Abstract:Samples of Pericarpium Citri Reticulatae (Citrus reticulata 'Chachi', CP-‘Chachi’), which collected in December of 2017, 2013, 2003 and 1993 with the natural aging process for 5, 10, 20 and 30 years, were used to compared the changes in total polyphenols, total flavonoids and fungal community composition. The relationship among these factors was analyzed. The content of flavonoids in CP-‘Chachi’ was qualitatively and quantitatively determined by HPLC system. ITS (Internal Transcribed spacer) sequencing techniques were used to detect the changes of microbial communities during the aging process of CP-‘Chachi’. The results indicated that the levels of flavonoids and total polyphenols, except for hesperidin, increased aging time, with the most significant growth observed in 5-O-desmethyl nobiletin at 298.36%, while hesperidin decreased by 32.52%. The three most abundant genera were Xeromyces, Symmetrospora and Cladosporium, and the highest number of genera was found in CP-2013. The community composition of CP-1993 and CP-2003 was highly similar, Xeromyces accounted for 84.04% and 83.58% in CP-1993 and CP-2003, respectively. The findings demonstrated that only the abundance of Xeromyces was positively correlated with the accumulation of active components during the aging process of CP-‘Chachi’. This indicated that Xeromyces was considered the dominant genus responsible for the aging quality of CP-'Chachi'. The relationship between the changes in flavonoids and fungal diveresity was preliminarily verified, which provided new insights into the study of the aging mechanism of CP-‘Chachi’ and its influencing factors on quality.

Abstract:Abstract Background: The mature pericarp of Citri Reticulatae 'Chachi' (PCRC) is one of the six traditional Chinese medicinal materials that should be used after long storage, and it was regarded that the longer the medicine was stored, the better. However, the aging mechanism of the medicine is not clear. Methods: In order to further investigate the effect of aging on the main active flavonoids of PCRC, ultra-performance liquid chromatography coupled with triple quadrupole mass spectrometry (UPLC-MS/MS) and metabolomics analysis were used to analyze the flavonoids of PCRC stored for different periods. Results: In the results, 219 flavonoids were detected. 5,7,3',4',5'-pentamethoxy dihydroflavone and 2'-hydroxy-3,4,5,3'4',6'-hexamethoxychalcone were found from PCRC for the first time. According to the clustering analysis of metabolites, aging times of 0, 1, and 2 were clustered into one group, and aging times of 3, 4, and 29 were clustered into the other group. Quantitative analysis showed that the former group contained a greater amount of 4 flavonoids than the latter group, while the latter group contained a greater amount of 15 polymethoxyflavonoids. The newly harvested PCRC was compared with the other 5 groups of PCRC (stored for 1, 2, 3, 4, and 29 years). Eight flavonoids, tectochrysin, apigenin, 2'-hydroxyisoflavone, luteolin, 6-hydroxyluteolin, gallocatechin, quercetin -O- acetylhexoside and apigenin -7-O-[β-D-glucuronide (1→2) -O-β-D-glucuronide], were used as marker components to discriminate newly harvested PCRC and aging PCRC. In addition, the antioxidant potency composite index (APC) indicated that the PCRC stored for three or four years had stronger antioxidant activity than the PCRC stored for other periods. By means of molecular docking, it was reviewed that the amount of antiviral components against SARS-CoV-2 in freshly harvested PCRC was significantly higher than that in aging PCRC. The results in this study supplied scientific data for quality control, evaluation, and rational utilization of PCRC and basic information for further analysis of the metabolic regulation of the active components of the PCRC.
Aged Pericarpium Citri Reticulatae 'Chachi' Attenuates Oxidative Damage Induced by tert-Butyl Hydroperoxide (t-BHP) in HepG2 Cells

Abstract:This study investigated the protective effects of aged Pericarpium Citri Reticulatae 'Chachi' (PCR-C) on tert-butyl hydroperoxide (t-BHP)-induced oxidative damage in HepG2 cells. According to HPLC analysis, PCR-C aged 10 years (PCR-C10) had the highest flavonoids content, especially polymethoxyflavones (PMFs), compared with the fresh peel of Citrus reticulata cv. 'Chachiensis' and PCR-C aged 1, 3, and 5 years. Then, flavonoids-rich PCR-C samples and non-flavonoids-rich PCR-C samples (NF) were prepared by extracting and purifying PCR-C of different aging periods, for further cell experiments. Pretreatment with flavonoids-rich PCR-C samples (particularly PCR-C10) considerably reversed t-BHP-induced oxidative damage in HepG2 cells by improving cell viability, increasing SOD activity and GSH levels and reducing the overproduction of ROS and MDA. Correlation analysis further indicated that the accumulation of PMFs, mainly 5,6,7,4'-tetramethoxyflavone and nobiletin, was the main reason that PCR-C10 maintained the redox balance in HepG2 cells. These findings provided direct evidence for the cellular antioxidant activity of aged PCR-C and a guide for PCR-C's classification, authentication and rational use.
Flavonoids contribute most to discriminating aged Guang Chenpi (Citrus reticulata 'Chachi') by spectrum-effect relationship analysis between LC-Q-Orbitrap/MS fingerprint and ameliorating spleen deficiency activity

Abstract:To further explore the mechanism of "the longer storage time, the better bioactivity" of aged Guang Chenpi, the dry pericarp of Citrus reticulata 'Chachi' (CRC), a series of activity assessments were performed on spleen deficiency mice. The constituents in CRC with different storage years were analyzed by LC-Q-Orbitrap/MS. A total of 53 compounds were identified, and CRC stored for more than 5 years showed higher flavonoid content, especially that of polymethoxyflavones. Anti-spleen deficiency bioactivity analysis among various CRC with different storage years showed aged CRC (stored for more than 3 years) could significantly alleviate fatigue and depression behaviors much better, increase D-xylose and gastrin secretion, and upregulate the expression of the linking protein occludin in the colon walls. Results from 16S rDNA sequencing showed that aged CRC could downregulate the abundance of Enterococcus, Gemmata, Citrobacter, Escherichia_Shigella, and Klebsiella, which were significantly overrepresented in the model group. Bacteroides, Muribaculum, Alloprevotella, Paraprevotella, Alistipes, Eisenbergiella, and Colidextribacter were downregulated in the model group but enriched in the CRC groups. At last, the spectrum-effect relationship analysis indicated that flavonoids such as citrusin III, homoeriodictyol, hesperidin, nobiletin, and isosinensetin in aged CRC showed the highest correlation with better activity in ameliorating spleen deficiency by regulating gut microbiota. Flavonoids contribute most to discriminating aged CRC and could disclose the basis of "the longer storage time, the better bioactivity" of aged Guang Chenpi.

Abstract:Citri Reticulatae ‘Chachi,’ also called “Guang Chen pi” (GCP), is a geoherb of Citri reticulatae pericarpium(CRP), mainly produced in Xinhui, Guangdong, China. It has been a food and traditional medicine for thousands of years. GCPs are popular green food, and can be divided into three picking times and different aging periods according to edible quality. Despite many pharmaceutical studies, there is little information on edible differences among GCP types. The dynamic changes in dietary value for different aging and picking times were investigated through quantifying polyphenols, soluble proteins, ascorbic acid, reducing sugars, polysaccharides, and β-carotene. The browning index (BI) and pH were also measured. Comprehensive analysis results showed that nutrient contents in brownish and reddish peels were close, and higher than in green peel. With aging time, ascorbic acid and β-carotene decreased, while dissolution of polysaccharides, polyphenols, and proteins increased, likely contributing to the aroma change. At the same time, pH of the system gradually decreased, but BI increased. Mathematical path analysis of related substrates and BI showed the greatest correlation of polyphenols with BI. The browning mechanism likely started with ascorbic acid degradation that caused reduced antioxidation, contributing to further reaction of polyphenols with other compounds to produce browning.

Abstract:Dry mature pericarp of Citrus reticulata "Chachi" (PCR), Pericarpium Citri Reticulatae Chachiensis, is a traditional Chinese medicine that displays characteristics of different usage at different harvest times in clinical use. The corresponding changes in the bioactive components in PCR from different harvest times remain unclear. Therefore, in this study, broadly targeted metabolomics technology was used to compare the differences in bioactive components among pericarps of PCR, which are the raw material of PCR at different growth stages. In the results, 210 kinds of flavonoid metabolites were detected. The content of hesperidin in red PCR harvested in December was higher than that in Citri Reticulatae Pericarpium Viride (CRPV) and reddish PCR harvested from July to November. Furthermore, the content of nobiletin, tangeretin, and 3,3 ',4 ',5,6,7,8-heptamethoxyflavone in CRPV from July to September was higher than that in the PCR harvested at other times. In addition, the result of cluster analysis and PCA showed that CRPV harvested from July to September had an obvious grouping pattern with the reddish PCR and the red PCR harvested from October to December. Differential metabolites in six comparison groups (A1 vs. A6, A1 vs. A2, A2 vs. A3, A3 vs. A4, A4 vs. A5, A5 vs. A6) were 67, 48, 14, 51, 42, and 40, respectively. The common differential metabolite of four comparison groups was 3 ',4 ',7-trihydroxyflavone (A1 vs. A2, A2 vs. A3, A3 vs. A4, A4 vs. A5). All the flavonoid differential metabolites screened were enriched in 16 metabolic pathways. Moreover, the results of the evaluation of the total antioxidant capacity indicated that CRPV in August was a suitable raw material for the production of antioxidants. Through molecular docking, the content of potential anti-SARS-CoV-2 components in the PCR in October was higher than that in the PCR in other periods. These results further proved that PCR at different harvest times was endowed with different efficacy and usage due to the difference in the accumulation of bioactive components.
Flavor characterization of Citri Reticulatae Pericarpium (Citrus reticulata 'Chachiensis') with different aging years via sensory and metabolomic approaches

Abstract:Guangchenpi (GCP), which is the peel of Citrus reticulata 'Chachiensis', is widely used as an herbal medicine, tea and food ingredient in southeast Asia. Prolonging its aging process results in a more pleasant flavor and increases its profitability. Through the integration of sensory evaluation with flavoromic analysis approaches, we evaluated the correlation between the flavor attributes and the profiles of the volatiles and flavonoids of GCP with various aging years. Notably, d-limonene, γ-terpinene, dimethyl anthranilate and α-phellandrene were the characteristic aroma compounds of GCP. Besides, α-phellandrene and nonanal were decisive for consumers' perception of GCP aging time due to changes of their odor activity values (OAVs). The flavor attributes of GCP tea liquid enhanced with the extension of aging time, and limonene-1,2-diol was identified as an important flavor enhancer. Combined with machine learning models, key flavor-related metabolites could be developed as efficient biomarkers for aging years to prevent GCP adulteration.
Identification of Crucial Polymethoxyflavones Tangeretin and 3,5,6,7,8,3′,4′-Heptamethoxyflavone and Evaluation of Their Contribution to Anticancer Effects of Pericarpium Citri Reticulatae ‘chachi’ During Storage

Abstract:Pericarpium Citri Reticulatae ‘Chachi’ (PCR-C), rich in polymethoxyflavones (PMFs), has potential anticancer bioactivity and its quality will be improved during storage. However, the main factors influencing the PCR-C quality during its storage remain unclear. In this study, multivariate analysis was performed to investigate free and bound PMFs of PCR-C during storage. The anticancer effects of purified PCR-C flavonoid extracts (PCR-CF) and the important PMFs were evaluated using A549 cells. The results showed that PCR-C samples exhibited remarkable differences in free PMFs during storage, which fell into three clusters: Cluster 1 included fresh (fresh peel) and PCR-C01 (year 1); Cluster 2 consisted of PCR-C03 (year 3) and PCR-C05 (year 5); and PCR-C10 (year 10) was Cluster 3. 3,5,6,7,8,3′,4′-heptamethoxyflavone, tangeretin, and isosinensetin were identified as the most important PMFs distinguishing the various types of PCR-C according to its storage periods. Moreover, PCR-CF inhibited A549 cell proliferation and induced cell cycle arrest at G2/M phase, cell apoptosis, and ROS accumulation, and all anticancer indices had an upward tendency during storage. Additionally, tangeretin and 3,5,6,7,8,3′,4′-heptamethoxyflavone exhibited anticancer effects on A549 cells, whereas isosinensetin displayed no anticancer effect, indicating that tangeretin and 3,5,6,7,8,3′,4′-heptamethoxyflavone jointly contributed to anticancer activity of PCR-C during storage. PCR-CF and the most important PMFs killed cancer cells (A549 cells) but had no cytotoxicity to normal lung fibroblast cells (MRC-5 cells). Overall, the high quality of long-term stored PCR-C might be due to the anticancer effects of tangeretin and 3,5,6,7,8,3′,4′-heptamethoxyflavone.
The effects of core microorganism community on flavor compounds and active substances during the aging process of Citri Reticulatae Pericarpium

Abstract:Citri Reticulatae Pericarpium (CRP) is a traditional herbal and food spice, the flavor and active compounds content of Xinhui CRP improves with aging. To investigate the pattern of microbial community succession during the aging of Xinhui CRP and its correlation with changes in flavor compounds, the high-throughput sequencing, HPLC, and GC-IMS were used to analyze the microbial community, flavonoids, and flavor compounds of five different aging years in this study. The results revealed different dominant microbial communities in Xinhui CRP at different aging time, and unclassified Bacteria were the predominant bacterial genus during 10-15 years of aging. As the aging time increases, the abundance of microbial community decreases and gradually stabilizes. At the fungal genus level, Xeromyces (>99 %) were the dominant genus during the 10-15 years aging time and had a significant correlation with polymethoxyflavones (PMFs), and the concentrations of PMFs increased with the progression of aging years. The GC-IMS results revealed distinctive flavor profiles in Xinhui CRP across different aging years, floral and fruity aromas, such as heptanal, 3-methyl-3-butenol, and 1-butanol, among others, with increasing aging years. A comprehensive correlation analysis further elucidates the close relationship between the core microorganism community and flavor formation in Xinhui CRP (p < 0.05). Notably, Pseudomonas and Escherichia Shigella exhibited significant correlations with beta-pinene and alpha-pinene, whereas Aureobasidium and Sarcopodium were associated with nerol and α-phellandrene (p < 0.05). This study provides new ideas for accelerating the good quality and flavor of Xinhui CRP during the aging process from the perspective of key microorganisms.
Influence of the Stage of Ripeness on the Phytochemical Profiles, Antioxidant and Antiproliferative Activities in Different Parts of Citrus Reticulata Blanco Cv. Chachiensis

Abstract:Citrus is a rich source of nutritive and bioactive compounds. Impact of fruit tissues and ripening stage on phytochemical profile, antioxidant capacity and antiproliferative activity was investigated in Citrus reticulate Blanco cv. Chachiensis (Chachi). Regardless of different ripening stage, peel had the highest contents of phenolics and flavonoids, strongest antioxidant activities resolved by PSC, ORAC and CAA assay and antiproliferative activities against MCF-7, HepG2 and SiHa cancer cell. Increased level of total phenolics, hesperidin and naringin, and elevated antioxidant capacities were detected in peel during maturity, whereas highest amount of nobiletin and strongest antiproliferative activities were explored in immature peel. In flesh the vitamin C content increased during ripening, while lower phenolic content, smallest ORAC value and weakest antiproliferative activity were resolved in mature flesh. The flavonoid content and antioxidant capacity in seed were comparable with flesh and relatively constant during ripening, suggesting seeds as a valuable source of flavonoid extraction for further utilization. The characteristic flavonoids, nobiletin, tangeretin, hesperetin and naringenin inhibited proliferation of SiHa, HepG2 and MCF-7 cell dose-dependently, thus suggesting their contribution and potential application. Abstract:The chemical composition, antioxidant and antimicrobial activity of Pericarpium Citri Reticulatae (PCR) essential oil obtained using an improved Clevenger type apparatus were studied. Among the five different PCRs examined the highest yield of essential oil was found in Chachi 2004 (harvested and stored in 2004) and the lowest in Chachi 2008 (harvested and stored in 2008). Fifty three different volatile compounds were determined, including terpenic hydrocarbons, alcohols, aldehydes, ketones and esters. D-limonene, one of terpenes, was the major constituent in PCR. The antioxidant capacity of PCR essential oil varied considerably with the duration of storage time, and the oil from Chachi 1994 has the strongest ferric-reducing antioxidant power. In addition, the essential oil possessed varying degrees of antimicrobial activity against Gram-positive bacteria (Staphylococcus aureus, Bacillus subtilis, Bacillus cereus), except Streptococcus faecalis, while had no effect on Gram-negative bacteria (Escherichia coli, Enterobacter cloacae).

Abstract:Pericarpium Citri Reticulatae ‘Chachi’ (PCR-C), abundant in flavonoids, is traditionally believed to enhance its pharmacological effects with long-term storage. The efficacy of these flavonoids is closely related to their intestinal absorption, which is primarily affected by P-glycoprotein (P-gp). This study explored the interaction between flavonoid absorption and P-gp in PCR-C flavonoid extracts (PCR-CF) over different storage periods using a Caco-2 monolayer model and KB/MDR cells, respectively. The transported concentrations (TC) of key flavonoids-hesperidin, isosinensetin, nobiletin, and tangeretin-significantly varied with the storage duration of PCR-CF. Notably, nobiletin and tangeretin enhanced the TC of these flavonoids by inhibiting P-gp activity, thereby influencing intestinal absorption of PCR-CF with different storage time. These findings provide a scientific basis for the traditional belief that long-stored PCR-C possesses superior medicinal properties.
Evaluation of chemical components and quality in Xinhui Chenpi (Citrus reticulata 'Chachi') with two different storage times by GC-MS and UPLC

Abstract:Xinhui Chenpi (XHCP) is a well-known type of Chenpi (CP) widely used as both a Chinese herb and a food ingredient. While previous studies have explored how the quality of CP changes over time, there has been limited research specifically on XHCP. This study aims to assess the chemical components and quality of XHCP based on total flavonoid content (TF), antioxidant activity (AA), and color value (CV) at two stages: freshly harvested (XHCP-0Y) and after 3 years of storage (XHCP-3Y). Thirty-eight common volatile compounds were identified, and the content of 17 compounds among them, nine nonvolatile compounds, which included one alkaloid (synephrine), three phenolic acids (PA, protocatechuic acid, vanillic acid, and ferulic acid), and five flavonoids (narirutin, hesperidin, sinensetin, nobiletin, and tangeretin), were firstly detected by the newly developed gas chromatograph-mass spectrometer (GC-MS) and ultra-performance liquid chromatography (UPLC) methods. Compared to XHCP-0Y, the content of 17 volatile compounds and synephrine decreased in XHCP-3Y to varying degrees, while the content of PA, five flavonoids, TF, AA, and CV increased. The reduction of dryness caused by volatile compounds and the enhancement of efficacy related to PA, flavonoids, and AA suggested improved quality of XHCP after 3 years of storage. The methods developed in this study show promise for evaluating the quality of XHCP during the aging process.

Abstract:The dried and aged pericarps of Citri Reticulatae are condiments and medicinal products in southeast and eastern Asia for hundreds of years, among which Pericarpium Citri Reticulatae ‘Chachiensis’ (PCR-C) is the premium one with obvious health benefits. In order to explore the microbiota in PCR-C and their relationship with the chemical components during aging, culture-independent methods were applied to investigate PCR-C microbiota for the first time. Here in different PCR-C samples, 16S rRNA gene amplicon sequencing revealed common central bacterial community, which were absent or only accounted for small proportion in fresh pericarps or jute bag controls. Bacillus and Lactococcus were the top two dominant genera in PCR-C with acidic pH (4.06–4.51) and low moisture (11.48%–19.13%). Several OTUs were found to closely relate with specific compositions in essential oils and phenolics, such as d-limonene and nobiletin, which contributed to PCR-C flavor and quality. As the first study to reveal the central bacterial communities in PCR-C, it provides new insights to improve the quality and aging process of traditional Pericarpium Citri Reticulatae, and lays foundation for functional characterization of the microbes within.
Inhibitory effects of Pericarpium Citri Reticulatae 'Chachi' flavonoid extracts during storage against NSCLC in vivo and in vitro

Abstract:Pericarpium Citri Reticulatae 'Chachi' (PCR-C), rich in flavonoids, shows antioxidative, anti-inflammatory, and antitumor effects, which was traditionally considered to be "the longer periods, the better". The bioactivity of flavonoids depends on gastrointestinal absorption and hepatic metabolism. This work aims to investigate which metabolites of flavonoids are the differential factors that mainly contributed to the variance of inhibitory effect of PCR-C flavonoid extracts (PCR-CF) against non-small cell lung cancer (NSCLC) during storage. The inhibitory effect of PCR-CF against NSCLC was evaluated by subcutaneous xenograft tumor model and Caco-2/HepG2 coculture model. Non-targeted metabolomics was employed to analyze the metabolic difference of PCR-CF during storage. Results showed that PCR-CF during storage inhibited tumor growth in vivo. Meanwhile, PCR-CF during storage inhibited A549 cell proliferation and induced cell apoptosis and cell cycle arrest at G2/M phase via ROS-dependent generation involving in MAPK pathway in vitro. Furthermore, 15 flavonoids metabo-lites were identified as marker differential metabolites affecting the variance of inhibitory effect of PCR-CF during storage against NSCLC. This study indicated that the inhibitory effects of PCR-CF against NSCLC were improved with the extended storage periods due to the variance of differential metabolites. The results would provide scientific guidance for quality control of PCR-C during storage.

Abstract:Citrus peel is a commonly used food-medicine material in the production of fast-moving consumer goods (FMCGs). For instance, Ganpu tea is manufactured by combining the peel of Citri Reticulatae 'Chachi' (PCRC) with Pu-erh tea. The alleviated irritation of PCRC through years of aging makes Citri reticulatae Pericarpium a traditional Chinese medicine. Herein, we introduced short-term steaming into the processing of PCRC to favor the quick removal of its irritation while retaining its food-medicine properties. Sensory evaluation and volatile component analysis showed that 60-s steaming reduced irritation of freshly prepared PCRC. Biological evaluations indicated no effects of steaming on the neuroprotective activity of PCRC. The process increased the contents of several bioactive ingredients, including hesperidin, nobiletin, tangeretin, and synephrine. In addition, physical indications of accelerating PCRC aging were observed. Taken together, our findings suggest that short-term steaming may offer a promising new possibility for enhancing the quality of citrus peel.

Abstract:Pericarpium Citri Reticulatae (PCR) is a natural citrus by-product with beneficial health and nutritive properties that is used widely in food and is an ingredient in traditional Chinese medicine. PCR improves gradually with aging. However, the present research has not yet revealed the reasons for this. Some data prove the important role of microorganisms in the quality of tobacco and fermented tea with the time of the aging of these foods. Our studies further proved that the coexisting Aspergillus niger plays an important role in the change of flavonoids and volatile oil in PCR during this process. Therefore, we put forward that longer storage is better for PCR and is highly correlated with the change of the coexisting microbial population structure caused by environmental factors. Samples of PCR aged in Beijing, Sichuan, Guangdong, and Yunnan were collected at different time points. Using GC/MS and high throughput 16S rDNA and ITS sequencing techniques, massive changes in volatile profile and microbial communities were observed during aging. Spearman correlation analysis indicated that Exobasidium, Xeromyces, Pseudocercospora, Russula, Aspergillus, Herbaspirillum, Sphingomonas , and Streptococcus , which are the dominant microbial genera in Sichuan and Guangdong showed strong connections with volatile components of chemical markers. It was preliminarily verified that the changes of volatile components for PCR are highly correlated with the change of the coexisting microbial population structure caused by environmental factors, providing a new idea for the research on the aging mechanism of PCR and key influencing factors of aging quality.

Abstract:The aging process endows Chenpi (Pericarpium Citri Reticulatae) with unique value and efficacy. This study investigated the dynamic changes in the fine structure, chain conformation, and morphology of water-soluble polysaccharides from Chenpi over time. In the initial storage period of Chenpi (1 year), Chenpi polysaccharides (CP) exhibited a triple-helical structure, with chains entangled and aggregated into rough spherical conformations. Galacturonic acid (GalA, 47.78 mol%) was the predominant monosaccharide. As aging progressed, the homogalacturonan (HG) regions gradually degraded, leading to decreases in molecular sizes (particle size, Mw, Mn, and Rg), a reduction in the degree of esterification, and a weakening of the semicrystalline structure. Consequently, arabinose (Ara) emerged as the main monosaccharide (41.38 mol%). Neutral sugars continuously enriched the rhamnogalacturonan (RG-I) side chains, forming highly branched single molecules that curled into spherical shapes. By 15 years, CP were fully degraded, adopting a compact molecular conformation with the triple-helix structure disappearing and sizes uniformly below 20 nm. However, AFM results indicated aggregation phenomena in 15-year CP. Additionally, CP viscosity decreased while thermal stability improved, reflecting the natural structural transformation of CP. This study provides scientific evidence supporting the application of Chenpi in the food and pharmaceutical industries.

Abstract:Citri Reticulatae Pericarpium, especially the pericarp of Citrus reticulata Blanco cv. Chachiensis (PCRC), is an important edible and medicinal ingredient for health and pharmacological properties. Citrus Huanglongbing, a devastating disease that currently threatens the citrus industry worldwide, is caused by a phloem-limited alpha-proteobacterium, "Candidatus Liberibacter asiaticus" (CLas). The industry of cultivar Chachiensis has been suffering from HLB. Although HLB affected the quality of citrus fruit, whether the quality of PCRC was affected by HLB remains unclear. In this study, we compared the metabolite profiles between HLB-affected and healthy PCRC from three sources: fresh, 6-month-old, and 9-year-old PCRC, through the untargeted LC-MS method. Compared to healthy controls, various types of bioactive compounds, mainly flavonoids, terpenoids, alkaloids, coumarins, polysaccharides, and phenolic acids, accumulated in HLB-affected PCRC, especially in the HLB-affected 9-year PCRC. In particular, isorhamnetin, isoliquiritigenin, luteolin 7-O-beta-D-glucoside, limonin, geniposide, pyrimidodiazepine, scoparone, chitobiose, m-coumaric acid, malonate, and pantothenic acid, which contributed to the pharmacological activity and health care effects of PCRC, were highly accumulated in HLB-affected 9-year-old PCRC compared to the healthy control. Multibioassay analyses revealed that HLB-affected 9-year-old PCRC had a higher content of total flavonoids and total polyphenols and exhibited similar antioxidant capacity as compared to healthy controls. The results of this study provided detailed information on the quality of HLB-affected PCRC.
Physicochemical characterization of puerh tea polysaccharides and their antioxidant and α-glycosidase inhibition

Abstract:Puerh tea is one of the most popular (or healthiest) beverages in Southeast Asia, because of its higher antioxidant, hypolipidemic and antidiabetic effects. One of the important active ingredients, tea polysaccharides (TPS) has long been recognized to possess immune modulatory and antidiabetic effects. In the present work, polysaccharides were isolated from puerh tea (PTPS) with aging (mild fermentation) for 1 year (PTPS-1), 3 years (PTPS-3) and 5 years (PTPS-5), respectively. The results obtained indicated that all of these three kinds of PTPS are acid heteropolysaccharides bound with proteins, with differences in molecular weights and in monosaccharide compositions. The longer aging process not only produced a higher yield of PTPS but also had higher contents of proteins and uronic acids which proportionally correlated with their antioxidant and α-glycosidase inhibitory activities (PTPS-5 > PTPS-3 > PTPS-1). The anti-α-glycosidase activities of PTPS-5 and PTPS-3 were found to be three times more and equally potent than that of the antidiabetic drug acarbose. All of these findings firstly indicate that the aging process not only induced changes in chemical compositions and proteins conjugation in PTPS, but also markedly enhanced antioxidant and anti-α-glycosidase activities. PTPS-5 exhibited potential antidiabetic effect and merits further studies.
