DECREASED EXPRESSION OF GM3 DETECTED BY HIGH PERFORMANCE LIQUID CHROMATOGRAPHY IN A TRANSGENIC MOUSE MODEL OF AD

Abstract:Gangliosides are expressed in the outer leaflet of the plasma membrane in all vertebrate cells and are particularly abundant in the nervous system. One of the major functions for gangliosides is the formation of lipid rafts and the abnormal metabolism of the gangliosides is closely associated with the pathology of Alzheimer's disease (AD), the most common neurodegenerative disorder characterized pathologically by β-amyloid (Aβ) deposit and tau hyperphosphorylation. Recently, a special Aβ species named G-Aβ was identified in an AD brain, which was featured by its bonding to GM1gangliosides, and the G-Aβ acted as a seed for Aβ fibrillogenesis in the AD brain. Here, we analyzed the changes of gangliosides in a transgenic mouse model of AD, i.e. 3xTg (B6;129-Psen1tm1MpmTg (APPSwe, tauP301L)1Lfa/Mmjax). The changes of gangliosides in hippocampus, cortex, cerebellum, and spinal cord of the mice at different ages were analyzed using high performance liquid chromatography (HPLC). Compared with the age-matched wild type mice, the levels of gangliosides, mainly GD1 and GT1, were significantly increased in hippocampus and significantly decreased in the spinal cord of 3xTg mice. As GM3 is the rimary ganglioside affecting synthesis of other gangliosides, we further investigated the role of GM3 on tau phosphorylation. We found that tau-5, PS404 and AT8 are reduced. Our data provide novel evidence that GD1 and GT1 are main gangliosides, GM3 play a key role in the rate of synthesis gangliosides.
