Abstract:The effects on the green alga Chlorella vulgaris of 40 herbicides in 19 different chemical structure classes and with 11 dissimilar modes of action were studied through 96-h acute toxicity tests. Experimental results indicated that the average acute toxicity of acetolactate synthase (ALS)-inhibiting herbicides to C. vulgaris was close to those of the acetyl-CoA carboxylase (ACCase)-inhibiting herbicides and the lipid synthesis-inhibiting protoporphyrinogen oxidase (Protox) inhibitor. Their acute toxicities were higher than those of the microtubule process inhibitor, mitotic process inhibitor, the glutamine synthase inhibitors, and 5-enolpyruvyl-shikimate-3-phosphate synthase (EPSP synthase) inhibitors. The acute toxicities of auxin herbicides to C. vulgaris were the lowest among all herbicides tested and that of the photosynthesis-inhibiting herbicides was the highest.

Abstract:The effects of 40 herbicides with nine modes of action on the green alga Raphidocelis subcapitata were studied by 96-h acute toxicity tests. Results showed that the EC50 of the herbicides with respect to the photosynthetic processes of R. subcapitata ranged from 0.0007 to 4.2286mgL−1. Photosynthesis was the process of the green alga most sensitive to the tested herbicides. The most toxic herbicides were atrazine, ametryme, simazine, prometryne, cyanazine, isoproturon, chlorotoluron, diuron, methabenzthiazuron, and paraquat. The EC50 of the protoporphyrinogen oxidase inhibitor oxadiargyl, which was the parameter least sensitive to the herbicides tested in this study, was 42.5mgL−1. The descending order of the average acute toxicity to R. subcapitata of herbicides with regard to the nine modes of action was as follows: photosynthetic process>cell division>lipid synthesis, acetyl-coenzyme A carboxylase>acetolactate synthase> 5-enolpyruvyl-shikimate-3-phosphate synthase, glutamine synthase, hormone synthesis>protoporphyrinogen oxidase.
Influence of inorganic arsenic on photosynthetic activity of Chlorella pyrenoidosa

Abstract:The arsenic contamination in aquatic environment has drawn significant attention to ecotoxic risk .The valence states of arsenic including arsenite [As] and arsenate [As(Ⅴ)] play an important role for their behavior and toxicity at different levels of exposure concentrations .In this study ,the toxicity of different concentrations of As(Ⅲ) and As(Ⅴ) to Chlorella pyrenoidosa was determined and compared based on four different test endpoints :The growth inhibition rate ,the chlorophyll a (Chl a) concentration ,the chlorophyll fluorescence parameters of photosystem Ⅱ (PSII) and the ultra-structural morphology of cells .The EC50 values of As(Ⅴ) were higher than As (Ⅲ) ,which indicated that As(Ⅲ) was more toxic to C .pyrenoidosa than As(Ⅴ) .The Chl a concentration of C . pyrenoidosa treated by As(V) was higher than that treated by As(Ⅲ) .In general ,the chlorophyll fluorescence parameters including the maximum quantum yields (Fv/Fm ) ,the effective quantum yields (Yield) ,the maximum electron transport rate (rETRmax ) and the linear section slope (α) of C .pyrenoidosa treated by 150 .0 mg · L -1 As (Ⅲ) for 96 h were decreased by 77％ ,91％ ,92％ and 85％ ,which was higher than that exposed to As (Ⅴ) of 19％ ,50％ , 51％ and 23％ , respectively , and chlorophyll fluorescence parameters were inhibited by high concentrations of both arsenic . The ultrastructural morphology of cells grown in the two species arsenic was observed by transmission electron microscopy .The cell wall separated from the cell membrane ,accumulated starch granules were observed in the chloroplast ,and diminishing pyrenoid and some lipid droplets were also observed in the cytoplasm .The mechanism of arsenite and arsenate toxicity to C .pyrenoidosa was explored ,and the inhibition of PSII photosynthetic efficiency to C . pyrenoidosa was determined . These results will help to develop an understanding of the biologically mediated environmental processes of arsenite and arsenate .

Abstract:The effects of three sulfonylurea herbicides, metsulfuron-methyl, chlorsulfuron and bensulfuron, and their main degradation products on the green algae (Chlorella pyrenoidosa) were studied through 96-hour algae growth inhibition tests and organic nitrogen bioavailability tests. Experimental results indicated that, the acute toxicity of these sulfonylurea herbicides was much higher (about 2 or 3 orders) than their corresponding degradation products, and nitrogen of the degradation products could be used by the algae as the only nitrogen source to stimulate their growth in the order .

Abstract:To reduce the leaching potential, to prevent groundwater contamination and to maintain the efficacy of a pesticide, natural polysaccharides have received increasing attention due to their biocompatibility and useful biological reactivity for controlled release formulations (CRFs) of pesticides. In this paper, the toxicities of the chiral herbicide dichlorprop (DCPP) and its complexes with chitosan molecules (DCPP-CS) and chitosan nanoparticles (DCPP-NP) to two different green algae were determined and compared. The inhibition rates of DCPP, DCPP-CS and DCPP-NP were determined at 24, 48, 72, 96, 120, 144, 168 h, and the results show that (S)-DCPP was more toxic to Chlorella vulgaris than (R)-DCPP, while the (R)-DCPP was more toxic to Scenedesmus obliquus than (S)-DCPP. The study also found that the chiral selectivity of DCPP to Chlorella vulgaris and Scenedesmus obliquus could be changed when DCPP was complexed with chitosan molecules (CS) or chitosan nanoparticles (NP). For Chlorella vulgaris, the order of inhibition was (R)-DCPP-CS > (S)-DCPP-CS and (R)-DCPP-NP > (S)-DCPP-NP; for Scenedesmus obliquus, the order was (S)-DCPP-CS > (R)-DCPP-CS and (S)DCPP-NP > (R)-DCPP-NP. This phenomenon suggests that the enantioselective behaviors of chiral compounds might shift when interactions with other chiral receptors coexist in different biological environments. Additionally, chitosan molecules and chitosan nanoparticles also showed different toxicities, which could be ascribed to the difference in the physicochemical properties between CS and NP or the differences in the cell walls of the two fresh water green algae.

Abstract:Growth-inhibiting tests were carried out for 12 pesticides (including 11 fungicides: fosetyl-aluminum, benomyl, metalaxyl, iprodione, dimetachlone, carbendazim, thiophanate-methyl, bismerthiazol, procymidone, zineb, chlorothalonil, and the acaricide abamectin) in the green algae Chlorella pyrenoidosa and Scenedesmus obliqnus and the differential sensitivities of the two green algae to those pesticides were compared. The results indicate that the acute toxicity of benomyl to C. pyrenoidosa and S. obliqnus is the highest among all of the pesticides tested and is close to that of the photosynthesis-inhibiting herbicides atrazine, simazine, and chlorotoluron. Meanwhile, algal species vary widely in their response to the pesticides. The results demonstrated that there was a differential response to various pesticides by the two species of algae and that the sensitivity of various species of algae exposed to chlorothalonil varied by nearly two orders of magnitude; sensitivity to thiophanate-methyl varied by more than one order. Investigations using different algal species as test organisms have demonstrated that algae vary greatly in their response to chemicals. Differential sensitivity of green species to the compounds could induce species shifts within communities.

Abstract:The toxicity of 7 herbicides to the three cyanobacteria was tested in this work. The results indicated that: (1) There was a highly significant relationship between dried weight or chlorophyll-a and OD680nm for tested cyanobacteria; (2) the toxicity of the tested herbicides with the order from high to low was: photosynthesis-inhibiting > ACCase inhibitor > protox inhibiting herbicides; (3) the sensitivity of various species exposed to cyanazine, diclofop, prometryn, simazine and simetryn varied by over one order of magnitude. The decreasing order of sensitivity of cyanobacteria to the selected herbicides was: M. Flosaquae > M. Aeruginosa > A. flos-aquae. Cyanobacteria can produce toxins including hepatotoxins e.g. microcystins and endotoxins e.g. lipopolysaccharides. Therefore, the research on comparing the differential sensitivity of cyanobacteria and green algae is of important scientific significance and realistic value

Abstract:This work examines the effects of six pesticides on five green algae: Pseudokirchneriella suhcapitata, Scenedesmus quadricauda, Scenedesmus obliquus, Chlorella vulgaris and Chlorella pyrenoidosa. The results indicate that the decreasing order of the average toxicity to five algae was: fluazinam > propineb > maneb > mancozeb > zineb > bromoxynil octanoate. However, according to sensitivity magnitude, the decreasing order of the ecological risk was: Maneb > bromoxynil octanoate > propineb/fluazinam > zineb/mancozeb. There was a strong variance between toxicity and ecological risk.

Abstract:Residues of triazophos in aquatic ecosystems due to extensive use for controlling pests in agriculture has became worldwide concern, while the toxic response of triazophos on the non-target green algae in aquatic environment is not well studied. Therefore, the acute (96 h) toxic effects of 1 and 10 mg/L triazophos on green algae Chlorella pyrenoidosa were evaluated in present study. The results showed that the growth was notably inhibited when treated with triazophos and the 96 h-EC50 (median inhibition concentration) were 12.79 mg/L. The content of photosynthetic pigments (including chl a, chl b, total-chl and carotinoids) clearly decreased under two treatments after 48 h and 96 h with exception for the values at 48 h exposure in 1 mg/L treatment. In addition, the transcript abundance of photosynthesis-related genes (psbA, psbC and rbcL) showed obvious decrease in above two treatments after exposure 96 h to triazophos. In response to 10 mg/L triazophos treatment, the morphology of thylakoid chloroplast of algal cells were obviously damaged. It was also found that starch granules increased with down-regulation of atpB gene expression in 10 mg/L treatment, which suggests that triazophos may inhibit the energy metabolism of C. pyrenoidosa. Moreover, the algal growth inhibition was along with the increase of intracellular reactive oxygen species (ROS), activity of antioxidant enzymes and malondialdehyde content indicating oxidative damage and lipid peroxidation in the algal cells. Our findings reveal that triazophos has potential toxicity and environmental risks to one of the primary producers green algae.
Toxic mechanism of three azole fungicides and their mixture to green alga Chlorella pyrenoidosa

Abstract:Currently, few studies have investigated the joint toxicity mechanism of azole fungicides at different exposure times and mixed at the relevant environmental concentrations. In this study, three common azole fungicides, namely, myclobutanil (MYC), propiconazole (PRO), and tebuconazole (TCZ), were used in studying the toxic mechanisms of a single substance and its ternary mixture exposed to ambient concentrations of Chlorella pyrenoidosa. Superoxide dismutase (SOD), catalase (CAT), chlorophyll a (Chla), and total protein (TP), were used as physiological indexes. Results showed that three azole fungicides and ternary mixture presented obvious time-dependent toxicities at high concentrations. MYC induced a hormetic effect on algal growth, whereas PRO and TCZ inhibit algal growth in the entire range of the tested concentrations. The toxicities of the three azole fungicides at 7 days followed the order PRO > TCZ > MYC. Three azole fungicides and their ternary mixture induced different levels of SOD and CAT activities in algae at high concentrations. The ternary mixture showed additive effects after 4 and 7 days exposure, but no effect was observed at actual environmental concentrations. The toxic mechanisms may be related to the continuous accumulation of reactive oxygen species, which not only affected protein structures and compositions but also damaged thylakoid membranes, hindered the synthesis of proteins and chlorophyll a, and eventually inhibited algal growth. These findings increase the understanding of the ecotoxicity of azole fungicides and use of azole fungicides in agricultural production.

Abstract:Topramezone is a new, highly selective herbicide of pyrazole structure for the post-emergence control of broadleaf and grass weeds in corn. In this study, the effects of topramezone on C. vulgaris, especially in relation to the cell growth, oxidative stress, cell morphology and photosynthetic activity were assessed. Results showed that topramezone treatment was detrimental to C. vulgaris growth during the 24-96h of exposure. The changes in cells pigments content and relative transcript of photosynthesis-related genes, which implies that topramezone disrupted the photosynthetic system. Moreover, topramezone induced membrane permeability in a significant proportion of cells with a maximum damage rate of 40.40%, and morphology of cells was more complicated than the control group. TEM images also revealed that topramezone compromised the integrity of the cells. The data corroborated topramezone induced ROS triggered oxidative stress, leading to an increase of MDA. These results suggested that topramezone could have significant effects on growth and physiological functions in algae species, and we supposed that this herbicide affected all of these parameters and the observed effects can be explained by the generation of oxidative stress. This research helps to understand how topramezone affects C. vulgaris and provides a scientific basis for applications of topramezone in aquatic environment.
Effects of Acetanilide Herbicides on Growth and Oxidative Damage of Microcystis aeruginosa

Abstract:The widespread use of acetanilide herbicides causes potential risks to aquatic ecological environment. In order to reveal their toxic effect on algae, we used Microcystis aeruginosa as the model organism, the impacts on biomass and oxidative damage of alachlor, acetochlor and butachlor to M. aeruginosa were investigated. The results showed an obvious lag effect and dose ̄response relationship. Lower concentrations of herbicides stimulated the growth of algae, but higher concentrations inhibited the growth. The oxidative pressure on M. aeruginosa increased with the extension of exposure time and the increasing concentrations of the three acetanilide herbicides. Levels of malondialdehyde (MDA) which is a lipid peroxide degradation product, were markedly increased, meanwhile, the 240 生态毒理学报第 11 卷 activities of superoxide dismutase (SOD) and peroxidase (POD) were also significantly increased. After 96 h expo ̄ sure, the relative MDA contents in alachlor (32.0 mg􀅰L), acetochlor (32.0 mg􀅰L) and butachlor (15.0 mg􀅰L) ex ̄ posure solution were 138% , 204% and 154% , the relative SOD activities were 116% , 87% and 115% , the relative POD activities were 475% , 278% and 627% , respectively. Combined the results of biomass and oxidative damage, the order of toxicity to the M. aeruginosa of the three acetanilide herbicides was butachlor > acetochlor > alachlor.
Effects of Glufosinate on Antioxidant Enzymes, Subcellular Structure, and Gene Expression in the Unicellular Green Alga Chlorella Vulgaris

Abstract:Greater exposure to herbicide increases the likelihood of harmful effects in humans and the environment. Glufosinate, a non-selective herbicide, inhibits glutamine synthetase (GS) and thus blocks ammonium assimilation in plants. In the present study, the aquatic unicellular alga Chlorella vulgaris was chosen to assess the effects of acute glufosinate toxicity. We observed physiological changes during 12–96h of exposure, and gene transcription during 6–48h of exposure. Exposure to glufosinate increased malondialdehyde content by up to 2.73 times compared with the control, suggesting that there was some oxidative damage. Electron microscopy also showed that there were some chloroplast abnormalities in response to glufosinate. The activities of the antioxidant enzymes superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) also increased markedly in the presence of glufosinate. Maximum activities of SOD, POD, and CAT were 2.90, 2.91, and 2.48 times that of the control, respectively. These elevated activities may help alleviate oxidative damage. A real-time polymerase chain reaction (PCR) assay showed changes in transcript abundances of three photosynthetic genes, psaB, psbC, and rbcL. The results showed that glufosinate reduced the transcript abundances of the three genes after 12h exposure. The lowest abundances of psaB, psbC and rbcL transcripts in response to glufosinate exposure were 38%, 16% and 43% of those of the control, respectively. Our results demonstrate that glufosinate affects the activities of antioxidant enzymes, disrupts chloroplast ultrastructure, and reduces transcription of photosynthesis-related genes in C. vulgaris.
The Toxic Effects of the Extracellular Active Components from One Algae-lysing Bacteria on Chlorella Pyrenoidosa

Abstract:Toxic effects of L7 lyophilized powder of extracellular active components (L7-LPEAC), extracted from the Algae-lysing bacteria L7, on Chlorella pyrenoidosa were studied according to the changes of effective photosynthesis rate (EPR), the protein content, the chlorophyll a content and the MDA content of algae. The results showed that the growth of alga was promoted at low concentrations of L7-LPEAC (0.80 g/L, 1.25 g/L). The 96 h-EC50 and 120 h-EC50 upon Chlorella pyrenoidosa are 5.75 g/L and 2.55 g/L, respec tively. The chlorophyll a content increased firstly and then decreased at high concentrations of L7-LPEAC (≥2 g/L), so did the protein content. Compared with the control group, there is a significant statistics diff erence (P0.05) in the treatment groups (2.00 g/L, 3.13 g/L, 4.90 g/L), while extreme significant statistics difference (P0.01) in the other treatment groups (7.67 g/L, 12.00 g/L) after 72 h. The inhibition rates of chlorophyll a content of Chlorella pyrenoidosa in all treatment groups reached over 60% after 120 h. For the remediation of eutrophic nature water body, it can not only kill the algae which caused the water eutrophication, but also maintain the balance of the aquatic ecosystem well by adding some proper concentrations of L7-LPEAC.

Abstract:Growth-inhibition tests for 14 pesticide adjuvants which are widely used to manufacture various pesticidal formulations in China, were performed on the green algae Scenedesmus quadricauda, and Chlorella vulgaris to compare differential sensitivity among populations of these algae to the adjuvants. The results showed that the acute toxicities of 700♯, 1601♯, By-140, and SOPA to S. quadricauda and C. vulgaris were the lowest among all the tested adjuvants. The acute toxicities of Tween 80, O-25, and AEO-13 phosphate to the selected two green algae were intermediate among the tested adjuvants. The acute toxicities of 602♯, 500♯, OT, NP-10, OP-10, and JFC were the highest. Meanwhile, the algal species vary widely in their response to those adjuvants. The results showed that there was a differential response to various adjuvants among the selected algal species and that the sensitivities of the various algal species exposed to 1601♯ and OT varied by nearly 1 order and to 700♯, and By-140 varied by over 1 order of magnitude. In addition, the NOEC and LOEC values of the selected 14 adjuvants to S. quadricauda and C. vulgaris were tested; the result showed that NOEC⩽EC10<EC20 for 27 of the 28 data sets (96%) and EC10⩽LOEC⩽EC20 for 20 of the 28 data sets (78%).

Abstract:This paper aims to acquire the experimental data on the eco-toxicological effects of agricultural pollutants on the aquatic plants and the data can support the assessment of toxicity on the phytoplankton. The pesticide of Chlorpyrifos used as a good model to investigate its eco-toxicological effect on the different microalgae in freshwater. In order to address the pollutants derived from forestry and agricultural applications, freshwater microalgae were considered as a good sample to investigate the impact of pesticides such as Chlorpyrifos on aquatic life species. Two microalgae of Chlorella pyrenoidosa and Merismopedia sp. were employed to evaluate toxicity of Chlorpyrifos in short time and long time by means of measuring the growth inhibition rate, the redox system and the content of chlorophyll a, respectively. In this study, the results showed that EC50 values ranging from 7.63 to 19.64mg/L, indicating the Chlorpyrifos had a relatively limited to the growth of algae during the period of the acute toxicity experiment. Moreover, when two kinds of algae were exposed to a medium level of Chlorpyrifos, SOD and CAT activities were importantly advanced. Therefore, the growth rate and SOD and CAT activities can be highly recommended for the eco-toxicological assessment. In addition, chlorophyll a also could be used as a targeted parameter for assessing the eco-toxicity of Chlorpyrifos on both Chlorella pyrenoidosa and Merismopedia sp.

Abstract:Triflumizole is one of imidazole fungicides that works by inhibiting ergosterol biosynthesis, and is widely used for the control of powdery mildew and scabs on various fruits and crops. Triflumizole residue has been frequently detected in soil and aquatic ecosystems. While many studies have focused on its toxic effect on terrestrial and aquatic animals, little attention has been paid to aquatic algae, the primary producers of aquatic environments. Therefore, we evaluated the acute (96 h) toxicity effects of triflumizole on the freshwater algae Chlorella vulgaris, by examining growth, cell morphology, photosynthesis, and oxidative stress. The results showed that the 96 h median inhibition concentration (96 h-EC50) was 0.82 mg/L (95% confidential interval 0.70-0.98 mg/L). The growth of algal cells was conspicuously inhibited by triflumizole exposure, and the cell surfaces appeared to be shrunkThe chlorophyll content (including Chl-a, Chl-b and T-Chl) dramatically decreased at triflumizole concentrations of 0.2 and 1.0 mg/L. In addition, the transcript abundance of photosynthesis-related genes (psaB, psbC and rbcL) showed obvious decreases in above treatments after 96 h of exposure to triflumizole. Moreover, the algal growth inhibition was accompanied by an increase in intracellular reactive oxygen species and malondialdehyde content, as well as increased activity of antioxidant enzymes such as superoxide dismutase and peroxidase, indicating oxidative stress and lipid peroxidation. Our findings reveal that triflumizole has potential toxicity to the primary producers (freshwater algae) in aquatic ecosystems.
Acute Toxicity of Three Typical Pollutants to Aquatic Organisms and Their Water Quality Criteria

Abstract:Two species of microalgae Chlorella pyrenoidosa, Scenedesmus obliqnus and a red worm Chironomidae larvae were selected as test organisms in determining the acute toxicity effects of Cr (VI), 2,4,6-trichlorophenol and nitrobenzene. The results were able to provide more information on water quality criteria and more data on their toxicity to indigenous aquatic organisms in China. The 96 h-EC50 values of Cr (VI), TCP and nitrobenzene on C. pyrenoidosa were 1.34 mg x L(-1, 4.55 mg x L(-1) and 86.58 mg x L(-1), respectively, while those of S. obliqnus were 19.52 mg x L(-1), 3.71 mg x L(-1) and 74.15 mg x L(-1), respectively. The mortality of C. larvae was 15% when the concentration of Cr(VI) was increased to 1,500 mg x L(-1). The 48 h-LC50 values of TCP and nitrobenzene on C. larvae were 9.29 mg x L(-1) and 98.34 mg x L(-1), respectively. These results indicated that Cr( VI) showed higher toxicity to C. pyrenoidosa, while only moderate toxicity to S. obliqnus; TCP had higher toxicity to C. pyrenoidosa and S. oblignus; while nitrobenzene was only moderately toxic to both species of microalgae. The toxicity among the three pollutants to C. larvae was in the order of TCP > nitrobenzene > Cr (VI).
Acute Toxicity of Quaternary Ammonium Cationic Surfactants to Chlorella vulgaris and Scenedesmus obliquus

Abstract:Chlorella vulgaris and Scenedesmus obliquus, two green freshwater alga in environment, have been widely used to evaluate the im- pacts of xenobiotics duo to their sensitivity to contaminants. In this study, we investigated the acute toxicity of quaternary ammonium cationic surfactants(QACS)with different length of alkyl chain(CL=8~18)to C. vulgaris and S. obliquus. The results indicated that the 96 h-EC50 value of thirteen kinds of QACS on C. vulgaris and S. obliquus were 0.108~9.472 mg·L-1 and 0.085~12.188 mg·L-1, respectively. Further- more, the acute toxicity of QACS with the same substituent group on the two species of algae gradually increased with the increasing of CL. However, the different substituent groups had little effect on the acute toxicity of QACS with the same CL to the two green algae. The relation- ship between acute toxicity[lg(1/EC50)] and octanol-water partition coefficients(lgKow), CL could be better fitted by parabola model than linear equation, and the fitting trends were same for the two green algae. The parabola equations for C. vulgaris and S. obliquus were lg(1/EC50)=-0.106 lgKo 2w +0.746 lgKow-0.830(R2=0.944), lg(1/EC50)=-0.034 CL2+1.051 CL-7.725(R2=0.945)and lg(1/EC50)=-0.119 lgK o2w +0.830 lgKow-0.812(R2=0.941), lg(1/EC50)=-0.039 CL2+1.218 CL-8.772(R2=0.945), respectively, which could provide references for the prediction of acute toxicity for similar compounds to freshwater algae.
