P3‐185: Prediction of MCI to Alzheimer's Conversion Based on Tensor‐based Morphometry and Kernel Support Vector Machine

Abstract:Magnetic resonance imaging (MRI) is a powerful medical imaging technique used in radiography to investigate the anatomy of the body. Alzheimer's disease (AD) and mild cognitive impairment (MCI) patients show significant changes in the whole brain structural integrity. We assessed the brain morphological measures from tensor-based morphometry (TBM) to predict conversion from MCI to AD. 3D MRI scans of 70 MCI-stable subjects and 40 MCI-converter subjects were procured from ADNI database. TBM approach was employed to obtain voxel-wise features, i.e., relative volume differences between individual's brain and a study-specific brain template, as a 3D matrix of determinant of Jacobian map. Principal component analysis is employed to reduce the features. Kernel Support Vector Machine (kSVM) was built to distinguish MCI-stable from MCI-converter subjects. The kernel is set to radial basis function. 50x10-fold cross validation was performed to evaluate the out-of-sample performance of the built classifier. With the kSVM, we obtained classification accuracies of 74.5% (sensitivity/specificity: 72.5%/75.7%) to detect MCI-converter from MCI-stable. The method shows promise in separating MCI-converter from MCI-stable subjects. We will test other kernels in future study.
