by capillary electrophoresis with reliable online immobilized enzyme microreactor (IMER). In addition, molecular docking study has been used for supporting inhibition interaction between enzyme and inhibitors. The IMER of tyrosinase was constructed at the outlet of the capillary by using glutaraldehyde as cross-linker. The parameters including enzyme reaction, separation of the substrate and product, and the performance of immobilized tyrosinase were investigated systematically. Because of using short-end injection procedure, the product and substrate were effectively separated within 2 min. The immobilized tyrosinase could remain 80% active for 30 days at 4°C. The Michaelis-Menten constant of tyrosinase was determined as 1.78 mM. Kojic acid, a known tyrosinase inhibitor, was used as a model compound for the validation of the inhibitors screening method. The half-maximal inhibitory concentration of kojic acid was 5.55 μM. The method was successfully applied for screening tyrosinase inhibitors from 15 compounds of TCM. Four compounds including quercetin, kaempferol, bavachinin, and bakuchiol were found having inhibitory potentials. The results obtained in this work were supported by molecular docking study.

Abstract:Sunitinib, N-desmethyl imatinib, dasatinib, imatinib, and bosutinib are tyrosine kinase inhibitors (TKIs) that are commonly employed in the treatment of a multitude of cancers. However, the inappropriate concentrations of TKIs can result in ineffective treatment or the emergence of multiple adverse effects. Consequently, the development of a rapid and sensitive analytical method for TKIs is of paramount importance for the safe administration of drugs. In this work, solid-phase microextraction (SPME) probe combined with an electrospray ionization mass spectrometry (ESI-MS) coupling platform was constructed for rapid and sensitive determination of TKIs. The covalent organic frameworks (COFs) coated SPME probe was made of 2,4,6-tris(4-aminophenyl)-1,3,5-triazine (TAPT) and 2,5-dibutoxyterephthalaldehyde (DBTA) by in-situ layer-by-layer chemical bonding synthesis strategy. The TAPT-DBTA-SPME probe exhibited several advantageous properties which rendered it suitable for the enrichment of TKIs. Under the optimal conditions, the developed analytical method demonstrated a broad linear range (0.05-500.00 µg/L), a low limit of detection (0.02 µg/L) and a high enrichment factor (51-203) for TKIs. The developed analytical method was successfully applied to a pharmacokinetic study of TKIs in mouse plasma and tissue matrix, demonstrating that the proposed analytical method has promise for clinical applications and metabolic monitoring.

Abstract:In the present study, the preparation of the magnetic phosphodiesterase-5 linked Fe3O4@ SiO2 nanoparticles was successfully achieved by amide reaction and the magnetic phosphodiesterase-5 linked Fe3O4@SiO2 nanoparticles were evaluated as a new adsorbent for magnetic dispersive solid-phase extraction of ligands from medicinal plant samples before the analysis by UHPLC-Q-TOF/MS. The prepared phosphodiesterase-5 linked Fe3O4@SiO2 nanoparticles were characterized by X-ray diffraction, Fourier transform infrared spectroscopy, transmission electron microscopy, vibration sample magnetometer and potential laser particle size analyzer. The effects of EDC concentration, incubation time and bead-protein ratio on the amount of immobilized protein were studied. The main experimental parameters affect extraction efficiency of ligands, such as wash times, wash solvents, incubation pH, ion strength and incubation temperature, were investigated and optimized by using echinacoside as a model compound. The absolute recovery of echinacoside was ranged from 98.36%-102.16% in Cistanche tubulosa sample under the optimal extraction conditions. Good linearity was observed in the investigated concentration range of 0.006 mgmL-1-0.97 mgmL-1(R2 = 0.9999). The limit of detection was 0.002 mgmL-1. The RSDs of within-day and between-day precision were less than 2.3%. Due to the excellent magnetic behavior of Fe3O4@SiO2 nanoparticles, the proposed method was shown to be simple and rapid. Remarkably, the magnetic phosphodiesterase-5 linked Fe3O4@SiO2 nanoparticles could be recycled for ten times with loss of 10% activity.
Construction of QSAR model based on cysteine-containing dipeptides and screening of natural tyrosinase inhibitors

Abstract:Considering that natural products as tyrosinase inhibitors are considered to be safe, with little or no toxic side effects and friendly to the environment, it is urgent to develop a new recognition strategy for natural tyrosinase inhibitors. In current study, an integrated computational analysis was conducted on Cys-containing dipeptides with high tyrosinase inhibitory activity. Firstly, molecular fingerprint similarity (FS) clustering analysis was performed on the target molecule using machine learning. Secondly, genetic algorithm was used to construct two kinds of highly accurate QSAR models (R2 = .978 and .984, respectively) with Cys at C-terminal and N-terminal. Finally, three novel natural candidate inhibitors (NP1, NP2, NP3) were discovered using Molnatsim natural product cluster library, automated screening process and QSAR based on the maximum common substructure (MCS) algorithm, their IC50pre were 260.96, 3.37 and 0.05 μm/mol. Pharmacokinetic predictions showed that NP2 and NP3 had high Bioavailability Score (BS) and Gastrointestinal (GI) absorption, and molecular dynamics simulations further validated the stability of these novel natural candidate inhibitors in binding to tyrosinase. In conclusion, our results provide new ideas for discovering new activities of natural products, and provide an accurate QSAR model for developing novel tyrosinase inhibitors based on MCS Cys-containing dipeptides. PRACTICAL APPLICATIONS: Tyrosinase is related to the occurrence of diseases such as excessive melanin deposition such as freckles and chloasma, and studies have shown that neurodegeneration associated with Parkinson's disease and Huntington's disease is also related. In addition, enzymatic browning on the surface of fresh fruit and vegetable slices will shorten the shelf life and affect their quality. Therefore, screening, designing and developing efficient tyrosinase inhibitors is very important in the fields of medicine, cosmetics, food and so on.
Determination of bioactive flavonoids using β-cyclodextrin combined with chitosan-modified magnetic nanoparticles

Abstract:s To accurately determine flavonoids (rutin, quercetin or kaempferol), it is necessary to extract them from complex matrices. The ultrasound-assisted magnetic dispersion microsolid phase extraction technique has been predominantly used for separation and enrichment of the target analytes. The combination of magnetic chitosan nanoparticles and a deep eutectic supramolecular solvent (DESP) is likely to enhance the efficiency of flavonoid extraction from food. In this study, adsorbents were prepared by modifying chitosan with magnetic nanoparticles, and the eluent was a DESP derived from β-cyclodextrin and an organic acid. The successful preparation of these materials was confirmed by FTIR, XRD, FE-SEM and 1 H NMR. The extraction recovery rates exceeded 93 %, with limits of detection and quantitation ranging from 0.9 to 2.4 μg/L and 2.7 to 7.2 μg/L, respectively, and the flavonoid clearance rates for ABTS and DPPH radicals reached 100 %. Therefore, the integration of magnetic chitosan nanoparticles with the DESP provides a new and efficient method for the extraction of flavonoids while also presenting a potential application of the DESP in separations.
