Significance of p38 mitogen-activated protein kinase and nuclear factor-kappa B in inflammatory epithelium of human nasal mucosa

Abstract:OBJECTIVE The present study was designed to investigate the role of p38 mitogenactivated protein kinase(MAPK) and nuclear factorkappa B(NF-κB) in human nasal epithelial cells stimulated with lipopolysaccharide(LPS). METHODS Western blot was performed to examine the levels of activated p38(phosphorylation p38) in human nasal epithelial cells treated with increasing concentration of LPS and the inhibitory effects of SB203580 on phosphorylation p38. The gel electrophoresis mobility shift assay was used to detect the influence of SB203580 on the DNA binding activity of NF-κB. RESULTS LPS treatment activated the p38 in nasal epithelial cells in a dose-dependent fashion and a maximum phosphorylated p38 was induced by 1 mg/L LPS(0.851±0.108)(t =9.484, P0.01). The activity of p38 MAPK was significantly inhibited by SB203580 in LPS-treated cells(0.248±0.055)(t =8.741, P 0.01). LPS treatment induced a significant increase(2.116±0.037) in the DNA binding activity of NF-κB, which can be partially reversed by SB203580(1.323±0.038)(t =10.662, P 0.01). CONCLUSION In LPS-treated nasal epithelial cells, the p38 MAPK was activated and it possibly partially regulated activation of NF-κB.
