Abstract:Xylanase produced from Trichoderma spp.can induce plant resistance,which is one of important biocontrol mechanisms of fungal Trichoderma.The effect of cultural condition on the producing xylanase by T.harzianum strains was investigated.Appropriate pH,temperature,concentration of surface active agent Tween 80,incubation period were optimized for enzyme production.The results show that the culture conditions of maximum enzyme activities produced from different T.harzianum strains were different in shaker flasks with semi-solid media.Maximum enzyme activities were at pH 5.0,30 ℃,0.7%(concentration of surface active agent Tween 80)and 72 h for NF9,pH 4.0,27 ℃,1.2% and 84 h for P1,pH 5.0,27 ℃,0.7% and 84 h for CT22,pH 4.0,30 ℃,0.7% and 96 h for TC3,respectively.

Abstract:In order to study rapidly on the storage stability of β-glucanase, the accelerated storage test was carried out to estimate the shelf life of β-glucanase at different temperature. The samples was kept in the water bath at 28℃, 36.2℃, 45℃ and its activity was determined at different time. The results of statistical treatment of data indicate that the activity of β-glucanase decreases by 10% after 1.36d, 4.37d, 20.09d, 30.7d, 67.0d, 145.9d, 2.21years, 4.14 years and decreased by 50% after 8.5d, 25.47d, 104.9d, 154.0d, 316.0d, 648d, 3154d when it was kept at 45℃, 36.2℃, 28℃, 25℃, 20℃, 15℃, 4℃ and 0℃, respectively. The results indicate that the stability of β-glucanase is competent for the storage, and the accelerated storage test can estimate the shelf life of β-glucanase.

Abstract:The technological conditions degrading corn stalk,such as dosage of enzyme,pH,reaction time,reaction temperature were studied with xylan.It was optimized by orthogonal experiments.The influential extent of affecting factors to degrading corn stalk is: pH>reaction temperature>dosage of enzyme>reaction time.The best condition of degrading corn stalk with xylanase is dosage of enzyme 65 ×104 U/g,pH 4.0,reaction temperature 50 ℃,reaction time 20 h.The best degradation rate is up to 26.61%.

Abstract:The enzymatic characteristics, enzyme kinetics and active site of xylanase were researched by using the methods of activity and chemical modification. The results showed that conditions of reaction temperature of 60 degrees C, pH of 5.0 was best for xylanase hydrolysis. And the activity of this enzyme had good stability at the temperature of 40-60 degrees C and pH of 5.0. The chemical modification of NBS, CDC, TNBS were used to react with xylanase. The result showed that the tryptophan residues and carboxyl made up of the active center of the protein of this enzyme, and was essential in the catalytic site.
Overall Evaluation of Xylanase As Feed Additives

Abstract:Activity of a kind of xylanase was assessed in this study by four respective experiments to evaluate whether it was acid-resistant or heat-resistant,whether the activity of xylanase can keep down after passing through the stomach which was with low pH and contains activated pepsin and whether it was also effective to raw materials such as wheat bran.As a result,the activity of this xylanase kept above 5 000 U/g when pH value of buffer varied between 5.4 and 6.8,while drops down fast when pH was below 5.4.The activity decreased fast when the xylanase was in gastric juice of finishing pigs,dropping down to 37.4% of the initial after 1.5 hours’ reaction.In heat-resistant experiment,it was observed that the activity of the xylanase loses no more than 5% of the initial when left in a 90℃ oven for 5 minutes.Further more,this xylanase could break down starch-free wheat bran by 5% after 4 hours’ reaction.These indexes could give a comparatively accurate evaluation on xylanases and therefore provide more reliable date to feed producers and farmers.
Characterisation and effects of a xylanase enzyme preparation extracted from Thermomyces lanuginosus cultures

Abstract:This paper describes the production of an enzyme preparation from the fungus Thermomyces lanuginosus. Thermal resistance, pH stability and lignocellulolytic activity of the enzyme preparation high in xylanase were studied on a variety of grains and forages. The enzyme preparation preserved more than 70% of its original xylanase activity for 4 and 1 h at 60 and 70 degrees C, respectively. The xylanase activity remained over 80% when the preparation was incubated for 30 min at pH 4.5. In vitro digestibility studies indicated that the enzyme digested 7.5, 8.5 and 8.0% of the dry matter (DM) of barley meal, wheat bran and oat meal samples, respectively. When applying 60-min incubation, 7.5, 7.3 and 8.4% of DM of the oat straw, alfalfa hay and triticale straw was digested, respectively. When the time of digestion was increased to 360 min, the sunflower hull showed 15.8% DM digestibility.

Abstract:Xylanase was separated and purified from a culture filtrate of Trichoderma Koningii through ammonium sulfate precipitation,Sephadex G-25 and Sephadex G-100 column chromatography.The purified xylanase was then proved by SDS-PAGE to be a single composition with a relative molecular mass of 55208.Good thermostability of xylanase with an optimal activity at 65℃ and pH 6.0 was also revealed.The results show that more than 83% or 85% of the relative activity of xylanase can be remained when respectively stored at 30~60℃ for 2h or in the pH range of 3.0~10.0.Moreover,Ba~(2+),Pb~(2+),Fe~(2+),Fe~(3+),Al~(3+) and 12mmol/L Cu~(2+) are strong inhibitors to xylanase,while Ca~(2+),Zn~(2+) and 4mmol/L Cu~(2+) are stimulators.The Michaelis-Menten constant of xylanases in the enzymatic hydrolysis of birchwood xylan is 5.37g/L,and the maximal reaction velocity is 0.94μmol/min.

Abstract:[Objective] The objective was to use xylanase to degrade the xylan in corn stalk and provide the reference for the biodegradation of corn stalk.[Method] Corn stalk was dried,grinded and sieved.A single factor test and orthogonal test were taken to study the effect of enzyme addition,pH value,enzymolysis time and temperature on the degradation rate of xylan in corn stalk.[Result] The effect of factors on degradation rate in order was pH valueenzymolysis temperatureenzyme additionenzymolysis time.When the enzymolysis temperature was 50 ℃,the degradation rate of xylan was the biggest.When the enzyme addition was 65×104 U/g,the enzymolysis time was 20 h and pH value was 4.0,the degradation rate of xylan was the biggest,being 27.80%,27.00%,26.37% resp.[Conclusion] The optimum conditions for the degradation of xylan in corn stalk by xylanase was enzyme addition of 65×104 U/g,pH value of 4.0,enzymolysis time of 20 h and enzymolysis temperature of 50 ℃,under which the degradation rate of xylan in corn stalk was 26.61%.

Abstract:Some properties of the immobilized α-glucosidase (ITGL), which was prepared by adsorbing and crosslinking on spherical microporous chitosan carrier, were compared with the nature one (TGL)'s. The optimum reaction pH of the ITGL was 4.5, while the TGL's was 5.0; its optimum reaction temperature was 70°C, which was 5°C higher than that of TGL, and the range of its optimum reaction temperature was 40 ∼ 70°C, and much larger than that of TGL. Its stability for enduring pH changing, heat, and long-time storing was much better than that of TGL. TGL and ITGL were both unstable to high ion intensity and urea. The low concentration of Ca 2+ and Mg 2+ could increase the activity of TGL and ITGL, but Fe 2+, Ag +, Cu 2+, and Hg 2+ obviously inhibited the activity of both enzymes. The reaction half-life of ITGL was 24 days. The kinetics constant of ITGL did not obviously change when the substrate was maltose.

Abstract:In this manuscript, the conditions for corn stalk by β-glucanase were carefullyinvestigated by single factors experiment and orthogonal experiments and described as follows:dosage of enzyme>pH>reaction temperature>reaction time. The best condition of degradingcorn stalk with β-glucanase is dosage of enzyme 25. 86×104 U/g, pH 4. 5, reaction temperature45 ℃, reaction time 15 h. With the optimum conditions, the degradation rate was achieved at41.96%.

Abstract:Xylanases have broad applications in the food industry to decompose the complex carbohydrate xylan. This is applicable to enhance juice clarity, improve dough softness, or reduce beer turbidity. It can also be used to produce prebiotics and increase the nutritional value in foodstuff. However, the low yield and poor stability of most natural xylanases hinders their further applications. Therefore, it is imperative to explore higher-quality xylanases to address the potential challenges that appear in the food industry and to comprehensively improve the production, modification, and utilization of xylanases. Xylanases, due to their various sources, exhibit diverse characteristics that affect production and activity. Most fungi are suitable for solid-state fermentation to produce xylanases, but in liquid fermentation, microbial metabolism is more vigorous, resulting in higher yield. Fungi produce higher xylanase activity, but bacterial xylanases perform better than fungal ones under certain extreme conditions (high temperature, extreme pH). Gene and protein engineering technology helps to improve the production efficiency of xylanases and enhances their thermal stability and catalytic properties.

Abstract:In order to improve the β-glucanase stability,the effect of different substances on β-glucanase stability was studied through single factor experiments,and the orthogonal experiment was used to optimize the complex protective agents.Finally,stability of enzyme with or without complex protective agents was measured.Results indicate that the albumin,glutin and CaCl2·H2O can enhance the enzyme stability.There is a remarkable synergistic effect when the three protective agents are mixed to use.The complex protective agents consists of 0.8% albumin(W/V),2% glutin(W/V) and 1 mmol·L-1 CaCl2·H2O.The t1/2 value of enzyme with protective agents is 4℃ higher than that of enzyme with no protective agent.The thermal degradation rate is-0.0398 for enzyme with protective agent and-0.084 for enzyme with no protective agent at 36.2℃.The results above suggest that the complex protective agents can effectively enhance the thermostability and storage stability of β-glucanase.

Abstract:In experimental plant science, research often faces large numbers of tissue samples resulting from sequential harvests of a larger number of genotypes and treatments combinations. Analyses of biological processes such as enzyme kinetics are often time-consuming or need specific sample preparation procedures before the actual measurements can be done. Time is thus often the critical factor and the possibility to store plant samples either as tissue or as extracts increases the available timeframe for analyses. Biological molecules such as enzymes often change their activities when stored and thus do not reflect the processes occurring in living tissue. We investigated the effect of different storage methods such as freeze-drying, freezing at −20 °C, and freezing at −80 °C on the activity of three enzymes known as antioxidants, namely ascorbate peroxidase, glutathione reductase, and superoxide dismutase from two rice varieties. Varieties differed in enzyme activity in extracts of fresh material from leaf blades, leaf sheaths, and roots. When subjected to different storage methods, there were no differences between varieties, but strong effects of the different storage methods on enzyme activities were found. The effects of the storage methods on enzyme activity strongly differed between extracts from stored tissue samples or extracts stored from freshly sampled material. We propose enzyme-specific storage methods and durations that allow for expanding the window for analyses in large experimental studies involving destructive samplings for enzyme kinetics.

Abstract:Dynamics and mode of acting on xylan of three parts of xylanases (Part A, Part B and Part C) separated and purified from a culture filtrate of Penicillium corylophilum No. P-3-31 were investigated. The Km and Vmax of the three purified enzymes with birchwood xylan as a substrate were 1.00 mg/mL and 0.159 U/mL for Part A ( pH 4.0, 50 ℃) , 1.59 mg/mL and 0.274 U/mL for Part B (pH 4.0,50 ℃) and 0.85 mg/mL and 0.200 U/mL for Part C (pH 5.5,50 ℃). Based on the dynamic studies, it was deduced that three purified enzymes had the same mode of action on xylan. Xylotriose and xylobiose were main hydrolysates of long-chain xylan by these enzymes. Xylooligosaccharides upwards from xylotetraose were immediately hydrolyzed, and xylotetraose was mainly hydrolyzed to xylobiose. Xylotriose was slowly hydrolyzed, but xylobiose was unable to be further hydrolyzed by these enzymes. The studies on the hydrolysis dynamics of xylan from corncob with crude enzyme showed that the hydrolysis of arabinose side-chain was almost simultaneous with the hydrolysis of xylan, therefore it could be concluded that there existed a α-L-arabinofuranosidase in crude enzyme.

Abstract:In order to screen strains that have high efficient xylan degrading ability,liquid fermentation and culture,phylogenetic and morphological analysis,SDS-PAGE and zymogram analysis and other methods were used.Comparative analysis revealed that in 10 strains,50647 showed the best xylanase producing time and activity.After 3 days,the activity of xylanase in the culture supernatant reached to 49.3 U/mL,special activity was 1066.2U/mg;This strain was identified as Pestalotiopsis sp.Culture supernatant contained at least six xylanases with different molecular mass.

Abstract:AbstractXynAF1 from Aspergillus fumigatus Z5 is an efficient thermophilic xylanase belonging to glycoside hydrolase family 10 (GH10). The non-catalytic amino acids N179 and R246 in its catalytic center formed one and three intermolecular H-bonds with the substrate in the aglycone region, respectively. Here we purified XynAF1-N179S and XynAF1-R246K, and obtained the protein-product complex structures by X-ray diffraction. The snapshots indicated that mutations at N179 and R246 had decreased the substrate-binding ability in the aglycone region. XynAF1-N179S, XynAF1-R246K, and XynAF1-N179S-R246K lost one, three, and four H-bonds with the substrate in comparison with the wild-type XynAF1, respectively, but this had little influence on the protein structure. As expected, N179S, R246K, and N179S-R246K led to a gradual decrease of substrate affinity of XynAF1. Interestingly, the enzyme assay showed that N179S increased catalytic efficiency, while both R246K and N179S-R246K had decreased catalytic efficiency.Key points• The non-catalytic amino acids of XynAF1 could form H-bonds with the substrate.• The protein-product complex structures were obtained by X-ray diffraction.• The enzyme-substrate-binding capacity could affect enzyme catalytic efficiency.

Abstract:he characteristics of the xylanase from Trichoderma reesei strainGXC were studied. The results showed that the optimal temperature was 55 ℃and pH 5.3 respectively , the range of pH stability was wide ,the enzyme was stable below 40 ℃. Cu2+ ,Zn2+,Fe2+and Fe3+ had inhibitory effect on the enzyme activity, but Co2+ and Mn2+ could stimulate the activity. Vmax was 526 μmol / (min·mg) , Km was 3.42 mg/mL.

Abstract:In this study,an enzyme activity comparasion was conducted to find the differences among four kinds of xylanase gene expressions in E.coli BL21,providing the basis for factories to screen the gene with high expression and heat resistance.A pair of primers was designed according to the published nucleotide sequences of putative xylanase genes.With the primers,four target fragments of the β-1,4-endo-xylanase gene was amplified from Bacillus macerans(B.macerans) B3,the lichens Bacillus(B.licheniformis) B6,Bacillus cereus(B.cereus) B10 and Bacillus(B.subtilis) B12,the sequencing results showed that the open reading frame of the four genes were all 642 bp.Three of which were first reported.The results of enzyme characteristics analysis showed that the four recombinant xylanase enzyme activity at 55 ℃ was 36.16,3.85,7.22 and 98.98 U/ml,respectively,in which the bacillus subtilis xylanase enzyme showed the highest enzyme activity.The heat resistance experiment results showed that the bacillus subtilis xylanase had a higher enzyme activity and better heat-resistance.
