Investigation of enzyme and serum protein protective effects, along with molecular docking studies of mixed ligand ruthenium(II) polypyridyl complexes

Abstract:Five new ruthenium(II) complexes [Ru(ƞ2-N,S-mpy)(ƞ1-S-mpy)(tptz)] (Ru1), [Ru(ƞ2-N,S-mbtz)(ƞ1-S-mbtz)(tptz)] (Ru2), [Ru(ƞ2-N,S-mpt)(ƞ1-S-mpt)(tpy)] (Ru3), [Ru(ƞ2-N,S-mbtz)(ƞ1-S-mbtz)(tpy)] (Ru4) and [Ru(ƞ2-N,S-mpy)( ƞ1-S-mpy) (tpy)] (Ru5) have been synthesized and characterized by elemental analyses, IR, 1H and 13C NMR, and electronic absorption spectroscopy. Molecular structure of the representative complex Ru1 was determined crystallographically. The coordination geometry in Ru1 is distorted octahedral where tptz is coordinated to ruthenium centre in a κ3-manner and two 2-marcaptopyridine ligands in κ2 and κ1-manner. The synthesized complexes were evaluated for inhibition studies for intracellular enzymes such as amylase, lipase, trypsin, pepsin, and serum protein protecting activity. Lipase, trypsin, pepsin displayed an inhibitory effect for the synthesized complex and the ligands with observed IC values of the order of 10-6 M, whereas amylase was found to be activated. As with other Ru(II) pharmacological active compounds, the serum protein binding studies were performed both the N,S′ and N,N′ donor ligands as well as ruthenium(II) complexes Ru1-Ru5. It was found that binding of the respective complexes was more effective as compared to the respective ligands.
