Abstract:Despite great achievement that has been made in the synthesis of covalent organic frameworks (COFs), precise construction of COFs with well-defined nano/microstructures poses a rigorous challenge. Herein, we introduce a simple template-free strategy for controllable synthesis of hollow microtubular COFs. The obtained COFs show a spontaneous morphology transformation from a microfiber to a hollow microtubular structure when the concentrations of catalytic acid are regulated elaborately. Furthermore, the as-prepared COFs exhibit high crystallinity, well-defined hollow tubular morphology, and high surface areas (∼2600 m2/g). Taking the advantages of the unique morphological structure, the hollow microtubular COFs can serve as an ideal host material for enzymes. The resultant biocomposites show high catalytic performance and can be successfully applied to rapid and high-efficiency proteolysis of proteins. This work blazes a trail for controllable synthesis of the hollow microtubular COFs through a template-free process and expands the application of COFs as a promising platform for enzyme immobilization.

Abstract:Due to their extraordinary features such as Large surface areas, high porosity, and active surface functional groups, protein-inorganic nanoflowers have gained increasing attention. We have for the first time synthesized collagen-zinc phosphate hybrid nanoflowers via a mild biomineralization process. The incubation of 1.8 mM Zn(NO3)(2) with 0.2 wt% of collagen protein in 20 mM PBS at 25 degrees C for 24 hours Leads to the formation of exquisite flower-like nanomaterials with branched petals. Collagen may initiate the nucleation of the zinc phosphate crystals to form a framework of petals, and then glue the petals together. The synthesized collagen-Zn-3(PO4)(2) nanoflowers display catalytic activity towards water oxidation. The triple helical structure and repetitive (GLy-X-Y)(n) sequence pattern may endow collagen with a distinct capability to provide a superior biotemplate for producing hierarchical hybrid nanomaterials. The creation of collagenzinc phosphate hybrid nanoflowers by our facile and green tactics significantly enhances our capability to construct hopeite-based functional materials.

Abstract:Hybrid nanomaterials have recently emerged as a new interface of nanobiocatalysis, serving as a host platform for enzyme immobilization. Enzyme immobilization in inorganic crystal nanoflowers and metal-organic frameworks (MOFs) has sparked the bulk of scientific interest due to their superior performances. Many breakthroughs have been achieved recently in the preparation of various types of enzyme@MOF and enzyme-hybrid nanoflower composites. However, it is unfortunate that there are few reviews in the literature related to enzyme@MOF and enzyme-hybrid nanoflower composites and their improved synthesis strategies and their applications in biotechnology. In this review, innovative synthetic strategies for enzyme@MOF composites and enzyme-hybrid nanoflower composites are discussed. Enzyme@MOF composites and enzyme-hybrid nanoflower composites are reviewed in terms of biotechnological applications and potential research directions. We are convinced that a fundamental study and application of enzyme@MOF composites and enzyme-hybrid nanoflower composites will be understood by the reader as a result of this work. The summary of different synthetic strategies for enzyme@MOF composites and enzyme-hybrid nanoflower composites and the improvement of their synthetic strategies will also benefit the readers and provide ideas and thoughts in the future research process.

Ionic Liquid‐Mediated Dynamic Polymerization for Facile Aqueous‐Phase Synthesis of Enzyme‐Covalent Organic Framework Biocatalysts

Abstract:Utilizing covalent organic frameworks (COFs) as a hypotoxic and porous scaffold to encapsulate enzyme (enzyme@COF) has inspired numerous interests at the intersection of chemistry, materials, and biological science. In this study, we report a convenient scheme for one-step, aqueous-phase synthesis of highly crystalline enzyme@COF biocatalysts. This facile approach relies on an ionic liquid (2 μL of imidazolium ionic liquid)-mediated dynamic polymerization mechanism, which can facilitate the in situ assembly of enzyme@COF under mild conditions. This green strategy is adaptive to synthesize different biocatalysts with highly crystalline COF "exoskeleton", as well evidenced by the low-dose cryo-EM and other characterizations. Attributing to the rigorous sieving effect of crystalline COF pore, the hosted lipase shows non-native selectivity for aliphatic acid hydrolysis. In addition, the highly crystalline linkage affords COF "exoskeleton" with higher photocatalytic activity for in situ production of H2O2, enabling us to construct a self-cascading photo-enzyme coupled reactor for pollutants degradation, with a 2.63-fold degradation rate as the poorly crystalline photo-enzyme reactor. This work showcases the great potentials of employing green and trace amounts of ionic liquid for one-step synthesis of crystalline enzyme@COF biocatalysts, and emphasizes the feasibility of diversifying enzyme functions by integrating the reticular chemistry of a COF.

Abstract:Integrating nanozymes with natural enzymes to form cascade reactions is one of the most promising ways to develop biocatalysts with versatile performance; however, the applicability of the cascade is typically hampered by the instability of enzymes and the hindrance of mass transfer in the host environment. Utilizing amorphous ZIF-90 (aZIF-90) as a host material, herein, we have reported a one-pot way to encapsulate glucose oxidase (GOx) and magnetic nanoparticles (MNP) to form GOx/MNP@aZIF-90. We reasoned that the amorphous structure of ZIF-90 not only provides a protected environment to confine the cascade reaction but also generates mesopores and internal voids to improve the performance of the enzymatic cascade. The catalytic activity of aZIF-90 was almost 4 times higher than that of crystalline composites, and the residual activity was higher than 80% after being stored for 9 days. This is the first time that GOx and MNP were simultaneously confined in aZIF-90 with mesopores, which suggested that an amorphous metal-organic framework is promising in the development of an enzymatic cascade.

Abstract:In recent years, enzyme immobilization on metal-organic frameworks (MOFs) and inorganic crystal nanoflowers has generated a lot of research interest due to their great promise of enhancing enzyme stability, activity and even selectivity. In the past few years, many studies have focused on this topic to prepare various types of enzyme-MOF and enzyme-nanoflower composites. In this review, we will introduce the preparation and special properties of these newly developed enzyme-inorganic crystal composites. We summarized the preparation methods as physical adsorption, covalent conjugation and self-assembly. The advantages and limitations of each method will also be discussed in this review.

Abstract:A significant challenge in chemistry is to create synthetic structures that mimic the complexity and function of natural systems. Here, a self-activated, enzyme-mimetic catalytic cascade has been realized by utilizing expanded mesoporous silica-encapsulated gold nanoparticles (EMSN-AuNPs) as both glucose oxidase- and peroxidase-like artificial enzymes. Specifically, EMSN helps the formation of a high degree of very small and well-dispersed AuNPs, which exhibit an extraordinarily stability and dual enzyme-like activities. Inspired by these unique and attractive properties, we further piece them together into a self-organized artificial cascade reaction, which is usually completed by the oxidase-peroxidase coupled enzyme system. Our finding may pave the way to use matrix as the structural component for the design and development of biomimetic catalysts and to apply enzyme mimics for realizing higher functions.

Abstract:A nature-inspired approach to encapsulate enzymes in spindle-like copper hydroxysulfate nanocrystals was developed by a biomimetic mineralization process. Several types of enzymes including glucose oxidase (GOx), horseradish peroxidase (HRP), Candida antarctica lipase B (CALB) and cytochrome c (Cyt c) were successfully encapsulated in copper hydroxysulfate nanocrystals quickly (within 1 hour) with maintained or even greatly enhanced catalytic efficiencies (k(cat)/K-M of Cyt c showed a 143-fold increase) and high stabilities, demonstrating the feasibility of utilizing copper hydroxysulfate nanocrystals as a novel type of nanocarrier for enzyme immobilization. In addition, by this approach, for the first time, we showed that an immobilized enzyme can be endowed with self-antibacterial activity by an inorganic component. This self-antibacterial performance together with the improved catalytic efficiencies and stabilities can greatly benefit the enzymatic catalysis in aqueous media and promote the future development of novel biosensors.

Abstract:The synthesis of ultrafine, uniform, well-dispersed functional nanoparticles (NPs) under mild conditions in a controlled manner remains a great challenge. In biological systems, a well-defined biomineralization process is exploited, in which the control over NPs' size, shape and distribution is temporally and spatially regulated by a variety of biomolecules in a confined space. Inspired by this, we embedded proteins into metal-organic frameworks (MOFs) and explored a novel approach to synthesize metallic NPs by taking the synergy of protein-induced biomineralization process and space-confined effect of MOFs. The generation and growth of ultrafine metal NPs (Ag or Au) was induced by the entrapped lysozyme molecules and confined by the ZIF-8 pores. Due to the narrow size distribution and homogeneous spatial distribution of metal NPs, the as-synthesized NPs exhibit remarkably elevated catalytic activity. These findings demonstrate that MOFs can be loaded with specific proteins to selectively deposit inorganic NPs via biomimetic mineralization and these novel kinds of nanohybrid materials may find applications in catalysis, sensing and optics.

Abstract:The integration of chemical catalyst and biocatalyst is of great interest due to the accompanying high designability of chemo-enzymatic cascade reactions for sustainable catalytic processes in chemical engineering. Here, a facile strategy is proposed by preparation of a copper ions coordinated cytidine monophosphate (CMP) encapsulated lipase composite via in-situ assembly in aqueous solution which followed by reduction to obtain an enzyme-copper hybrid catalyst. The as-synthesized enzyme-copper nucleotide composite exhibited both high enzymatic activity (6.05 U/mg) and copper-catalytic activity (with reaction rate constant of 8.85 min-1) in the chemo-enzymatic cascade reaction for hydrolysis of p-nitrophenyl butyrate (p-NPB) and generation of p-aminophenol (p-AP), which is a pharmaceutical intermediate for production of analgesics and antipyretics. This study thus sheds light on fabrication of hybrid catalyst under facile conditions, exhibiting potential in development of biosensors, industrial catalysis and biomanufacturing.

Abstract:Here, we describe a versatile protein-directed assembly strategy that enables the organization of different types of proteins and organic linkers into a highly crystalline hybrid framework through hydrogen-bond interaction. The whole assembly process is protein actuated but is independent of the protein-surface property. Advanced low-electron-dose cryoelectron microscopy techniques clearly witness the crystallographic structure of hybrid framework at a single-molecular level, and we demonstrate that the proteins are independently and tightly isolated in the crystalline frameworks, with a record-high protein content in the reported biohybrid framework materials. In addition, the hybrid framework has ultrahigh chemical stability, and its aperture structure and protein confinement tightness are controllable through modulating the organic linkers. When using enzymes as the building block, the obtained enzyme framework shows significantly improved stability compared with the free enzymes and displays notable advantages for biocatalysis compared with the burgeoning enzyme-MOF biohybrids in terms of active ingredient content, robustness, and catalytic efficiency.

Abstract:An understanding of controlled formation of biomimetic mesocrystals is of great importance in materials chemistry and engineering. Here we report that organic-inorganic hybrid plates and even mesocrystals can be conveniently synthesized using a one-pot reaction in a mixed system of protein (bovine serum albumin (BSA)), surfactant (sodium bis(2-ethylhexyl) sulfosuccinate (AOT)) and supersaturated calcium phosphate solution. The morphologies of calcium-phosphate-based products are analogous to the general inorganic crystals but they have abnormal and interesting substructures. The hybrids are constructed by the alternate stacking of organic layer (thickness of 1.31 nm) and well-crystallized inorganic mineral layer (thickness of 2.13 nm) at the nanoscale. Their morphologies (spindle, rhomboid and round) and sizes (200 nm-2 mu m) can be tuned gradually by changing BSA, AOT and calcium phosphate concentrations. This modulation effect can be explained by a competition between the anisotropic and isotropic assembly of the ultrathin plate-like units. The anisotropic assembly confers mesocrystal characteristics on the hybrids while the round ones are the results of isotropic assembly. However, the basic lamellar organic-inorganic substructure remains unchanged during the hybrid formation, which is a key factor to ensure the self-assembly from molecule to micrometre scale. A morphological ternary diagram of BSA-AOT-calcium phosphate is used to describe this controlled formation process, providing a feasible strategy to prepare the required materials. This study highlights the cooperative effect of macromolecule (frame structure), small biomolecule (binding sites) and mineral phase (main component) on the generation and regulation of biomimetic hybrid mesocrystals.

Abstract:Therapeutic enzymes have been considered as a promising candidate for various medical applications due to their high specificity. However, how to maintain the enzymatic stability in the long-term, while performing the tunable activity of a certain enzyme for different physiological requirements, is still a challenge. Herein, a time and ratio associated method was employed to form a silica shell with controllable nanopores on the surface of the enzyme as a nanoreactor for preserving and freely tailoring its activity. The model enzymes (a-amylase and glucose oxidase) entrapped in these silica nanoreactors are able to perform a distinct activity as designed with up to a 10-fold difference, while the lifecycle and tolerability to harsh conditions (pH and temperature) are all dramatically enhanced compared with those of free enzymes. Moreover, this modified enzyme was used for tumor therapy and presented excellent performance. The silica-covered glucose oxidase (GO) associating with Larginine (Arg) modification could be easily delivered to tumor cells and then exhibited high persistence of the activity in cytoplasm, which is able to convert glucose to H2O, (GO) and finally generate NO (Arg) in tumor cells, resulting in high cell cytotoxicity for tumor therapy. The high efficacy of our device in tumor therapy indicates that this approach could be broadly used to engineer enzymes for different biological applications.

Abstract:Hybrid materials that combine organic polymers and biomacromolecules offer unique opportunities for precisely controlling 3D chemical environments. Although biological or organic templates have been separately used to control the growth of inorganic nanoclusters, hybrid structures represent a relatively unexplored approach to tailoring nanocluster properties. Here, we demonstrate that a molecularly defined lysozyme-polymer resin material acts as a structural scaffold for the synthesis of copper nanoclusters (CuNCs) with well controlled size distributions. The resulting CuNCs have significantly enhanced fluorescence compared with syntheses based on polymeric or biological templates alone. The synergistic approach described here is appealing for the synthesis of biocompatible fluorescent labels with improved photostability.

Abstract:The enzyme-triggered self-assembly of peptides has flourished in controlling the self-assembly kinetics and producing nanostructures that are typically inaccessible by conventional self-assembly pathways. However, the diffusion and nanoscale chemical gradient of self-assembling peptides generated by the enzyme also significantly affect the outcome of self-assembly, which has not been reported yet. In this work, we demonstrated for the first time a spatiotemporal control of enzyme-triggered peptide self-assembly. By simply adjusting the temperature, we could change both the catalytic activity of the enzyme of phosphatase and their aggregation states. The strategy kinetically controls the production rate of self-assembling peptides and spatially controls their distribution in the system, leading to the formation of nanoparticles at 37 °C and nanofibers at 4 °C. The nanofibers showed ∼10 times higher cellular uptake by 3T3 cells than the nanoparticles, thanks to their higher stability and more ordered structures. Using such spatiotemporal control, we could prepare optimized nanoprobes with low background fluorescence, rapid and high cellular uptake, and high sensitivity. We postulate that this strategy would be very useful in general for preparing self-assembled nanomaterials with controllable morphology and function.

Abstract:Metal-organic frameworks (MOFs) for enzyme encapsulation-induced biomimetic mineralization under mild reaction conditions are commonly microporous and hydrophobic, which result in a rather high mass transfer resistance of the reactants and restrain the enzyme catalytic activity. Herein, we prepared a type of hierarchical porous and hydrophilic MOF through the biomimetic mineralization of enzymes, zinc ions, 2-methylimidazole, and lithocholic acid. The hierarchical porous structure accelerated the diffusion process of the reactants and the increased hydrophilicity conferred interfacial activity and increased the enzyme catalytic activity. The immobilized enzyme retained higher catalytic activity than the free enzyme and exhibited enhanced resistance to alkaline, organic, and high-temperature conditions. The nanobiocatalyst was reusable and showed long-term storage stability.
Enzyme/Nanocopper Hybrid Nanozymes: Modulating Enzyme-like Activity by the Protein Structure for Biosensing and Tumor Catalytic Therapy

Abstract:Artificial enzymes with modulated enzyme-mimicking activities of natural systems represent a challenge in catalytic applications. Here, we show the creation of artificial Cu metalloenzymes based on the generation of Cu nanoparticles in an enzyme matrix. Different enzymes were used, and the structural differences between the enzymes especially influenced the controlled the size of the nanoparticles and the environment that surrounds them. Herein, we demonstrated that the oxidase-like catalytic activity of these copper nanozymes was rationally modulated by enzyme used as a scaffold, with a special role in the nanoparticle size and their environment. In this sense, these nanocopper hybrids have confirmed the ability to mimic a unique enzymatic activity completely different from the natural activity of the enzyme used as a scaffold, such as tyrosinase-like activity or as Fenton catalyst, which has extremely higher stability than natural mushroom tyrosinase. More interestingly, the oxidoreductase-like activity of nanocopper hybrids was cooperatively modulated with the synergistic effect between the enzyme and the nanoparticles improving the catalase activity (no peroxidase activity). Additionally, a novel dual (metallic and enzymatic activity) of the nanozyme made the highly improved catechol-like activity interesting for the design of 3,4-dihydroxy-l-phenylalanine (l-DOPA) biosensor for detection of tyrosinase.
