Abstract:A novel cascade radical addition/cyclization reaction of non-activated olefins and oxamic acids has been proposed. Under transition metal-free conditions, 36 quinazolinone derivatives containing an amide moiety were successfully synthesized, with the highest yield being 81%. This method involves the preparation of aminoacyl fused quinazolinone derivatives under mild conditions, offering advantages such as a high yield, a broad substrate compatibility, and a high atom economy.

Abstract:2-Perfluoroalkylated quinolines were efficiently synthesized via the Michael addition of perfluoroalk-2-ynoates with isatins, followed by the reaction with alcohols through an intramolecular rearrangement cyclization in the presence of Na2CO3. Under the mild and metal-free reaction conditions, a broad scope of quinolines was synthesized and the mechanism was proposed.

Abstract:Atom- and step-economy in IBX assisted diversity-oriented synthesis is achieved with a versatile AQ auxiliary α-amino acid analogs offering rapid access to polycyclic spiro-quinolines featuring a quaternary stereocenter in 20%–91% yields under mild conditions via 7,8-dearomatization of quinolines. Free of a preinstalled activation group is highlight of this intramolecular oxidation spiroannulation tandem reaction. This type of N-heterospirocycles, traditionally difficult to access, may open the door to a potentially interest scaffold for synthetic and medicinal chemistry.

Abstract:An efficient method for the synthesis of substituted quinolines is described. By FeCl3-promoted C–C bond formation, 2,3-disubstituted and 2,3,4-trisubstituted quinolines were successfully synthesized from the reaction between imines and electron-deficient alkynes in good yields. The reaction can also be performed in a sequential three-component manner by using aromatic amines and aromatic aldehydes instead of the corresponding imines.

Abstract:A wide range of isoquinoline ring-modified Quinap oxides with different steric and electronic variations have been constructed through a palladium-catalyzed imidoylative cyclization of arylethenyl isocyanides with 2-diphenylphosphinyl-1-naphthyl bromides. The Pd-catalysis plays dual roles in the formation of both axial C–C bond and isoquinoline ring. This de novo synthetic strategy features good functional group tolerance, high yields and easy scale-up, providing Quinap derivatives with substitution patterns that could not be obtained using coupling reactions. Chiral ligands 7 and 12 can be readily prepared by transformation of the resulting Quinap oxide to their BINOL esters, and have been proven to be superb chiral ligands for the copper-catalyzed enantioselective A3-coupling and alkynylation of quinoline reactions. In general, the enantioselectivies obtained using ligands 7 and 12 are excellent, and the ee values are higher than those using Quinap as ligand, even three times higher in some cases. Mechanism studies revealed that a monomeric copper(I) complex bearing a single chiral ligand was formed and served as the catalytically active species.

Abstract:C-N bond formation is crucial for the preparation of synthetic organic nitrogen-containing heterocyclic compounds. This work provides a facile and highly efficient strategy (ca. similar to 92 %) for the synthesis of quinazoline compounds, which involves tandem C-H/N-H bond functionalization using metal-free conditions without the need for an external oxidant. Meanwhile, the reaction system exhibits good functional group tolerance and a wide substrate scope. Furthermore, our corresponding isotope tracking experiments have shown that dimethyl sulfoxide acts as a carbon source. Finally, this work affords a new strategy toward the further design and synthesis of heteroaromatic rings.

Abstract:An iodine‐promoted one‐pot cascade oxidative annulation reaction has been developed for the synthesis of [1,2,3]triazolo[1,5‐a]quinolines from methyl azaarenes and N‐tosylhydrazines. The reaction has a broad substrate scope and can be easily scaled up to gram‐scale. 1,2,3‐Triazoles are an important skeletal structure for the construction of C‐C and C‐P bonds, 2‐cyclopropylquinolines and imidazo[1,5‐a]quinolines, for which different synthetic applications were explored.

Abstract:This research work entails single‐pot construction of 4‐aryl imidazoquinoxaline scaffolds with high yields in the presence of molecular iodine‐DMSO as a green catalyst under classical heating at 100–110 °C. The structures of the synthesized 4‐aryl imidazoquinoxalines were established by Mass, 1H NMR, and 13C NMR spectroscopic analysis. The prominent features of this protocol include broad compatibility of various arylacetic acids, commercially viable and inexpensive catalyst, user friendly solvent, low catalyst loading, no additional oxidizing agents, and scale amicable procedure. A one‐pot synthetic protocol has been developed using aryl acetic acid as new and alternate surrogate for the synthesis of 4‐aryl imidazoquinoxalines with molecular iodine‐DMSO green catalytic system under classical heating at 100–110 °C. A variety of quinoxaline derivatives containing quinoline are prepared by using this methodology. This new protocol has a potential to tolerate broad substrates, that is, different amines and aryl acetic acids.

Abstract:Multi-heteroatom heterocycle synthesis through direct C-H bond activation is methodologically appealing but synthetically challenging. An efficient double C-N bond formation sequence to prepare quinazolinones utilizing primary amides and oxadiazolones in a catalytic redox-neutral [CoCp*(CO)I-2]/AgSbF6 system, where oxadiazolone could function as an internal oxidant to maintain the catalytic cycle, is reported. Amide-directed C-H bond activation and oxadiazolone decarboxylation are key to the success of this traceless, atom- and step-economic, and cascade approach for the construction of the quinazolinone skeleton.

Abstract:A series of benzimidazo [1,2-c] quinazolines have been synthesized via phenyliodine(III) diacetate (PIDA)-mediated intramolecular C-H bond cycloamination reaction. This method results in a direct oxidative C-N bond formation in a complex molecule by using a metal-free protocol. A plausible reaction mechanism was described on the basis of the experiments. Some new compounds were evaluated for their antitumor activity against HUH 7 and SK-HEP-1 hepatocarcinoma cell line. Among the compounds screened, 4m was found to be the most active compound against HUH 7.

Abstract:A hypervalent iodine(III)-mediated intramolecular decarboxylative Heck-type reaction of 2-vinyl-phenyl oxamic acids has been developed. The unique ring-strain-enabled radical decarboxylation mechanism is preliminarily revealed. This protocol features metal-free reaction conditions and operational simplicity, allowing the lactamization of 2-vinylanilines using a readily accessible carbonyl source and the synthesis of various 2-quinolinones with excellent chemoselectivity at room temperature.

Abstract:An efficient I-2-catalyzed cascade coupling protocol was developed for the synthesis of pyrrolo[1,2-a]-quinoxaline and imidazo[1,5-a]quinoxaline derivatives via sp(3) and sp(2) C-H cross-dehydrogenative coupling. A nontoxic, readily available catalyst, I-2, and an oxidant, DMSO, were used in this metal-free process. The target compounds were obtained in good-to-excellent yields with a broad substrate scope.
