Abstract:The synthesis of a new kind of amphiphilic poly(vinyl-co-ester)s via simple one-pot radical ring-opening polymerization (RROP) is reported. A series of biodegradable, biocompatible, and cross-linkable copolymers P(MDO-co-PEGMA-co-CMA) with different compositions were synthesized by copolymerization of 2-methylene-1,3-dioxepane (MDO), poly(ethylene glycol) methyl ether methacrylate (PEGMA) and 7-(2-methacryloyloxyethoxy)-4-methylcoumarin methacrylate (CMA). All the copolymers were well characterized by H-1 NMR, C-13 NMR, and gel permeation chromatography (GPC). The hydrolytic degradation and enzymatic degradation of the copolymers were investigated. The amphiphilic P(MDO-co-PEGMA-co-CMA) can self-assemble into micelles in aqueous solution. Polymeric micelles with different sizes were obtained through tuning the compositions of the copolymers, i.e., the hydrophilic/hydrophobic ratio. The size and morphology of the micelles were measured by dynamic light scattering (DLS) and transmission electron microscopy (TEM). Because of the coumarin side groups in the micellar core, the micelles can be cross-linked upon exposure to long-wavelength UV light (lambda = 365 nm). These micelles also showed excellent biocompatibility by the in vitro cytotoxicity experiments. Furthermore, the biodegradable micelles were applied for the delivery of an anticancer drug doxorubicin (DOX). The cell viability of DOX-loading micelles compared to free DOX was discussed. The effects of cross-linking and the copolymer composition on the drug release behavior were well studied. The results indicate that this new kind of amphiphilic poly(vinyl-co-ester) s could serve as promising nanocarriers for the delivery of anticancer drugs.
Enhanced Percutaneous Delivery of Methotrexate Using Micelles Prepared with Novel Cationic Amphipathic Material

Abstract:BACKGROUND:Methotrexate (MTX) is an antiproliferative drug widely used to treat inflammatory diseases and autoimmune diseases. The application of percutaneous administration is hindered due to its poor transdermal penetration. To reduce side effects and enhanced percutaneous delivery of MTX, novel methotrexate (MTX)-loaded micelles prepared with a amphiphilic cationic material, N,N-dimethyl-(N',N'-di-stearoyl-1-ethyl)1,3-diaminopropane (DMSAP), was designed.MATERIALS AND METHODS:DMSAP was synthesized via three steps using simple chemical agents. H nuclear magnetic resonance and mass spectroscopy were used to confirm the successful synthesis of DMSAP. A safe and non-toxic phosphatidylcholine, soybean phosphatidylcholine (SPC), was added to DMSAP at different ratios to form P/D-micelles. Then, MTX-entrapped micelles (M/P/D-micelles) were prepared by electrostatic adsorption. The physicochemical properties and blood stability of micelles were examined thoroughly. In addition, the transdermal potential of the micelles was evaluated by permeation experiments.RESULTS:In aqueous environments, DMSAP conjugates could self-assemble spontaneously into micelles with a low critical micelle concentration (CMC) of 0.056 mg/mL. Stable, spherical MTX-entrapped micelles (M/P/D-micelles) with a size of 100-120 nm and high zeta potential of +36.26 mV were prepared. In vitro permeation studies showed that M/P/D-micelles exhibited superior skin permeability and deposition of MTX in the epidermis and dermis compared with that of free MTX.CONCLUSION:These special novel cationic M/P/D-micelles can enhance the permeability of MTX and are expected to be a promising percutaneous delivery system for therapy skin diseases.

Abstract:Nanoparticles are appealing drug delivery systems since they promise prolonged circulation time and predictable release behaviors. The current work reports a novel hyperbranched amphiphilic block copolymer synthesized using deactivation-enhanced atom transfer radical polymerization (DE-ATRP) for smart drug delivery. PEG2000-Br was applied as a macroinitiator to initiate acid-cleavable divinyl (ACD) monomers linked by acetal groups, formulating the hydrophilic (PEG) and hydrophobic (ACD) segments. The polymer appeared to self-assemble into micelles with diameters in the range of 70-100 nm, and was examined for the controlled release of doxorubicin (DOX). Results showed that DOX-loaded micelles (similar to 90 nm) could achieve drug loading as high as 8.2 wt%, with interesting pH-dependent release behaviors. Studies with flow cytometry (FCM) and confocal laser scanning microscopy (CLSM) showed that DOX-loaded micelles exhibited a high cellular uptake performance by HeLa cells, which indicated promising antitumor efficacy for such a drug delivery system. Additionally, such DOX-loaded micelles exhibited remarkable cytotoxicity against HeLa cells in a dose-and time-dependent manner due to the enhanced cell uptake behavior of micelles. These results indicated that the polymeric micelles might be used as a promising candidate for a pH-responsive drug delivery for cancer therapy.
Fabrication and in vitro drug release study of microsphere drug delivery systems based on amphiphilic poly-α,β-[N-(2-hydroxyethyl)-l-aspartamide]-g-poly(l-lactide) graft copolymers

Abstract:Biodegradable amphiphilic graft copolymers with different compositions were synthesized by grafting poly(l-lactide) (PLLA) sequences onto a water-soluble poly-α,β-[N-(2-hydroxyethyl)-l-aspartamide] (PHEA) backbone. The critical micelle concentration (CMC) of the graft polymers was determined by fluorescence probe technique. Using the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay, the graft polymers were proved to have low cytotoxicity. Based on the specific physicochemical property of the graft copolymers, submicron sized microsphere drug delivery systems were prepared by a very convenient “ultrasonic dispersion method”, which did not involve toxic organic solvents. The drug-loaded microspheres had a regular spherical shape with a narrow size distribution. A hydrophobic drug, prednisone acetate, was encapsulated into polymeric microspheres and the in vitro drug release was studied.
In situ grafting of PEG Acrylate on drugs with aliphatic hydroxyl functionalities via RAFT polymerization to synthesize drug/polymer conjugates with improved water solubility

Abstract:Despite the fact that mitoxantrone (MTX) is an effective prescription drug for cancer therapy, its poor water solubility is a great challenge that limits its practical applications. Herein, to overcome this shortcoming, a drug/polymer conjugate of MTX with poly (ethyleneglycol) acrylate (PEG-A) (MTX/PPEG-A) was developed via in situ reversible addition-fragmentation chain transfer (RAFT) polymerization. Impressively, the MTX/PPEG-A conjugate shows highly improved water solubility, which is about 550% that of the unmodified MTX. The synthesized MTX/PPEG-A conjugate drug efficacy can be maintained and cytotoxicity reduced slightly compared with the unmodified MTX. In addition, the MTX/PPEG-A can be hydrolyzed by the esterase into pristine MTX with restored drug efficacy in vivo. All in all, the significant improvement of the water solubility and the slightly improved biocompatibility of MTX/PPEG-A will be well-suited for the administration and delivery of anti-cancer drugs in cancer therapy.

Abstract:Amphiphilic copolymers that can assemble into core-shell micelles in aqueous media have potential application as drug carriers for controlled-release. In this article an amphiphilic triblock copolymer poly(ethylene oxide)-block-poly(lactic acid)-block-poly(2-hydroxyethyl methacrylate) (PEG-b-PLA-b-PHEMA) was synthesized by ring-opening reaction and atom transfer radical polymerization. The polymers were characterized by H-1 NMR, FT-IR and gel permeation chromatography. In aqueous solution, PEG-b-PLA-b-PHEMA can self-assemble into micelles. Characterized by DLS and TEM, micelles are found to be spherical with uniform size. Large numbers of hydroxyl groups on the PHEMA chain offer reaction sites for subsequent functional modification, which make it a promising platforms for drug delivery systems.
Synthesis and Self-Assembly of Amphiphilic ABA Type Triblock Copolymer with Well-Defined Glycopolymer Segments

Abstract:A novel amphiphilic ABA type triblock copolymer with well-defined glycopolymer segments was successfully synthesized via the atom transfer radical polymerization (ATRP) technique, using a bromo-terminated difunctional polysulfone as macroinitiator. The difunctional polysulfone macroinitiator was prepared by esterifying the phenolic end groups of polysulfone to α-haloesters. This macroinitiator was then used to initiate the polymerization of a glucose-carrying monomer, 3-O-methacryloyl-1,2:5,6-di-O-isopropylidene-D-glucofuranose (MAIpG), resulting an ABA type triblock copolymer. After acidolysis treatment, the isopropylidenyl groups of the protected sugar residues were removed, thus the amphiphilic ABA type triblock copolymer with well-defined glycopolymer segments was obtained. The polymers obtained were identified by FT-IR, 1H-NMR, GPC, and TGA. The self-assembly behavior of the amphiphilic glycopolymer in selective solvent (e.g. DMF/H2O) system was also preliminarily explored. The resultant amphiphilic glycopolymer shows potential applications in the fields of controlled release and delivery of drugs, micro-reactors, nano-materials, medical devices, and so on.

Abstract:A series of poly(l-lactide)-grafted gelatin (Gel-g-PLLA) copolymers are synthesized by coupling the amino groups of gelatin with different molar masses and the carboxyl endgroup of poly(l-lactide) in the presence of 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride and N-hydroxysuccinimide. Fourier transform infrared and nuclear magnetic resonance (H-1 NMR) data confirm the successful bonding between gelatin and PLLA. Self-assembled micelles of copolymers are prepared by using the direct dissolution method. The size and micellar morphology are determined from dynamic light scattering as well as transmission electron microscopy measurements. Meanwhile, in vitro drug release reveals that Gel-g-PLLA micelles show excellent sustained-release properties when used as the carrier of the anticancer drug paclitaxel. A burst release of about 70% is observed in the first 24 h. All these features, together with the outstanding biocompatibility, make Gel-g-PLLA micelles a promising drug carrier for cancer therapy.

Abstract:A well-defined double hydrophilic graft copolymer, with polyacrylate as backbone, hydrophilic poly(ethylene glycol) and poly(methacrylic acid) as side chains, was synthesized via successive atom transfer radical polymerization followed by the selective hydrolysis of poly(methoxymethyl methacrylate) side chains. The grafting-through strategy was first used to prepare poly[poly(ethylene glycol) methyl ether acrylate] comb copolymer. The obtained comb copolymer was transformed into macroinitiator by reacting with lithium diisopropylamine and 2-bromopropionyl chloride. Afterwards, grafting-from route was employed for the synthesis of poly[poly(ethylene glycol) methyl ether acrylatel-g-poly(methoxymethyl methacrylate) amphiphilic graft copolymer. The molecular weight distribution of this amphiphilic graft copolymer was narrow. Poly(methoxymethyl methacrylate) side chains were connected to polyacrylate backbone through stable C-C bonds instead of ester connections. The final product, poly[poly(ethylene glycol) methyl ether acrylatel-g-poly(methacrylate acid), was obtained by selective hydrolysis of poly(methoxymethyl methaerylate) side chains under mild conditions without affecting the polyacrylate backbone. A new Br-containing acrylate monomer, tert-butyl 2-((2-bromopropanoyloxy)methyl) acrylate, was first prepared, and it call be polymerized via RAFT in a controlled way to obtain a well-defined homopolymer with narrow molecular weight distribution (M-w/M-n = 1.06). Grafting-onto strategy was employed to synthesize PtBA-g-PPO well-defined graft copolymers with narrow molecular weight distributions (M-w/M-n = 1.05-1.23) via ATNRC reaction between Br-containing PtBA-based backbone and poly(propylene oxide) with 2,2,6, 6-tetramethylpiperidine-1-oxyl (TEMPO) end group using CuBr/PMDETA or Cu/PMDETA as catalytic system, The final PAA-g-PPO amphiphilic graft copolymers were obtained by the selective acidic hydrolysis of PtBA backbone in acidic environment without affecting the side chains. The critical micelle concentrations in aqueous media were determined by a fluorescence probe technique. Diverse micellar morphologies were formed with varying the content of hydrophobic PPO segment.

Abstract:Amphiphilic graft copolymers composed of poly(hydroxyethyl aspartamide)-co-propyl aspartamide) (PHEA-PA) as the hydrophilic backbone and poly(lactide) (PLA) as the hydrophobic side chain were successfully synthesized by grafting lactide (LTD) onto PHEA-PA. Polysuccinimide (PSI) was firstly synthesized from l-aspartic acid through thermal condensation polymerization method, and then followed by two-step aminolysis processes to form PHEA-PA with different amounts of hydroxyl groups. The hydroxyl groups were then used as initial points to graft PLA onto PHEA-PA backbone via a ring-opening polymerization in the presence of Sn(Oct)2. The grafting reaction of PLA onto PHEA-PA was confirmed by 1H NMR, FT-IR, and DSC. Micelle-like aggregates were obtained when the amphiphilic comb-like copolymer solution of DMF was added into an excessive amount of water. Strong hydrophobic intramolecular and intermolecular association of PLA chains led to aggregates with hydrophobic microdomains. The dispersion of the self-aggregates of poly(hydroxyethyl aspartamide-co-propyl aspartamide)-g-poly(d,l-lactide) (PHEA-PA-g-PLA) copolymers is stable in aqueous medium. Spherical aggregates of PHEA-PA-g-PLA were observed by transmission electron microscopy (TEM) and scanning electron microscopy (SEM), the diameters were about 100nm.

Abstract:Encapsulating anticancer drugs to synthetic polymer is a promising approach to improve the efficiency and reduce the side effects of anticancer drugs. In this study, novel chitosan derivatives with polyamidoamine moieties (CS-PAMAM) were synthesized and characterized by morphology, particle size, and zeta potential. Then the anticancer drug-methotrexate-encapsulated CS-PAMAM was prepared by hydrophobic-hydrophilic interactions. The drug release assay showed that the amount of the methotrexate release from CS-PAMAM was pH depended. Meanwhile, the cell viability assay illustrated that CS-PAMAM was suitable for the drug delivery because of its low cytotoxicity on cells. Moreover, our results showed that the CS-PAMAM could significantly improve the cytotoxicity of free methotrexate on A549 cells. These results demonstrate that CS-PAMAM may provide a suitable platform for the water-insoluble drug delivery.

Abstract:A series of well-defined amphiphilic graft copolymer bearing a hydrophilic poly(2-hydroxyethyl acrylate) (PHEA) backbone and hydrophobic poly(methyl methacrylate) (PMMA) side chains was synthesized by successive reversible addition-fragmentation chain transfer (RAFT) polymerization and atom transfer radical polymerization (ATRP) through the grafting-from strategy. A well-defined PHEA-based backbone with Cl-containing ATRP initiating group in every repeated unit ( M w / M n = 1.08), poly(2-hydroxyethyl 2-((2-chloropropanoyloxy)methyl)acrylate) (PHECPMA), was first prepared by RAFT homopolymerization of 2-hydroxyethyl 2-((2-chloropropanoyloxy)methyl)acrylate (HECPMA), a Cl-containing trifunctional acrylate. ATRP of methyl methacrylate was subsequently initiated by PHECPMA homopolymer to afford the target well-defined poly(2-hydroxyethyl acrylate)- graft -poly(methyl methacrylate) (PHEA- g -PMMA) graft copolymers ( M w / M n ≤ 1.36) with 34 PMMA side chains and 34 pendant hydroxyls in PHEA backbone using CuCl/dHbpy as catalytic system. The critical micelle concentration ( cmc ) of the obtained graft copolymer was determined by fluorescence spectroscopy using N -phenyl-1-naphthylamine as probe while micellar morphologies in aqueous media were visualized by transmission electron microscopy. Interestingly, PHEA- g -PMMA graft copolymer could self-assemble into large compound micelles rather than common spherical micelles, which can encapsulate hydrophilic rhodamine 6 G and hydrophobic pyrene separately or simultaneously.

Abstract:Well-defined amphiphilic poly(ethylene glycol) and poly(dehydroabietic ethyl methacrylate) block copolymers (PEG-b-PDAEMA) were prepared by atom transfer radical polymerization. The methacrylate block contains a characteristic hydrophobic, biocompatible and economical dehydroabietic moiety. PEG-b-PDAEMA block copolymer micellar nanoparticles loaded with piperlongumine (PLGM) were successfully prepared by a nanoprecipitation method. In vitro and in vivo behaviors of these nanoparticles were thoroughly examined by a set of characterization techniques. Confocal laser scanning microscopy study revealed that these nanoparticles could be well taken up by cancer cells. In vivo near-infrared fluorescence imaging showed that the PLGM-loaded nanoparticles effectively targeted the tumor site by the enhanced permeability and retention (EPR) effect in H22 tumor-bearing mice. The in vivo antitumor examination found that PLGM-loaded nanoparticles exhibited superior efficacy in impeding the tumor growth compared to the commercially available Taxol (R) and free PLGM formulation. Abstract:ABA-type triblock copolymers poly (trimethylene carbonate)-poly (ethylene glycol)-poly (trimethylene carbonate) were synthesized by ring-opening polymerization of trimethylene carbonate initiated by dihydroxyl poly (ethylene glycol). The critical micelle concentration of amphiphilic triblock copolymers in aqueous solution was determined by fluorescence spectroscopy using 9-chloromethyl anthracene as fluorescence probe. Core-shell-type nanoparticles were prepared by the dialysis technique. Transmission electron microscopy images showed that these nanoparticles were regularly spherical in shape. Micelle size determined by dynamic light scattering is 50-160 nm. Anticancer drug methotrexate (MTX) as model drug was loaded in the polymeric nanoparticles. X-ray powder diffraction spectra showed that model drugs were molecularly dispersed in the core. In vitro release behavior of MTX was investigated.

Abstract:The amphiphilic heterograft copolymers bearing biocompatible/biodegradable grafts [poly(2-methyl-2-oxazoline-co-2-pentyl-2-oxazoline)-g-poly(d-l-lactic acid)/poly(2-ethyl-2-oxazoline)] were synthesized successfully by the combination of cationic ring-opening polymerization and click chemistry via the ⟨"grafting to"⟩ approach. The challenge of this synthesis was to graft together hydrophobic and hydrophilic chains on a hydrophilic platform based on PMeOx. The efficiency of grafting depends on the chemical nature of the grafts and of the length of the macromolecular chains. The self-assembly of these polymers in aqueous media was investigated by DLS, cryo-TEM, and SANS. The results demonstrated that different morphologies were obtained from nanospheres and vesicles to filaments depending on the hydrophilic weight ratio in the heterograft copolymer varying from 0.38 until 0.84. As poly(2-ethyl-2-oxazoline) is known to be thermoresponsive, the influence of temperature rise on the nanoassembly stability was studied in water and in a physiological medium. SANS and DLS measurements during a temperature ramp allowed to show that nanoassemblies start to self-assemble in "raspberry like" primary structures at 50 °C, and these structures grow and get denser as the temperature is increased further. These amphiphilic heterograft copolymers may include hydrophobic drugs and should find important applications for biomedical applications which require stealth properties.

Abstract:The amphiphilic heterograft copolymers poly(methyl methacrylate-co-2-(2-bromoisobutyryloxy)ethyl methacrylate)-graft-(poly(acrylic acid)/polystyrene) (P(MMA-co-BIEM)-g-(PAA/PS)) were synthesized successfully by the combination of single electron transfer-living radical polymerization (SET-LRP), single electron transfer-nitroxide radical coupling (SET-NRC), atom transfer radical polymerization (ATRP), and nitroxide-mediated polymerization (NMP) via the “grafting from” approach. First, the linear polymer backbones poly(methyl methacrylate-co-2-(2-bromoisobutyryloxy)ethyl methacrylate) (P(MMA-co-BIEM)) were prepared by ATRP of methyl methacrylate (MMA) and 2-hydroxyethyl methacrylate (HEMA) and subsequent esterification of the hydroxyl groups of the HEMA units with 2-bromoisobutyryl bromide. Then the graft copolymers poly(methyl methacrylate-co-2-(2-bromoisobutyryloxy)ethyl methacrylate)-graft-poly(t-butyl acrylate) (P(MMA-co-BIEM)-g-PtBA) were prepared by SET-LRP of t-butyl acrylate (tBA) at room temperature in the presence of 2,2,6,6-tetramethylpiperidin-1-yloxyl (TEMPO), where the capping efficiency of TEMPO was so high that nearly every TEMPO trapped one polymer radicals formed by SET. Finally, the formed alkoxyamines via SET-NRC in the main chain were used to initiate NMP of styrene and following selectively cleavage of t-butyl esters of the PtBA side chains afforded the amphiphilic heterograft copolymers poly(methyl methacrylate-co-2-(2-bromoisobutyryloxy)ethyl methacrylate)-graft-(poly(t-butyl acrylate)/polystyrene) (P(MMA-co–BIEM)-g-(PtBA/PS)). The self-assembly behaviors of the amphiphilic heterograft copolymers P(MMA-co–BIEM)-g-(PAA/PS) in aqueous solution were investigated by AFM and DLS, and the results demonstrated that the morphologies of the formed micelles were dependent on the grafting density. © 2011 Wiley Periodicals, Inc. Abstract:A series of well-defined amphiphilic triblock copolymers, poly(ethylene glycol)-b-poly(tert-butyl acrylate)-b-poly(2-hydroxyethyl methacrylate) (PEG-b-PtBA-b-PHEMA), were synthesized via successive atom transfer radical polymerization (ATRP). ATRP of tBA was first initiated by PEG-Br macroinitiator using CuBr/N,N,N',N",N"-pentamethyldiethylenetriamine as catalytic system to give PEG-b-PtBA diblock copolymer. This copolymer was then used as macroinitiator to initiate ATRP of HEMA, which afforded the target triblock copolymer, PEG-b-PtBA-b-PHEMA. The critical micelle concentrations of obtained amphiphilic triblock copolymers were determined by fluorescence spectroscopy using N-phenyl-1-naphthylamine as probe. The morphology and size of formed aggregates were investigated by transmission electron microscopy and dynamic light scattering, respectively. Finally, an acid-sensitive PEG-b-PtBA-b-P(HEMA-CAD) prodrug via cis-aconityl linkage between doxorubicin and hydroxyls of triblock copolymers with a high drug loading content up to 38%, was prepared to preliminarily explore the application of triblock copolymer in drug delivery. Abstract:Highlights • MSNs were successfully modified by MPTES. • Poly (acrylic acid- co -allylamine) (PAA) was grafted onto modified MSNs. • Folic acid was successfully conjugated to PAA-g-MSNs to produce a DDS. • DDS is a promising pH-responsive nanocarrier for MTX. A new drug delivery system (DDS) was designed using mesoporous silica nanoparticles functionalized with a copolymer of poly(acrylic acid- co -allylamine) conjugated with folic acid. DDS was developed to enable targeted and controlled delivery of methotrexate to cancer cells. The DDS pores were utilized for loading methotrexate through multilayer physical adsorption, which occurred spontaneously and exothermically. The methotrexate adsorption capacity of the DDS was 673.001 ± 4.518 mg/g. The copolymer acted as a gatekeeper, regulating the entry and exit of methotrexate from the pores. The DDS exhibited minimal methotrexate leakage at high pH levels but showed a significantly increased release at lower pH levels, demonstrating a pH-controlled drug release pattern. Mathematical modeling indicated that diffusion played a major role in drug release, and the DDS functioned as a reservoir system with a declining release rate over time. Importantly, the DDS displayed significant cytocompatibility, as demonstrated by MTT assays. Furthermore, the methotrexate-loaded DDS showed remarkable efficacy in killing MCF-7 cells, suggesting that folic acid facilitates DDS-specific targeting of MCF-7 cells via folic acid receptors.
