Adipose-Derived Stromal Cells (adscs) Promote Bone Formation in Vivo Not by Transforming to Osteoblasts

Abstract:Background: ADSCs (adipose-derived stromal cells) were deemed as perfect seed cells for bone tissue engineering. However, the mechanism of ADSCs promoting bone defect repair remains to be investigated. The present study aimed to verify if ADSCs were transformed to osteoblasts in vivo bone defect environment. Materials and Methods: ADSCs were infected with GFP adenoviral vectors. Based on porous hydroxyapatite/beta-tricalcium phosphate scaffolds and ADSCs, we prepared tissue-engineered constructs. Empty defects and scaffolds without ADSCs were set as controls. Histomorphometry was used to analyze bone formation four weeks after their implantation in critical calvarial defects of rats. Results: The difference between the scaffold group and cell-loaded group was statistically significant, and ADSCs can promote bone defect formation in vivo. Meanwhile, immunohistochemistry of GFP showed that ADSCs were not transformed to osteoblasts. To further verify the reliability of the experiment, ADSCs obtained from GFP transgenic C57 mice were used to repeat the experiment. It was also shown that ADSCs were not transformed to osteoblasts in vivo. Conclusion: ADSCs can promote the bone formation in vivo not by transforming to osteoblasts, and the exact processes involved are still largely unknown.
