Abstract:Near-infrared (NIR, 700-900 nm) fluorescent quantum dots are highly promising as NIR bioprobes for high-resolution and high-sensitivity bioimaging applications. In this article, we present a class of NIR-emitting CdTe/CdS/ZnS core-shell-shell quantum dots (QDs), which are directly prepared in aqueous phase via a facile microwave synthesis. Significantly, the prepared NIR-emitting QDs possess excellent aqueous dispersibility, strong photoluminescence, favorable biocompatibility, robust storage-, chemical-, and photo-stability, and finely tunable emission in the NIR range (700-800 nm). The QDs are readily functionalized with antibodies for use in immunofluorescent bioimaging, yielding highly spectrally and spatially resolved emission for in vitro and in vivo imaging. In comparison to the large size of 15-30 nm of the conventional NIR QDs, the extremely small size (similar to 4.2 nm or 7.5 nm measured by TEM or DLS, respectively) of our QDs offers great opportunities for high-efficiency and high-sensitivity targeted imaging in cells and animals. Abstract:Carbon dots (CDots) are brightly fluorescent, especially in the same green spectral region overlapping that of the green fluorescent protein, and also benign and nontoxic. For imaging invivo, however, red/near-IR probes are required for the high optical transmittance through tissues and the contrast against interference from autofluorescence background. Herein, high-performance red/near-IR CDots are designed and prepared by modifying the core carbon nanoparticles in CDots with the incorporation of selected dye species into the nanoparticle structure. The representative modified CDots thus obtained are used as fluorescence probes in the invivo imaging of normal and tumor-bearing nude mice via various exposure pathways. The results suggest that these red/near-IR CDots are indeed excellent probes for fluorescence imaging invivo, especially with their enhanced tumor uptake. The potentially far-reaching implications of being able to design and synthesize the modified CDots of strong optical absorptions and bright fluorescence emissions in the red/near-IR are discussed in terms of their broad applicability to invivo imaging and theranostics.

Abstract:In this paper, we report the use of near-infrared (NIR)-emitting alloyed quantum dots (QDs) as efficient optical probes for high contrast in vivo imaging of tumors. Alloyed CdTe1-xSex/CdS QDs were prepared in the non-aqueous phase using the hot colloidal synthesis approach. Water dispersion of the QDs were accomplished by their encapsulation within polyethyleneglycol (PEG)-grafted phospholipid micelles. For tumor-specific delivery in vivo, the micelle-encapsulated QDs were conjugated with the cyclic arginine-glycine-aspartic acid (cRGD) peptide, which targets the alpha(nu)beta(3) integrins overexpressed in the angiogenic tumor vasculatures. Using in vivo NIR optical imaging of mice bearing pancreatic cancer xenografts, implanted both subcutaneously and orthotopically, we have demonstrated that systemically delivered cRGD-conjugated QDs, but not the unconjugated ones, can efficiently target and label the tumors with high signal-to-noise ratio. Histopathological analysis of major organs of the treated mice showed no evidence of systemic toxicity associated with these QDs. These experiments suggest that cRGD-conjugated NIR QDs can serve as safe and efficient probes for optical bioimaging of tumors in vivo. Furthermore, by co-encapsulating these QDs and anticancer drugs within these micelles, we have demonstrated a promising theranostic, nanosized platform for both cancer imaging and therapy.

Abstract:The rapid development of fluorescence imaging technologies requires concurrent improvements in the performance of fluorescent probes. Quantum dots have been extensively used as an imaging probe in various research areas because of their inherent advantages based on unique optical and electronic properties. However, their clinical translation has been limited by the potential toxicity especially from cadmium. Here, a versatile bioimaging probe is developed by using highly luminescent cadmium-free CuInSe2/ZnS core/shell quantum dots conjugated with CGKRK (Cys-Gly-Lys-Arg-Lys) tumor-targeting peptides. This probe exhibits excellent photostability, reasonably long circulation time, minimal toxicity, and strong tumor-specific homing property. The most important feature of this probe is that it shows distinctive versatility in tumor-targeted multimodal imaging including near-infrared, time-gated, and two-photon imaging in different tumor models. In a glioblastoma mouse model, the targeted probe clearly denotes tumor boundaries and positively labels a population of diffusely infiltrating tumor cells, suggesting its utility in precise tumor detection during surgery. This work lays a foundation for potential clinical translation of the probe.

Abstract:Molecular imaging plays a key role in personalized medicine, which is the goal and future of patient management. Among the various molecular imaging modalities, optical imaging may be the fastest growing area for bioanalysis, and the major reason is the research on fluorescence semiconductor quantum dots (QDs) and dyes have evolved over the past two decades. The great efforts on the synthesis of QDs with fluorescence emission from UV to near-infrared (NIR) regions speed up the studies of QDs as optical probes for in vitro and in vivo molecular imaging. For in vivo applications, the fluorescent emission wavelength ideally should be in a region of the spectrum where blood and tissue absorb minimally and tissue penetration reach maximally, which is NIR region (typically 700-1000 nm). The goal of this review is to provide readers the basics of NIR-emitting QDs, the bioconjugate chemistry of QDs, and their applications for diagnostic tumor imaging. We will also discuss the benefits, challenges, limitations, perspective, and the future scope of NIR-emitting QDs for tumor imaging applications.

Abstract:Accurate diagnosis and treatment of tumors, one of the top global health problems, has always been the research focus of scientists and doctors. Near-infrared (NIR) emissive semiconducting polymers dots (Pdots) have demonstrated bright prospects in field of in vivo tumor fluorescence imaging owing to some of their intrinsic advantages, including good water-dispersibility, facile surface-functionalization, easily tunable optical properties, and good biocompatibility. During recent years, much effort has been devoted to developing Pdots with emission bands located in the second near-infrared (NIR-II, 1000-1700 nm) region, which hold great advantages of higher spatial resolution, better signal-to-background ratios (SBR), and deeper tissue penetration for solid-tumor imaging in comparison with the visible region (400-680 nm) and the first near-infrared (NIR-I, 680-900 nm) window, by virtue of the reduced tissue autofluorescence, minimal photon scattering, and low photon absorption. In this review, we mainly summarize the latest advances of NIR-II emissive semiconducting Pdots for in vivo tumor fluorescence imaging, including molecular engineering to improve the fluorescence quantum yields and surface functionalization to elevate the tumor-targeting capability. We also present several NIR-II theranostic Pdots used for integrated tumor fluorescence diagnosis and photothermal/photodynamic therapy. Finally, we give our perspectives on future developments in this field.

Abstract:Specific labeling of biomarkers with bright and high photostable fluorophores is vital in fluorescent imaging applications. Here, we report a general strategy to develop single-molecule dendritic nanodots with finely tunable optical properties for in vivo fluorescent imaging. The well-defined nanodots are based on the divergent growth of biodegradable polylysine dendrimers with fluorophore as the core. By tuning the size and surface chemistry, we obtained fluorescent nanodots with excellent brightness and photostability, favorable pharmacokinetics, and multivalent tumor-targeting capability. The nanodots provided robust, stable, long-lasting, and specific fluorescence enhancement in tumor tissue with an in situ tumor-to-normal ratio (TNR) of ~3 and lasting over 5 days, and ex vivo TNR up to ~17, holding considerable promise for cancer imaging and image-guided surgery. This strategy significantly improves the in vivo performance of fluorophores and can be applied to other modality imaging probes.

Abstract:Early detection and subsequent complete surgical resection are among the most efficient methods for treating cancer. However, low detection sensitivity and incomplete tumor resection are two challenging issues. Nanoparticle-based imaging-guided surgery has proven promising for cancer-targeted imaging and subsequent debulking surgery. Particularly, the use of near infrared (NIR) fluorescent probes such as NIR quantum dots (QDs) allows deep penetration and high sensitivity for tumor detection. In this study, NIR-emitting CdTe QDs (maximum fluorescence emission peak at 728 nm) were synthesized with a high quantum yield (QY) of 38%. The tumor-specific QD bioconjugates were obtained by attaching cyclic Arg-Gly-Asp peptide (cRGD) to the surface of synthesized QDs, and then injected into U87 MG tumor-bearing mice via tail veins for tumor-targeted imaging. The tumor and its margins were visualized and distinguished by NIR QD bioconjugates, and tumor resection was successfully accomplished via NIR guidance using a Fluobeam-700 NIR imaging system. Our work indicates that the synthesized tumor-specific NIR QDs hold great promise as a potential fluorescent indicator for intraoperative tumor imaging.

Abstract:Fluorescence probes emitting in the second near-infrared (NIR-II, 1000–1700 nm) window with the ability for deep-tissue imaging in mammals herald a new era in surgical methodology. However, the brightness of these NIR-II probes is still far from satisfactory due to their low fluorescence quantum yields (QYs), preventing the observation of high-resolution images such as whole-organ vascular networks in real time. Described here is the molecular engineering of a series of semiconducting polymer dots (Pdots) incorporated with aggregation-induced emission moieties to exhibit the QYs as high as 14% in the NIR-II window. Benefiting from the ultrahigh brightness, a 1400 nm long-pass filter is utilized to realize in vivo 3D tumor mapping in mice. To further understand how the geometrical and electron structures of the semiconducting polymers affect their optical properties, the in-depth and thorough density-functional theory calculations are performed to interpret the experimental results. This study lays the groundwork for further molecular design of highly bright NIR-II Pdots.

Abstract:Quantum dot (QD),also named "artificial atom",is a synthetic nanomaterial using atomic,which can emit different wavelengths of fluorescence under UV excitation;the small diameter of QD as well as the narrow peak and the lasting brightness of the fluorescence emitted by QD make QD a potential dye substituted with organic dye for the di agnosis and treatment of tumor.The study aimed to summarize the research progress of quantum dots (QDs) in fluorescence imaging and tumor diagnosis and treatment in recent years.METHODS PubMed and CNKI database retrieval system in China were used to retrieve key words quantum dots,fluorescence imaging,oncotherapy,tumor therapy,cancer treatment,swollen diagnosis and fluorescence imaging from 2005-01 to 2016-03 and 391 references were selected.Inclusion criteria:usable QDs in bioluminescence imaging with low toxicity;functionalized QDs with target capacity.According to the inclusion criteria analysis,31 articles were analyzed in the end.RESULTS The studies of fluorescence imaging of low toxicity quantum dots in cell imaging,target imaging in vivo,lymph node imaging,tumor imaging in vivo,tumor cells tricking in vivo and the like have set the stage for rightly lymph nodes locating,using deep imaging property of QDs to find small tumors and thoroughly resecting tumor tissues.Functionalized QDs could do much more in early tumor diagnosis and treatment,QDs mediated drug targeting treatment and thermal therapy also have promising prospects in clinic applications.CONCLUSION Quantum dots in biological fluorescence imaging and tumor diagnosis and treatment field have great potential,further exploration and development of QDs utilization in clinic are valuable.

Abstract:Quantum dots (QDs) represent a class of nanoscale wide bandgap semiconductors, and are primarily composed of metals, lipids, or polymers. Their unique electronic and optical properties, which stem from their wide bandgap characteristics, offer significant advantages for early cancer detection and treatment. Metal QDs have already demonstrated therapeutic potential in early tumor imaging and therapy. However, biological toxicity has led to the development of various non-functionalized QDs, such as carbon QDs (CQDs), graphene QDs (GQDs), black phosphorus QDs (BPQDs) and perovskite quantum dots (PQDs). To meet the diverse needs of clinical cancer treatment, functionalized QDs with an array of modifications (lipid, protein, organic, and inorganic) have been further developed. These advancements combine the unique material properties of QDs with the targeted capabilities of biological therapy to effectively kill tumors through photodynamic therapy, chemotherapy, immunotherapy, and other means. In addition to tumor-specific therapy, the fluorescence quantum yield of QDs has gradually increased with technological progress, enabling their significant application in both in vivo and in vitro imaging. This review delves into the role of QDs in the development and improvement of clinical cancer treatments, emphasizing their wide bandgap semiconductor properties.
Engineered G-C3n4 Quantum Dots for Tunable Two-Photon Imaging and Photodynamic Therapy

Abstract:Near-infrared (NIR)-light-triggered diagnostic and therapeutic systems normally are the integration of several components, whose complex structures and low reproducibility restrict their further applications. Herein, we proposed a single component of defect graphitic phase carbon nitride quantum dots (g-C3N4 QDs) as a dual-functional nanoplatform, which could synchronously achieve two-photon imaging (TPI) and two-photon excited photodynamic therapy (TPE-PDT) under an 800 nm NIR laser. In order to regulate the competitive capability between TPI and PDT, three kinds of engineered g-C3N4 QDs (CN, CN-DPT, and CN-THDT QDs) with different two-photon catalytic capabilities to generate reactive oxygen species (ROS) were prepared through copolymerization melamine with selected monomers. In the evaluation of in vitro cytotoxicity, three g-C3N4 QDs possessed very good biocompatibilities, which could be delivered to tumor cells for imaging and therapy. Under an 800 nm NIR light, CN-DPT QDs exhibited a bright TPI effect and produced an efficient ROS to achieve TPE-PDT, thus suggesting that the single component of CN-DPT QDs could perform as an appropriate dual-functional TPI and TPE-PDT probe triggered by an 800 nm NIR light. The engineered g-C3N4 QDs with a high stability and excellent biocompatibility have contributed to the method of TPI and TPE-PDT, which may lead to g-C3N4-based nanomaterials as novel imaging and therapeutic agents in cancer treatment.

Abstract:The rapidly developing field of quantum dots (QDs) provides researchers with more options for imaging modalities and therapeutic strategies. In recent years, QDs were widely used as multifunctional materials for tumor imaging and therapy due to their characteristic properties such as semiconductive, zero-dimension and strong fluorescence. Nevertheless, there still exist the challenges of employing these properties of QDs for clinical diagnosis and therapy. Herein, we briefly review the development, properties and applications of QDs in tumor imaging and therapy. Future perspectives in these areas are also proposed as well.

Abstract:Many theranostic nanomedicines have been fabricated by packaging imaging and therapeutic moieties together. However, concerns about their potential architecture instability and pharmacokinetic complexity remain major obstacles to their clinical translation. Herein, we demonstrated the use of CuInS/ZnS quantum dots (ZCIS QDs) as "all-in-one" theranostic nanomedicines that possess intrinsic imaging and therapeutic capabilities within a well-defined nanostructure. ZCIS QDs were exploited for the first time for multispectral optical tomography (MSOT) imaging and synergistic PTT/PDT therapy. Due to the intrinsic fluorescence/MSOT imaging ability of the ZCIS QDs, their size-dependent distribution profiles were successfully visualized at tumor sites in vivo. Our results showed that the smaller nanomedicines (ZCIS NMs-25) have longer tumor retention times, higher tumor uptake, and deeper tumor penetration than the larger nanomedicines (ZCIS NMs-80). The ability of ZCIS QDs to mediate photo-induced tumor ablation was also explored. Our results verified that under a single 660 nm laser irradiation, the ZCIS NMs had simultaneous inherent photothermal and photodynamic effects, resulting high therapy efficacy against tumor. In summary, the ZCIS QDs as "all-in-one" versatile nanomedicines allow high therapeutic efficacy as well as noninvasively monitoring tumor site localization profile by imaging techniques, and thus hold great potential as precision theranostic nanomedicines.

Abstract:Theranostic agents that integrated chemodynamic therapy (CDT) and imaging functions have great potential application in personalized cancer therapy. However, most theranostic agents were fabricated by chemically coupling two or more independent functional units with diagnostic or therapeutic capabilities, and therefore have a large size. To date, one-step synthesis of unmodified ultrasmall quantum dots (QDs) integrating CDT and fluorescence imaging capabilities remains a challenge. Herein, we reported a simple one-step synthesis method of ultrasmall (2.46 nm) Zn-doped Cu2S (Zn:Cu2S) QDs with inherent properties of both high CDT activity and near-infrared fluorescence imaging capability. The fluorescence of Cu2S QDs was significantly enhanced approximately tenfold after Zn doping due to the compensation of defects. In vitro and in vivo experiments demonstrated that the Zn:Cu2S QDs could specifically and significantly inhibit the cancer cell growth (inhibition rate exceeded 65%) without damaging the normal cells. Furthermore, the CDT mechanism study suggested that a Fenton-like reaction occurred after the Zn:Cu2S QDs entered the tumor cells, inducing apoptosis via the mitochondrial signaling pathway, and activating the production of reactive oxygen species (ROS) and autophagy to selectively eliminate tumor cells to achieve CDT. This work proposed a simple one-step synthesis of unmodified ultrasmall QDs with fluorescence imaging and CDT, which provides a promising strategy for QDs to act as multi-functional theranostic agents.

Abstract:The synthesis of water-soluble near-infrared (NIR)-emitting quantum dots (QDs) has recently received extensive attention for non-invasive detection of biological information in living subjects. Highly fluorescent CdTeS alloyed QDs for biological application are introduced in this paper. QDs were synthesized by a hydrothermal method and coated with N-acetyl-l-cysteine (NAC) as both bioactive ligand and sulfur source for biocompatibility and biological stability. The optical properties, morphology and structure of CdTeS alloyed QDs were characterized. The in vitro and in vivo toxicity was intensively investigated. Furthermore, the dynamics and bio-distribution of CdTeS alloyed QDs on living mice were studied. To explore biomedical application, folate-polyethylene glycol (FA-PEG) was used to decorate the CdTeS alloyed QDs (FP-CdTeS QDs) for targeted imaging of tumors over-expressing the folate receptor (FR). The tumor targeting capability of FP-CdTeS QDs on tumor bearing nude mice was demonstrated. The results showed that the prepared CdTeS QDs have excellent optical properties and low toxicity, which makes them an ideal inorganic material for biomedical imaging. In addition, the folate-PEG conjugated NIR-QDs displayed good biocompatibility as well as excellent sensitivity and specificity for optical imaging of tumors which can extend the application of CdTeS QDs.

Abstract:As light-emitting nanocrystals, quantum dots (QDs) have created a new realm of bioscience by combining nanomaterials with biology. They also have been a major focus of research and development during the past decade, which will profoundly influence future biological as well as biomedical research. In recent years, near-infrared (NIR) quantum dots have emerged in analytical applications, especially for in vitro and in vivo imaging. The impetus behind such endeavors can be attributed to their unique optical and chemical properties, with size-tunable light emission, high photo stability, and manifold fluorescence colors. In this review, we focus on fluorescent imaging with near-infrared (NIR) quantum dots (QDs) both in vitro and in vivo, and the advantages of QDs and potential problems to their use in practical biomedical applications. The ultimate targets aim at decreasing the cytotoxicity of QDs and the future outlook of QD applications in biomedical fields.
