TLR4-NF-κB Activation Inducing Colon Cancer Biological Diversity Related with Different APC Genotypes.

Abstract:e14698 Background: Previous investigations have shown TLRs/NF-κB activation related with contrary biological effects in colon cancer. APC gene mutation is frequent events in carcinogenesis of colon cancer. Here we investigate the function and cross-talk mechanism of TLRs/ NF-κB pathway activation and APC/GSK-3β/β-catenin though NF-κB and β-catenin, which related with cell growth and apoptosis. Methods: APC was knocked down in human colon cancer cell line HCT116 and human normal colon cell line NCM460 with lenti-viral packaging system. Different APC status cell lines were stimulated with LPS (lipopolysaccharide, TLR4 agonist). The molecular events from two pathway were investigated. Results: Thecell proliferation was promotedboth in HCT116-SiAPC and HCT116-ShAPC treated with LPS, while that was inhibited in HCT116 cells. The activation of TLRs/NF-κB signaling pathway by LPS was regulated with APC/GSK-3β/β-catenin pathway differently in cells with different APC status and ultimately influenced cells apoptosis. In vivo experiments showed that LPS promoted HCT116-ShAPC tumor growth while inhabited tumor growth of HCT116, the difference of which was significant (P < 0.05). The related molecular events were confirmed in tumor tissue. Conclusions: In summary, changes of APC phenotype produced different biological behavior by regulating the levels of β-catenin, which interacted with NF-κB in activated TLR4-NF-κB pathway. Our study also suggests that it is important of APC genotype for targeting TLR4-NF-κB pathway in prevention or therapy of colon cancer.
