Effect of panaxadiol saponins on fluidity of red blood cell membrane in heavy load swimming trained rats

Abstract:AIM: To study effects of prolonged intensive endurance exercise and Panaxadiol saponins (PDS) on red blood cells (RBC) membrane fluidity in rats, as well as the mechanism. METHODS: By the membrane fluorence polarization, Pyrollol-NBT, TBA methods and Scanning Electron Microscope, RBC membrane fluidity,RBC-SOD and serum MDA were measured after 7 weeks prolonged intensive swimming. RESULTS: Fluorence polarization(P value) were significant higher and fluidity were lower( P 0 05), at the same time RBC-SOD activities lower and MDA contents higher in exercise-saline group than that of normal control group( P 0 05). P value were lower and fluidity were higher ( P 0 05), at the same time, RBC-SOD activities lower and MDA contents higher in exercise-PDS group than that of exercise- saline group( P 0 05). RBC count, Hb, MCHC and Hb index in exercise- saline group were significant lower than that of normal control group( P 0 05) and those parameter in exercise- PDS group were significant lower than that of exercise- saline group( P 0 05).Morphological study by scanning electron microscope showed that abnormal erythrocytes were found in exercise-saline group, while seldomly found in exercise-PDS group and normal control groups. CONCLUSION: PDS might reduce the lipid peroxidation in serum and RBC and has a good protective effect against RBC injury induced by prolonged intensive endurance exercise.
