Abstract:An unprecedented [2,3]-sigmatropic rearrangement reaction of quaternary 2,3-allenylammonium ylides, generated in situ from tertiary 2,3-allenylamines and arynes, has been established. With 2-(trimethylsilyl)aryl triflates as aryne precursors, a range of tertiary 2,3-allenylamines bearing an electron-withdrawing group at the alpha-position smoothly participated in the aryne-mediated [2,3]-sigmatropic rearrangement at room temperature, delivering structurally diverse 2-vinylallyamines or 1-amino-1,3-dienes in moderate to excellent yields. Abstract:A new strategy has been established for the [2,3]-sigmatropic rearrangement of quaternary allylammonium ylides, generated in situ from tertiary allylamines and a bicyclo[1.1.0]butane via strain-release C-C bond cleavage. A range of tertiary allylamines with an acidic α-C-H bond participated in the 1-sulfonylbicyclo[1.1.0]butane-mediated [2,3]-sigmatropic rearrangement, delivering structurally diverse N-cyclobutyl homoallylamines in moderate to excellent yields.

Abstract:A highly efficient asymmetric [1,3]- and [3,3]-O-to-C sigmatropic rearrangement of 3-allyloxy-4 H -chromenones and their analogues was developed. Chiral N , N ′-dioxide complexes of 3d late transition metal complexes enabled two mechanistically different processes, giving a series of optically active 2,2-disubstituted chromane-3,4-diones and 2-allyl-3-hydroxy-4 H -chromen-4-ones as well as their related compounds in excellent yield and enantioselectivity. Systemic mechanistic studies and DFT calculation revealed the nature of the vinyl ether unit of the substrate, which biased regioselectivity via a stepwise tight ion pair pathway and a concerted pericyclic pathway, respectively. The enantioselectivity of the two processes is also disclosed.

Abstract:Recent advantages in the field of aryl sulfonium salts made these salts readily avaible in one chemical step from common functional groups. A new class of aryl sulfonium salts was developed to improve the site-selectivity of the aryne formation via deprotonation/elimination sequence. Mechanism study revealed an intramolecular aryne translaoction pathway enabled by the reversible proton transfer. Current method could enable late stage (hetero)aryne formation on the pharmaceuticals and greatly reduce the synthetic effort to access structurally highly diversed (hetero)aryne. Mild condition and broad scope pro-vide orthogonal reactivity compared with Kobayashi aryne precursors.

Abstract:Give Me an Ar, give Me an N! Arylation of the methyl group in a simple derivative of readily available alanine under palladium catalysis was followed by intramolecular amidation at the same position to give chiral α-amino-β-lactams with a wide range of aryl substituents (see scheme; Phth=phthaloyl). The α-amino-β-lactams were obtained in moderate to high yields with good functional-group tolerance and high diastereoselectivity.

Abstract:The radical Truce–Smiles rearrangement is a straightforward strategy for incorporating aryl groups into organic molecules for which asymmetric processes remains rare. By employing a readily available and non-expensive chiral auxiliary, we developed a highly efficient asymmetric photocatalytic acyl and alkyl radical Truce–Smiles rearrangement of α-substituted acrylamides using tetrabutylammonium decatungstate (TBADT) as a hydrogen atom–transfer photocatalyst, along with aldehydes or C-H containing precursors. The rearranged products exhibited excellent diastereoselectivities (7:1 to >98:2 d.r.) and chiral auxiliary was easily removed. Mechanistic studies allowed understanding the transformation in which density functional theory (DFT) calculations provided insights into the stereochemistry-determining step.

Abstract:An efficient asymmetric remote arylation of C(sp 3 )−H bonds under photoredox conditions is described here. The reaction features the addition radicals to a double bond followed by a site-selective radical translocation (1, n -hydrogen atom transfer) as well as a stereocontrolled aryl migration via sulfinyl-Smiles rearrangement furnishing a wide range of chiral α -arylated amides with up to >99 : 1 er. Mechanistic studies indicate that the sulfinamide group governs the stereochemistry of the product with the aryl migration being the rate determining step preceded by a kinetically favored 1, n -HAT process.

Abstract:Transition-metal catalyzed asymmetric allylic substitution with alkyl and heteroaryl carbon nucleophiles has been well-established. However, the asymmetric allylic arylation of acyclic internal alkenes with aryl nucleophiles remains challenging and underdeveloped. Herein we report a stereospecific 3-aza-Cope rearrangement interrupted asymmetric allylic substitution-isomerization (Int-AASI) that enables asymmetric allylic arylation. By means of this stepwise strategy, both enantioenriched allylic arylation products and axially chiral alkenes could be readily obtained in high enantioselectivities. Experimental studies support a mechanism involving a cascade of asymmetric allylic amination, stereospecific aryl 3-aza-Cope rearrangement and alkene isomerization. Density functional theory studies detailed the reasons of achieving the high chemoselectivity, regioselectivity, stereoselectivity and stereospecificity, respectively.

Abstract:Starting with vinyl sulfoxides and propargyl silane, the Au(I)-catalyzed asymmetric [3,3]-sigmatropic rearrangement of sulfonium intermediate furnished, after protodemetallation step and hydrolysis of thionium ion, the corresponding 4-oxo-2-aryl-2-alkyl-pentanal derivatives. From the fine analysis of crude mixtures representative of this transformation, formal α-arylated products of acetone (1-arylpropan-2-one derivatives) and aryl sulfanes were also isolated and characterized, depending on the substrates and reaction conditions used. Therefore, a tentative mechanistic explanation of the formation of these unexpected products was highlighted and DFT calculations have also streamlined the reactivity of these cationic gold(I)-catalyzed transformations.

Abstract:The diastereoselective [2,3]-sigmatropic rearrangements of lithium N-benzyl-O-allylhydroxylamides bearing a stereogenic centre adjacent to the migration terminus are examined. (E)-N-Benzyl-O-(4-phenylpent-2-enyl)hydroxylamine rearranges in 30% de to afford syn-(3RS,4RS)-3-(N-benzyl-N-hydroxy)-4-phenylpent-1-ene as the major diastereoisomer, consistent with the rearrangement proceeding under moderate steric control. Rearrangements of both lithium (E)- and (Z)-N-benzyl-O-(4-methoxy-4-phenylbut-2-enyl)hydroxylamides furnish syn-(1RS,2RS)-1-phenyl-1-methoxy-3-(N-benzylamino)but-3-ene in ≥90% and 88% de respectively, consistent with these rearrangements proceeding under chelation control.

Abstract:The use of a chiral ligand for stereocontrol has assisted the development of a number of asymmetric functionalization of proximal C–H bonds. Herein, we report a chiral ligand-controlled, asymmetric remote meta-C–H activation of arenes, leading to asymmetric C–H olefination and arylation of hydrocinnamic acid derivatives through desymmetrization with Ac-L-Phe-OH as the chiral ligand using a Pd(II) catalyst. The origins of the enantioselectivity were explained with density functional theory calculations. The larger distortion energy of the substrate part in the C–H bond activation transition structure S-TS1 is the major controlling factor that disfavors the formation of the S-enantiomer product.

Abstract:The charge-accelerated aza-Claisen rearrangement of ammonium salts serves as a key step in the construction of complex nitrogen-containing molecules. However, much less attention has been paid to the aromatic aza-Claisen rearrangement than to the aliphatic one. Herein, we report an unprecedented aromatic aza-Claisen rearrangement of arylpropargylammonium salts, generated in situ from arynes and tertiary propargylamines, delivering structurally diverse 2-propargylanilines in moderate to good yields with high regioselectivity. This rearrangement proceeds in the absence of strong bases or transition metals, is compatible with moisture and air, tolerates a wide variety of functional groups, and is amenable to forming 11- to 13-membered heterocycles with a triple bond. The 2-propargylaniline products were treated with aluminum chloride in ethanol to afford multisubstituted indoles in moderate to excellent yields. Finally, a series of deuterium-labeling experiments was performed to elucidate the reaction mechanism.

Abstract:Highly enantioselective alpha-arylation of simple ketones has been achieved by chiral primary amine catalyzed asymmetric retro-Claisen cleavage of beta-diketones. This mild organocatalytic strategy enables the construction of alpha-aryl tertiary carbon stereocenters in good yields and excellent enantioselectivities (up to 98 % ee) with the para-quinone monoimines as aryl sources. Furthermore, oxidative catalytic asymmetric alpha-arylation has also been realized with free p-aminophenols.

Abstract:Arylethylamines are crucial elements in pharmaceutical molecules, making methods for their synthesis highly significant. The Truce-Smiles rearrangement is a well-developed strategy to synthesize arylethylamine motifs via aryl migration. However, most examples require amide substrates to activate the alkene to attack by a radical precursor. This strategy both limits the product scope to amide containing compounds as well as necessitating the incorporation of specific functional groups arising from the initial radical addition. In this work, we overcome these limitations, delivering a hydrogen-atom-transfer from a cobalt catalyst to unactivated alkenes to yield -arylethylamines with simple alkyl chains. DFT studies reveal that increasing the steric hindrance in at least one of the ortho positions on the migrating aromatic group promotes ipso over ortho addition, selectivity that contrasts with previous methods.

Abstract:A ternary catalytic system comprising a chiral aldehyde, a transition metal, and a Lewis acid is rationally designed for the asymmetric α-allylic alkylation reaction of aza-aryl methylamines and π-allylmetal electrophiles. Structural diversity chiral amines bearing carbon–carbon double bonds and aza-heterocycles are produced in moderate to good yields with good to excellent enantioselectivities. These products can be readily converted into other chiral amines without the loss of enantioselectivity.
