Abstract:Overall enamel is the hard tissue overlying teeth that is vulnerable to caries, congenital defects, and damage due to trauma. Not only is enamel incapable of self-repair in most species, but it is also subject to attrition. Besides the use of artificial materials to restore enamel, enamel regeneration is a promising approach to repair enamel damage. In order to comprehend the progression and challenges in tissue-engineered enamel, this article elaborates alternative stem cells potential for enamel secretion and expounds fined strategies for enamel regeneration in bioengineered teeth. Consequently, more and more cell types have been induced to differentiate into ameloblasts and to secrete enamel, and an increasing number of reports have emerged to provide various potential approaches to induce cells to secrete enamel based on recombination experiments, artificial bioactive nano-materials, or gene manipulation. Accordingly, it is expected to further project more optimal conditions for enamel formation in bioengineering based on a more thorough knowledge of reciprocal epithelial-mesenchymal interactions, by which the procedures of enamel regeneration are able to be practically recapitulated and widely spread for the potential clinical value of enamel repair.

Abstract:Tooth enamel, the outer shell of the tooth measuring several millimeters, is the hardest tissue of human body to masticate food and protect the inner dentin. The all-around mechanical properties and multiscale-oriented aligned nanoprism structure of tooth enamel have attracted wide interests from both clinical applications and materials scientists. Here, we focus on analyzing the structure-property relationship of tooth enamel and summarize the tooth-enamel analogs based on detailed mimicking of their hierarchy. The underlying principles relating to the formation process and hierarchical structure to excellent mechanical properties in enamel are first introduced to explain the design concept of enamel analogs. Based on this, the synthesis of enamel analogs by biomimetic remineralization and physical-chemical processes-assisted assembly are discussed. Furthermore, the promising applications of these enamel analogs are exhibited. Finally, a brief summary of present enamel analogs and perspective for the next generation of enamel analogs are proposed.

Abstract:This study presents a biomimetic method for remineralizing dental hard tissues using an amyloid‐like protein matrix composite with amorphous calcium phosphate. This approach mimics natural mineralization processes, enhancing the regeneration and mechanical strength of demineralized enamel and dentine, providing a promising material for the treatment of early enamel lesions and dentine hypersensitivity. Numerous remineralizing coatings aim to prevent or treat early enamel lesions and occlude exposed dentinal tubules (DTs). Nevertheless, the pace of remineralization is inadequate, and the mechanical robustness of the newly established mineral layer fails to match the inherent strength. In this study, a biomimetic mineralization strategy aimed at replicating key events in biological mineralization, specifically focusing on the organic–inorganic composite matrix, is proposed. The material utilizes Tris(2‐carboxyethyl)phosphine (TCEP), which serves a dual role: stabilized amorphous calcium phosphate (ACP) (ACP@TCEP) nanoparticles as its inorganic component, and catalyzing the cleavage of intramolecular disulfide bonds in poly(ethylene glycol) (PEG) grafted lysozyme (lyso‐PEG) to facilitate the formation of an amyloid‐like protein matrix composite with ACP (ACP@lyso‐PEG nanocomplexes). ACP@lyso‐PEG nanocomplexes can rapidly and efficiently form an enamel‐like remineralization layer on the surface of damaged dental hard tissue, reaching ≈4.205 μm thickness after 3 days of acid‐etched enamel. Furthermore, achieving a depth of DTs occlusion exceeding 60 μm after 5 days, using a simple immersion process. The resulting mineralized layer exhibits mechanical strength comparable to natural teeth. This study introduces a conceptual biomimetic mineralization strategy for effective enamel repair or DTs occlusion in clinical practices, and offers potential insights into the mechanisms of biomineral formation.

Abstract:Tooth enamel, which is a biological tissue mainly composed of well-aligned hydroxyapatite nanocrystals and an interlaced protein matrix, has remarkable mechanical and aesthetic behaviors. Nevertheless, it is challenging to regenerate enamel naturally, and potential pulp involvement and tooth loss may occur. As the hardest biogenic composite material, enamel has long been regarded as a promising load-bearing material. Thus, understanding the enamel formation process and enamel structural motif mechanisms is important for the design and engineering of high-performance biomimetic composites with high strength and physical resilience. Extensive studies have been conducted on mimicking the microstructure and mechanical properties of tooth enamel, and various enamel-like material synthesis protocols have been developed. In light of the engineering fabrication of enamel-like materials, this review focuses on recent progress in synthetic strategies for enamel-mimetic materials and provides a discussion of the potential applications of these materials.

Abstract:Tooth enamel, renowned for its high stiffness, hardness, and viscoelasticity, is an ideal model for designing biomimetic materials, but accurate replication of complex hierarchical organization of high-performance biomaterials in scalable abiological composites is challenging. We engineered an enamel analog with the essential hierarchical structure at multiple scales through assembly of amorphous intergranular phase (AIP)–coated hydroxyapatite nanowires intertwined with polyvinyl alcohol. The nanocomposite simultaneously exhibited high stiffness, hardness, strength, viscoelasticity, and toughness, exceeding the properties of enamel and previously manufactured bulk enamel-inspired materials. The presence of AIP, polymer confinement, and strong interfacial adhesion are all needed for high mechanical performance. This multiscale design is suitable for scalable production of high-performance materials.

Abstract:Background: This study focused on the aging mechanism and degradation of mechanical and structural features of elastodontic appliances (EA) under artificial and intraoral aging to achieve oral myofunctional therapy with particular removable silicone elastomer devices. Materials and methods: EAs artificially aged in saliva with different pH values were investigated through cyclic compression testing along with characterization techniques (Scanning electron microscopy, X-ray diffraction, and Fourier transform infrared spectroscopy), and characterization analysis was also performed on clinically retrieved EAs. Results: Artificial aging was found to have minimal effect on the structural properties of EAs, and intraorally aged samples showed perceptible micro-morphology. The Mullins index and peak stress decreased (P<0.01), while the compression set increased with prolonged aging time. Samples in alkaline saliva showed the largest Mullins effect (P<0.05). Conclusions: The aging mechanism of the elastomer was found to be the crosslinking of main chains and scission of side chains. The presence of OH- enhanced the rupture degree of side bonds. The decline in viscoelastic properties was shown to be more severe with longer service durations. Clinical relevance: Research on how the salivary environment and pH affect the aging characteristics of EAs is vital for guiding clinical applications and future modifications to extend their clinical lifetime.

Abstract:Tooth enamel, and especially the outer tooth enamel, is a load-resistant shell that benefits mastication but is easily damaged, driving the need for enamel-restorative materials with comparable properties to restore the mastication function and protect the teeth. Synthesizing an enamel analog that mimics the components and hierarchical structure of natural tooth enamel is a promising way to achieve these comparable mechanical properties, but it is still challenging to realize. Herein, we fabricate a hierarchical enamel analog with comparable stiffness, hardness, and viscoelasticity as natural enamel by incorporating three hierarchies of outer tooth enamel based on hierarchical assembly of enamel-like hydroxyapatite hybrid nanowires with polyvinyl alcohol as a matrix. This enamel analog possesses enamel-similar inorganic components and a nanowire-microbundle-macroarray hierarchical structure. It exhibits toughness of 19.80 MPa m1/2, which is 3.4 times higher than natural tooth enamel, giving it long-term fatigue durability. This hierarchical design is promising for scalable production of enamel-restorative materials and for optimizing the mechanical performance of engineering composites.
Anisotropic Demineralization and Oriented Assembly of Hydroxyapatite Crystals in Enamel: Smart Structures of Biominerals

Abstract:It is interesting to note that the demineralization of natural enamel does not happen as readily as that of the synthesized hydroxyapatite (HAP), although they share a similar chemical composition. We suggest that the hierarchical structure of enamel is an important factor in the preservation of the natural material against dissolution. The anisotropic demineralization of HAP is revealed experimentally, and this phenomenon is understood by the different interfacial structures of HAP-water at the atomic level. It is found that HAP {001} facets can be more resistant against dissolution than {100} under acidic conditions. Although {100} is the largest surface of the typical HAP crystal, it is {001}, the smallest habit face, that is chosen by the living organisms to build the outer surface of enamel by an oriented assembly of the rodlike crystals. We reveal that such a biological construction can confer on enamel protections against erosion, since {001} is relatively dissolution-insensitive. Thus, the spontaneous dissolution of enamel surface can be retarded in biological milieu by such a smart construction. The current study demonstrates the importance of hierarchical structures in the functional biomaterials.

Abstract:Biological materials are well worth learning for our own synthetic materials. It' s of great interest that the growth rate of teeth is different in species. The growth rate of human teeth is slow, but faster for rat and shark. The microstructure and composition difference may have a great effect on the growth rate. In this study, the microstructure and composition from human teeth, rat teeth and shark teeth were well characterized by scanning electron microscopy and X-ray diffraction. It is demonstrated that the size of individual apatite crystal and the growth direction of apatites in those enamel or enameloid display different types. The diameter of shark enameloid is the biggest. Three growth directions can be observed in shark enameloid but only two in human and rat. The main inorganic composition of the three teeth is also different. It is hydroxyapatite (HAP) for human and rat teeth, while fluorapatite (FAP) for shark. The XRD patterns also indicate that the assemble degree of human teeth is the most sophisticated. The results may give useful insight into new materials synthesis technology.
Epigenetic alterations in aging tooth and the reprogramming potential

Abstract:Tooth compartments and associated supportive tissues exhibit significant alterations during aging, leading to their impaired functioning. Aging not only affects the structure and function of dental tissue but also reduces its capacity to maintain physiological homeostasis and the healing process. Decreased cementocyte viability; diminished regenerative potential of stem cells residing in the pulp, alveolar bone and periodontal ligament; and impaired osteogenic and odontogenic differentiation capacity of progenitor cells are among the cellular impacts associated with oral aging. Various physiological and pathological phenomena are regulated by the epigenome, and hence, changes in epigenetic markers due to external stimuli have been reported in aging oral tissues and are considered a possible molecular mechanism underlying dental aging. The role of nutri-epigenetics in aging has emerged as an attractive research area. Thus far, various nutrients and bioactive compounds have been identified to have a modulatory effect on the epigenetic machinery, showing a promising response in dental aging. The human microbiota is another key player in aging and can be a target for anti-aging interventions in dental tissue. Considering the reversible characteristics of epigenetic markers and the potential for environmental factors to manipulate the epigenome, to minimize the deteriorative effects of aging, it is important to evaluate the linkage between external stimuli and their effects in terms of age-related epigenetic modifications.

Abstract:Dental Enamel is the hardest mineralized tissue in the human body which is comprised of nanorod-like hydroxyapatite crystals arranged into a highly organized micro-architectural unit called an enamel prism. In this paper the direct growth of human enamel-like structures on human tooth using fluorapatite/phosphoric acid pastes is explored. SEM images show that the newly formed calcium phosphate crystals can be self-assembled into a similar ordered microstructure as those seen in human enamel. The mechanism of how these structures form is discussed. This work demonstrates the potential of applying nanotechnology to regenerate dental enamel clinically without cells.

Abstract:Human dentin is a highly organized dental tissue displaying a complex microarchitecture consisting of micrometer-sized tubules encased in a mineralized type-I collagen matrix. As such, it serves as an important substrate for the adhesion of microbial colonizers and oral biofilm formation in the context of dental caries disease, including root caries in the elderly. Despite this issue, there remains a current lack of effective biomimetic in vitro dentin models that facilitate the study of oral microbial adhesion by considering the surface architecture at the micro- and nanoscales. Therefore, the aim of this study was to develop a novel in vitro microfabricated biomimetic dentin surface that simulates the complex surface microarchitecture of exposed dentin. For this, a combination of soft lithography microfabrication and biomaterial science approaches were employed to construct a micropitted PDMS substrate functionalized with mineralized type-I collagen. These dentin analogs were subsequently glycated with methylglyoxal (MGO) to simulate dentin matrix aging in vitro and analyzed utilizing an interdisciplinary array of techniques including atomic force microscopy (AFM), elemental analysis, and electron microscopy. AFM force-mapping demonstrated that the nanomechanical properties of the biomimetic constructs were within the expected biological parameters, and that mineralization was mostly predominated by hydroxyapatite deposition. Finally, dual-species biofilms of Streptococcus mutans and Candida albicans were grown and characterized on the biofunctionalized PDMS microchips, demonstrating biofilm-specific morphologic characteristics and confirming the suitability of this model for the study of early biofilm formation under controlled conditions. Overall, we expect that this novel biomimetic dentin model could serve as an in vitro platform to study oral biofilm formation or dentin-biomaterial bonding in the laboratory without the need for animal or human tooth samples in the future.

Abstract:Dental enamel biomimetic mineralization is a process to form the enamel-like mineral structures, which possess unique microstructure and exceptional physic-chemical properties, by mimicking the mechanism of natural enamel formation and biomineralization. Varieties of techniques such as molecular mimetic synthesis and molecular self-assembling were used to accomplish the microenvironment and molecular conditions similar to that of natural tooth enamel within human body. Early remineralization and biomineralization is the future of restoration for enamel defect, research on such products have huge potential in clinical applications, with speedy advancement in recent two decades. This review summarizes the major advances in researches on enamel biomimetic mineralization in recent years.

Abstract:Objective To explore the wear mechanisms of natural enamel and dentine. Methods Samples of human teeth in this study had been taken from teenagers undergoing orthodontics treatment,with the teeth freshly extracted and wear-free.A variety of mechanical factors,such as friction coefficients,wear scar width,element concentrations and wear surface evolution were considered under dry/artificial saliva lubrication.The wear scars were characterized by scanning electron microscopy(SEM).Energy dispersion spectrometry(EDS) was used to examine the element concentrations. Results The statistical differences were observed in wear width and hardness of samples(P0.05).SEM results showed that abrasive particles and plough were main features,little crack could be seen in enamel.EDS results showed that the element concentration of Fe was obviously found on the samples. Conclusions Enamel and dentine displayed good wear resistances.The main wear patterns are abrasive,adhesive and fatigue wear.The resistance ability of enamel against wear is better than that of dentine,with obvious cracking in the wear surface of enamel.The dentine wear is more humid and stable.The artificial saliva plays an important role in the wear behavior of enamel and dentine.
A Biomimetic Tooth Replicate That Is Hard, Damage-Tolerant, and Self-Healable

Abstract:Hard materials typically lack the mechanism of energy dissipation and cannot self-heal. Nature addresses this challenge by creating multiscale interfaces between high-contrast materials, namely minerals and biopolymers. Inspired by the enamel-dentin junction in nature, an enamel-like crown consisting of beta- FeOOH nanocolumns is interdigitated with a flexible self-healing layer. The iron oxide top layer has exceptionally high modulus and hardness, which is more resistant to cyclic deformation than the bottom layer. The latter however provides an additional pathway for viscous and plastic energy dissipation and enables self-healing by allowing upward polymer diffusion to seal the damage. Picture-frame crack patterns were observed under large loading conditions using microindentation, which localizes the damage at the indentation site. The bending properties can be optimized by varying the thickness of the bottom layer, and the crack induced by bending can be effectively captured at the interface without any delamination. The biomimetic tooth replicate is highly adhesive to a ceramic surface and shows an obvious inhibition effect against Streptococcus mutans, a significant contributor to tooth decay. Combined with ultralow thermal diffusivity, this has great potential as dental material. Learning from nature, our work thus provides a powerful pathway to broadening the scope of synthetic materials for dental replicates.
Biomimetic remineralization as a progressive dehydration mechanism of collagen matrices – Implications in the aging of resin–dentin bonds

Abstract:Biomineralization is a dehydration process in which water from the intrafibrillar compartments of collagen fibrils are progressively replaced by apatites. As water is an important element that induces a lack of durability of resin–dentin bonds, this study has examined the use of a biomimetic remineralization strategy as a progressive dehydration mechanism to preserve joint integrity and maintain adhesive strength after ageing. Human dentin surfaces were bonded with dentin adhesives, restored with resin composites and sectioned into sticks containing the adhesive joint. Experimental specimens were aged in a biomimetic analog-containing remineralizing medium and control specimens in simulated body fluid for up to 12 months. Specimens retrieved after the designated periods were examined by transmission electron microscopy for the presence of water-rich regions using a silver tracer and for collagen degradation within the adhesive joints. Tensile testing was performed to determine the potential loss of bond integrity after ageing. Control specimens exhibited severe collagen degradation within the adhesive joint after ageing. Remineralized specimens exhibited progressive dehydration, as manifested by silver tracer reduction and partial remineralization of water-filled microchannels within the adhesive joint, as well as intrafibrillar remineralization of collagen fibrils that were demineralized initially as part of the bonding procedure. Biomimetic remineralization as a progressive dehydration mechanism of water-rich, resin-sparse collagen matrices enables these adhesive joints to resist degradation over a 12-month ageing period, as verified by the conservation of their tensile bond strength. The ability of the proof of concept biomimetic remineralization strategy to prevent bond degradation warrants further development of clinically relevant delivery systems.

Abstract:Tooth enamel is a distinctive nanocomposite with a highly organized hierarchical structure made of nanometer- and micrometer-scale building blocks. This structure has an excellent mechanical function that can last for decades thanks to an effective but underexploited interfacial chemical bonding between the building blocks. In this study, the nanomechanical system test (NST), scanning electron microscope (SEM), X-ray diffraction (XRD, including powder XRD or PXRD, small angle XRD or SAXRD, and grazing incidence small angle XRD or GISAXRD), and atomic force microscope (AFM) have been employed to analyze the water-mediated bonding on the enamel surface. Via the cycling between hydration, dehydration, and rehydration treatments, a reversible change in the interfacial distance (i.e., d-space in the XRD pattern) between hydroxyapatite (HAP) nanocrystallites have been found switchable between the embrittling and toughening on the enamel surface. From the hydrated to the dehydrated conditions, an energy dissipation to deform a unit volume (1 μm3) of biocomposite on the enamel surface and subsurface has decreased by 20%. This finding can help quantify and predict biomineral-surface properties in all humidity and develop new methods to protect tooth enamel of "dry-mouth" patients.

Abstract:Tooth defect due to caries, trauma, or acid corrosion are common in mankind. Ceramics, metal and resin were used to repaired tooth defect in the past hundred years, but they cannot instead enamel and dentin in depth in clinic usage for the difference in structure and element. So the formation of organized nanocrystals that resemble enamel is crucial for successful enamel remineralization. Now synthesizing a mimicking structure of human enamel using acellular methods has attracted much interest from research groups who have tried using recombinant enamel making proteins like amelogenin, surfactants, to mimic the biomineralization process to restore the enamel layer. Since amelogenin can be used in the assembly of functional nanostructures, we hypothesis that rationally designed β-sheet-forming peptides that spontaneously form three-dimensional fibrillar scaffolds in response to specific environmental triggers may potentially be used in inducing tooth-like hydroxyapatite crystal ex vivo which important to treatment/prevention of dental caries, via bioactive surface groups.

Abstract:The oral cavity is an environment with diverse bacteria; thus, antibacterial materials are crucial for treating and preventing dental diseases. There is a high demand for materials with an enamel-like architecture because of the high failure rate of dental restorations, due to the physical differences between dental materials and enamel. However, recreating the distinctive apatite composition and hierarchical architecture of enamel is challenging. The aim of this study was to synthesize a novel material with an enamel-like structure and antibacterial ability. We established a non-cell biomimetic method of evaporation-based bottom-up self-assembly combined with a layer-by-layer technique and introduced an antibacterial agent (graphene oxide) to fabricate a biofunctional material with an enamel-like architecture and antibacterial ability. Specifically, enamel-like graphene oxide-hydroxyapatite crystals, formed on a customized mineralization template, were assembled into an enamel-like prismatic structure with a highly organized orientation preferentially along the c-axis through evaporation-based bottom-up self-assembly. With the aid of layer-by-layer absorption, we then fabricated a bulk macroscopic multilayered biofunctional material with a hierarchical enamel-like architecture. This enamel-inspired biomaterial could effectively resolve the problem in dental restoration and brings new prospects for the synthesis of other enamel-inspired biomaterials.
