Human Periodontal Ligament Stem Cell Spheroids Afford Enhanced Osteogenic Potency In Vitro and In Vivo

Abstract:The aim of this study was to investigate the osteogenic potency of human periodontal ligament stem cell (hPDLSC) spheroids generated in pre-formed agarose hydrogel microwells. hPDLSCs were cultured in a microwell-based agarose plate or a traditional tissue culture dish. The formation, characteristics, and proliferation of hPDLSCs were investigated. The cellular viability of hPDLSCs in spheroids was evaluated. In vitro, osteogenic mRNA marker expression and osteogenic potential were compared between spheroid- and monolayer-cultured hPDLSCs by qRT-PCR assay, Alizarin Red-S staining assay and alkaline phosphatase activity assay. In vivo, the subcutaneous transplantations of hPDLSC spheroids and monolayer-cultured hPDLSCs on calcium phosphate ceramic were performed in Balb/c nude mice. The potency of the spheroids for mineralized osteogenic tissue formation was assessed using HE and Masson's trichrome staining. hPDLSCs in microwell-based agarose plates were able to form spheroids 24 h after seeding and remained viable with approximately 83-90% cell viability within 7 days. The mRNA expression levels of osteogenic genes were significantly up-regulated in spheroid- derived hPDLSCs. Spheroid-derived hPDLSCs exhibited significant mineral matrix deposition and enhanced alkaline phosphatase activity. The in vivo study demonstrated the formation of bone-like tissue. The generation of hPDLSC spheroids presents a promising approach for the enhanced osteogenic potency of cells.
