LB822 Β-Catenin Activation in Hair Follicle Dermal Sheath Induces Ectopic Hair Outgrowth and Skin Fibrosis

Abstract:Hair follicle dermal sheath (DS) cells have long been speculated as stem cells with high plasticity. A recent study shows that a subset of dermal sheath cells around hair follicle can be mobilized to regenerate dermal sheath, maintain/supplement the cell number of dermal papilla and modulate hair type. Wnt/β-catenin signaling is required for hair follicle development and regeneration. However, little is known about how Wnt/β-catenin signaling regulates dermal sheath cells and progeny behaviors. In this study, we report that activation of β-catenin in dermal sheath cells, to some extent, endows them with hair inducing ability by through reprogramming terminal differentiated outer root sheath cells to generate new outgrowth. The generation of the new outgrowth resembles embryonic hair development and the new formed dermal condensates/dermal papilla cells originated from dermal sheath. We found that hair follicle bulge stem cells are activated and lose their stemness gradully. Interestingly, simultaniously progressive skin fibrosis is characterized by loss of subcutaneous adipose tissue and excessive accumulation of extracellular matrix released by pathologically activated fibroblasts in hypodermis. To investigate the underlying molecular mechanisms, gene expression profiling of purified dermal sheath cells was analyzed. After activation of β-catenin in dermal sheath cells, 44% dermal condensates signature genes are regulated, most of which are up-regulated. We found elevated expression of several growth factors, including Noggin, Fgf7 and Fgf10, which were previously implicated in hair follicle induction. In summary, we confirm the high plasticity of dermal sheath cells and report a potential mechanism by which Wnt/β-catenin signaling controls dermal sheath cells behaviors.
