Association analysis of dopamine β-hydroxylase gene and hyperactivity impulsive subtype of attention-deficit/hyperactivity disorder

Abstract:Objective:To examine the association between dopamine β-hydroxylase gene and hyperactivity impulsive subtype of attention-deficit /hyperactivity disorder. Methods: Blood DNA samples from 99 unrelated ADHDHI children( including 77 trios,from 2092 ADHD children) and 599 unaffected( healthy) controls were included in this study. The diagnosis was based on the Diagnostic and Statistical Manual of Mental Disorders,Fourth Edition( DSM-IV). Five single nucleotide polymorphisms( SNPs) of DBH gene were genotyped implementing Taq-ManTM real-time polymerase chain reaction( PCR) genotyping technique. The chi-square test and transmission disequilibrium test( TDT) were employed for statistical analyses using plink software; false discovery rate( FDR) control was used to evaluate gene-wide statistical significance. Results: In case-control analysis,the frequencies of the T allele of rs1076150( case-control frequencies 0. 933 vs. 0. 880) / rs1611115( 0. 231 vs. 0. 154) / rs2873804( 0. 885vs. 0. 803),and the frequencies of the A allele of rs1548364( 0. 882 vs. 0. 807) were significantly higher in ADHDHI after FDR test( empirical P 0. 05). In TDT analysis, T allele of rs1076150, rs2873804, the A allele of rs1548364 was preferentially transmitted in ADHD-HI( empirical P 0. 05) and T allele of rs1611115 showed a trend of over-transmission after FDR test( empirical P = 0. 054). Conclusion: These results provide some evidence that DBH gene may be associated with attention-deficit /hyperactivity disorder-hyperactivity impulsive subtype.
