Cyclosporine A inhibits NF-κB activity and down-regulates interleukin-2 gene expression after liver transplantation in rat

Abstract:Objective To investigate the relationship between the activity of NF-κB and the expression of IL-2 gene with or without cyclosporine A treatment after liver transplantation. Methods The rat orthotopic liver transplantation model was applied. The rats were divided into three groups: GroupⅠ: syngeneic control (Wistar-to-Wistar); Group Ⅱ: allograft (SD-to-Wistar); Group Ⅲ: allograft treated with CsA by intramuscular route (SD-to-Wistar+ CsA). EMSA and RT-PCR were used to analyze NF-κB activity of splenocytes and IL-2 gene transcription expression of grafted liver with or without CsA treatment after liver transplantation. Histopathological examination was done. Results Low NF-κB activity was only detected at day 5 and day 7 in Wistar-to-Wistar group after transplantation, meanwhile low IL-2 gene expression was detected at any time point in this group. In contrast, high activity of NF-κB was detected in SD-to-Wistar group and high level of IL-2 gene expression was detected at all time points in this group. The activity of NF-κB and IL-2 gene expression were significantly inhibited in SD-to-Wistar+CsA group as compared with those in SD-to-Wistar group( P 0.05 ). A good correlation was found between activity of NF-κB and IL-2 gene expression. Conclusion The expression change of IL-2 gene is at least partially due to the activity change of NF-κB after orthotopic liver transplantation. CsA can inhibit NF-κB activity and further down-regulate IL-2 gene expression. Blockade of NF-κB molecule pathway may be a benefit measurement to liver transplantation rejection.
