Abstract:Aqueous polymer two-phase system provides a powerful method for separation and purification of biomaterials. Among various factors, polymer concentration and polymer molecular weight are essential and have strong impact on the protein partitioning in these systems. Based on the modified Pitzer's model, a simple expression has been obtained for correlating protein partitioning in aqueous polymer two-phase systems with varying polymer concentration and different polymer molecular weights. Using only one group of parameters for each target protein, the partition coefficients of five proteins (lysozyme, chymotrypsinogen-A, bovine serum albumin, transferrin and catalase) in 16 sets of polyethylene glycol (PEG)/dextran systems were correlated. In addition, partition behavior of lactate dehydrogenase in PEG/hydroxypropyl starch systems were measured, correlated and partially predicted. A comparison of calculated and experimental data indicated that the model provides good correlation and prediction abilities on the protein partitioning in aqueous polymer two-phase systems with a wide range of polymer concentration and molecular weight.
Affinity Partition of Soybean Hull Peroxidase in Aqueous Two-phase Systems

Abstract:Metal chelating affinity partition with aqueous two-phase system was studied to separate the peroxidase fromsoybean hull extract. The effects of affinity ligand (polyethylene glycol (PEG)- iminodiacetic acid (IDA)-Cu(II)) on the affinitypartition of soybean hull peroxidase (SHP) were investigated in different types of aqueous two-phase systems. The resultsindicated that PEG/ hydroxypropyl starch (PES) aqueous two-phase system was suitable for affinity extraction of SHP. Severalfactors, such as adding amount of affinity ligand, molecular weight of phase-forming polymer, system compositions, pH, etc.,were optimized. The best separation conditions were: PEG2000 9%, PES200 14% and PED-IDA-Cu(II) 1%. The partitioncoefficient of SHP could reach 40, and purification factor was 2.8 with the recovery of 93%.
Separation of glutathione by aqueous two-phase extraction combined with temperature-induced phase separation

Abstract:The techniques for separation of glutathione (GSH) by aqueous two-phase extraction combined with temperature-induced phase separation were studied. The partition behavior of GSH in ethylene oxide-propylene oxide random copolymer (EOPO)/hydroxypropyl starch (PES)/water systems was investigated. The effects of such factors, as concentration of polymer, pH, on the partition coefficients in two separation processes (aqueous two-phase extraction and temperature-induced phase separation) were studied. The optimum conditions are EOPO4000 13%, PES100 10%, pH=10.5. The separation technique for GSH from yeast cell was also investigated. A feasible separation scheme was suggested, and the results show that the total extraction yield can reach over 80%.

Abstract:A novel aqueous two-phase system containing hydrophobically modified ethylene oxide (HM-EO) and sodium dodecyl sulphate (SDS) was developed to enhance the selectivity of protein partitioning in two phases. Phase diagrams of HM-EO/H2O and HM-EO/SDS/H2O were measured, and the mechanism of interaction between HM-EO polymer and the anionic surfactant sodium dodecyl sulphate (SDS) was also discussed. It was found that the improvement of selectivity of protein partitioning was related to the increase of electrostatic potential difference between the two phases because of the charged network formed by mixed micelles of HM-EO and SDS in the bottom phase. With bovine serum albumin (BSA) and lysozyme as model proteins, some factors, such as pH, SDS concentration, conductivity and temperature of the system, were investigated for the influences of protein partition in HM-EO/SDS/H2O systems. The results showed that the addition of SDS not only changed the phase behaviour, but also played an important role in protein partitioning.

Abstract:Extraction of alpha-Amylase Inhibitor by Aqueous Two-phase Extraction was studied. Effect of the Concentration of PEG, (NH 4) 2SO 4 and NaCl on extraction was investigated. The result indicated that the partition coefficient of 4.40 and the recovery of total activity of 71.41% could be obtained under the conditions of PEG2000(12.0%), (NH 4) 2SO 4(13.3%)and NaCl(0.003%).

Abstract:A cationic hydrophobically modified ethylene oxide polymer (HM-EO) was used to form aqueous two-phase system with only one phase-forming composition. It was found that the phase separation could be achieved under room temperature. The phase behavior and charge properties of HM-EO aqueous two-phase systems were studied systematically. The amphiphilic property of HM-EO molecule resulted in the micellization and aggregation of micelles in water. The micellar aggregates formed in the bottom phase could be charged, and the net-charge could be adjusted by the pH and conductivity of bulk solution and adding sodium dodecyl sulfate (SDS) The increase of pH would improve the phase separation of HM-EO aqueous two-phase system. The effect of salts on the phase behavior was observed. The influence of cations varied as K+ > Na+ and that of anions as SO42- > F- > Cl- > Br- > I-. In addition,the interaction between HM-EO polymer and anion surfactant SDS was studied.The results indicated that the addition of SDS caused the formation of mixed micellar aggregates of SDS and HM-EO which changed significantly the charge property of HM-EO aqueous two-phase systems.

Abstract:The partitioning behavior of four amino acids, cysteine, phenylalanine, methionine, and lysine in 15 aqueous two-phase systems (ATPSs) with different polyethylene glycol (PEG) molecular weights and phosphate buffers has been studied in the present paper. The phase diagrams of the systems are investigated together with the effect of the PEG molecular weight and pH of the phosphate solutions. The composition of these systems and some parameters such as density and refractive index are determined. The influences of salts in ATPSs, side chain structure of the amino acids, pH of ATPSs, and the PEG molecular weight on the distribution ratios of the amino acids have been studied. This work is useful for the purification of amino acids and the separation of some proteins whose main surface exposed amino acid residues are these four amino acids, respectively.

Abstract:Aqueous stabilizer-free two-phase copolymerization of acrylamide (AM), 2-methylacryloylxyethyl trimethylammonium chloride (DMC) and acryloyloxyethyl dimethylbenzyl ammonium chloride (AODBAC) with initiator 2,2′-azobis[2-(2-imidazolin-2-yl) propane]-dihydrochloride was studied. UV spectrophotometer was used to monitor in-line transmittance change of reaction mixture and to detect critical point of phase separation. The conversion rate (Xc) and molecular weight (Mc) at the critical point were measured by improved brominating and viscometric methods, respectively, and were investigated on effect of various factors. The cationic surfmer, AODBAC, was found to have a good performance on promoting phase separation of the system. Change of initiator amount and molar fraction of AODBAC had minimal effect on Xc, which however increased linearly at increase of temperature. The increase of initiator amount, temperature, and molar fraction of AODBAC caused linear decrease of Mc. Xc decreased but Mc increased significantly when molar fraction of AM and overall monomer amount were increased.

Abstract:The partition behaviors of β-1,3-1,4-glucanase, α-amylase and neutral proteases from clarified and whole fermentation broths ofBacillus subtilis ZJF-1A5 were investigated. An aqueous two-phase system (polyethylene glycol (PEG)/MgSO4) was examined with regard to the effects of PEG molecular weight (MW) and concentration, MgSO4 concentration, pH and NaCl concentration on enzyme partition and extraction. The MW and concentration of PEG were found to have significant effects on enzyme partition and extraction with low MW PEG showing the greatest benefit in the partition and extraction of β-glucanase with the PEG/MgSO4 system. MgSO4 concentration influenced the partition and extraction of β-glucanase significantly. pH had little effect on β-glucanase or proteases partition but affected α-amylase partition when pH was over 7.0. The addition of NaCl had little effect on the partition behavior of β-glucanase but had very significant effects on the partitioning of α-amylase and on the neutral proteases. The partition behaviors of β-glucanase, α-amylase and proteases in whole broth were also investigated and results were similar to those obtained with clarified fermentation broth. A two-step process for purifying β-glucanase was developed, which achieved β-glucanase recovery of 65.3% and specific activity of 14027 U/mg, 6.6 times improvement over the whole broth.

Abstract:Based on two-film theory the kinetic model about α-amylase extraction by CTAB-n-butanol-isooctane solution as the reversed micellar extraction system was established.Varied conditions including initial enzyme activity,stirring speed,pH and ionic strength in aqueous phase,concentration of surfactant and co-solvent were performed on a stirred transfer cell with constant interface to determine the overall transfer coefficients(kL).The relationship of control type and interfacial area was also studied.The results showed that the slope about the common logarithm of residual enzyme activity to extraction time was constant,which suggested that this kinetic model could agree with extraction process and the relationship of kL with initial enzyme activity were uncorrelated when its activity was at 1105U·L-1 to 4420U·L-1.The process of transfer mechanism was affected by the stirring speed,especially by the conditions such as pH and ionic strength in aqueous phase,and concentration of surfactant and co-solvent.The relative slope of extraction rate to interfacial area was minished contrarily with the increase of pH.Moreover,the mechanism of extraction types was mainly controlled by resistance of interface,cooperation and diffusion when pH was 9.9,10.8 and 11.9 respectively which was performed at the appointed conditions.It also indicated that the relative slope of extraction rate to interfacial area could be used to evaluate the degree of influence of interface resistance.

Abstract:PEG/KH2PO4,PEG/(NH4)2SO4 and PEG/MgSO4 aqueous two-phase systems were studied.Result indicated that PEG/(NH4)2SO4 aqueous two-phase system was best.Then the glucoamylase was extracted and purified by PEG/(NH4)2SO4 aqueous two-phase system.The effects of the molecular of PEG,the concentration of PEG and PEG/(NH4)2SO4 solution on distribution coefficient and recycling rate were studied.The optimum conditions for glucoamylase extraction were determined.When the molecular of PEG was 20000,the concentration of PEG20000 solution was 28% and that of(NH4)2SO4 solution was 20%,the smallest distribution coefficient and highest recycling rate were obtained as 0.15 and 96.1%,respectively.

Abstract:The interfacial behavior of soy hull polysaccharide (SHP) at the oil-water interface and the stabilization mechanism of high internal phase emulsion (HIPE) with three enzymes (α-amylase, trypsin and papain) were investigated. The diameter of the α-amylase-treated emulsion was the minimum at 40 min, indicating that the carbohydrate portions of SHP form a thick layer on the surface of the droplet to prevent aggregation. Moreover, Raman spectroscopy revealed significantly higher levels of disordered content of SHP emulsion treated with α-amylase at 60 min, potentially affecting the directional movement of SHP molecules in the emulsion. Conversely, the content of β-sheet and β-turn was lower than trypsin and papain, possibly due to ion-dipole interaction between the polar group residues within SHP and ions, or protonation with H+.

Abstract:In the present study, an aqueous two-phase system (ATPS) composed of polyethylene glycol (PEG) and ammonium sulphate ((NH4)2SO4) was used to purify a thermostable xylanase produced by the thermophilic fungus Paecilomyces thermophila J18 in solid-state fermentation (SSF). The effects of PEG, (NH4)2SO4, loading mass and pH on xylanase partition in ATPS were investigated. Xylanase partitioned in the top polymer-rich phase, while the remainder of proteins partitioned in the bottom salt-rich phase. The ATPS composition (w/w) of 12.5% PEG-4000, 25% (NH4)2SO4 and 50% enzyme solution at pH 7.2 was proved to be an excellent system for purification of xylanase in a single-step operation. SDS-PAGE analysis revealed that the purified xylanase was near homogeneity. A purification factor of 5.54 and a 98.7% enzyme yield were achieved in the top phase of this system. However, the xylanase produced in SSF was purified to homogeneity with a recovery yield of 34.5% by three steps. Therefore, ATPS provides an efficient and attractive method for purifying the xylanase in SSF.

Abstract:A copolymer of 50% ethylene oxide and 50% propylene oxide (EOPO(1:1)) has been used in aqueous two-phase partitioning of amino acids. This EOPO(1:1) copolymer has a cloud point of 70 degrees C, at which it separates from water. The cloud point can be lowered to 45 degrees C by the addition of 0.2 M sodium sulfate. Amino acids are first partitioned at 25 degrees C in a system composed of EOPO(1:1) and hydroxypropyl starch (PES). EOPO(1:1) is enriched in the top phase and PES is in the bottom phase. Then, the top phase can be removed and its temperature increases above the cloud point of the EOPO(1:1). This results in formation of a new two-phase system with a top aqueous phase containing most of the amino acids and a bottom EOPO(1:1)-rich phase, thus allowing the lower EOPO(1:1)-rich phase to be removed and recycled for a second extraction. Amino acids are recovered exclusively in top aqueous phase.

Abstract:BSA partitioning was performed in the PEG/phosphate aqueous two-phase system. The effects of the phase compositions concentrations, addition of NaCl and the initial concentration of BSA on the partition coefficient K and the fraction of BSA in the bottom phase Y were investigated. The results showed that BSA was enriched to the bottom phase. The phase compositions of PEG 6000 mass fraction is 10%/PO3-4 mass fraction is 6% and PEG 10000 mass fraction is 6%/PO3-4 mass fraction is 6% respectively were desirable for partitioning. The addition of NaCl showed no influence on BSA partitioning in PEG 6000 mass fraction is 10% /PO3-4 mass fraction is 6% system. The mathematical model for K was derived from the initial mass concentration of BSA and well fitting with the experimental data.

Abstract:Effective extraction of flavones from persimmon leaves is an important technique in bioresearch.In the present study,extraction and purification of flavones of persimmon leaves were carried out simultaneously by using an aqueous partitioning(method.) This two-phase aqueous system was prepared by dissolving suitable amounts of polyethylene glycol(PEG)and(NH_4)_2SO_4 in(aqueous) solution.Flavones of persimmon leaves were recovered with optimum high yield and high concentration in the upper(polymer-rich) phase when(25%) PEG 600and(25%)(NH_4)_2SO_4 were applied.The optimum extraction conditions were pH value 11,MgCl_2 3% and temperature 25℃.

Abstract:Aqueous two-phase systems (ATPS) formed by polymer and salt have been utilized to enrich the desired biomolecule into one of the phase with higher yield and purity. The eco-friendly, biodegradable poly ethylene glycol (PEG) and different citrate salts were chosen as ATPS phase components to investigate the partitioning behavior of α-lactalbumin (α-La). System factors and process parameters such as type and concentration of salt, molecular weight and concentration of PEG, pH, temperature and the effect of additives were studied and the results are discussed in detail. PEG 1000–tri-potassium citrate system yields high partition coefficient of 20 with a better yield of 98 % in the top phase. The addition of NaCl as an additive and acidic pH lowers the yield of α-La in the top phase. Influence of phase volume ratio (Vr) on partitioning was studied and found that the partition coefficient remains almost constant along the tie line. High yield was achieved at a Vr of 3.5 at the tie line length of 50.63 (%, w/w).

Abstract:Aqueous two-phase systems (ATPS) have the advantages of environmentally friendly, high mass transfer efficiency, and mild extraction conditions. However, it is difficult to recycle these polymers, which limits the large-scale application of ATPS. In this study, a novel recyclable ATPS was constructed with thermo-responsive polymer PN and pH-responsive polymer PADB4.78 for the partition of tea saponin. PN represents poly-(N-isopropylacrylamide), and PADB4.78 represents poly-(acrylic acid-dimethylamine ethyl methacrylate-butyl methacrylate), where 4.78 in the subscript indicate the isoelectric point of the polymer. The recoveries of PN and PADB4.78 were 95.36% and 93.48%, respectively, after two cycles. Meanwhile, the phase formation mechanism of ATPS was studied by surface tension and low-field nuclear magnetic resonance (LF-NMR). The effects of polymer concentration, pH, temperature, types and concentrations of salt were investigated on tea saponin partition. In the 1.5% (w/v) PN/3.5% (w/v) PADB4.78 ATPS, the optimal partition coefficient (K) of crude tea saponin were 0.15 in the presence of 1.5 mM KCl at pH 7.6 and 25 °C while the extraction recovery (ERb) reached 92.13%. The K and ERb of tea saponin from tea seeds were 0.12 and 94.50% with 7.5 mM LiBr at pH 8.0 and 25 °C, respectively.
