Abstract:Objective To investigate the effect of different dosage and placement of calcium hydroxide on dentine pH in root canal and pulp chamber. Methods Twenty-five freshly extracted single-rooted human teeth were prepared and root canals were instrumented by using a conventional technique. Three cavities were drilled by 0.5mm from the root surface at the cervical, middle, and apical thirds. All samples were randomly divided into five groups. Group 1: pure aqueous calcium hydroxide paste (calcium hydroxide/distilled water solution) was placed into the rood canal; Group 2:the same aqueous calcium hydroxide paste was placed into the pulp chamber; Group 3: Hycal, a new form of calcium hydroxide paste was placed into the root canal; Group 4: the same form of calcium hydroxide paste was place into the pulp chamber; Group 5: calcium hydroxide containing gutta-percha points was placed in the root canal; access cavities and apical ends were sealed. The pH was measured in the dentinal cavities at 1, 2, and 3 hours, and 1, 3, 7, 14, and 21 days by using a calibrated microelectrode. Results Calcium hydroxide paste placed either in the pulp chamber or in the root canal could increase the root dentinal pH. There was no significant difference between the two groups except at the 7d. They all maintained an alkaline environment through three weeks. At 7d, the group 1 had the highest pH value ( 9.13 ), significantly difference from the other groups ( P 0.05 ), The effect of placing calcium hydroxide gutta-percha points in root canal on the increase of root dentinal pH was the most significant. At 3h, there was significant difference among the group 5 and the other groups ( P 0.001 ),but alkalinity in the dentine was maintained for 7 days only. The cervical pH was similar for all product or placement location at middle pH and apical pH. There was no significant difference between the three WHAT? when all groups were combined. Conclusion ① The aqueous calcium hydroxide paste and Hycal paste placed in the pulp chamber or root canal had the same effect on pH increase in root surface. They all maintained an alkaline environment for three weeks. ② The effect of root dentinal pH increase by placing calcium hydroxide Gutta-percha points in the root canal was the fastest but alkalinity in the dentine was maintained for 7days only.
