Abstract:OBJECTIVE:The aim of this study was to observe the suppressive effect of bortezomib alone and the synergistic suppressive effect of bortezomib and adriamycin on the proliferation of the cell line Jurkat cells, and to discuss the mechanism of apoptosis induced by bortezomib.METHODS:The suppressive effect of bortezomib and adriamycin on the proliferation of Jurkat cells in vitro was detected by MTT colorimetry, and the morphology of the cells was examined by histology. The cell apoptosis was measured by flow cytometry with Annexin V/PI staining and cell cycle analysis. The effect of bortezomib and adriamycin on the expression levels of caspase-3, caspase-8 and PARP were measured by Nestern blot.RESULTS:The proliferation of Jurkat cells was significantly inhibited by bortezomib treatment (between 10 - 320 ng/ml) for 24 h, 48 h and 72 h, and the growing inhibition ratio showed a positive correlation with the drug concentration (r(24 h) = 0.900, P < 0.01; r(48 h) = 0.849, P < 0.01; r(72 h) = 0.679, P < 0.01), in a concentration-dependent manner. The IC(50) of Jurkat cells treated with bortezomib in a dose of 10 - 320 ng/ml was 137.64 +/- 6.82 ng/ml, but the IC(50) of Jurkat cells treated with bortezomib combined with adriamycin (125 ng/ml) for 24 h was significantly decreased to 20.44 +/- 2.85 ng/ml. The apoptosis rate had a positive correlation with the concentration of bortezomib (P < 0.01). After the Jurkat cells were treated with bortezomib, apparent shear bands of caspase-8, caspase-3 and PARP proteins were observed.CONCLUSION:There is an effect of Bortezomib to induce apoptosis in Jurkat cells, and the extrinsic pathway is one of the apoptosis-inducing mechanisms. There is a synergistic suppressive effect of the combination of bortezomib and adriamycin on the proliferation of Jurkat cells and enhances their chemosensitivity.
Apoptosis of HL-60 Cells Induced by Dihydroartemisinin

Abstract:AIM To investigate the therapeutic effect of dihydroartemisinin on leukemia.METHODS Trypan blue exclusion method and MTT assay were employed to evaluate the survival of HL-60 cells.The induction of apoptosis was detected by acridine orange/ethidium bromide dual staining as well as DNA electrophoresis.The ratios of apoptosis in HL-60 cells were assessed by using flow cytometry.The protein expressions related to apoptosis in HL-60 cells were detected with Western blot.RESULTS Dihydroartemisinin effectively inhibited HL-60 cell viability in vitro and induced HL-60 cells apoptosis.Down-regulation of Bcl-2 expression and over-expression of Bax and caspase-3 mediated by dihydroartemisinin in a concentration-dependent pattern were observed.CONCLUSION Dihydroartemisinin induced apoptosis of HL-60 cells,and the mitochondrial pathway mediated by Bcl-2 protein family might be involved in dihydroartemisinin induced apoptosis.

Abstract:Objective To investigate the effect of homoharringtonine (HHT) combined with BRD4 inhibitor JQ1 on apoptosis of acute myeloid leukemia (AML) cells. Methods AML Kasumi-1 and Mv4-11 cells were treated with HTT, JQ1 and HTT plus JQ1 in vitro, respectively. Cell proliferation was measured by MTS, and cell apoptosis was examined by flow cytometry. The expression of apoptosis-related proteins Bcl-2, Caspase-3 and Parp was detected by Western blot. Results Compared with JQ1 and HHT alone, the combination of the two drugs significantly increased the inhibition effect on the proliferation of Kasumi-1 and Mv4-11 cells. The combination of two drugs also significantly enhanced the effect on cell apoptosis, and enhanced the inhibition on the Bcl-2 expression, and the activation on caspase3 expression and Parp cleavage. Conclusion HHT combined with BRD4 inhibitor JQ1 can enhance the effect of two drugs against AML cells, inducing cell apoptosis.

Abstract:Homoharringtonine (HHT) is a plant alkaloid with antileukemia activity that is currently being used for treatment of acute, chronic leukemias and MDS. In this study, we show that HHT can induce apoptosis in a variety of human myeloid leukemia cell lines (U937, HL-60, HEL, THP, and K562). U937 and HL60 cells undergo rapid apoptosis on treatment with HHT, as indicated by increased annexin V binding capacity, caspase-3 activation, and cleavage of poly(ADP-ribose) polymerase (PARP). In addition, the expression of bax is upregulated during HHT-induced cell death, whereas the expression of bcl-2 is only slightly decreased. Importantly, treatment of primary leukemic cells, obtained from acute myeloid leukemia patients, resulted in rapid apoptosis. Thus, our data provide the mechanism of HHT and justify the use of HHT in the treatment of human myeloid leukemia. (C) 2004 Wiley-Liss, Inc.
[Effect of Bortezomib on Sensitization of HL-60 Cells to TRAIL].

Abstract:This study was aimed to explore whether bortezomib can sensitize HL-60 cells to TNF-related apoptosis-inducing ligand (TRAIL) and to investigate its possible mechanism. The HL-60 cells were treated by different concentrations of TRAIL combined with subtoxic concentration of bortezomib. The proliferative inhibition of treated HL-60 cell was analysed by MTT assay. The cell apoptosis was determined by flow cytometry with Annexin V/PI double staining and the expression of caspase-8 was detected by Western blot. The results showed that the subtoxic concentration of bortezomib combined with 10 ng/ml of TRAIL enhanced apoptosis of HL-60 cells, as compared with TRAIL used alone; the expression of caspase-8 increased correspondingly. It is concluded that subtoxic concentration of bortezomib can sensitize HL-60 cells to TRAIL and its mechanism may be related to upregulation of caspase-8 expression.
Significance of Expression of Bcl-2 Mrna and Bax Mrna in Arsenic Trioxide-Induced Apoptosis of Hepatocellular Carcinoma Cells

Abstract:ObjectiveTo explore of significance of expression of bcl-2 mRNA and bax mRNA in arsenic trioxide-induced apoptosis of hepatocellular carcinoma (HCC) cells. MethodsAfter human HCC cell line bel-7402 was treated with arsenic trioxide at various concentrations, the apoptosis was detected with transmission electron microscopy, flow cytometry and DNA laddering. Meanwhile, the expression of bcl-2 mRNA and bax mRNA was determined with semiquantitative reverse transcription polymerase chain reaction (RT-PCR). ResultsCell apoptosis was not detected in 0.5 μmol/L As_2O_3 group but in 8 μmol/L As_2O_3 group. The expression of bcl-2 mRNA were not seen in the control group and drug-treated group. Enhancement of expression of bax mRNA along with increase of As_2O_3 concentration. ConclusionsProliferation is inhibited via inducing apoptosis by As_2O_3 in bel-7402 cell line. Enhancement of bax mRNA expression but not that of bcl-2 mRNA expression is related to this process.
Homoharringtonine is Synergistically Lethal with BCL-2 Inhibitor APG-2575 in Acute Myeloid Leukemia

Abstract:Background: Despite advances in targeted agent development, effective treatment of acute myeloid leukemia (AML) remains a major clinical challenge. The B-cell lymphoma-2 (BCL-2) inhibitor exhibited promising clinical activity in AML, acute lymphoblastic leukemia (ALL) and diffuse large B-cell lymphoma (DLBCL) treatment. APG-2575 is a novel BCL-2 selective inhibitor, which has demonstrated anti-tumor activity in hematologic malignancies. Homoharringtonine (HHT), an alkaloid, exhibited anti-AML activity. Methods: The synergistic effects of APG-2575 and HHT were studied in AML cell lines and primary samples. MTS was used to measure the cell viability. Annexin V/propidium iodide staining was used to measure the apoptosis rate by flow cytometry. AML cell xenografted mouse models were established to evaluate the anti-leukemic effect of BCL-2 inhibitor, HHT and their combination in vivo. Western blot was used to determine the expression of related proteins. Results: APG-2575 showed comparable anti-leukemic effect to the FDA-approved BCL-2 inhibitor ABT-199 in vitro and in vivo. Combined treatment of HHT with APG-2575 synergistically inhibited AML cell growth and engraftment. Mechanistically, HHT promoted degradation of myeloid cell leukemia-1 (MCL-1), which was reported to induce BCL-2 inhibitor resistant, through the PI3K/AKT/GSK3 beta signaling pathway. Conclusion: Our results provide an effective AML treatment strategy through combination of APG-2575 and HHT, which is worthy of further clinical research.

Abstract:3208 Myelodysplastic syndrome(MDS) is a group of monoclonal hematopoietic stem cell disorders characterized by ineffective hematopoiesis and peripheral blood cytopenias. The clinical course of the disease and response to therapy vary considerably among individuals, therefore no single agent is expected to benefit all MDS patients universally. Recently arsenic trioxide (As2O3) was used to treat patients with MDS. To determine the effection of arsenic trioxide on MDS, MUTZ-1 cell line which was isolated from the peripheral blood of a MDS-RAEB patient, was treated by 0.5–8μmol/l As2O3. The inhibition of cell growth was detected by MTT assay. Apoptosis was assessed by morphology, DNA ladder, and the apoptotic peak before G1 phase cells by flow cytometry. The results revealed that As2O3 inhibited MUTZ-1 cell growth with IC50 of 5.7μmol/l, 1.87μmol/l, 0.91μmol/l when treated after 24h,48h and 72h respectively. As2O3 induces apoptosis in MUTZ-1 cells in a dose- and time-dependent manner. 2.0μmol/l is an effective concentration in the action of As2O3 induces apoptosis in MUTZ-1cells. Caspase protease family is a group of key apoptosis regulatory molecule that becomes activated during the early stages of apoptosis. We demonstrated that in the process of apoptosis of MUTZ-1 cells induced by As2O3, some members of the caspase protease family include caspase-8, caspase-3 and its substrate PARP were activated. It means that the activation of caspase protease is an important way in the apoptosis of MUTZ-1 cells induced by As2O3. In addition to the activation of caspase protease, we detected the expression of hTERT mRNA by RT-PCR meanwhile. We found that the expression of hTERT mRNA was down regulated by 2–8μmol/l As2O3 in MUTZ-1cells. We concluded that As2O3 should be an effective agent in the treatment of MDS. It induces apoptosis in MDS cell line MUTZ-1 cells in vitro via the activation of caspases and down-regulation of hTERT mRNA expression. No significant financial relationships to disclose.

Abstract:OBJECTIVE:[corrected] To investigate the effect of apoptosis induced in human SGC-7901 cells by Harmine.METHODS:The effect of Harmine on human SGC-7901 cell survival and apoptosis was determined by MTT assay, light microscopy and flow cytometry. Cell genomic DNA was detected by agarose electrophoresis.RESULTS:The survival of human SGC-7901 cells decreased; Apoptotic cells were observed by fluorescent microscope; FCM analysis showed that the peak of apoptosis increased. Typical DNA Ladder were detected in DNA agarose gel electrophoresis.CONCLUSION:HM can induce apoptosis in human SGC-7901 cells.

Abstract:The effect and possible mechanism of tripterine on inducing human acute myelocytic leukemia HL-60 cell apoptosis were investigated.By using trypan-blue staining method,transmission electron microscopy(TEM),and flow cytometry(FCM),the effects of triperine with different dosages and durations to induce the apoptosis as well as the expression of Fas,FasL and NF-κBP65 of HL-60 cells were also studied.All the apoptic rates of HL-60 cells treated with 0.5~2.5 μmol·L-1 tripterine for 24 hours were remarkably higher than that with no drug treatment(P0.05).When the used dosage of tripterine was 1.5 μmol·L-1,the maximal apoptic rate could be found as a time-dependent pattern.Typical apoptic morphological changes in the tripterine-treated HL-60 cells were found.Tripterine with a dosage of 1.5 μmol·L-1 could increase the positive rates of Fas and FasL positve HL-60 cells(P0.05),but inhibit the expression of NF-κBP65 in the cells(P0.05). Tripterine can efficiently induce the apoptosis of HL-60 cells.The anti-tumor mechanism of tripterine is probably involved to up-regulate the expression of Fas and FasL genes and to down-regulate the expression of NF-κB gene in the treated HL-60 cells.

Abstract:BACKGROUND:Myelodysplastic syndromes (MDS) are clonal marrow stem-cell disorders with a high risk of progression to acute myeloid leukemia (AML). Treatment options are limited and targeted therapies are not available for MDS. In the present study, we investigated the cytotoxicity and the molecular mechanism of Homoharringtonine (HHT) and Bortezomib towards high-risk MDS cell line SKM-1 in vitro and the role of miR-3151 was first evaluated in SKM-1 cells.METHODS:SKM-1 cells were treated with different concentrations of HHT or Bortezomib, and cell viability was analyzed with CCK-8 assay. The influence on cell proliferation, cell cycle distribution and the percentage of apoptosis cells were analyzed by flow cytometry. Calcusyn software was used to calculate combination index (CI) values. Western blot was used to analysis phosphorylation of Akt and nuclear NF-κB protein expression in SKM-1 cells. Mature miR-3151 level and p53 protein level were detected after HHT or Bortezomib treatment. The cell proliferation and p53 protein level were reassessed in SKM-1 cells infected with lentivirus to overexpress miR-3151.RESULTS:Simultaneous exposure to HHT and Bortezomib (10.4:1) resulted in a significant reduction of cell proliferation in SKM-1 cells (P < 0.05). Cell cycle arrest at G0/G1 and G2/M phase was observed (P < 0.05). HHT and Bortezomib synergistically induced cell apoptosis by regulating members of caspase 9, caspase 3 and Bcl-2 family (P < 0.01). The mechanisms of the synergy involved Akt and NF-κB signaling pathway inhibition, downregulation of mature miR-3151 and increment of downstream p53 protein level. Overexpression of miR-3151 promoted cell proliferation and inhibited p53 protein expression in SKM-1 (P < 0.01).CONCLUSIONS:HHT and Bortezomib synergistically inhibit SKM-1 cell proliferation and induce apoptosis in vitro. Inhibition of Akt and NF-κB pathway signaling contribute to molecular mechanism of HHT and Bortezomib. miR-3151 abundance is implicated in SKM-1 cell viability, cell proliferation and p53 protein expression.
Research on joint application of chrysanthemum morifolium and aclarubicin in promoting HL-60 cells apoptosis

Abstract:Objective To explore the effects of combined use of chrysanthemum morifolium and aclarubicin on inducing apoptosis of leukemia cells( HL-60). Methods Leukemia cells( HL-60) were treated with aclarubicin at the dose of 0. 05 μg /ml,0. 1 μg / ml,0. 25 μg / ml,chrysanthemum morifolium at the concentration of 500 μM,and chrysanthemum morifolium at the concentration of 10 μM,100 μM,500 μM in the presence of low dose of aclarubicin( 0. 1 μg / mL) for 20 h respectively.Then the apoptotic rate was analyzed by flow cytometry,the protein expression of COX-2,NF-kB,caspase 3 and caspase 9were estimated by Western-blot method. Results Low-concentration aclarubicin had little effect on apoptotic rate of HL-60cells,as well as caspase 3,caspase 9,COX-2,NF-κB expressions on HL-60 cells. Combined treatment of chrysanthemum morifolium and lower-concentration aclarubicin can enhance the apoptotic rate,expression of caspase 3 and caspase 9,down-regulate COX-2 expression. This effect was highly correlated with the concentration of chrysanthemum morifolium. NF-κB activity was obviously suppressed by combination of chrysanthemum morifolium and aclarubicin. Conclusion Chrysanthemum morifolium can enhance the aclarubicin-mediated apoptotic effect on HL-60 cells,up-regulate expression of caspase 3 and caspase 9,down-regulate COX-2 expression in HL-60 cells via suppressing activity of NF-κB.

Abstract:Objective To investigate the effects of bortezomib on human osteosarcoma cells from the HOS cell line, and the underlying associated mechanisms. Methods Viability of HOS cells treated with bortezomib (5–20 nM) for different time periods was measured and changes in the cell cycle were assessed. Apoptosis and autophagy in HOS cells treated with bortezomib were analysed using annexin V-fluorescein isothiocyanate assay, transmission electron microscopy and Western blotting. Surges in mitogen-activated protein kinase (MAPK) pathways including MAPK/extracellular signal-regulated kinase (ERK) kinase (MEK1/2), ERK1/2, c-Jun N-terminal kinase (JNK) and p38 MAPK were analysed using Western blotting. Results Bortezomib induced growth inhibition in a time- and dose-dependent manner, and autophagy and apoptosis in a dose-dependent manner, in HOS cells. HOS cell autophagy and apoptosis in response to bortezomib, corresponded with changing levels of intracellular MAPK signalling molecules. Conclusions This study provided new insights into the mechanisms underlying bortezomib-induced apoptosis in human osteosarcoma HOS cells, and suggests that bortezomib could be a potent chemotherapeutic agent in the treatment of osteosarcoma.
Effect of Bortezomib Combined with Doxorubicin on Lymphoblastic Lymphoma Molt-4 Cells

Abstract:Objective To investigate the effect of bortezomib combined with doxorubicin on lymphoblastic lymphoma cell line Molt-4.Methods Molt-4 cells were cultured in the presence of bortezomib and doxorubicin,cell viability was monitored by CCK-8 and trypan-blue exclusion.Apoptosis was detected by flow cytometry and mitochondrial membrane potential,expression of Fas was also measured with flow cytometry.Results Molt-4 cell proliferation was substantially inhibited in concentration-dependent manners when treated with either bortezomib or doxorubicin.The combination of both drugs synergistically inhibited Molt-4 cell proliferation at 48 hours [(57.24±0.10) ％].Combination therapy further enhanced bortezomib and doxorubicin induced apoptosis [48 h (23.08±1.25) ％] (P ＜ 0.05).Detection of mitochondrial membrane potential showed that combination therapy could promote apoptosis (15.84 ％,5.38 ％,5.52 ％) but did not significantly change the level of Fas expression (P ＞ 0.05).Conclusion Combination therapy of bortezomib with doxorubicin efficiently inhibits proliferation and induces apoptosis of Molt-4 cells.Activation of mitochondrial and intrinsic apoptotic pathway may play important roles.

Abstract:Sepin-1, a potent non-competitive inhibitor of separase, inhibits cancer cell growth, but the mechanisms of Sepin-1-mediated growth inhibition are not fully understood. Here we report that Sepin-1 hinders growth of breast cancer cells, cell migration, and wound healing. Inhibition of cell growth induced by Sepin-1 in vitro doesn’t appear to be through apoptosis but rather due to growth inhibition. Following Sepin-1 treatment caspases 3 and 7 are not activated and Poly (ADP-ribose) polymerase (Parp) is not cleaved. The expression of Forkhead box protein M1 (FoxM1), a transcription factor, and its target genes in the cell cycle, including Plk1, Cdk1, Aurora A, and Lamin B1, are reduced in a Sepin-1-dependent manner. Expressions of Raf kinase family members A-Raf, B-Raf, and C-Raf also are inhibited following treatment with Sepin-1. Raf is an intermediator in the Raf-Mek-Erk signaling pathway that phosphorylates FoxM1. Activated FoxM1 can promote its own transcription via a positive feedback loop. Sepin-1-induced downregulation of Raf and FoxM1 may inhibit expression of cell cycle-driving genes, resulting in inhibition of cell growth.

Methods Viability of HOS cells treated with bortezomib (5-20nM) for different time periods was measured and changes in the cell cycle were assessed. Apoptosis and autophagy in HOS cells treated with bortezomib were analysed using annexin V-fluorescein isothiocyanate assay, transmission electron microscopy and Western blotting. Surges in mitogen-activated protein kinase (MAPK) pathways including MAPK/extracellular signal-regulated kinase (ERK) kinase (MEK1/2), ERK1/2, c-Jun N-terminal kinase (JNK) and p38 MAPK were analysed using Western blotting. Results Bortezomib induced growth inhibition in a time- and dose-dependent manner, and autophagy and apoptosis in a dose-dependent manner, in HOS cells. HOS cell autophagy and apoptosis in response to bortezomib, corresponded with changing levels of intracellular MAPK signalling molecules. Conclusions This study provided new insights into the mechanisms underlying bortezomib-induced apoptosis in human osteosarcoma HOS cells, and suggests that bortezomib could be a potent chemotherapeutic agent in the treatment of osteosarcoma.

Abstract:Objective:To explore the machanism of apoptosis induced by Ara-C in HL-60 cells.Methods:Cell viability was determined by trypan blue exclusion,the growth inhibition was analyzed by MTT assay;cell apoptosis by morphology and flow cytometry;Bcl-2 and caspase-3 by western blot.Results:Ara-C(1-100μM)inhibited cellular proliferation and induced apoptosis and reducde the levels of Bcl-2 proteins and activated caspase-3 in a dose and time dependent fashion in HL-60 cells.PD98059(25μM)(a inhibitor of MEK)enhanced apoptosis and activity of caspase-3 induced by Ara-C without altering Bcl-2 protein.However,SB203580(20μM)(a inhibitor of P38MAPK)reduced the apoptosis and activity of caspase-3 induced by Ara-C without altering Bcl-2 protein.SP600125(a inhibitor of JNK)did not affected effects of Ara-C.Conclusion:Ara-C downregulates Bcl-2 and activates caspase-3 during apoptosis in HL-60 cells;a inhibitor of MEK enhances apoptosis induced by Ara-C.However,a inhibitor of P38MAPK reduces the effects of Ara-C in HL-60 cells.
Bortezomib Potentiates Antitumor Activity of Mitoxantrone Through Dampening Wnt/β-catenin Signal Pathway in Prostate Cancer Cells

Abstract:Background Bortezomib (BZM), alone or in combination with other chemotherapies, has displayed strong anticancer effects in several cancers. The efficacy of the combination of BZM and mitoxantrone (MTX) in treating prostate cancer remains unknown. Methods Anticancer effects of combination of BZM and MTX were determined by apoptosis and proliferation assay in vivo and in vitro. Expression of β-Catenin and its target genes were characterized by western blot and Real-time PCR. Results BZM significantly enhanced MTX-induced antiproliferation in vivo and in vitro. Mice administered a combination of BZM and MTX displayed attenuated tumor growth and prolonged survival. BZM significantly attenuated MTX-induced apoptosis. Moreover, the combination of BZM and MTX contributed to inhibition of the Wnt/β-Catenin signaling pathway compared to monotherapy. Conclusions This study demonstrates that BZM enhances MTX-induced anti-tumor effects by inhibiting the Wnt/β-Catenin signaling pathway in prostate cancer cells.
[Beta-Elemene Enhances Aclarubicin-Induced Apoptotic Effect in HL-60 Cells and Its Mechanism.].

Abstract:OBJECTIVE:To explore the effects of beta-elemene combined with aclarubicin on the induction of HL-60 cell apoptosis and its mechanisms in antileukemia therapy.METHODS:HL-60 cells were treated for 20 hours with different dose of aclarubicin (0.05, 0.10, 0.25 microg/ml) or with different concentrations of beta-elemene (10, 20, 40 microg/ml) in the presence or absence of aclarubicin (0.10 microg/m). The apoptotic rate was analyzed by flow cytometry (FCM), the productions of PGE2 in culture supernatants was detected by competitive ELISA and the expressions of COX-2 and NF-kappaB activity in HL-60 cells by Western blot.RESULTS:Lower concentration of aclarubicin (0.05, 0.10 microg/ml) didn't affect apoptotic rate, and COX-2, NF-kappa B and PGE2 expression on HL-60 cells. Combined treatment of beta-elemene and aclarubicin (0.10 microg/ml) enhanced the apoptotic effect and down-regulated COX-2, NF-kappaB and PGE2 expressions. There was a positive correlation between the effects and beta-elemene concentrations.CONCLUSION:beta-elemene enhances aclarubicin-mediated apoptotic effect, down-regulation of COX-2 and their inducing products PGE2 in HL-60 cells by suppressing activitation of NF-kappaB.
