Abstract:Compared to conventional fluorescence imaging in the visible (400-700 nm) and NIR-I regions (700-900 nm), optical fluorescence imaging in the second near infrared window (NIR-II, 1000-1400 nm) offers reduced photon scattering, deeper tissue penetration and lower auto-fluorescence. Despite excellent imaging capabilities, current NIR-II probes have not yet reached their full potential due to weak quantum yield, low water solubility and suboptimal biocompatibility. To address these problems, we report herein a new NIR-II fluorescent PbS quantum dots (QDs) that are fabricated in water using β-lactoglobulin (LG) as a biological template. The LG-PbS QDs exhibit satisfactory dispersibility, relatively high quantum yield and favorable biocompatibility, and therefore are suitable for high-resolution in vivo imaging applications.

Abstract:The rapid advance and growth of the point-of-care diagnosis industry has provided an impetus for the development of novel signal labels for highly sensitive bio-molecule detections. Colloidal quantum dots (QDs) exhibit superior brightness, facile surface functionalization and exceptional photostability, making them the preferred option for these biological applications. However, the significantly reduced fluorescence intensity and limited photochemical stability in complex biological environments have greatly hampered their further use. Herein, with an innovative alloying engineering strategy, the high-quality water-soluble CdSe/CdxZn1−xS QDs with near-unity PL quantum yield and monoexponential PL decay dynamics are obtained. Notably, for the first time, a record-breaking stability at the single QD level in water with nonblinking behavior persisting for an hour is achieved, which approaching those of state-of-the-art hydrophobic QDs. Furthermore, the nanocomposites formulated with these novel alloyed QDs demonstrate a remarkable PL QY of 96 % and provide an ultrasensitive detection for prostate specific antigen on lateral flow immunoassays. These findings presented here shed new light on the design of high-brightness water-soluble QDs in single-molecule level and QDs-based nanocomposites, significantly pushing ahead toward high-sensitivity biomedical detections and diagnosis.
In situ aqueous synthesis of genetically engineered polypeptide-capped Ag2S quantum dots for second near-infrared fluorescence/photoacoustic imaging and photothermal therapy

Abstract:Ag2S quantum dots have received extensive attention as theranostic agents for second near-infrared (NIR-II) fluorescence and photoacoustic dual-mode imaging, and photothermal therapy. However, it is still greatly challenging to synthesize Ag2S quantum dots using aqueous synthesis. In this study, genetically engineered polypeptide-capped Ag2S quantum dots were successfully synthesized. Three cysteines were integrated to the C-terminal and N-terminal of RGDPC10A to enhance the stability and brightness of the synthesized Ag2S quantum dots. The RGDPC10A-capped Ag2S quantum dots exhibited excellent stability, outstanding resistance to photobleaching, and a superior quantum yield of up to 3.78% in the NIR-II biological window. The in vitro and in vivo results showed that the RGDPC10A-capped Ag2S quantum dots possessed typical NIR-II fluorescence, photoacoustic imaging, and photothermal therapeutic effectiveness against tumors. Moreover, the results of toxicity assays suggested that the RGDPC10A-capped Ag2S quantum dots have negligible long-term toxicity. These findings open up the possibility for synthesizing theranostic agents by using this aqueous method.

Abstract:Fluorescent imaging in the second near-infrared region (NIR-II, 1000-1700 nm) is optimal for in vivo imaging due to their low autofluorescence and weak tissue scattering. Recently, many efforts have been paid in search of NIR-II emitting agents. Herein, we report a facile one-pot approach for the synthesis of NIR-II emissive Ag2S quantum dots (QDs). The effect of ligand was carefully investigated to improve the quality of QDs. It is found that the QDs tend to form clusters when 1-dodecanethiol was used as the only ligand. The addition of oleic acid can greatly improve the dispersity of the nanocrystals but lead to a significantly decrease of the emission intensity. Meanwhile, the present of small amount of oleylamine can prevent the formation of undesired silver nanoparticles and improve the fluorescent intensity at high temperature. Whole-body blood vessel imaging and tumor imaging were performed with these biocompatible Ag2S QDs after PEGylation to demonstrate their potentials in vivo.

Abstract:The second near-infrared window (NIR-II, wavelength of 1.0–1.4 μm) is optimal for the bioimaging of live animals due to their low albedo and endogenous autofluorescence. Herein, we report a facile and one-pot biomimetic synthesis approach to prepare water-dispersible NIR-II-emitting ultrasmall Ag2S quantum dots (QDs). Photoluminescence spectra showed that the emission peaks could be tuned from 1294 to 1050 nm as the size of the Ag2S QDs varied from 6.8 to 1.6 nm. The x-ray diffraction patterns and x-ray photoelectron spectra confirmed that the products were monoclinic α-Ag2S. Fourier transform infrared spectrograph analysis indicated that the products were protein-conjugated Ag2S QDs. Examination of cytotoxicity and the hemolysis test showed that the obtained Ag2S QDs had good biocompatibility, indicating that such a nanomaterial could be a new kind of fluorescent label for in vivo imaging.

Abstract:Ribonuclease-A (RNase-A) encapsulated PbS quantum dots (RNase-A@PbS Qdots) which emit in the second near-infrared biological window (NIR-II, ca. 1000–1400 nm) are rapidly synthesized under microwave heating. Photoluminescence (PL) spectra of the Qdots can be tuned across the entire NIR-II range by simply controlling synthesis temperature. The size and morphology of the Qdots are examined by transmission electron microscopy (TEM), atomic force microscopy (AFM), and dynamic light scattering (DLS). Quantum yield (Φf) measurement confirms that the prepared Qdots are one of the brightest water-soluble NIR-II emitters for in vivo imaging. Their high Φf (∼17.3%) and peak emission at ∼1300 nm ensure deep optical penetration to muscle tissues (up to 1.5 cm) and excellent imaging contrast at an extremely low threshold dose of ∼5.2 pmol (∼1 μg) per mouse. Importantly, this protein coated Qdot displays no signs of toxicity toward model neuron, normal, and cancer cells in vitro. In addition, the animal’s metabolism results in thorough elimination of intravenously injected Qdots from the body within several days via the reticuloendothelial system (RES), which minimizes potential long-term toxicity in vivo from possible release of lead content. With a combination of attractive properties of high brightness, robust photostability, and excellent biocompatibility, this new NIR-II emitting Qdot is highly promising in accurate disease screening and diagnostic applications.

Abstract:Lanthanide hybridized carbon quantum dots (Ln-CQDs) were synthesized by a facile one-pot hydrothermal method using citric acid as a carbon precursor and Yb3+ or Nd3+ as a doping ion. The morphology and chemical structures of Ln-CQDs were investigated by TEM, XRD, XPS, and FTIR spectroscopy. The obtained Ln-CQDs are spherical and well dispersed in water, and their aqueous solutions emit strong blue emission under UV excitation. The Ln-CQDs exhibit excitation-dependent PL behavior with the emission maximum ranging from 443 to 552 nm under 300-520 nm excitation. Moreover, the carbon quantum dots can not only act as visible imaging agents, but also as antennae for photoluminescence (PL) of lanthanide ions. Hence, CQDs hybridized with Yb3+ or Nd3+ ions exhibit the characteristic emission in the near-infrared region with the emission maximum centered at about 998 and 1068 nm, respectively. The MTT assay against HeLa cells verified the low cytotoxicity of Ln-CQDs. They have been used as excellent optical probes for multicolor cell-imaging, demonstrating their great potential for both visible/NIR bioimaging and biomedical applications in vivo.

Abstract:With significantly decreased light scattering and tissue autofluorescence, fluorescence imaging in the second near infrared (NIR-II, 1000-1700 nm) region has been heavily explored in biomedical field recently. Silver sulfide quantum dots (Ag2S QDs) with unique optical properties were one of the most classic NIR-II imaging probes. However, the Ag2S QDs for in vivo purpose were mainly obtain by oil phase-based high-temperature route at present. Here, we proposed a mild aqueous route to prepare NIR-II emissive Ag2S QDs for in vivo tumor imaging. Original Ag2S QDs was obtained by mixing sodium sulfide and silver nitrate in a thiol-terminated polyethylene glycol (mPEG-SH) solution. Treating the original Ag2S QDs with extra mPEG-SH ligands produced highly PEGyalted Ag2S QDs. These re-PEGylated Ag2S QDs exhibited much better blood circulation and tumor accumulation in vivo comparing with the original ones, which can serve as excellent tumor imaging probes. The whole-body blood vessel imaging of living mice was achieved with high resolution, the bio-distribution of these QDs were studied by NIR-II imaging as well. This work also highlighted the importance of ligand density for tumor targeting.

Abstract:Near-infrared-II (NIR-II, 1000-1700 nm) bioimaging features high penetration depth and high spatio-temporal resolution compared to traditional fluorescence imaging, but the key is to develop stable and biocompatible NIR-II fluorophores suitable for in vivo applications. Silver sulfide quantum dots (Ag2S QDs) have been demonstrated to be excellent for in vivo NIR-II imaging with unique optical properties and decent biocompatibility, but they often require complex post modifications for in vivo applications. Herein we demonstrate a facile one-pot strategy to synthesize PEGylated dendrimer-encapsulated Ag2S QDs useful for in vivo NIR-II imaging. Silver ions were first loaded into the core of an acylthiourea-functionalized dendrimer (PEG-PATU) through coordination between silver ions and acylthiourea groups, followed by the addition of sodium sulfide to form Ag2S QDs in situ. The resulting PEG-PATU Ag2S QDs exhibit excellent NIR-II fluorescence signals, and thus could be used for high efficiency labelling and tracking of A549 cancer cell mobility in vivo and real time visualization of the vast circulatory network of a mouse.
Ultrasmall near-infrared Ag2Se quantum dots with tunable fluorescence for in vivo imaging

Abstract:A strategy is presented that involes coupling Na(2)SeO(3) reduction with the binding of silver ions and alanine in a quasi-biosystem to obtain ultrasmall, near-infrared Ag(2)Se quantum dots (QDs) with tunable fluorescence at 90 °C in aqueous solution. This strategy avoids high temperatures, high pressures, and organic solvents so that water-dispersible sub-3 nm Ag(2)Se QDs can be directly obtained. The photoluminescence of the Ag(2)Se QDs was size-dependent over a wavelength range from 700 to 820 nm, corresponding to sizes from 1.5 ± 0.4 to 2.4 ± 0.5 nm, with good monodispersity. The Ag(2)Se QDs are less cytotoxic than other nanomaterials used for similar applications. Furthermore, the NIR fluorescence of the Ag(2)Se QDs could penetrate through the abdominal cavity of a living nude mouse and could be detected on its back side, demonstrating the potential applications of these less toxic NIR Ag(2)Se QDs in bioimaging.
In situ growth of Ag2S quantum dots on cellulose nanocrystals and their near-infrared bioimaging performance

Abstract:Elongated nanoparticles show distinct advantages over spherical nanoparticles in bioimaging because of surface area-to-volume, rate of clearance from the body and elimination mechanism. In this work, we investigated the fluorescence emission properties of the hybrid system by decorating silver sulfide quantum dots (Ag2S QDs) in situ on the surface of cellulose nanocrystal (CNC) with unique rod shape, modifiability and biocompatibility. This water-dispersible fluorescent probe has both absorption and fluorescence in near-infrared (NIR) region. By varying the amount of surface ligands, uniformly dispersed Ag2S QDs with different crystalline states but similar sizes were prepared due to the anchoring effect of CNC. The fluorescence quantum yield of fluorescent probes can be improved up to 109-fold (from 0.04 % to 4.36 %). In addition, the CNC-restricted interparticle spacing of Ag2S QDs (< 10 nm), in combination with the overlap of wide fluorescence emission and ultraviolet absorption, significantly enhanced the 1070 nm emission in the NIR-II region via fluorescence resonance energy transfer (FRET). Further conjugation of these CNC probes with folic acid-polyethylene glycol-amino (FA-PEG-NH2) enables in vitro bioimaging of Hela cells, which are potentially applicable for in vivo cancer detection system. The synthetic strategy provides a new way for one-pot preparation of fluorescent probes with both high NIR-I absorption and NIR-II fluorescence.
Fluorescent Nitrogen and Sulfur Co-Doped Carbon Dots from Casein and Their Applications for Sensitive Detection of Hg2+ and Biothiols and Cellular Imaging.

Abstract:Fluorescent nitrogen and sulfur co-doped carbon dots (NSCDs) were synthesized by a one-step pyrolysis strategy using casein as carbon, nitrogen and sulfur sources, and characterized by UVevis spectrum, fluorescent spectrum, X-ray photoelectron spectroscopy (XPS) and FT-IR, etc. The synthesized NSCDs displayed a blue emission under ultraviolet illumination with a quantum yield of 31.8%, and a good aqueous solubility, photostability and biocompatibility. It was found that the fluorescence intensity of NSCDs could be selectively quenched by Hg2+, so NSCDs was used as an effective probe for the detection of Hg2+. The linear range and the limit of detection (LOD) of the fluorescent sensor based on NSCDs for the detection of Hg2+ were 0.01-0.25 mu M and 6.5 mu M, respectively. Spiked water samples were detected by the sensor with the recovery of 95.4-106.3% and relative standard deviation (RSD) of 3.6-8.6%. It was also observed that the quenched NSCDs-Hg2+ system could be restored by the addition of biothiols such as L-cysteine (Lcy), homocysteine (Hcy) and glutathione (GSH), thus NSCDs-Hg2+ system was employed as a fluorescent sensor for the detection of biothiols. The linear range and LOD of the NSCDs-Hg2+ system were 1-10 mM and 23.6 nM for Lcy, 0.2-2.5 mM and 12.3 nM for Hcy, and 0.1-2.0 mM and 16.8 nM for GSH, respectively. The NSCDs-Hg2+ system was applied for the detection of biothiols in serum samples with satisfied results. In addition, the study in vitro imaging HeLa cells revealed that the synthesized NSCDs could be used as effective fluorescent probes in cellular imaging without noticeable cytotoxicity. In Vivo Tumor-Targeted Fluorescence Imaging Using Near-Infrared Non-Cadmium Quantum Dots.

Abstract:The high tumor uptake of ultrasmall near-infrared quantum dots (QDs) attributed to the enhanced permeability and retention effect is reported. InAs/InP/ZnSe QDs coated by mercaptopropionic acid (MPA) exhibit an emission wavelength of about 800 nm (QD800-MPA) with very small hydrodynamic diameter (<10 nm). Using 22B and LS174T tumor xenograft models, in vivo and ex vivo imaging studies show that QD800-MPA is highly accumulated in the tumor area, which is very promising for tumor detection in living mice. The ex vivo elemental analysis (Indium) using inductively coupled plasma (ICP) spectrometry confirm the tumor uptake of QDs. The ICP data are consistent with the in vivo and ex vivo fluorescence imaging. Human serum albumin (HSA)-coated QD800-MPA nanoparticles (QD800-MPA-HSA) show reduced localization in mononuclear phagocytic system-related organs over QD800-MPA plausibly due to the low uptake of QD800-MPA-HSA in macrophage cells. QD800-MPA-HSA may have great potential for in vivo fluorescence imaging.

Abstract:Quantum dots (QDs) with near-infrared fluorescence (NIR) are an emerging class of QDs with unique capabilities owing to the deeper tissue penetrability of NIR light compared with visible light. NIR light also effectively overcomes organism autofluorescence, making NIR QDs particularly attractive in biological imaging applications for disease diagnosis. Considering latest developments, Ag2S QDs are a rising star among NIR QDs due to their excellent NIR fluorescence properties and biocompatibility. This review presents the various methods to synthesize NIR Ag2S QDs, and systematically discusses their applications in biosensing, bioimaging, and theranostics. Major challenges and future perspectives concerning the synthesis and bioapplications of NIR Ag2S QDs are discussed.

Abstract:In this paper, PbS semiconductor quantum dots (QDs) with near-infrared (NIR) photoluminescence were synthesized in oleic acid and paraffin liquid mixture by using an easily handled and 'green' approach. Surface functionalization of the QDs was accomplished with a silica and polyethylene glycol (PEG) phospholipid dual-layer coating and the excellent chemical stability of the nanoparticles is demonstrated. We then successfully applied the ultrastable PbS QDs to in vivo sentinel lymph node (SLN) mapping of mice. Histological analyses were also carried out to ensure that the intravenously injected nanoparticles did not produce any toxicity to the organism of mice. These experimental results suggested that our ultrastable NIR PbS QDs can serve as biocompatible and efficient probes for in vivo optical bioimaging and has great potentials for disease diagnosis and clinical therapies in the future.

Abstract:Fluorescence in the near infrared IIb window (NIR-IIb, 1500–1700 nm) shows higher signal-to-noise ratio, better space-time resolution, and deeper tissue penetration. However, materials with excellent fluorescence properties in the NIR-IIb region were rarely reported. Ag2Te quantum dots (QDs) could emit variable fluorescence in the NIR-II window due to their narrow band gap. Unfortunately, the fluorescence in the NIR-IIb region was not strong enough to image in vivo because the lattice defects on the surface of Ag2Te increased non-radiative transitions and decreased fluorescence performance. In this work, we coated a Ag2Te core with a Ag2Se shell to passivate the lattice defects and enhance its fluorescence efficiency. The fluorescence property of the as-prepared Ag2Te/Ag2Se core/shell QDs is more than 10 times stronger than that of Ag2Te. The brighter Ag2Te/Ag2Se QDs showed high-resolution in vivo imaging in the NIR-IIb region to distinguish thin blood vessels up to 66 μm. Overall, Ag2Te/Ag2Se QDs with excellent fluorescence in the NIR-IIb region were prepared and showed great potential for biomedical imaging.

Molecular Engineering of an Organic NIR-II Fluorophore with Aggregation-Induced Emission Characteristics for In Vivo Imaging.

Abstract:Design and synthesis of new fluorophores with emission in the second near-infrared window (NIR-II, 1000-1700 nm) have fueled the advancement of in vivo fluorescence imaging. Organic NIR-II probes particularly attract tremendous attention due to excellent stability and biocompatibility, which facilitate clinical translation. However, reported organic NIR-II fluorescent agents often suffer from low quantum yield and complicated design. In this study, the acceptor unit of a known NIR-I aggregation-induced emission (AIE) luminogen (AIEgen) is molecularly engineered by varying a single atom from sulfur to selenium, leading to redshifted absorption and emission spectra. After formulation of the newly prepared AIEgen, the resultant AIE nanoparticles (referred as L897 NPs) have an emission tail extending to 1200 nm with a high quantum yield of 5.8%. Based on the L897 NPs, noninvasive vessel imaging and lymphatic imaging are achieved with high signal-to-background ratio and deep penetration. Furthermore, the L897 NPs can be used as good contrast agents for tumor imaging and image-guided surgery due to the high tumor/normal tissue ratio, which peaks at 9.0 ± 0.6. This work suggests a simple strategy for designing and manufacturing NIR-II AIEgens and demonstrates the potential of NIR-II AIEgens in vessel, lymphatic, and tumor imaging.

Au:Cdhgte Quantum Dots for in Vivo Tumor-Targeted Multispectral Fluorescence Imaging

Abstract:Near-infrared gold-doped CdHgTe quantum dots (QDs) with improved photoluminescence and biocompatibility were developed using an aqueous solution route with l-glutathione and l-cysteine as stabilizers. As-prepared Au:CdHgTe QDs were covalently linked to arginine-glycine-aspartic acid (RGD) peptide, anti-epidermal growth factor receptor (EGFR) monoclonal antibody (MAb), and anti- carcinoembryonic antigen-related cell adhesion molecule-1 (CEACAM1) MAb separately. Three Au:CdHgTe QD bioconjugates (QD800-RGD, QD820-anti-CEACAM1, and QD840-anti-EGFR) were successfully used as probes for in vivo tumor-targeted multispectral fluorescence imaging of xenografts. Fluorescence signals from the QD bioconjugates used to detect three tumor markers were spectrally unmixed, and their co-localization was analyzed. The results indicate that multiple tumor markers could be simultaneously detected by multispectral fluorescence imaging in vivo using QD bioconjugates as probes. This approach has excellent potential as an imaging method for the noninvasive exploration and detection of multiple tumor markers in vivo, thereby substantially aiding the diagnosis of cancer.
