Abstract:Time-resolved fluorescence lifetime imaging (FLIM) in the near-infrared region of 900–1700 nm not only allows a deep tissue penetration depth but also offers the unique benefit of the quantitative visualization of molecular events in vivo and is independent of local luminescence intensity and fluorophore concentration. Herein, we report the design of a wide-range pH sensitivemolecular probe based on Yb porphyrinate. The Yb probe shows increasing NIR emission and lifetime with pKa values of ca. 6.6 from pH 9.0 and 5.0 and also displays an elongated lifetime from ca. 135 to 170 ms at lower pH values (5.0–1.0) due to aggregation and reduced exposure to water at low pH values. Importantly, the probe is able tomonitor a wide range of in vivo gastrointestinal pH values in mice models and the potential applications in imaging-guided gastrointestinal diagnostics and therapeutics were revealed. This study shows that lifetime contrast is important for preclinical imaging; lanthanide complexes could be successfully used in the design of stimuli-responsive NIR s probes for advanced in vivo imaging.
Development of an Au Nanoclusters Based Activatable Nanoprobe for NIR-II Fluorescence Imaging of Gastric Acid

Abstract:Gastric acid is an important functional substance secreted by the stomach of the living organisms, reflecting the gastric physiological condition. The sensing of gastric acid in vivo is of great significance for evaluation of gastric function, diagnosis and treatment of gastric diseases and maintenance of organism health but remains challenging due to the harsh acid and digestive environment of stomach. This study developed an activatable nanoprobe based on Au nanoclusters (Au NCs) for sensitive and real-time noninvasive near-infrared II (NIR-II) fluorescence imaging detection of gastric acid in vivo for the first time. The Au NCs were encapsulated by polydopamine to have enhanced NIR-II luminescence and high stability and combined with methylene blue to possess the pH responsiveness for gastric acid imaging. The developed nanoprobe could not only monitor gastric acid secretion in vivo but also imaging the changes of gastric acid caused by feeding, acid-inhibition drugs and gastric ulcer disease. This study provides a promising avenue for the improvement of the application performance of Au NCs and imaging analysis of gastric acid and related gastric diseases.

Abstract:Near-infrared (NIR, 700-1700 nm) bioimaging has gained considerable attention in recent years owing to its high imaging resolution and increasing penetration depth of tissues. NIR emissive lanthanide(iii) complexes have emerged as alluring candidates for the design of NIR molecular probes in clinical imaging and diagnosis because of their desirable temporal-spatial resolution, negligible photobleaching effect, long lifetime and low biotoxicity. In this mini-review, we summarize our efforts in the molecular design of NIR luminescent lanthanide porphyrinoids and their biological applications in bioimaging and further clinical diagnosis and treatment. Remarkably, beta-fluorinated porphyrin Yb3+ probes exhibit high luminescence (5-13% quantum yields in H2O), long decay lifetimes (>100 mu s) and enable in vivo fluorescence intensity imaging and time-resolved fluorescence lifetime imaging (FLIM). Finally, the main concerns and challenges for NIR emissive Ln biolabels such as the diversity of the NIR Ln(3+) optical toolbox and their biocompatibility and targeting capability in biological media are discussed so as to facilitate the long-term development of lanthanide chemical biology.

Abstract:High-resolution imaging is a powerful technique in theranostics and staging of tumors. Dual-modal imaging using lanthanide hybrid nanoparticles as probes has been attracting increasing attention due to their specific and intrinsic physical properties. The present work reports a novel water-soluble micelle of an amphiphilic block copolymer with diketone pendants chelating gadolinium (Gd3+) and europium (Eu3+) cations as a dual probe for magnetic resonance imaging (MRI) and fluorescence imaging for clinical detection. The Gd3+ and Eu3+ hybrid micelles have homogeneous size distribution and hydrodynamic diameters tunable between 80 and 220 nm. The macromolecular diketone ligand enhances the relaxation rate of Gd3+ and sensitizes Eu3+ luminescence efficiently. Further sensitized by the co-doped Gd3+, the Eu3+ complex exhibits a long-lived emission with a large Stokes shift. A strong red light at a wavelength of 614 nm is observed with an excitation in the range of 350 and 405 nm. Cellular uptake experiments reveal excellent biocompatibility and rapid uptake of the nanoparticles by MCF-7 cells. The robust dual-modal probes are promising in applications for early diagnosis of tumors, presurgical planning, and intraoperative fluorescence-guided surgery.

Abstract:Over the past few years, significant efforts have been made to create new fluorescent probes operating at longer wavelengths, particularly in the second near-infrared (NIR-II) window from 1000 to 1700 nm, offering enhanced tissue penetration compared to light in the visible and first near-infrared window (700-900 nm). However, most of the reported NIR-II fluorophores meet such dilemmas; they are excreted slowly and largely retained within the reticuloendothelial system. Here, we report a rapidly excreted NIR-II lanthanide complex Nd-DOTA (over 50% excreted through the kidneys within 3 h postinjection) with a molecular mass only 0.54 kDa. The NIR-II imaging quality of Nd-DOTA was far superior to that of clinically approved ICG with good photostability and deep tissue penetration (7 mm). Superior tumor-to-normal tissue ratio was successfully achieved to facilitate the abdominal ovarian metastases surgical delineation. Metastases with ≤1 mm can be completely excised under NIR-II bioimaging guidance. Significantly, since the Nd-DOTA structure is same to the clinically approved magnetic resonance imaging (MRI) contrast Gd-DOTA, it will speed up the clinical translation for this novel kind of NIR-II probes in the future.

Abstract:The accurate and sensitive in situ imaging of inflammatory bowel disease (IBD) is critical for the precise diagnosis and treatment of this disease, but it remains a challenge for fluorescent probes because of a more dynamical shift of pH in the intestinal tract than under other physical conditions. To address this issue, we report here that the rational introduction of the ortho -halogen bond at the electron acceptor can regulate the protonation and deprotonation of fluorescent dye, resulting in a pH-insensitive dye (pH 5–12). Therefore, constructing a near-infrared (NIR) probe, 2F-RBH , which effectively avoids this pH effect with a stable fluorescent signal and selectively indicates sites of inflammation in situ , by tracking peroxynitrite (ONOO − ) in an acute colitis mouse model. Collectively, our results highlight that 2F-RBH is a promising probe for the imaging of IBD and that our pH-insensitive dye has potentially desirable properties for in vivo bioimaging.

Abstract:pH homeostasis is strictly controlled at a subcellular level. A deregulation of the intra/extra/subcellular pH environment is associated with a number of diseases and as such, the monitoring of the pH state of cells and tissues is a valuable diagnostic tool. To date, only a few tools have been developed to measure the pH in living cells with the spatial resolution needed for intracellular imaging. Among the techniques available, only optical imaging offers enough resolution and biocompatibility to be proposed for subcellular pH monitoring. We present herein a ratiometric probe based on upconversion nanoparticles modified with a pH sensitive moiety for the quantitative imaging of pH at the subcellular level in living cells. This system provides the properties required for live cell quantitative imaging i.e. positive cellular uptake, biocompatibility, long wavelength excitation, sensitive response to pH within a biologically relevant range, and self-referenced signal.

Abstract:Recent advances in NIR detector bloom the design of NIR fluorophores for bioimaging guided diagnosis and therapeutics, with reduced light-tissue interaction, enhanced signal-to-noise ratio and increased penetration depth. NIR lanthanide molecular probes represent an important and emerging group of NIR imaging and sensing materials with attractive structural and photophysical characteristics including small sizes, metal-centered emission, long decay lifetime, large Stokes shift and high resistance to photobleaching. In this review, the sensitization of lanthanides and design principle of lanthanide molecular probes were described. We summarized the recent progresses in last decade on NIR luminescent lanthanide molecular probes by precisely tuning the antenna ligands. The development tendency of applying lanthanide molecular probes for in vivo bioimaging and biosensing would also be discussed, showing the unique and attractive properties of lanthanide coordination compounds compared to organic molecules and inorganic quantum nanoparticles.

Abstract:We report here the development of clickable and highly near-infrared (NIR) fluorescent lanthanide (Ln) complexes for bioorthogonal labeling of biomolecules. These azide- or alkyne-functionalized Ln complexes are hydrophilic and fluorogenic, exhibiting a strong increase of NIR fluorescence upon conjugation with biomolecules. Metabolic labeling of biomolecules with azide or alkyne, followed by click labeling with the Ln complexes, enables NIR fluorescence (NIRF) imaging of DNA, RNA, proteins, and glycans in cells. Furthermore, multicolor imaging is performed by combining click-labeling with the Ln complexes and immunostaining. In addition, the Ln complexes is compatible with click-expansion microscopy (click-ExM), which enables high-resolution NIRF imaging of cellular glycoproteins. Finally, the Ln complexes can be used for time-of-flight secondary-ion mass spectrometry (ToF-SIMS) imaging, thus achieving the first example of dual-modal imaging combining NIRF and SIMS microscopies.

Abstract:Applying a single molecular probe to monitor enzymatic activities in multiple, complementary imaging modalities is highly desirable to ascertain detection and to avoid the complexity associated with the use of agents of different chemical entities. We demonstrate here the versatility of lanthanide (Ln3+) complexes with respect to their optical and magnetic properties and their potential for enzymatic detection in NIR luminescence, CEST and T1 MR imaging, controlled by the nature of the Ln3+ ion, while using a unique chelator. Based on X-ray structural, photophysical, and solution NMR investigations of a family of Ln3+ DO3A-pyridine model complexes, we could rationalize the luminescence (Eu3+, Yb3+), CEST (Yb3+) and relaxation (Gd3+) properties and their variations between carbamate and amine derivatives. This allowed the design of probes which undergo enzyme-mediated changes detectable in NIR luminescence, CEST and T1-weighted MRI, respectively governed by variations in their absorption energy, in their exchanging proton pool and in their size, thus relaxation efficacy. We demonstrate that these properties can be exploited for the visualization of b-galactosidase activity in phantom samples by different imaging modalities: NIR optical imaging, CEST and T1-weighted MRI.

Abstract:Fluorescence sensing has the advantages of being real time, noninvasive, and convenient and having a low impact on the original environment for in vivo detection. Here, a reversible time-gated ratiometric in vivo detection method that could eliminate the interferences from probe amount, photon scattering, and absorption is proposed. Correspondingly, the composite probe must be able to reversibly respond to changes in the microenvironment and emit two luminescence signals at the same working wavelength but different lifetimes. Benefitting from the reversible detection mechanism, the probes could be used to monitor a dynamic biological process and the ratio signal value could be determined only by the concentration of analytes, independent of the probe concentration. Furthermore, benefitting from the same working wavelength, the read-out errors from photon absorption and scattering could be minimized. This method is very suitable for in vivo detection in which the probe distribution and depth are unknown and variable. As a typical model, different pH values in the gastrointestinal area and pH changes caused by drugs and fasting are successfully monitored.

Abstract:Lanthanide‐based NIR‐IIb probes are revolutionizing in vivo biomedical imaging with their enhanced tissue penetration and resolution. This review summarizes recent advancements and applications of lanthanide‐based NIR‐IIb probes, including ratiometric and multiplexed imaging, lifetime sensing and multimodal imaging. Future research directions and challenges in probe development and imaging technology are also discussed. The past decades have witnessed the significant development and practical interest of in vivo biomedical imaging technologies and optical materials in the second‐near infrared (NIR‐II, 1000–1700 nm) window. Imaging with the extended emission wavelength toward the long‐wavelength end (NIR‐IIb, 1500–1700 nm) further offers micrometer imaging resolution and centimeter tissue penetration depth by taking advantage of the much‐reduced photon scattering and near‐zero tissue autofluorescence background, which have become a very hot research area. This review focuses on the recent advances in the development of lanthanide‐based NIR‐IIb probes for in vivo biomedical applications. The progress including ratiometric imaging, multiplexed imaging for wide‐field and microscopy, lifetime multiplexing and sensing, persistent luminescence, and multimodal imaging is summarized. Challenges and future directions concerning the investigation of the photophysical and photochemical properties of NIR‐IIb probes, the selection of near‐infrared cameras as well as the potential extension of the NIR‐IIb imaging sub‐window are pointed out. This review will inspire readers who have a strong interest in developing optical imaging technology and long‐wavelength fluorescence probes for high‐contrast in vivo biomedical applications.

Abstract:In the second near-infrared spectral window (NIR-II; with wavelengths of 1,000–1,700 nm), in vivo fluorescence imaging can take advantage of reduced tissue autofluorescence and lower light absorption and scattering by tissue. Here, we report the development and in vivo application of a NIR-II phosphorescent probe that has lifetimes of hundreds of microseconds and a Stokes shift of 430 nm. The probe is made of glutathione-capped copper–indium–selenium nanotubes, and in acidic environments (pH 5.5–6.5) switches from displaying fluorescence to phosphorescence. In xenograft models of osteosarcoma and breast cancer, intravenous or intratumoral injections of the probe enabled phosphorescence imaging at signal-to-background ratios, spatial resolutions and sensitivities higher than NIR-II fluorescence imaging with polymer-stabilized copper–indium–sulfide nanorods. Phosphorescence imaging may offer superior imaging performance for a range of biomedical uses.

Abstract:<p>Fluorescence imaging plays an important role in early diagnosis and surgical resection of cancer because of its real-time, noninvasiveness, high sensitivity, and resolution. Compared with the traditional visible and first near-infrared (NIR-I) region, fluorescence imaging with NIR-II character has obvious advantages in superior imaging depth and contrast. Because in the NIR region, the auto-fluorescence of biological tissues and the absorption and scattering of photons by tissues will be significantly weakened. Lanthanide-doped nanoparticles (LDNPs) is a promising candidate for NIR fluorescence probe because of its easily adjustable emission wavelength and fluorescence lifetime (FL). In this review, we summarize the general design mechanism of LDNPs from the aspects of preparation method, surface modification and bioconjugation, then introduce the development of LDNPs on bioimaging, biosensing, and multifunctional nano-platform in recent years. The challenges and efforts of LDNPs in this field have further prospected.</p>

Abstract:The luminescence intensity of near-infrared (NIR) emitting lanthanide nanoparticles (LnNPs) is usually limited, owing to their small absorption cross section. Although dye sensitization has been proven to be an effective way to improve the luminescence intensity of LnNPs, the sensitization effect is fairly limited, owing to the simplicity of the sensitizers used and the complexity of the energy transfer process, typically involving three steps. In this study, a more efficient sensitizer (Cy7) was chosen to replace a commonly used one (ICG) and the energy transfer process was also optimized through using Yb3+ ions as emitter ions and Nd3+ ions as intermediate ions. With Cy7 as a sensitizer, the sensitization effect was assessed to be better than with ICG, owing to the higher quantum yield of Cy7. Meanwhile, the Cy7-sensitized NIR lanthanide nanomaterial was proven to be good for deep tissue penetration and low-power excitation bioimaging. Furthermore, the highly-enhanced NIR signal was successfully used in blood vessel imaging and fluorescence-guided peritumoral lymph node dissection in a mouse model.
