Abstract:The Biaryl-bridged Salen Ti complexes are a high active catalyst at substrate-to-catalyst ratios as high as 1 000:1 in the reaction of asymmetric addition of trimethylsilyl cyanide to aldehydes,and the optimal catalyst 4 derived from(R,R)-1,2-diaminocyclohexane and 3,5-di-tert-butyl-2-hydroxybenzaldehyde produces trimethylsilyl ethers of cyanohydrins with up to 87% enantiomeric excess at ambient temperature.

Abstract:A dimeric salen ligand derived from L-tartaric acid was synthesized through linking two salen units with 1,6-dibromohexane. The corresponding Ti complex was proved to be an efficient catalyst for asymmetric addition of trimethylsilyl cyanide to aldehydes, providing the corresponding optically active trimethylsilyl ether of cyanohydrins with moderate to good enantiomeric excesses and excellent yields under relatively low catalyst loading condition.

Abstract:Herein, we report an unprecedented strategy for the simultaneous construction of a remote C-N stereogenic axis and three contiguous stereogenic carbon centers via silver-catalyzed desymmetrizative [3 + 2] cycloaddition of prochiral N-aryl maleimides with activated isocyanides. This method features operational simplicity, wide substrate scope, high efficiency, and good to excellent stereoselectivity. Notably, it represents the first example of catalytic enantioselective synthesis of C-N atropisomers with the use of activated isocyanides.

Abstract:Chiral organosilanes are of great value in asymmetric synthesis, functional materials, and medicinal chemistry. Compared with single-silyl compounds, bis(silyl) ones are understudied because of the lack of the efficient synthetic protocols. The development of efficient synthetic approaches to access bis(silyl) compounds is highly desirable for studying their basic properties and potential utilities. Here, a cobalt-catalyzed sequential double hydrosilylation of aliphatic alkynes was developed to synthesize highly enantioenriched gem-bis(silyl)alkanes. This protocol used simple aliphatic alkynes and silanes to construct valuable chiral gem-bis(silyl)alkanes. The control experiments, isotopic labeling experiments, kinetic studies, and density functional theory calculations were conducted to elucidate the reaction mechanism. The synthetic versatility of gem-bis(silyl)alkanes was demonstrated by the synthesis of chiral organosilanols, α-hydroxysilanes through selective C–Si bond transformation and hydrosilylation of alkynes to construct chiral silanes containing adjacent C-stereocenter and Si-stereocenter.

Abstract:A novel chiral spiro-pyrrolidine silyl ether organocatalyst has been designed. Its catalytic asymmetric effect is demonstrated by the Michael addition reaction, which affords the desired products with an all-carbon quaternary center in up to 99% ee and 87% yield. Furthermore, the reaction process has been investigated via in situ NMR experiments.

Abstract:A catalytic enantioselective addition reaction of silyl enol ethers to ketomalonate in the presence of C-2-symmetric copper(II) bisoxazolines as catalysts is presented; the reaction has been studied for different ligands and substrates under various reaction conditions and proceeds in good yield and with ee's > 90% and a procedure for the preparation of an optically active hydroxy acid is shown.

Abstract:We report a new thiourea-Bronsted acid cooperative catalytic system for the enantioselective cyanosilylation of aldehydes with yields up to 90% and enamioselectivities up to 88%. The addition of an achiral acid was found to be crucial for high asymmetric induction. Mechanistic investigations using a combination of NMR, ESI-MS, and density functional theory computations (including solvent corrections) at the M06/6-31G(d,p) level of theory suggest that the key catalytic species results from the cooperative interaction of bifunctional thioureas and an achiral acid that form well-defined chiral hydrogen-bonding environments.

Abstract:In contrast with abundant methods for the asymmetric functionalization of alkyl radicals to generate stereogenic centres at reaction sites, catalytic enantioselective desymmetrizing functionalization of alkyl radicals for forging multiple stereocentres—including positions that are remote from the reaction sites—with both high enantio- and diastereoselectivity remains largely unexplored. The major challenge for such reactions is the high reactivity of open-shell alkyl radicals. Here, we describe a strategy to address this challenge: the use of Cu( ii ) phosphate to immediately associate with the in situ-generated reactive alkyl radical species, creating a compact and confined chiral microenvironment for effective stereocontrol. With this strategy, we have developed a general and efficient catalytic enantioselective desymmetrizing functionalization of alkene-tethered 1,3-diols. It provides various tetrahydrofurans and analogues bearing multiple stereocentres with remarkably high levels of enantio- and diastereocontrol. Density functional theory calculations and mechanistic experiments revealed a reaction mechanism involving an enantiodetermining outer-sphere C–O bond formation step.

Abstract:An efficient enantioselective addition of thiols to acyclic trifluoromethyl ketimines has been established by using a bifunctional squaramide catalyst, which was derived from quinine, and the reaction was completed in 5 to 10 min. The construction of chiral tetrasubstituted carbon centers bearing trifluoromethylated N,S-ketals has been achieved in high yields (up to 96% yield) with excellent enantioselectivities (up to 99% ee).

Abstract:An approach to construct tetrasubstituted carbon stereocenters bearing both a trifluoromethylthio (SCF3) group and a cyano group has been realized, through asymmetric organophosphine‐catalyzed Mannich‐type reactions. The products were obtained in high yields and moderate to high enantioselectivities. Two diastereoisomers could be isolated from each reaction (overall 25 examples), rendering this approach a viable opportunity for molecular diversity generation and improvement of the bioactivity of related drug candidates.

Abstract:A highly enantioselective sulfa-Michael addition (SMA) of various thiols to alpha,beta-unsaturated N-acylated oxazolidinones has been achieved with a chiral squaramide catalyst under very mild reaction conditions. In addition, the conversion of the beta-thio-substituted adduct to the corresponding methyl beta-tosylbutanoate was demonstrated in high yield through a methanolysis/oxidation sequence.

Abstract:Asymmetric functionalizations of α‐sulfanyl cyclic ketones were realized via chiral phosphoric acid catalyzed enantioselective addition reactions with aldimines, azodicarboxylates and allenamides. A series of chiral organosulfur compounds possessing sulfur‐containing tetrasubstituted stereocenters were accessed via these methods, with excellent regioselectivities and high stereoselectivities.

Abstract:Silacycles have exhibited significant potential for application in the fields of medicinal chemistry, agrochemistry, and materials science. Accordingly, the development of effective methods for synthesizing these compounds has attracted increasing attention. Here, we report an efficient Cu‐catalyzed enantioselective hydrosilylation of arylmethylenecyclopropanes with hydrosilanes, that allows the rapid assembly of various enantioenriched carbon‐ and silicon‐stereogenic silacyclopentanes in good yields with excellent enantioselectivities and diastereoselectivities under mild conditions. Further stereospecific transformation of the Si−H bond on the chiral silicon center expands the diversity of these C‐ and Si‐stereogenic silacyclopentanes.

Abstract:The development of new strategy for construction of chiral cyclic sulfide-containing multiple stereogenic centers is highly desirable. Herein, by combination of base-promoted retro-sulfa-Michael addition and palladium-catalyzed asymmetric allenylic alkylation, the streamlined synthesis of chiral thiochromanones containing two central chiralities (including a quaternary stereogenic center) and an axial chirality (allene unit) was successfully realized with up to 98% yield, 49.0:1 dr and >99% ee.

Abstract:Stereoselective synthesis utilizing small-molecule catalysts, particularly N-heterocyclic carbene (NHC), has facilitated swift access to enantioenriched molecules through diverse activation modes and NHC-bound reactive intermediates. While carbonyl derivatives, imines, and "activated" alkenes have been extensively investigated, the exploration of heteroatom-centered analogues of NHC-bound intermediates has long been neglected, despite the significant potential for novel chemical transformations they offer once recognized. Herein, we disclose a carbene-catalyzed new activation mode by generating unique sulfinyl azolium intermediates from carbene nucleophilic addition to in situ-generated mixed sulfinic anhydride intermediates. Combined experimental and computational mechanistic investigations pinpoint the chiral NHC-catalyzed formation of sulfinyl azolium intermediate as the enantio-determining step. The novel "S"-based carbene reactive intermediate imparts high efficiency for the catalytic construction of sulfur-stereogenic compounds, giving rise to sulfinate esters with high yields and enantioselectivities under mild conditions. Notably, distinct from most of the NHC-catalyzed enantioselective transformations focusing on the "C" central chiral products, our study realizes a unique carbene-catalyst control over chiral "S" stereocenters via direct asymmetric S-O bond formation for the first time. Furthermore, these sulfinyl-containing products could serve as versatile synthetic platforms for enantioenriched S-stereogenic functional molecules and exhibit remarkable antibacterial activities against rice plant pathogens, which is valuable for the development of novel agrochemical agents.

Abstract:Herein, we report the discovery of the catalytic asymmetric conjugated addition of allylstannanes to indol-2-ones promoted by Ni(II)/chiral N,N′-dioxide. This method enables the stereodivergent construction of contiguous quaternary carbon centers (CQCCs), which can be readily achieved by a suitable combination of a chiral catalyst with stereospecific allylstannanes. This protocol allows for a unified access to all stereoisomers of 3,3-disubstituted oxindoles with CQCCs in good to excellent enantioselectivities and diastereoselectivities.

Abstract:Organic molecules bearing chiral sulfur stereocenters exert a great impact on asymmetric catalysis and synthesis, chiral drugs, and chiral materials. Compared with acyclic ones, the catalytic asymmetric synthesis of thio-heterocycles has largely lagged behind due to the lack of efficient synthetic strategies. Here we establish the first modular platform to access chiral thio-oxazolidinones via Pd-catalyzed asymmetric [3+2] annulations of vinylethylene carbonates with sulfinylanilines. This protocol is featured by readily available starting materials, and high enantio- and diastereoselectivity. In particular, an unusual effect of a non-chiral supporting ligand on the diastereoselectivity was observed. Possible reaction mechanisms and stereocontrol models were proposed.

Abstract:Although recent progress has been made in introducing an SCF3 functionality into a variety of molecules, enantioselective trifluoromethylthiolation remains challenging, especially the -trifluoromethylthiolation of carbonyl compounds. The present study describes a diastereo and enantioselective Cu-catalyzed tandem 1,4-addition/trifluoromethylthiolation of acyclic enones. The tandem reaction enables the asymmetric integration of the -SCF3 group to carbonyl compounds, establishing chiral tertiary -carbon centers and affording -SCF3--substituted carbonyl compounds in 50-92% yields with up to 20:1 dr and 96% ee.
