Abstract:Scientists have been attracted by polymersomes as versatile drug delivery systems since the last two decades. Polymersomes have the potential to be versatile drug delivery systems because of their tunable membrane formulations, stabilities in vivo, various physicochemical properties, controlled release mechanisms, targeting abilities, and capacities to encapsulate a wide range of drugs and other molecules. Asymmetrical polymersomes are nano- to micro-sized polymeric capsules with asymmetrical membranes, which means, they have different outer and inner coronas so that they can exhibit better endocytosis rate and endosomal escape ability than other polymeric systems with symmetrical membranes. Hence, asymmetrical polymersomes are highly promising as self-assembled nano-delivery systems in the future for in vivo therapeutics delivery and diagnostic imaging applications. In this review, we prepared a summary about recent research progresses of asymmetrical polymersomes in the following aspects: synthesis, preparation, applications in drug delivery and others.

Abstract:Polymersomes provide a highly promising platform for mimicking biological membranes as well as for controlled delivery of various pharmaceuticals and biopharmaceuticals. The recent advancement has made it possible to prepare small to giant polymersomes spanning from nano- to microscales, stimuli-sensitive polymersomes that swell or collapse in response to an external or internal stimulus, chimaeric polymersomes that contain distinct interior environments separated from the outside by an asymmetric membrane, porous polymersomes with tailored permeability, biomimetic and targeting polymersomes that selectively deliver drugs, proteins, and/or imaging probes to the sites of action. In this Perspective, we will give a brief introduction to polymersomes, highlight recent design and preparation of several sophisticated polymersomes, and discuss future challenges.

Abstract:Polymersomes are self-assembled spherical vesicles based on amphiphilic block copolymers. This review presents a summary of the achievements in the field of polymersome researches to date. Polymersomes have been applied as versatile drug carriers. Some polymersomes, which have well-known stimuli-responsibility, can release drugs in a controlled manner at the target site when they are given a specific stimulation such as pH, temperature, light, magnetic field, hydrogen bond actions, electrostatic force or ultrasound. The preparation methods of polymersomes are similar to that of liposomes, including the thin film rehydration technique, solvent method, direct dissolution, double emulsion in microfluidic device, and electroformation. In addition, biologically active ligands, such as antibodies, can be readily conjugated onto the exterior brush surface of polymersomes to target the vesicles or to provide a therapeutic response. Polymersomes offer superior advantages for future clinical therapeutic and diagnostic imaging applications.

Abstract:Polymersomes, composed of amphiphilic block copolymers, are self-assembled vesicles that have gained attention as potential drug delivery systems due to their good biocompatibility, stability, and versatility. Various experimental techniques have been employed to characterize the self-assembly behaviors and properties of polymersomes. However, they have limitations in revealing molecular details and underlying mechanisms. Computational modeling techniques have emerged as powerful tools to complement experimental studies and enabled researchers to examine drug delivery mechanisms at molecular resolution. This review aims to provide a comprehensive overview of the state of the art in the field of polymersome-based drug delivery systems, with an emphasis on insights gained from both experimental and computational studies. Specifically, we focus on polymersome morphologies, self-assembly kinetics, fusion and fission, behaviors in flow, as well as drug encapsulation and release mechanisms. Furthermore, we also identify existing challenges and limitations in this rapidly evolving field and suggest possible directions for future research.

Abstract:Polymersomes are artificial nanoparticles formed by the self-assembly process of amphiphilic block copolymers composed of hydrophobic and hydrophilic blocks. They can encapsulate hydrophilic molecules in the aqueous core and hydrophobic molecules within the membrane. The composition of block copolymers can be tuned, enabling control of characteristics and properties of formed polymersomes and, thus, their application in areas such as drug delivery, diagnostics, or bioimaging. The preparation methods of polymersomes can also impact their characteristics and the preservation of the encapsulated drugs. Many methods have been described, including direct hydration, thin film hydration, electroporation, the pH-switch method, solvent shift method, single and double emulsion method, flash nanoprecipitation, and microfluidic synthesis. Considering polymersome structure and composition, there are several types of polymersomes including theranostic polymersomes, polymersomes decorated with targeting ligands for selective delivery, stimuli-responsive polymersomes, or porous polymersomes with multiple promising applications. Due to the shortcomings related to the stability, efficacy, and safety of some therapeutics in the human body, polymersomes as drug delivery systems have been good candidates to improve the quality of therapies against a wide range of diseases, including cancer. Chemotherapy and immunotherapy can be improved by using polymersomes to deliver the drugs, protecting and directing them to the exact site of action. Moreover, this approach is also promising for targeted delivery of biologics since they represent a class of drugs with poor stability and high susceptibility to in vivo clearance. However, the lack of a well-defined regulatory plan for polymersome formulations has hampered their follow-up to clinical trials and subsequent market entry.

The assembled spherical micelle structures exhibit recognition properties defined by their DNA sequences and can be used to build higher-ordered structures through hybridization with materials that possess complementary DNA. This work is an important step toward creating DNA modified structures that can act as phase transfer agents and carriers of nanovesicles.

Abstract:Polymersomes with inhomogeneous membranes in composition and structure have generated widespread interest for the preparation of functionalized nanocarriers. We propose a simple but versatile strategy to manipulate inhomogeneous subdomains on polymersome membranes by the co-assembly of block copolymer blends with varied molecular architectures and chemistries. Both binary and ternary copolymer blends are considered to construct polymersomes, and the subdomains of the membranes are formed by controlling the difference in the flexibility and rigidity of different blocks. This difference contributes to the formation of disk-like domains (by rigid blocks) and soft domains (by flexible blocks) on the membrane. An interesting effect of this structure is that in response to external stimuli, the soft membrane domain becomes worm-like or porous to "open" the polymersome for matter exchange, while the rigid domain stays undecomposed and acts like an anchor binding all flexible copolymers. Once the external stimuli disappear, all flexible copolymers can be pulled back to restore the original polymersome morphology (i.e., "close" the polymersome). The specific morphological reversibility of hybrid polymersomes holds great potential for practical applications where changeable membrane permeability or shape under environmental stimuli is highly needed.

Abstract:Due to a strong retardation effect of o-nitrobenzyl ester on polymerization, it is still a great challenge to prepare amphiphilic block copolymers for polymersomes with a o-nitrobenzyl ester-based hydrophobic block. Herein, we present one such solution to prepare amphiphilic block copolymers with pure poly (o-nitrobenzyl acrylate) (PNBA) as the hydrophobic block and poly (N,N’-dimethylacrylamide) (PDMA) as the hydrophilic block using bulk reversible addition-fragmentation chain transfer (RAFT) polymerization of o-nitrobenzyl acrylate using a PDMA macro-RAFT agent. The developed amphiphilic block copolymers have a suitable hydrophobic/hydrophilic ratio and can self-assemble into photoresponsive polymersomes for co-loading hydrophobic and hydrophilic cargos into hydrophobic membranes and aqueous compartments of the polymersomes. The polymersomes demonstrate a clear photo-responsive characteristic. Exposure to light irradiation at 365 nm can trigger a photocleavage reaction of o-nitrobenzyl groups, which results in dissociation of the polymersomes with simultaneous co-release of hydrophilic and hydrophobic cargoes on demand. Therefore, these polymersomes have great potential as a smart drug delivery nanocarrier for controllable loading and releasing of hydrophilic and hydrophobic drug molecules. Moreover, taking advantage of the conditional releasing of hydrophilic and hydrophobic drugs, the drug delivery system has potential use in medical applications such as cancer therapy.

Abstract:Polymersomes are a class of artificial liposomes, assembled from amphiphilic synthetic block copolymers, holding great promise toward applications in nanomedicine. The diversity in polymersome morphological shapes and, in particular, the precise control of these shapes, which is an important aspect in drug delivery studies, remains a great challenge. This is due to a lack of general methodologies that can be applied and the inability to capture the morphologies at the nanometer scale. Here, we present a methodology that can accurately control the shape of polymersomes via the addition of polyethylene glycol (PEG) under nonequilibrium conditions. Various shapes including spheres, ellipsoids, tubes, discs, stomatocytes, nests, stomatocyte-in-stomatocytes, disc-in-discs, and large compound vesicles (LCVs) can be uniformly captured by adjusting the water content and the PEG concentration. Moreover, these shapes undergo nonequilibrium changes in time, which is reflected in their phase diagram changes. This research provides a universal tool to fabricate all shapes of polymersomes by controlling three variables: water content, PEG concentration, and time. The use of the biofriendly polymer PEG enables the application of this methodology in the field of nanomedicine.

Abstract:Polymersomes, the structural analogues of liposomes, are hollow structures enclosed by a bilayer membrane made from amphiphilic copolymers. Polymersomes have been proposed to mimic the structure and properties of cellular membranes and viral capsids. Excellent robustness and stability, chemical versatility for tunable membrane properties and surface functionalization make polymersomes attractive candidates for drug delivery, diagnostic imaging, nanoreactor vessels, and artificial organelles. In further biomimetic strategies, stimuli-responsive polymersomes that can recognize various external physical or internal biological environmental stimuli and conduct "on demand" release in dose-, spatial-, and temporal-controlled fashions have been widely developed. This Perspective focuses on recent advances in stimuli-responsive polymersomes and their potential biomedical applications. Representative examples of each stimulus, the advantages and limitations of different strategies, and the future opportunities and challenges are discussed.

Abstract:Hollow block copolymer particles called polymer vesicles (polymersomes) serve as versatile containers for compartmentalization in synthetic biology and drug delivery. Recently, there has been growing interest in using polymersomes as colloidal building blocks for creating higher-order clustered structures. Most reports thus far rely on the use of DNA base-pairing interactions to "glue" polymersomes with other colloidal components. In this study, we present two alternative electrostatically driven approaches to assemble polymersomes and model colloids (micelles) into hybrid clusters. The first approach uses pH to manipulate electrostatic interactions and effectively control the clustering extent of micellar subunits on polymersomes, while the second approach relies on the hydrolysis of an acid trigger, glucono delta-lactone (GDL), to introduce temporal control over clustering. We envisage our approaches and structure reported herein will help inspire the creation of new prospects for materials science and biomedical applications.

Abstract:Encapsulating delicate biomolecules into submicron-sized polymer particulate systems with preserved native conformation and sufficient loading efficiency is of great challenge. To address this issue, we developed a unique polymersome which differs from reported polymersome in that its bilayer membrane was formed of two different amphiphilic diblock copolymers in an “asymmetric” way. By adding two diblock copolymers, poly (ethylene glycol)-block-poly (ε-caprolactone) (PEG-PCL) and dextran-block-poly (ε-caprolactone) (DEX-PCL), into a so-called dextran-in-PEG aqueous two-phase system, DEX-PCL formed the inner leaflet around the dispersed dextran phase and PEG-PCL formed the outer leaflet with the PEG block facing the PEG continuous phase. We name this unique assembly process as “phase-guided assembly”. Polymersomes of asymmetric bilayer membrane possess a series of advantages over “symmetric” polymer bilayer vesicles previously reported. The asymmetric bilayer created a different chemical environment of the interior to which proteins were encapsulated highly efficiently (up to 90%) by thermodynamically favored partition. Probably due to the thermodynamic preference, erythropoietin (EPO) encapsulated in this system showed a well-preserved bioactivity in cell proliferation assay. The core of the polymersomes may be cross-linked to enhance their mechanical strength. Phase-guided assembly system and asymmetric bilayer polymersomes demonstrated in this study may serve the high demands for delivering nucleotide and protein medicines and other biological applications.
Combining Nanoparticle Shape Modulation and Polymersome Technology in Drug Delivery

Abstract:This paper highlights the potential benefits of using self-assembled polymeric nanoparticles of various shapes to enhance drug uptake. First, we highlight the growth and development of the polymersome, using a liposome as a blueprint for amphiphilic codelivery. Then, we focus on the advantages of nanoparticle elongation, drawing from the field of solid nanoparticles, as opposed to self-assembled vesicles which have not yet been extensively explored in shape-modulated drug delivery applications. Notably, regardless of the material used in the solid nanoparticle systems, more elongated shapes lead to greater cellular uptake, decreased interaction with the reticuloendothelial system macrophages, and increased circulation times. Finally, we highlight the methods currently being developed to modulate polymersome shape, thus providing a drug delivery system with the benefits derived from amphiphilicity and elongated structures. Current methods employed to modulate polymersome shape involve osmotic pressure gradients, solvent switching, and the use of cross-linking agents. Although these methods are successful in modulating polymersome shapes and the benefits of elongated nanoparticles in therapeutic targeting are clear, these methods have not yet been explored for applications in drug delivery.
Challenges and future directions for next-generation biomedical polymersomes

Abstract:As a major transport system in cells, naturally occurring vesicles play an important role to ensure that the right cargo is shipped to the right destination at the right time. Similarly, artificial vesicles, such as polymersomes with similar structures and functions, have been developed for various biological applications due to the advantages of polymersomes such as excellent membrane and colloidal stability, high loading efficiency of hydrophobic drugs, low membrane permeability, high functional modifiability, and long circulation time. However, relatively complicated synthesis and preparation methods, unpredictable biocompatibility and limited functions and delivery routes, have largely hindered the clinic translation of polymersomes. In this review, we propose five key scientific questions for next-generation polymersomes, in the aspects of synthesis of building blocks, self-assembly techniques, and potential biomedical applications, to emphasize on the future challenges and directions, such as simplifying the design and synthetic process, streamlining and standardizing the self-assembly procedure, and broadening the horizon of potential applications.

Abstract:Drug delivery systems made based on nanotechnology represent a novel drug carrier system that can change the face of therapeutics and diagnosis. Among all the available nanoforms polymersomes have wider applications due to their unique characteristic features like drug loading carriers for both hydrophilic and hydrophobic drugs, excellent biocompatibility, biodegradability, longer shelf life in the bloodstream and ease of surface modification by ligands. Polymersomes are defined as the artificial vesicles which are enclosed in a central aqueous cavity which are composed of self-assembly with a block of amphiphilic copolymer. Various techniques like film rehydration, direct hydration, nanoprecipitation, double emulsion technique and microfluidic technique are mostly used in formulating polymersomes employing different polymers like PEO-b-PLA, poly (fumaric/sebacic acid), poly(N-isopropylacrylamide) (PNIPAM), poly (dimethylsiloxane) (PDMS), and poly(butadiene) (PBD), PTMC-b-PGA (poly (dimethyl aminoethyl methacrylate)-b-poly(l-glutamic acid)) etc. Polymersomes have been extensively considered for the conveyance of therapeutic agents for diagnosis, targeting, treatment of cancer, diabetes etc. This review focuses on a comprehensive description of polymersomes with suitable case studies under the following headings: chemical structure, polymers used in the formulation, formulation methods, characterization methods and their application in the therapeutic, and medicinal filed.

Abstract:Polymersomes, also called polymeric vesicles, are self-assembled by amphiphilic copolymers. Due to their unique characters, polymersomes are attracting more and more interest as an important class of vehicles for nanopharmaceuticals. In this chapter, various methods to prepare and characterize polymersomes are introduced systematically with several applicable examples. In addition, the advantages and disadvantages of each method were compared and analyzed with the aim to help readers choose the appropriate method in the process of experiments. Although some methods we introduced here are effective in preparing and characterizing polymersomes, the remaining challenge in this filed is to develop new tools. The reason is that polymersome is a kind of complex nanostructure, and some minor factors can affect the formation of polymersome. Meanwhile, more advanced technology should be developed to precisely determine the structure of some complex polymersomes such as multilayer polymersomes.
Polymersomes from Dual Responsive Block Copolymers: Drug Encapsulation by Heating and Acid-Triggered Release

Abstract:A series of well-defined thermoresponsive diblock copolymers (PEO45-b-PtNEAn, n=22, 44, 63, 91, 172) were prepared by the atom transfer radical polymerization of trans-N-(2-ethoxy-1,3-dioxan-5-yl) acrylamide (tNEA) using a poly(ethylene oxide) (PEO45) macroinitiator. All copolymers are water-soluble at low temperature, but upon quickly heating to 37 °C, laser light scattering (LLS) and transmission electron microscopy (TEM) characterizations indicate that these copolymers self-assemble into aggregates with different morphologies depending on the chain length of PtNEA and the polymer concentration; the morphologies gradually evolved from spherical solid nanoparticles to a polymersome as the degree of polymerization ("n") of PtNEA block increased from 22 to 172, with the formation of clusters with rod-like structure at the intermediate PtNEA length. Both the spherical nanoparticle and the polymersome are stable at physiological pH but susceptible to the mildly acidic medium. Acid-triggered hydrolysis behaviors of the aggregates were investigated by LLS, Nile red fluorescence, TEM, and (1)H NMR spectroscopy. The results revealed that the spherical nanoparticles formed from PEO45-b-PtNEA44 dissociated faster than the polymersomes of PEO45-b-PtNEA172, and both aggregates showed an enhanced hydrolysis under acidic conditions. Both the spherical nanoparticle and polymersome are able to efficiently load the hydrophobic doxorubicin (DOX), and water-soluble fluorescein isothiocyanate-lysozyme (FITC-Lys) can be conveniently encapsulated into the polymersome without using any organic solvent. Moreover, FITC-Lys and DOX could be coloaded in the polymersome. The drugs loaded either in the polymersome or in the spherical nanoparticle could be released by acid triggering. Finally, the DOX-loaded assemblies display concentration-dependent cytotoxicity to HepG2 cells, while the copolymers themselves are nontoxic.

Abstract:Polymer vesicles have an important application prospect in biomedical and other fields due to their unique lumen-membrane-corona structure. In this feature article, we highlighted recent advance in polymer vesicles for biomedical applications, including new mechanism and methods for self-assembly, and new biomedical applications. The former includes two principles of acid-induced adsorption and affinity-enhanced attraction for preparing vesicles with ultrahigh biomacromolecular loading efficiency, "micelle to vesicle transition" strategy for preparing asymmetric vesicles, fusion-induced particle assembly to build hierarchical vesicles, and ring-opening polymerization of N-carboxyanhydride-induced self-assembly to fabricate vesicles on a large scale; the latter includes vesicles for blood glucose control, vesicles to treat diabetic ulcers, antibacterial vesicles, anticancer vesicles, bone-targeting vesicles to treat osteoporosis and renoprotective angiographic vesicles. Abstract:In the past decade, polymersomes (also referred to as polymeric vesicles) have attracted rapidly growing interest based on their intriguing aggregation phenomena, cell and virus-mimicking dimensions and functions, as well as tremendous potential applications in medicine, pharmacy, and biotechnology. Unlike liposomes self-assembled from low molecular weight lipids, polymersomes are in general prepared from macromolecular amphiphiles of various architectures including amphiphilic diblock, triblock, graft and dendritic copolymers. Polymersomes exhibit very unique features highlighted with high stability, tunable membrane properties, versatility, and capacity of transporting hydrophilic as well as hydrophobic species such as anticancer drugs, genes, proteins, and diagnostic probes. Recently, much effort has been directed to the development of intelligent polymersomes; that respond to internal or external stimuli, in particular, pH, temperature, redox potential, light, magnetic field, and ultrasound, either reversibly or nonreversibly. Stimuli-sensitive polymersomes have emerged as novel programmable delivery systems in which the release of the encapsulated contents can be readily modulated by the stimulus. The stimuliresponsive release may result in significantly enhanced therapeutic efficacy and minimized possible side effects. It is also feasible to form and disassemble polymersomes in water simply by applying an appropriate stimulus. In this article, recent advances in stimuli-sensitive polymersomes have been reviewed, and perspectives on future developments have been discussed.

Abstract:The current challenge for polymersomes is to prepare amphiphilic block polymers with not only well-defined molecular weight, but also precisely high-order multiblock structure in terms of the distribution of monomeric units along the chain. Here, we describe a synthesis of temperature-responsive polymersomes with precisely defined membrane structures using high-precision amphiphilic multiblock polymer via aqueous single electron transfer living radical polymerization (SET-LRP) in which poly(ethylene glycol) (PEG) macromolecules are used as initiators. We develop a one-pot, rapid and multistep sequential polymerization process with yields >99% within 30 min for each block, giving access to a wide range of high-precision multiblock polymers with very narrow molecular weight distributions (Ð ≤ 1.14). Synthesized multiblock polymers are used to form nano-sized polymersomes which are highly promising as smart carriers for high loading and triggered release of biopharmaceutics such as pharmaceutical proteins and peptides. This synthesis approach is environmentally friendly, fully translation and thus represents a significant advance in the design and synthesis of a new generation of polymer nanomaterials with precisely defined structures, which is highly attractive for applications in nanotechnology.
