Abstract:Hydrogen polysulfides (H2Sn, n >= 2) is recently regarded as a potential signaling molecule which shows a higher efficiency than hydrogen sulfides (H2S) in regulating enzymes and ion channels. However, the development of specific fluorescent probes for H2Sn with long-wavelength emission (>600 nm) are still rare. In this work, a semi-naphthorhodafluor-based red-emitting fluorescent probe SNARF-H2Sn containing a phenyl 2-(benzoylthio) benzoate responsive unit was constructed. SNARF-H2Sn was capable of selectively detecting H2Sn over other reactive sulfur species. Treatment with H2Sn would result in a > 1000-fold fluorescence enhancement within 10 min. SNARF-H2Sn showed a low limit of detection down to 6.7 nM, and further enabled to visualize exogenous/endogenous H2Sn in living A549 cells and zebrafish. (C) 2020 Elsevier B.V. All rights reserved.

Abstract:Due to the crucial biological roles of hydrogen polysulfides (H2Sn) in living systems, the selective determination of H2Sn is essential. In this work, we reported a turn-on fluorescent probe, JSI-Sn, for H2Sn with high sensitivity and good selectivity. Probe JSI-Sn displayed a near-infrared emission (λmaxEm = 701 nm) and a large Stokes shift (123 nm) in the presence of H2Sn in solution. Using probe JSI-Sn as an indispensable tool, the monitoring of intracellular H2Sn in living cells and zebrafish were realized.

Abstract:In order to quantitatively detect hydrogen polysulfides (H2Sn, n > 1) in biosystems, we propose a new two-photon turnon fluorescent probe, named Np–Sn, based on the D–π–A skeleton two-photon fluorophore of 4 -hydroxy-1,8-naphthalimide derivative and deprotection of 2-fluoro-5-nitrobenzoic ester by H2Sn–Np–Sn displayed a more than 80-fold enhancement towards H2Sn in 550 nm and high sensitivity with a detection limit as low as 33 nM. Additionally, the probe Np–Sn was further used for fluorescence imaging of H2Sn in living cells under two-photon excitation (820 nm), which showed a high-resolution imaging, thus demonstrating its practical application in biological systems for the study of physiological and pathological functions of H2Sn.

Abstract:Hydrogen polysulfides (H2Sn, n > 1) belong to sulfane sulfur in the reactive sulfur species (RSS) family and play significant roles in maintaining biological homeostasis in organisms. The detection of H2Sn in living systems is essential, but further understanding of its "intact" function in living cells has been limited, owing to the lack of appropriate analytical tools. In this work, a new fluorescent probe PP-PS was designed for the detection of endogenous H2Sn. The probe has a large Stokes shift (178 nm), low detection limit (1 nM), and short response time (1 minute). Besides, PP-PS was successfully applied in the imaging of endogenous H2Sn in lysosomes of living cancer cells, xenograft mouse tumor tissues, and LPS-induced mouse inflammation tissues. These results revealed that the probe PP-PS could act as a new fluorescence imaging tool to study the function of intracellular hydropolysulfides.

Abstract:Hydrogen polysulfide (H2Sn, n≥2), as a kind of active sulfur species (RSS), has become a hot topic in RSS. It can regulate the biological activity of many proteins through S-sulfhydrylation of cysteine residues (protein Cys-SSH), and has a protective effect on cells. Although there have been some studies on hydrogen polysulfide, its production, degradation pathway and regulation mechanism still need further be researched. In presented study, an original lysosome-localized fluorescent probe for determining H2Sn was developed utilizing rhodamine as the fluorogen. The probe used morpholine as the locating unit of lysosomes and chose 2-fluoro-5-nitrobenzoate as the recognizing group. Before adding H2Sn, the proposed probe displayed a spironolactone structure and emitted very weak fluorescence. After adding H2Sn, a conjugated xanthene was formed and the probe demonstrated green fluorescence. When the H2Sn concentration was varied from 6.0×10-7 mol·L-1 to 10.0×10-5 mol·L-1, the fluorescence intensity of the probe was linearly dependent on the H2Sn concentration. And the detection limit was 1.5×10-7 mol·L-1. The presented probe owned a fast response speed, good selectivity, excellent sensitivity and broad pH work scope. In addition, the probe had been well utilized to sense endogenic and exogenic H2Sn in lysosomes.

Abstract:We have developed a novel colorimetric and ratiometric fluorescence probe for the selective and sensitive monitoring of hydrogen sulfide based on a dicyanoisophorone platform. An excellent linear relationship of fluorescence intensity ratio (I-637/I-558) (R-2 = 0.9867) versus hydrogen sulfide concentration in the range of 1-12 mu M was obtained. This probe exhibited a remarkable fluorescence response to hydrogen sulfide over other physiological thiols or biological species, which fluoresces in the red region with a large Stokes shift (172 nm). This probe was successfully utilized to monitor H2S under in vitro physiological conditions and for imaging H2S in living cells and living zebrafish in vivo.

With the addition of hydrogen sulfide, the absorption and fluorescence spectra of the probe show regular changes. Abstract:As a common gaseous signaling molecule, hydrogen sulfide (H2S) plays a vital role in physiology and pathology. The development of fluorescent probes for detecting H2S has attracted widespread attention. However, most of the reported fluorescent probes with nitrobenzoxadiazole (NBD) as the recognition group have been widely used to simultaneously detect biothiols and H2S, instead of specifically detecting H2S. Herein, a novel NBD-based near-infrared (NIR) fluorescent probe named CX-N for the detection of H2S is synthesized. The selectivity of CX-N for H2S is significantly higher than that for biothiols and other potential interferences. After reacting with H2S, CX-N shows a significant increase in NIR fluorescence (75-fold), large Stokes shift (155 nm) and fast response (4 min). And the possible response mechanism of CX-N to H2S is given and confirmed by HPLC and HRMS. Based on the low cytotoxicity of CX-N, it has been used for H2S imaging in live cells and zebrafish. More importantly, CX-N has also been successfully applied for the real-time imaging of H2S in inflammatory and tumor mice based on its NIR emission, which provides a reliable platform for the specific recognition of H2S in complex biological systems.

Abstract:Since Hydrogen Sulfide (H2S) was recognized as a gas transmitter, its detection and quantification have become a hot research topic among chemists and biologists. In this area, fluorescent probes have shown great advantages: fast and strong response, low detection limit and easy manipulation. Here we developed a new fluorescent probe that detected H2S selectively among various bioactive and inorganic salts. This probe was based on the core structure of fluorescein and reacted with H2S through azide-reduction. Great linearity was achieved correlating fluorescence intensity and H2S concentrations in solution. The detection of H2S in cancer cells was also achieved.

Abstract:Hydrogen polysulfides (H2Sn, n>1) as important intracellular reactive sulfur species (RSS) are believe to be responsible for cellular redox regulation. Lots of researches about H2Sn focusing on their formation, detection and bio-function in signalling regulation are spring up but with poor understanding, especially for biosynthesis and bio-function remain complicated and confusing. Recent studies reveal that thionitrous acid (HSNO) as potential intermediate linked signalling molecules of nitrogenous and sulphureous during biotic redox regulation. However, there are limited evidences for supporting the interrelation and bioeffect between HSNO and H2Sn. Herein, we have successfully designed a near-infrared (NIR) fluorescent probe ((2-fluoro-5-nitrobenzoyl)oxy)-Benzo[e]cyanine (BCy-FN) for detection H2Sn and for the first time observing HSNO-mediated H2Sn generation in cells and in vivo. The probe is harvested from fluorophore BCy-Keto and 2-fluoro-5-nitrobenzoic acid in one step, featuring mitochondria localization. The unique Enol-Keto tautomerization of fluorophore enables the probe becomes more sensitive and has powerful application. Hypoxia model has been constructed and powerfully interpreted the pretreatment of HSNO for zebrafish hypoxia process effectively improves H2Sn levels and defends the hypoxia induced brain damage. We believe the present studies will help environmentalist and biologist for better understanding of biosynthesis and bio-function in HSNO-mediated H2Sn formation process under hypoxia stress.

Abstract:A dicyanoisophorone-based near-infrared fluorescent probe (NIR-NP) was successfully designed for hydrogen sulfide (H2S) detection. 7-Nitro-1,2,3-benzoxadiazole (NBD) amine, an electron-withdrawing group, was applied to quench the fluorescence of dicyanoisophorone-based fluorescent dyes and could be removed by H2S-specific thiolysis of an NBD amine bond, leading to a significant fluorescence turn-on response. The probe could quantitatively detect H2S with a lower detection limit (0.03 mu M) in vitro and in living cells. The probe with a large Stokes shift (186 nm) showed a high selectivity to H2S in the presence of other biothiols, including glutathione, homocysteine, and cysteine. Moreover, the probe was successfully applied to image endogenous H2S in different tumor cells and in liver tissues. The excellent properties of the probe and its applications in cell and tissue imaging indicated its potential for further exploration and application of H2S in the pathophysiology of cancers and even in the diagnosis of H2S-related diseases.

Abstract:Hydrogen sulfide (H2S) has been recognized as an important endogenous gasotransmitter associated with biological signaling transduction. However, recent biological studies implied that the H2S-related cellular signaling might actually be mediated by hydrogen polysulfides (H2S n , n > 1), not H2S itself. Unraveling such a mystery strongly demanded the quantification of endogenous H2S n in living systems. However, endogenous H2S n has been undetectable thus far, due to its extremely low concentration within cells. Herein, we demonstrated a strategy to detect ultra-trace endogenous H2S nvia a fluorescent τ-probe, through changes of fluorescence lifetime instead of fluorescence intensity. This τ-probe exhibited an ultrasensitive response to H2S n , bringing about the lowest value of the detection limit (2 nM) and a lower limit of quantification (10 nM) to date. With such merits, we quantified and mapped endogenous H2S n within cells and zebrafish. The quantitative information about endogenous H2S n in cells and in vivo may have a significant implication for future research on the role of H2S n in biology. The methodology of the τ-probe established here might provide a general insight into the design and application of any fluorescent probes, beyond the limit of utilizing fluorescence intensity.

Abstract:In this work, we reported a fluorescent probe Fur-SH, a derivative of benzofuranone, which was used to detect H2S in living cells and zebrafish. Based on the three structural characteristics of the probe, the effects of different structural modifications on the optical properties of the fluorophore were compared. Then, the fluorophore FurOH was synthesized by modifying diethylamino group with benzofuranone as the main skeleton. With 2,4-dinitrofluorobenzene as the recognition group and diethylamino as the electron donor, the push-pull electron effect occurred with nitro group, which led to fluorescence quenching, and an openable fluorescent probe Fur-SH was formed. The probe Fur-SH (lambda ex = 510 nm; lambda em = 570 nm) had the advantages of smaller full width at half maxima, rapid response (5 min) and wide pH window. The quantitative properties of the probe were excellent, reaching saturation at 50 equivalents of substrate. The probe Fur-SH showed high sensitivity to H2S, with LOD of 48.9 nM and LOQ of 50 nM. At present, the probe Fur-SH had been applied to fluorescence imaging of MCF-7 cells and zebrafish. By comparing the effects of different structures on the optical properties of fluorophores, this work was expected to be helpful to the development of fluorescent probes in the future.

Abstract:Novel cyanine-based fluorescent probes for the detection of H2S were developed. The probes developed are stable under physiological conditions. The water soluble fluorescent probe 2 displayed ultrafast and specific response to H2S displaying NIR fluorescence of 115-fold turn-on with the detection limit of 11 nM without assistance of organic solvent or surfactant. Cell imaging experiments indicated that probe 2 was cell-permeable and was able to detect H2S sensitively in lysosomes. Moreover, our probe was able to detect H2S intrinsically produced H2S through enzymatic/non-enzymatic biosynthetic pathway from Cys/GSH. Moreover, we applied probe 2 to detect H2S in living mice and demonstrated the fast metabolism of H2S. Thus, probe 2 shows great promise as a reporter for H2S.
