Abstract:Barcode particles have a demonstrated value for multiplexed high-throughput bioassays. Attempts to develop this technology tend to focus on the generation of featured barcodes both with a large number of identifications to increase the throughput and with novel functions to improve the assays. Here, we report a new class of barcodes that are composed of multiple photonic crystal or magnetic-tagged ethoxylated trimethylolpropane triacrylate (ETPTA) cores and polyethylene glycol (PEG) hydrogel shells. These barcodes are prepared by polymerizing microfluidic multiple double emulsions. As the photonic crystal cores possess distinct reflection peaks, our barcodes allow for a substantial number of coding levels for multiplexing applications. The hydrogel shells surrounding the barcodes enable the creation of three-dimensional scaffolds for immobilizing probes. Moreover, the presence of magnetism in the barcodes confers their controllable movement under magnetic fields, which can be used to significantly increase the sensitivity of the bioassays and to simplify the processing. These features make photonic crystal barcodes ideal for biomedical applications. Biological systems are often studied through the use of assays, in which different experimental parameters are varied systematically. One method uses so-called barcode particles to track different samples. However this approach is complicated by the difficulty of producing barcode systems that allow for a sufficient number of configurations. Zhongze Gu and colleagues from Southeast University in Nanjing, China, have now developed a barcode system based on photonic crystals. These crystals, whose color reflection can be controlled during fabrication, are prepared with the help of a microfluidic system that also enables the simultaneous incorporation of the biomolecules to be probed. The barcodes particles also contain a magnetic tagged component that allows them to be moved in a magnetic field for easier read-out. By combining several types of photonic crystals in one unit, up to 1.28 million marker combinations are possible. Barcodes that composed of multiple colloidal crystal or magnetic-tagged ETPTA cores and PEG hydrogel shells were developed by using microfluidics. As the cores are with distinct reflection peaks, the barcodes allow for substantial number of coding levels for multiplexing. The hydrogel shells surrounding the barcodes enable the creation of three-dimensional scaffolds for immobilizing probes. Abstract:We developed a microfluidic method to form chromatic-pattern barcodes without using photomasks as photolithography methods. Two different aqueous quantum dot solutions were loaded as liquid-state cores to form two-component microcapsules, which present as chromatic patterns under UV illumination for barcoding. This microfluidic method highly simplifies the formation of patterns without using any expensive alignment instruments and allows creating the easily discernible pattern-type barcodes with a high coding capacity up to tens of thousands. The easily discernible chromatic-pattern barcodes with a small size down to 40 μm are very promising to conduct multiplexed biomolecular assays under microscopes in general labs.
Heteroepitaxial Growth of Multiblock Ln-MOF Microrods for Photonic Barcodes.

Abstract:Micro/nanoscale multicolor barcodes with unique identifiability and a small footprint play significant roles in applications such as multiplexed labeling and tracking systems. Now, a strategy is reported to design multicolor photonic barcodes based on 1D Ln-MOF multiblock heterostructures, where the domain-controlled emissive colors and different block lengths constitute the fingerprint of a corresponding heterostructure. The excellent heteroepitaxial growth characteristics of MOFs enable the effective modulation of the coding structures, thereby remarkably increasing the encoding capacity. The as-prepared multicolor barcodes enable an efficient authentication and exhibit great potential in fulfilling the functions of anti-counterfeiting, information security, and so on. The results will pave an avenue to novel hybrid MOFs for optical data recording and security labels.

Abstract:Barcode particles have a demonstrated value for multiplexed high-throughput bioassays. Here, a novel photonic crystal (PhC) barcode is presented that consists of hollow colloidal nanospheres assembled through microfluidic droplet templates. Due to their gas-filled core, the resultant barcode particles not only show increased refractive index contrast, but also remain in suspension by adjusting the overall density of the PhC to match that of a detection solution. In addition, magnetic nanoparticles can be integrated to give the barcodes a magnetically controllable motion ability. The encoding ability of the barcodes is demonstrated in microRNA detection with high specificity and sensitivity, and the excellent features of the barcodes make them potentially very useful for biomedical applications.

Abstract:Photonic crystal (PC) barcodes are a new type of spectrum-encoding microcarriers used in multiplex high-throughput bioassays, such as broad analysis of biomarkers for clinical diagnosis, gene expression, and cell culture. Unfortunately, most of these existing PC barcodes suffered from undesired features, including difficult spectrum-signal acquisition, weak mechanical strength, and high ontology fluorescence, which limited their development to real applications. To address these limitations, we report a new type of structural color-encoded PC barcodes. The barcodes are fabricated by the assembly of monodisperse polydopamine- (PDA-) coated silica (PDA@SiO2) nanoparticles using a droplet-based microfluidic technique and followed by pyrolysis of PDA@SiO2 (C@SiO2) barcodes. Because of the templated carbonization of adhesive PDA, the prepared C@SiO2 PC beads were endowed with simultaneous easy-to-identify structural color, high mechanical strength, and ultralow ontology fluorescence. We demonstrated that the structural colored C@SiO2 barcodes not only maintained a high structural stability and good biocompatibility during the coculturing with fibroblasts and tumor cells capture but also achieved an enhanced fluorescent-reading signal-to-noise ratio in the fluorescence-reading detection. These features make the C@SiO2 PC barcodes versatile for expansive application in fluorescence-reading-based multibioassays.

Abstract:Micron-scale barcode particles enable labelling of small objects. Here, we demonstrate high-throughput barcode fabrication inside a microfluidic chip that can embed multiple, dye-doped high quality-factor whispering gallery mode cavities inside aqueous droplets at kilohertz rates. These droplets are then cured to form polyacrylamide hydrogel beads as small as 30 μm in diameter. Optical resonance spectra of the embedded cavities provide the hydrogels with unique barcodes with their diversity combinatorically scaled with the number of embedded cavities. Using 3 cavities per hydrogel, we obtain approximately one million uniquely identifiable, optically readable barcode microparticles.

Abstract:Micro/nanoscale photonic barcodes hold great potential for broad applications in items tracking, multiplexed bioassays and anti-counterfeiting. The ever-increasing demand in advanced anti-counterfeiting applications calls for micro/nanoscale barcodes with accurate recognition, large encoding capacity and high security level. Here, we proposed a strategy to construct the dual-stimuli responsive photonic barcodes based on the perovskite quantum dots (PQDs) doped polymer whispering-gallery-mode (WGM) microcavities via swelling-deswelling method. Benefiting from the well-defined spherical microcavities, the photoluminescence (PL) spectra of as-prepared composites exhibit a series of sharp peaks characteristics resulting from the effective WGM modulation, which constitutes the fingerprint of a specific resonator and thus allows a definition of photonic barcodes. On this basis, we achieved responsive photonic barcodes based on the volatile polar-solvent-controlled luminescence in the microspheres benefitting from the space-confined microcavities and the ionic feature of the PQDs. Moreover, the light-controlled photonic barcodes have further been acquired through reversibly regulating the inactivation and activation of the energy transfer (ET) process between the PQDs and photochromic dyes. The well-established protocols of PQDs@WGM enable the development of distinct responsive barcodes with multi-responsive features, which will pave an avenue to new types of flexible WGM-based components for optical data recording and security labels.

Abstract:The design of nanosegregated fluorescent tags/barcodes by geometrical patterning with precise dimensions and hierarchies could integrate multilevel optical information within one carrier and enhance microsized barcoding techniques for ultrahigh-density optical data storage and encryption. However, precise control of the spatial distribution in micro/nanosized matrices intrinsically limits the accessible barcoding applications in terms of material design and construction. Here, crystallization forces are leveraged to enable a rapid, programmable molecular packing and rapid epitaxial growth of fluorescent units in 2D via crystallization-driven self-assembly. The fluorescence encoding density, scalability, information storage capacity, and decoding techniques of the robust 2D polymeric barcoding platform are explored systematically. These results provide both a theoretical and an experimental foundation for expanding the fluorescence storage capacity, which is a longstanding challenge in state-of-the-art microbarcoding techniques and establish a generalized and adaptable coding platform for high-throughput analysis and optical multiplexing.
Predefinable Colorimetric Quantum-Dot Barcodes with Simple and Express Identification Algorithm.

Abstract:Specific fluorescent profiles were created by loading quantum-dot (Qdot) mixtures in liquid cores of monodispersed polymer microcapsules, which were used as colorimetric barcodes for small object identification. Since the emission intensities of Qdot-loaded liquid cores maintain a linear relation to the Qdot concentrations and the Qdots in the liquid cores with different emission peaks have no obvious interference, the colorimetric barcodes can be predefined by the compositions of Qdot mixtures. The colorimetric barcodes can be identified easily by recording the emission intensities of the encoded microcapsules at respective Qdot emission peaks with a simple and express algorithm, which are suitable to conduct high-throughput multiplexed assays by flow cytometer for biological screening applications.

Abstract:Multiplexed optical encoding is emerging as a powerful technique for high-throughput cellular analysis and molecular assays. Most of the developed optical barcodes, however, either suffer from large particle size or are incompatible with most commercial optical instruments. Here, a new type of nanoscale fluorescent barcode (Pdot barcodes) was prepared from semiconducting polymers. The Pdot barcodes possess the merits of small size (∼20 nm in diameter), narrow emission bands (full-width-at-half-maximum (fwhm) of 30-40 nm), three-color emissions (blue, green, and red) under single-wavelength excitation, a high brightness, good pH and thermal stability, and efficient cellular uptake. The Pdot barcodes were prepared using a three-color and six-intensity encoding strategy; for ratiometric readout of the barcodes, one of the colors might be used as an internal reference. We used the Pdot barcodes to label 20 sets of cancer cells and then distinguished and identified each set based on the Pdot barcodes using flow cytometry. We also monitored and tracked single cells labeled with different Pdot barcodes, even through rounds of cell division. These results suggest Pdot barcodes are strong candidates for discriminating different labeled cell and for long-term cell tracking.

Abstract:With the rapid advancement of fluorescence microscopy, there is a growing interest in the multiplexed detection and identification of various bioanalytes (e.g., nucleic acids and proteins) for efficient sample processing and analysis. We introduce in this work a simple and robust method to provide combinations for micrometer-scale fluorescent DNA barcodes of hierarchically assembled DNA origami superstructures for multiplexed molecular probing. In addition to optically resolvable dots, we placed fluorescent loci on adjacent origami within the diffraction limit of each other, rendering them as unresolvable bars of measurable lengths. We created a basic set of barcodes and trained a machine learning algorithm to process and identify individual barcodes from raw images with high accuracy. Moreover, we demonstrated that the number of combinations can be increased exponentially by generating longer barcodes, by controlling the number of incorporated fluorophores to create multiple levels of fluorescence intensity, and by employing super-resolution imaging. To showcase the readiness of the barcodes for applications, we used our barcodes to capture and identify target nucleic acid sequences and for simultaneous multiplexed characterization of binding kinetics of several orthogonal complementary nucleic acids.

Abstract:A novel strategy for constructing photonic barcodes with encoding multi‐level complexity, based on dye@metal‐organic framework (MOF) heterostructures, is developed. Dual‐wavelength polarized near‐infrared (NIR) lasing can be switched and manipulated orientationally, thus converted to the dynamic photonic barcodes for high‐level security. Developing functional photonic tags is of great importance to meet the globally increasing demand for information security and anti‐counterfeiting. However, it still suffers from the restricted encoding capacity, low complexity, and unsatisfactory confidentiality. Herein, a strategy is demonstrated to construct multi‐level complex photonic barcodes in near‐infrared (NIR) region based on a composition‐graded metal‐organic framework (MOF) heterostructure with the hierarchical encapsulation of different dye molecules. Such MOF microstructure exhibits switchable dual‐wavelength NIR lasing by adjusting the excited position, polarization direction, and pump energy. Thus, through converting the laser spectra with dynamic and regular changes to solid bars, the multi‐block photonic barcodes with three‐level encoding system are constructed, dramatically increasing the complexity and capacity. It makes the MOF‐based heterostructure with switchable lasing properties become a hard‐to‐counterfeit potential material for advanced anticounterfeiting of high‐level security.

Abstract:Perovskite quantum dots (PQDs) are new class of optoelectronic materials, which have been widely studied for their extraordinary physical properties. Attempts to develop these materials are tending to make their fabrication much controllable and extend their values in different areas. Here, we present a novel strategy for one-step in situ synthesis of PQD-encapsulated barcode particles with the assistance of microfluidic technique. By changing the halide ratio in perovskite precursor solutions that emulsified in microfluidic devices, a series of PQDs with different colors have been successfully fabricated, which made them ideal materials as barcodes. Because of the stable encapsulation of ethyleneglycol dimethacrylate (EGDMA) resin, the PQD-encapsulated barcode particles were with no cytotoxicity and could be anti-quenched. It was demonstrated for the first time that the microfluidic encapsulated PQDs barcode particles were valuable for multiplex biomolecular encoding and assays. These features indicate that the microfluidic encapsulated PQDs barcode particles are ideal for many practical applications and have a broad prospect in biomedical field.

Structural Color Barcodes for Biodiagnostics

Abstract:Suspension array technology has revolutionized the field of biodiagnostics due to its capability of multiplex assays. Construction of the barcodes is essential, where structural color barcode is one of the most satisfying candidates because of its stable encoding and convenient decoding features. Benefiting from microfluidics technique, structural color barcode particles have been synthesized with finely tuned size, configuration, and surface reactive moieties. This allows for the barcode's interactions with disease‐specific biomarkers at multiscale, from the molecular to the tissue/organ level. In this minireview, we present the very latest progress on structural color barcodes for biodiagnostic applications. We discuss in depth through typical examples of how the sensitive and efficient assays of different types of analytes are accomplished concerning the specific features entailed in the barcodes. We also provide critical thinking about the remaining challenges and future development of structural color barcodes from a biodiagnostics perspective.
Expanded Single-Color Barcoding in Microspheres with Fluorescence Anisotropy for Multiplexed Biochemical Detection.

Abstract:Single-color barcoding strategies could break the limits of spectral crosstalk in conventional intensity-based fluorescence barcodes. Fluorescence anisotropy (FA), a self-referencing quantity able to differentiate spectrally similar fluorophores, is highly attractive in designing fluorescent barcodes within a limited emission window. In this study, FA-based encoding of polystyrene (PS) microspheres was realized for the first time. The FA signals of fluorophores were stabilized inside PS microspheres owing to hampered rotational motion. Fluorescent labels were incorporated with similar emission but different structures, symmetries, and lifetimes. On the one hand, Förster Resonance Energy Transfer (FRET) including homo-FRET and hetero-FRET resulted in a decrease of steady-state FA with increasing dye loading, converting conventional intensity-based codes into FA-based codes. On the other hand, mixing dyes with different intrinsic FA values generated different FA values at the same fluorescence intensity level. Single color 5-plex FA-encoded microspheres were demonstrated and decoded on a homemade microscopic FA imaging platform in real time. The FA-encoded microspheres were successfully applied to detect the oligonucleotide of the foodborne bacterium, Bacillus cereus, without spectral crosstalk between the encoding and reporting dyes. Overall, FA-based encoding with an expanded coding capacity in the FA dimension holds great potential in multiplexed high-throughput chemical and biological analyses.

Abstract:Microprinting is changing anti-counterfeit technology by producing complex micro-/nano-scale labels to increase the security level. However, the current microprinted anti-counterfeiting labels only contain 2D features, thought of as single coding (e.g. mostly in texts or graphs), unable to satisfy the high-end anti-counterfeiting demands of producing multi-coding, high-storage-density labels. Here, we introduce an approach for fluorescence printing to produce a 3D colourful, sophisticated anti-counterfeiting label. In the method, pre-synthetic QDs are printed with photoresists based on two-photon polymerisation, allowing for accurate regulation of fluorescence in both 2D and 3D. The fabricated photoresists exhibit immediate and postprocessing-free colour emissions under UV light. We demonstrated the ultra-high free design and accurate manufacturing capability of the approach by fabricating multi-colour, multi-layer patterns containing both texts and graphs on a 50μm × 50μm × 30μm portable label. The approach can also be applied to multi-dimensional optical data storage and colourful microdisplays.

Precisely Encoded Barcodes Using Tetrapod CdSe/CdS Quantum Dots with a Large Stokes Shift for Multiplexed Detection

Abstract:A serious obstacle to the construction of high‐capacity optical barcodes in suspension array technology is energy transfer, which can prompt unpredictable barcode signals, limited barcode numbers, and the need for an unfeasible number of experimental iterations. This work reports an effective and simple way to eliminate energy transfer in multicolor quantum dots (QDs)‐encoded microbeads by incorporating tetrapod CdSe/CdS QDs with a large Stokes shift (about 180 nm). Exploiting this unique feature enables the facile realization of a theoretical 7 × 7‐1 barcoding matrix combining two colors and seven intensity levels. As such, microbeads containing tetrapod CdSe/CdS QDs are demonstrated to possess a powerful encoding capacity which allows for precise barcode design. The ability of the Shirasu porous glass membrane emulsification method to easily control microbead size facilitates the establishment of a 3D barcode library of 144 distinguishable barcodes, indicating the enormous potential to enable large‐scale multiplexed detection. Moreover, when applied for the multiplexed detection of five common allergens, these barcodes exhibit superior detection performance (limit of detection: 0.01–0.02 IU mL−1) for both spiked and patient serum samples. Therefore, this new coding strategy helps to expand barcoding capacity while simultaneously reducing the technical and economic barriers to the optical encoding of microbeads for high‐throughput multiplexed detection.
Fluorescence-profile pre-definable quantum-dot barcodes in liquid-core microcapsules

Abstract:Microparticles incorporated with quantum-dot (QD) barcodes for multiplexed bioassays attract a great attention due to their potential applications in drug discovery, gene profiling and clinic diagnostics. However, the existing QD barcodes lack a necessary optical stability to ambient fluids or a repeatability of fluorescent profiles. We developed a new QD barcode by loading an aqueous QD mixture as a liquid core into a monodispersed polymer microcapsule by a microfluidic method to avoid those problems. We found that the QDs in the liquid cores were able to maintain their original characteristics, especially the linear relation between photoluminescence intensity and concentration. In addition, we found that the fluorescent profiles of the QD-loaded liquid cores were the same as those of the QD mixtures before being loaded inside the microcapsules. With these two properties, the QD barcodes can be predefined directly by multiplexing the emission peaks and concentrations of the QDs in the liquid cores. Furthermore, the graphical information from fluorescent images of the microcapsules, such as the sizes and numbers of the QD-loaded liquid cores, offers another dimension for barcoding to increase the coding capacity. We also presented a microfluidic method to manufacture the QD-barcoded microcapsules of size ~30 μm. These pre-definable QD barcodes with stable fluorescent profiles can be used as a platform for various high-throughput screening applications in different bioassay buffers.

Abstract:We develop a new strategy to prepare quantum dot (QD) barcode particles by polymerizing double-emulsion droplets prepared in capillary microfluidic devices. The resultant barcode particles are composed of stable QD-tagged core particles surrounded by hydrogel shells. These particles exhibit uniform spectral characteristics and excellent coding capability, as confirmed by photoluminescence analyses. By using double-emulsion droplets with two inner droplets of distinct phases as templates, we have also fabricated anisotropic magnetic barcode particles with two separate cores or with a Janus core. These particles enable optical encoding and magnetic separation, thus making them excellent functional barcode particles in biomedical applications.
