Abstract:A novel antioxidant peptide EDEQKFWGK from porcine plasma hydrolysate (PPH) was separated by chromatography, HPLC, and identified by LC-MS/MS. Results showed that EDEQKFWGK had better antioxidant ability (Hydroxyl RAS 32.19%, ABTS RAS 92.93% and DPPH RAS 26.76%) compared with glutathione (30.11%, 82.01%, 26.44%) due to the presence of hydrophobic, aromatic acids (F, W) and acidic amino acids (E, D), decreasing ROS by providing hydrogen atom and chelating metal ions. Furthermore, the antioxidant properties of synthetic EDEQKFWGK still significant despite in vitro digestion because of the production of smaller active peptide. Additionally, it could increase SOD, CAT, GSH-Px to resist oxidative damage in HepG2 cells by inhibiting ROS (O2- , OH·), forming complexes to prevent OH· from destroying DNA and binding to ARE to promote antioxidase expression. Thereby, the novel peptide EDEQKFWGK from porcine plasma had much stable antioxidant properties and hade great potential in formulating functional foods. PRACTICAL APPLICATIONS: This research isolated a novel antioxidant peptide. Moreover, the antioxidant effects of peptide were confirmed under the in vitro digestion model and oxidative damage HepG2 cells model. The results showed the antioxidant peptide could play better effect after digestion and protect the cells from oxidative damage. These data could expand the sequence data of antioxidant peptides and promote the high-value utilization of PPH.

Abstract:Caseins with known sequences were simulated peptide-cut into small peptides by 39 kinds of proteases using the software of peptidecutter,and the suitable effective proteases were chosen in terms of the amount of oligo-peptides and enzymatic peptide library was established.The most effective protease was determined in the confirmatory experiment.Angiotensin Ⅰ-converting enzyme inhibitory peptides(ACEIPs) and CPPs were obtained after ultra-filtration,and the inhibitory activity of ACEIP and the N/P ratio of CPP were used to assess the activity of ACEIP and the purity of CPP.The results implied that:the trypsin was chosen as the most suitable protease.Compared with the mixture before ultra-filtration,the activity of ACEIP and the purity of CPP were extremely significant improved(p<0.01).In this way,higher activity of ACEIPs and higher purity of CPPs were obtained.

Abstract:Various bioactive peptides are continuously being identified from casein hydrolysates. In this work, a novel angiotensin I-converting enzyme (ACE)-inhibitory (ACEI) peptide, NMAINPSKENLCSTFCK, derived from the alpha(s2)-casein fragment residues 25-41, was screened and identified by UPLC-ESI-Q-TOF-MS/MS from tryptic casein hydrolysate. The IC50 value of the peptide, determined by an HPLC method, was 129.07 mu M. The Lineweaver-Burk plot showed that this peptide acted as a mixed-type inhibitor against ACE, which might be attributed to the peptide being susceptible to degradation by ACE, indicating that the mixed-type inhibition could partly be a result of newly generated peptide fragments. The physicochemical characteristics and the secondary structure were evaluated by circular dichroism analysis and online prediction software (Expasy, PepDraw, and ProtParam) to identify the basic characteristics of this peptide. Moreover, molecular docking was simulated by Discovery Studio 2017 R2 software to provide the potential mechanisms underlying the ACEI activity of the peptides.

Abstract:Casein is an excellent source for producing anticoagulant and angiotensin I-converting enzyme inhibitory (ACEI) peptides. Here, the anticoagulant and ACEI activities of the casein hydrolysate produced by in vitro simulated gastrointestinal (GI) digestion were evaluated. The casein hydrolysate showed potent anticoagulant activity by prolonging the thrombin time (TT) and activated partial thromboplastin time (APTT), and also presenting great ACEI activity, with an IC50 value of 0.52 mg mL(-1). Subsequently, the transepithelial transport properties of the casein hydrolysate were analyzed by using the Caco-2 cell monolayer model. The peptides profile of the casein hydrolysate before and after it passed across the Caco-2 cell monolayer were identified by NanoLC-Q-TOF-MS/MS. The results showed that a total of 121 and 184 peptides were identified before and after casein hydrolysate moved through the Caco-2 cell monolayer, respectively. Eighty peptides were presented at both time points of the transport study. Among the 80 peptides, 26 of them were screened with a high possibility of exerting physiological roles after they were absorbed into the blood by in silico methods, and the physicochemical characteristics, e.g., hydrophobicity, net charge, and toxicity of the peptides also be evaluated. Our results provided a new prospect and approach for producing bioactive peptides from casein with anticoagulant and ACEI activities.

Abstract:Various bioactive peptides are identified from casein hydrolysates. YQEPVLGPVR (PICA), a novel antithrombotic peptide derived from beta-casein (fragment 193-202), was identified by high-performance liquid chromatography - liquid chromatography-mass spectrometry/mass spectrometry. The anticoagulation activity assay showed that this peptide has a strong anticoagulant activity. It was proved that the peptide did not interact with the active site of thrombin to inhibit thrombin, and that it inhibited thrombin activity by binding the exosite-1 of thrombin, which was also confirmed by the fibrinogen clotting time assay. It was shown that PICA prolonged fibrinogen clotting time in a dose-dependent manner. Secondary structures of the thrombin-PICA complex were also measured by circular dichroism to prove that PICA can combine with thrombin. Moreover, Discovery Studio 2017 R2 software was used for molecular docking to provide the potential mechanism for the antithrombotic activity of the peptide. These results suggested that PICA probably can be used as an antithrombotic ingredient in the functional food industry.

Abstract:There is no study on food-derived peptide with both anticoagulant and angiotensin I-converting enzyme inhibitory (ACEI) activities yet. In this work, the anticoagulant and ACEI activities of the casein hydrolysates released by pepsin digestion were evaluated for the first time to the best of our knowledge. Results indicated that the casein hydrolysate exhibited potent anticoagulant activity by prolonging the thrombin time (TT) and the activated partial thromboplastin time (APTT). Compared with control samples, at 10mg/mL, the TT and APTT of casein hydrolysate were 186.0 % ± 6.6 % and 163.5 % ± 7.4 %, respectively. The casein hydrolysate also showed a strong ACEI activity with an IC50 value of 1.775mg/mL. The components of the bioactive casein hydrolysate were analyzed by nanoscale liquid chromatography quadrupole time-of-flight tandem mass spectrometry (NanoLC-Q-TOF-MS/MS). Total of 115 peptides were identified, among which 34, 9, 55 and 17 peptides were derived from αs1-, αs2-, β-, and κ-casein, respectively. The results of PeptideRanker and PepSite 2 analysis showed that 6 peptides (FRQFYQL, NENLLRF, NPWDQVKR, PVVVPPFLQ, PVRGPFPIIV, and ARHPHPHLSF) have both ACEI and anticoagulant activities by binding to the active sites of ACE and thrombin. This study indicated that casein is a potential functional food supplement that can be used for medical purposes.

Abstract:The present study here establishes a complete and effective method for isolating, purifying and identifying extracellular and intracellular peptides, and also describes the characters and bioactivities of peptides from fermented casein with Lactobacillus helveticus. Intracellular peptides are much larger in quantity and more complex in composition than extracellular peptides, between which the correlation reveals proteolytic and metabolic mechanisms.In addition, totally 241 different peptide sequences were identified by Nano LC-MS/MS from casein (212) and Lactobacillus helveticus proteins (29). These casein-derived peptides mostly originated from beta-casein, followed by (61-casein, K-casein, and alpha 52-casein, and came from extracell (69) and intracell (143), in which common peptides have a total of 27. Forty-four of the identified peptides were previously described as bioactive, including angiotensin-converting enzyme (ACE)-inhibitory, antioxidant, immunomodulating, antimicrobial, DPP-IV inhibitory, antiamnesic and anticancer effects and so on. Thirteen peptides with the potential of some biological activities are obtained, which were described in previous studies. A total of 47 novel peptides of 5 to 26 amino acids that were not disclosed were obtained. The new sources of natural bioactive peptides may have the very high application value as potential new peptide drugs for treatment human diseases. The product peptide DELQDKIHPF found in both extracell and intracell was quantitatively analyzed using the MRM mode of UPLC-U3Q, 23.1 and 9.76 ng/mL, respectively. Abstract:Casein hydrolysates exert various biological activities, and the responsible functional peptides are being identified from them continuously. In this study, the tryptic casein hydrolysate was fractionated by an ultrafiltration membrane (3 kDa), and the peptides were identified by capillary electrophoresis-quadrupole-time-of-flight-tandem mass spectrometry. Meanwhile, in silico methods were used to analyze the toxicity, solubility, stability, and affinity between the peptides and angiotensin-I-converting enzyme (ACE). Finally, a new angiotensin-I-converting enzyme inhibitory (ACEI) peptide, EKVNELSK, derived from alpha(s1)-casein (fragment 35-42) was screened. The half maximal inhibitory concentration value of the peptide is 5.998 mM, which was determined by a high-performance liquid chromatography method. The Lineweaver-Burk plot indicated that this peptide is a mixed-type inhibitor against ACE. Moreover, Discovery Studio 2017 R2 software was adopted to perform molecular docking to propose the potential mechanisms underlying the ACEI activity of the peptide. These results indicated that EKVNELSK is a new ACEI peptide identified from casein hydrolysate.
Identification of a Novel ACE-inhibitory Peptide from Sheep Casein and Evaluation of the Molecular Binding Mechanism

Abstract:The purpose of this study was to prepare a novel Angiotensin-I-Converting Enzyme(ACE) inhibitory peptide by hydrolyzing sheep casein with protease and analyze its molecular inhibitory mechanism, so as to provide technical support for the development of functional sheep milk polypeptide dairy products. In this experiment, sheep casein was used as raw material, with hydrolysis degree, ACE inhibition rate and molecular weight distribution as indexes, the most suitable protease was selected from four proteases, and the components with less than <3 kDa was identified by LTQ Orbitrap Velos mass spectrometer. the potential ACE inhibitory peptides were selected for artificial synthesis and it half inhibitory concentration (IC50) was measured. The inhibition kinetics of enzyme was determined by Linewaver-Burk mapping, and the inhibition mechanism of peptide segment was further explained by molecular docking. The results showed that alkaline protease was the best hydrolytic protease. A new ACE inhibitory peptide KYIPIQY was screened from κ -casein and its IC50 was 5.73 μmol/L. Linewaver-Burk diagram showed that the peptide showed a mixed inhibition mode on ACE. Molecular docking showed that KYIPIQY can form hydrogen bonds with S1 and S2 active pockets of ACE, and closely combine with hydrophobic force. KYIPIQY can distort the Zn2+ tetrahedron of ACE, and inhibit the deactivation of ACE catalytic activity, thus exerting high-efficiency ACE inhibitory activity.
Bioactive hydrolysates from casein: Generation, identification, and in silico toxicity and allergenicity prediction of peptides.

Abstract:BACKGROUNDBioactive casein peptides have attracted considerable attention for their applications in industry. However, there is little clarity regarding mass spectrometric profiles for peptides in enzymatic hydrolysates of casein produced under varying conditions. In this study, the compositions of the peptides from casein hydrolysates were compared for different enzyme/substrate ratio (E/S) and hydrolysis times. The toxicity, allergenicity and bioactivity of the identified peptides were assessed in silico. RESULTSA total of 70 unique peptides were identified, and there were 28, 21, 13 and 8 peptides from (s1)-casein, (s2)-casein, -casein and -casein respectively. The peptide number decreased with the increase in E/S and hydrolysis time. Moreover, peptides with relative molecular mass M-r ranging from 1000 to 1500 Da occupied the highest proportion of 31.43%, and almost all of the peptides showed M-r less than 5000 Da. In silico analysis showed that all of the peptides were non-toxic and non-allergenic, and several of them were assessed by PeptideRanker as having a relatively high likelihood of being bioactive peptides. CONCLUSIONSComposition of the peptides in the casein hydrolysates varied with the enzymolysis conditions. This study's results may facilitate the production of target bioactive peptides by controlling E/S and hydrolysis time, which is beneficial for the application of casein peptides in the functional food industry. Abstract:Casein has been recognized as a good source of bioactive peptides that can be used for the production of functional food. In this study, thrombin inhibitory peptides from casein were screened by bioinformatics based on sequences predicted from in silico enzymatic digestion with trypsin, and identified by UPLC-Q-TOF-MS/MS with Mascot analysis. Molecular docking was conducted using the software of Discovery Studio 2017 to provide the potential mechanism underlying the thrombin inhibitory activity of the peptides. A total of 35 novel peptides with thrombin inhibitory activity were identified. Moreover, FQSEEQQQTEDELQDK, derived from beta-casein (fragment 33–48), showed a remarkable sequence similarity with fragment 54–65 of hirudin, and exhibited strong inhibitory activity against thrombin. FQSEEQQQTEDELQDK had similar target domain (Lys36-Gln38-Arg73-Thr74-Lys81-Ile82-Lys110) on the stereo structure of thrombin compared with hirudin. This domain belongs to the partial structure of the active site 2 of thrombin (PDB: 2BVR), and it also located at the exosite I motif buried in the external surface of thrombin molecule, with potential inhibitory activity of the peptide.

Abstract:In this study, casein derived peptides were hydrolyzed with trypsin and then transported across Caco-2 cell monolayers to choose intestinal absorbable antioxidant peptides. E/S ratio at 5.45 %, pH at 7.13 and temperature at 51.99 degrees C were found to be the optimal conditions for enzymatic hydrolysis of casein by trypsin by Response Surface Method. The fraction below 10 kDa purified by the hollow fiber ultrafiltration membrane was applied to Sephadex G-10 gel filtration chromatography, and the obtained components were transported across the Caco-2 cell monolayer. The transported peptides were detected on the basolateral side of the Caco-2 cell monolayer using ultra-performance liquid chromatography-tandem mass spectrometry. It was found that LHSMK was an unreported pentapeptide. The synthetic peptide LHSMK had free radical scavenging ability, and the antioxidant activity exhibited a dose-dependent increase. Furthermore, LHSMK can also regulate antioxidant enzymes activity in oxidative-damage cells. This study provides a method for the preparation of intestinal absorbable peptides, and provides basic research for the development of functional food market.

Abstract:A novel anticoagulant peptide (IEELEEELEAER) derived from oyster (Crassostrea gigas) was discovered by combining the emerging bioinformatics with the classical enzymolysis approach. The anticoagulant peptide drastically reduced the extrinsic clotting activity (49% residual PT activity) and impaired the intrinsic clotting activity (77% residual PT activity). Consistent with the clotting data, the thrombin peak height reduced to 88.7 from 123.4 nM, and the thrombin generation time delayed to 5.32 from 4.42 min when an extrinsic trigger was applied. The inhibitory kinetics of FXIa, FIXa, FXa, FIIa, and APC in a purified component system rationally explained the reduction of the extrinsic clotting activity and impairment of thrombin generation. Besides the inhibition of FXa and FIIa activity, the activation processes of FX and FII by an intrinsic/extrinsic tenase complex and prothrombinase were also damaged. The anticoagulant activity in the plasma system was the result of comprehensive inhibition of various factors. The research provided a frame for anticoagulant evaluation and inhibitory mechanism of bioactive peptides from food products.

Abstract:In this study, we identified a novel angiotensin-I-converting enzyme (ACE) inhibitory peptide, YQKFPQYLQY (YQK), derived from bovine casein. Casein was hydrolyzed using pepsin and trypsin. The target peptide, YQK, was separated from the hydrolysate by ultrafiltration and Sephadex G-15chromatography. The IC<sub>50</sub> value of YQK was 11.1μM. YQK retained its ACE inhibitory activity under various temperature and pH conditions. It was also stable against the digestive enzymes pepsin and trypsin. The Lineweaver-Burk plot suggested that the inhibitory mode of YQK was competitive. Furthermore, its antihypertensive effects in spontaneously hypertensive rats (SHRs) also revealed that oral administration of YQK can significantly decrease systolic blood pressure. These results suggested that YQK may have potential applications in functional foods or pharmaceuticals as an antihypertensive agent.

Abstract:A newly discovered anticoagulant peptide was isolated, purified and identified from the pepsin hydrolysate of oyster (Crassostrea gigas) which could potently prolong the activated partial thromboplastin time and the thrombin time. The anticoagulant peptide with a 1264.36 Da molecular mass was similar to the amino acid sequence of the C-terminal segment (DFEEIPEEYLQ) of hirudin (a potent thrombin inhibitor). The peptide specifically inhibited a vital blood coagulation factor: thrombin. The molecular docking energy scores of the anticoagulant peptide with the active site, exosite-I and exosite-II of thrombin were 132.355 kcal mol-1, 151.266 kcal mol-1 and 147.317 kcal mol-1, respectively. The anticoagulant peptide interacted with thrombin by competing with fibrinogen for an anion-binding exosite I. In the anticoagulant peptide-thrombin complex, there are seven hydrogen bonds and reciprocity exists between hydrogen atoms and oxygen atoms, and electrostatic and hydrophobic interactions are also involved. Such abundant interactions may be accountable for the high affinity and specificity of the anticoagulant peptide.

Abstract:Angiotensin I-converting enzyme (ACE) inhibitory activities in untreated koumiss and koumiss digested with ACE, pepsin, trypsinase, and chymotrypsin were compared and analyzed. Four novel ACE inhibitory peptides (PI, PK, PM, and PP) were purified using ultrafiltration and high performance liquid chromatography (HPLC). The classification study showed that these 4 peptides were of the true inhibitor type. The amino acid sequences of these peptides are YQDPRLGPTGELDPATQPIVAVHNPVIV, PKDLREN, LLLAHLL, and NHRNRMMDHVH, respectively. Their individual IC50 (50% inhibitory concentration) values were as follows: 14.53±0.21μM, 9.82±0.37μM, 5.19±0.18μM, and 13.42±0.17μM. From sequence analysis, we determined that PI was part of β-casein in mare's milk. The 3 peptides PK, PM, and PP did not correspond with any known milk protein. The results suggest that koumiss is rich in ACE inhibitory peptides, and the ACE inhibitors in koumiss are of the pro-drug type or a mixture of the pro-drug type and the true inhibitor type. These results may provide evidence about the beneficial effects of koumiss, especially on cardiovascular health.
Determination of A1 and A2 β-Casein in Milk Using Characteristic Thermolytic Peptides via Liquid Chromatography-Mass Spectrometry

Abstract:β-casein, a protein in milk and dairy products, has two main variant forms termed as A1 and A2. A1 β-casein may have adverse effects on humans. The fact that there is only one amino acid variation at the 67th position between A1 and A2 β-casein makes it difficult to distinguish between them. In this study, a novel method using characteristic thermolytic peptides is developed for the determination of A1 and A2 β-casein in milk. Firstly, caseins extracted from milk samples are thermolytic digested at 60 °C without any denaturing reagents required for unfolding proteins, which simplifies the sample pretreatment procedure. The characteristic thermolytic peptides (i.e., fragments 66–76 and 59–76 for A1 and A2 β-casein, respectively) selected to specifically distinguish A1 and A2 β-casein only have eleven or eighteen amino acid moieties. Compared with tryptic characteristic peptides with a length of 49 amino acid moieties, these shorter thermolytic characteristic peptides are more suitable for LC-MS analysis. This novel method, with the advantages of high specificity, high sensitivity, and high efficiency, was successfully applied for the analysis of six milk samples collected from a local supermarket. After further investigation, it is found that this method would contribute to the development of A2 dairy products for a company and the quality inspection of A2 dairy products for a government.

Abstract:An enzymatic hydrolysis approach was proposed for the preparation of bioactive hydrolysate of scorpion Buthus martensii Karsch (BmK) protein. Results showed that the anticoagulant activity of the hydrolysates of BmK protein was closely related to both the enzyme type and the degree of hydrolysis. Alcalase AF showed to be the best enzymes for the hydrolysis. And the hydrolysis degree (DH) was closely related with the anticoagulant activity of the hydrolyzate. At a DH value of 18% with Alcalase AF, the hydrolyzate exhibited the highest activity. The hydrolysate was then separated and purified by consecutive chromatographic procedures, giving a novel anticoagulant peptide consisting of ten amino acids (MW: 1119.8Da) with its sequence of Val-Glu-Pro-Val-Thr-Val-Asn-Pro- His-Glu identified by MALDI-TOF/TOF MS. The negatively charged amino acids and hydrophobic amino acids may contribute to the anticoagulant activity of the prepared peptides. And the Val residue in N-terminal was also critical for the anticoagulant activity of the BmK peptide. Furthermore, the anticoagulant activity kept stable after in vitro digestive simulation. This research provided a promising bioprocessing route for production of anticoagulant peptides using BmK protein as a potentially valuable bioresource.

Abstract:A comprehensive method was applied to evaluate the anticoagulant activity of a novel anticoagulant peptide (NAESLRK) derived from oyster (Crassostrea gigas). The anticoagulant peptide drastically reduced the extrinsic clotting activity and also impaired the intrinsic clotting activity slightly. Consistent with clotting data, the thrombin peak height was reduced to 84.7 nmol/L from 123.4 nmol/L, and thrombin generation time was delayed to 4.67 min from 4.42 min when the extrinsic trigger was applied. The inhibitory kinetics of FXIa, FIXa, FXa, FIIa, and APC in a purified component system rationally explained the reduction of extrinsic clotting activity and impairment of thrombin generation. Besides the inhibition of FXa and FIIa activity, the activation processes of FX and FII by intrinsic/extrinsic tenase complex and prothrombinase were also damaged. The anticoagulant activity in the plasma system was the result of comprehensive inhibition of various factors. The research provided a novel method for anticoagulant evaluation and inhibitory mechanism of bioactive peptides from food products.
