Abstract:A visible-light-induced highly efficient C(sp3)-H amination of ethers with amides and azoles has been presented under mild conditions via a nitrogen- and carbon-centered radical coupling process. This protocol successfully utilizes 2,3-dichloro-5,6-dicyano-p-benzoquinone (DDQ) and tert-butyl nitrite (TBN) as cocatalysts to deliver the aminated products of ethers under aerobic conditions. Notably, the developed reaction features the corresponding products in good yields (up to 93%) with a wide substrate scope. The mechanistic study indicates that C-N bond formation proceeds via a direct radical cross-coupling process. Preliminary biological activity analysis indicates that the resulting products have good and selective inhibitory activity on osteosarcoma (OS) cell lines and are promising for use as hits for drug discovery.

Abstract:A protocol of visible-light-promoted C2 selective arylation of quinoline and pyridine N-oxides, with diaryliodonium tetrafluoroborate as an arylation reagent, using eosin Y as a photocatalyst for the construction of N-heterobiaryls was presented. This methodology provided an efficient way for the synthesis of 2-aryl-substituted quinoline and pyridine N-oxides. This strategy has the following advantages: specific regioselectivity, simple operation, good functional group tolerance, and high to moderate yields under mild conditions.

Abstract:An efficient method for the synthesis of isoquinolinone derivatives via photopromoted carboamination of alkynes is developed. Starting from the readily available propargyl alcohol derivatives, the polycyclic isoquinolinone derivatives could be obtained with good aryl and heterocycle tolerance. Both terminal and alkyl substituted alkynes could be employed. This protocol is operationally easy, and easily conducted on a gram-scale. A possible mechanism involving radical addition and cyclization following aromatization was proposed.

Abstract:Herein, we describe a visible light‐induced C(sp2)−H arylation method for quinoxalin‐2(1H)‐ones and coumarins using iodonium ylides without the need for external photocatalysts. The protocol demonstrates a broad substrate scope, enabling the arylation of diverse heterocycles through a simple and mild procedure. Furthermore, the photochemical reaction showcases its applicability in the efficient synthesis of biologically active molecules. Computational investigations at the CASPT2//CASSCF/PCM level of theory revealed that the excited state of quinoxalin‐2(1H)‐one facilitates electron transfer from its π bond to the antibonding orbital of the C−I bond in the iodonium ylide, ultimately leading to the formation of an aryl radical, which subsequently participates in the C‐H arylation process. In addition, our calculations reveal that via single‐ electron transfer (SET) process, the C−I bond cleavage in iodonium ylide and new C−C bond formation between resultant aryl radical and cationic quinoxaline species take place in a concerned manner. This enables the arylation reaction to efficiently proceed along an energy‐efficient route.

Abstract:Simple and practical strategies for visible-light-induced C-H alkylation of 2-amino-1,4-naphthoquinones with cyclobutanone oxime esters and hydroxamic acid derivatives have been developed under mild and redox-neutral conditions. These two reactions can be carried out at room temperature and obtain a variety of 2-amino-1,4-naphthoquinone derivatives with cyano and amide groups. Moreover, the cyanoalkylation reaction of 2-amino-1,4-naphthoquinones can proceed smoothly in the absence of photocatalysts.

Abstract:A unified strategy involving visible-light-induced iminyl-radical formation has been established for the construction of pyridines, quinolines, and phenanthridines from acyl oximes. With fac[Ir(ppy)(3)] as a photoredox catalyst, the acyl oximes were converted by 1e(-) reduction into iminyl radical intermediates, which then underwent intramolecular homolytic aromatic substitution (HAS) to give the N-containing arenes. These reactions proceeded with a broad range of substrates at room temperature in high yield. This strategy of visible-lightinduced iminyl-radical formation was successfully applied to a five-step concise synthesis of benzo[c]-phenanthridine alkaloids.

Abstract:We describe an efficient method for α‐functionalization of N‐aryl‐tetrahydroisoquinolines under visible‐light‐irradiation catalyzed by organic photocatalyst. This protocol provides a concise and environmental approach for the rapid allylation and benzylation of N‐aryl‐tetrahydroisoquinolines, and shows broad substrate scope. Stable organoboron reagents have shown their ability in the construction of challenging Csp3–Csp3 bond. The load of the photocatalyst is low and the oxidant is inexpensive and less toxic.

Abstract:Due to the high C–H bond dissociation energy of alkanes, the utilization of alkanes as alkyl radical precursors for C–H functionalization of heteroarenes is synthetically captivating but practically challenging especially under metal- and photocatalysts-free conditions. We report herein a mild and practical visible-light-mediated method for C–H alkylation of quinoxalin-2(1H)-ones with trifluoroacetic acid as hydrogen atom transfer reagents and air as the oxidant. This mild protocol performed under metal- and photocatalysts-free circumstances and represents good functional-group tolerance as well as a broad substrate scope.

Abstract:A visible‐light‐catalyzed formal [3 + 2] annulation‐aromatization reaction for the synthesis of various substituted imidazo[2,1‐a]isoquinolines from amidines with stabilized isoquinolinium N‐ylides in the presence of bases is developed. The procedure reported here involves a direct C‐H activation and the formation of C‐C/C‐N bonds in one‐pot. The reaction mechanism is probed by radical‐trapping experiment, fluorescence quenching, and light on/off experiments. The late‐stage modification experiments provide the potential applications in the field of organic and medicinal chemistry for chemists.

Abstract:A general visible-light-induced photocatalyst-/additive-free strategy was developed for the construction of various nitrogen-heterocycles (42 examples, up to 97% yield) such as γ-lactams, δ-lactams, pyrrolidines, indolones, quinolinones, and fused polycyclic structures at room temperature. The prominent features of this protocol are mild and environmentally friendly conditions, broad substrate scope, and good functional group tolerance. Importantly, the reaction can be performed under natural sunlight, the most sustainable energy source imaginable. Furthermore, this strategy provides straightforward access to a range of derivatives through subsequent elaboration of the xanthate group.

Abstract:A metal-free three-component approach has been developed to prepare 2,4-disubstituted quinazolines from o-acylanilines, trialkylamines and ammonium chloride under visible-light using Eosin Y as the photocatalyst. The notable features of this work include, (i) tertiaryamines as alkyl- and triethanolamine as a C2-OH synthon; (ii) good functional group tolerance with 38% - 98% yields; (iii) proof of concept with o-amino benzaldehyde as a substrate to deliver 2-methyl quinazoline 3pa; and (iv) gram-scale synthesis of compounds 3ga, 3ja and 3ma. A reductive quenching mechanism was proposed based on the control studies and redox potential values.

Abstract:Visible light-induced C-3 arylation of quinoxalin-2(1H)-ones with abundantly available aryl iodides with good yields via an EDA-complex formation has been accomplished. Both aryl/heteroaryl iodides and quinoxalin-2(1H)-ones possessing electron-donating as well as electron-withdrawing groups were coupled well to access the desired products in good yields. The radical scavenging, EPR, UV-visible experiments, and quantum yield revealed that the reaction went through a radical pathway via a SET process. Furthermore, the protocol could also be applied for late-stage functionalization, illustrated the practicability of the present protocol.

Abstract:A visible‐light‐induced, external photocatalyst‐free, site‐selective C‐H arylation of 4H‐pyrido[1,2‐a]pyrimidin‐4‐ones and various 2‐aryl heteroarenes has been developed to provide ortho arylated products 41‐ 95% yield at room temperature. A mechanistic study using controlled experiments, UV‐visible study, DFT study, and observation of colored aggregates suggest that in situ generated Ag‐palladacycle complex of 2‐aryl heteroarenes is responsible for absorbing the visible light and generating aryl radicals from aryldiazonium salts. Further, this methodology demonstrates its versatility by transforming various 2‐aryl heteroarenes, including 2‐phenylquinazolin‐4(3H)‐one, 3‐phenylquinoxalin‐2(1H)‐one, 2‐phenylpyridines, pyrazolo[1,5‐a]pyrimidines, 2‐phenylquinolines, 2‐phenylisoquinolines, and 2‐phenylbenzo[d]thiazoles, into their respective regio‐selective C‐H arylated products under mild reaction conditions.

Abstract:Aryl halides are one of the most important chemical feedstocks in pharmaceuticals due to its easy accessibility, inexpensiveness, and widely utilized as aryl radical precursors in organic synthetic chemistry. Conventionally, the stoichiometric reagents such as AIBN/n‐Bu3SnH were used for generation of aryl radical from aryl halides, and suffered from requirement of toxic radical initiators, high temperature and thus, the development of simple, mild strategies for the generation of aryl radical from aryl halides are highly desirable. Recently, visible light mediated strategies received considerable attention, allowing the generation of aryl radical from aryl halides under mild reaction conditions. The present review described the recent breakthroughs and advancements in visible light mediated photocatalyst‐free C‐B/C/O/P/Se/S bond formation of aryl halides.

Abstract:Herein a metal-free approach for the synthesis of isoquinolone derivatives by means of photoinitiated deaminative [4+2] annulation of alkynes and N-amidepyridinium salts is described. This protocol exhibits a broad scope and good functional group tolerance and regioselectivity under benign reaction conditions. Preliminary studies suggest that the critical amide radical is derived from the photocatalytic cleavage of the N-N bond of the N-amidepyridinium salt, which adds to the triple bond of the alkyne and undergoes the annulation process to afford the desired isoquinolones.
