Abstract:Herein, we present a novel strategy for synthesizing polyfluorinated compounds by the fluorotrifluoromethylation of olefins, which was achieved through a new trifluoromethyl-iodine(III) reagent TFNI-1. TFNI-1 was readily synthesized via a three-step process, and its structure was characterized by NMR spectroscopy and X-ray crystallography. It is shown by radical trapping and radical clock experiments that the reaction involves the CF(3) radical intermediate.

Abstract:Atom transfer radical addition and Heck-type reaction between alkenes and ethyl iododifluoroacetate by using organophosphine compounds as catalysts have been developed. The reaction proceeds under mild reaction conditions with excellent functional group tolerance and high chemo- and regioselectivities. Mechanistic study indicates that the reaction might be initiated via noncovalent interactions between an organophosphine catalyst and a carbon–iodine bond.

Abstract:Hexafluoroisopropanol (HFIP)-catalyzed direct dehydroxydifluoro-alkylation of benzylic and allylic alcohols with difluoroenoxysilanes is developed. This procedure enables the synthesis of a broad range of alpha,alpha-difluoroketones, a class of highly valuable intermediates and building blocks in medicinal and organic chemistry. Here, we have demonstrated for the first time that HFIP could act as a powerful catalyst for fluorinated carbon-carbon bond formation. The application of this protocol in late-stage dehydroxydifluoroalkylation of potentially bioactive drugs and natural products has also been carried out.

Abstract:Abstract We report a simple protocol for (ethoxycarbonyl)difluoromethylthiolation of nucleophilic compounds using a difluoroalkyl sulfonium salt which can be prepared in situ via Tf2O-triggered electrophilic activation of a benzyl difluoroalkyl sulfoxide. With the protocol, difluoroalkylthiolated arenes, heteroarenes, α-difluoroalkylthiolated carbonyl compounds, etc. were obtained smoothly with good to excellent yields. Merits of the reaction include the readily available difluoroalkylthiolation reagent and substrates, mild conditions, and excellent regioselectivity.

Abstract:AbstractS‐((Phenylsulfonyl)difluoromethyl)thiophenium salts were designed and prepared by a triflic acid catalyzed intramolecular cyclization of ortho‐ethynyl aryldifluoromethyl sulfanes. The thiophenium salts were found to be efficient as electrophilic difluoromehtylating reagents for introduction of a CF2H group to sp3‐hybridized carbon nucleophiles such as of β‐ketoesters and dicyanoalkylidenes. The (phenylsulfonyl)difluoromethyl group can be readily transformed into CF2H under mild reaction conditions. Enantioselective electrophilic difluoromethylation was also achieved in the presence of bis(cinchona) alkaloids.

Abstract:Compounds bearing fluorinated moieties are pervasive in a wide range of pharmaceuticals and agrochemicals. The installation of fluorinated units is a persistently vital task in synthetic chemistry, where facile and manipulable assays are highly demanding. Herein, we establish a general and programmable fluorination strategy for the modular assembly of mono- and difluoromethylarenes through the controllable deprotonation and fluorination of phosphonium ylides. Abstract:Under visible light irradiation, o-phosphinophenolate functions as an easily accessible photoredox catalyst to activate trifluoromethyl groups in trifluoroacetamides, trifluoroacetates, and trifluoromethyl (hetero)arenes to deliver corresponding difluoromethyl radicals. It works in relay with a thiol hydrogen atom transfer (HAT) catalyst to enable selective defluoroalkylation and hydrodefluorination. The reaction allows for the facile synthesis of a broad scope of difluoromethylene-incorporated carbonyl and (hetero)aromatic compounds, which are valuable fluorinated intermediates of interest in the pharmaceutical industry. The ortho-diphenylphosphino substituent, which is believed to facilitate photoinduced electron transfer, plays an essential role in the redox reactivity of phenolate. In addition to trifluoromethyl groups, pentafluoroethyl groups could also be selectively defluoroalkylated.

Abstract:Herein, we describe the design and synthesis of a difluoromethylsulfonyl imidazolium salt, which can act as a radical difluoromethylation reagent to achieve the challenging amino- and oxy-difluoromethylation of alkenes. Notably, the three steps for the synthesis of the imidazolium salt do not require any tedious distillation or column chromatography purification process, and the amino- and oxy-difluoromethylation paths are simply determined by the selection of reaction solvents.

Abstract:A simple and robust method for electrochemical alkyl C–H fluorination is presented. Using a simple nitrate additive, a widely available fluorine source (Selectfluor), and carbon-based electrodes, a wide variety of activated and unactivated C–H bonds are converted into their C–F congeners. The scalability of the reaction is also demonstrated with a 100 gram preparation of fluorovaline.

Abstract:Herein, we report a simple and efficient method for the direct installation of chlorodifluoroethyl group onto aromatic molecules of various aromatic amides with a new 2-chloro,2,2-difluoroethyl(mesityl)iodonium salt (CDFI). Abstract:A general approach towards diverse fluorinated phosphonates via geminal difunctionalization reactions of α-diazo arylmethylphosphonates is described. The diazo functionality (RR'C[double bond, length as m-dash]N) is successfully converted to RR'CF, RR'CHF, RR'CFBr or RR'CFNR'' groups by employing different fluorination reagents. A variety of fluorinated organophosphorus compounds were readily accessed in good to excellent yields from a common type of precursor.

Abstract:Fluorinated small molecules are commonly utilized in the realm of functional small-molecule chemistry, and N-difluoromethyl (N-CF2H) compounds are especially fascinating because of their unique and unexplored physiochemical properties. However, despite limited progress, a general applicable methodological approach to the synthesis of N-CF2H compounds remains elusive. Herein, guided by computation, we present an operationally simple and practical protocol to access N-CF2H amides and related carbonyl derivatives. The protocol utilizes a one-pot conversion of thioformamides via desulfurization-fluorination and acylation, providing N-difluoromethylcarbamoyl fluoride building blocks that can be further diversified to a diversity of unexplored N-CF2H carbonyl compounds with rich functionality. Furthermore, preliminary studies on their properties and stability showcased their application in pharmaceuticals and agrochemicals.

Abstract:The direct 1,6-nucleophilic difluoromethylation, trifluoromethylation, and difluoroalkylation of para-quinone methides (p-QMs) with Me3SiRf (Rf = CF2H, CF3, CF2CF3, CF2COOEt, and CF2SPh) under mild conditions are described. Although Me3SiCF2H shows lower reactivity than Me3SiCF3, it can react with p-QMs promoted by CsF/18-Crown-6 to give structurally diverse difluoromethyl products in good yields. The products can then be further converted into fluoroalkylated para-quinone methides and α-fluoroalkylated diarylmethanes.
