Abstract:Paracheirodon innesi was exposed (simultaneously or sequentially) to copper and lead in varying concentrations in a synthetic water solution. Metal accumulations in fish gills and the fish (without gills) were measured at the end of the exposure period. Results of the exposure experiment indicated that lead seems to facilitate the uptake of copper and vice versa when the exposure process is simultaneous. In the case of sequential exposure first to copper for 7 days then to lead for 20 days, lead accumulations in the fish and the gills were enhanced by copper, indicating a possible alteration of the physiological absorption process. The synergistic effects observed during either simultaneous or sequential exposure experiments can be described using logistic equations.

Abstract:Carassius auratus were exposed to lead in synthetic water. Mucus secreted from the fish was measured and characterized for its lead binding capacity. The increase in mucus concentration in the water depended on secretion and degradation rates and the former was speeded up by lead. The complexation between mucus and lead can be described based on a single-site interaction model. Lead speciation in the system with or without mucus was calculated using chemical equilibrium modeling. The dominant species of lead was PbCO(3)(0). Other major species included Pb(OH)(+), Pb(OH)(2)(0), Pb(CO(3))(2)(2-), Pb-mucus, and Pb(2+), among which free lead and lead hydroxo complexes were available to fish uptake.

Abstract:mu Aquaculture industries supply half of the global production, leading to an increasing concern about health risk of the aquaculture products because of widespread distribution of heavy metals in the environment. Considering high dependence of toxicity and bioaccumulation of one metal not merely upon total content but mainly upon its chemical forms, recent investigations into risk assessment of aquaculture products based on total contents of heavy metals may be unreasonable and unfair. In this study, we conducted reasonable assessment to bioaccumulation and health risk of aquaculture fish and crustacean collected from eight aquaculture ponds in Zhejiang Province of East China in terms of chemical forms of two model heavy metals (Sn(II), Pb(II), four organotin compounds, trimethyl lead -TML and triethyl lead -TEL). Although all metal compounds were presented below 0.2-4.1 mu g kg(-1) in aquatic food, they were at moderate contents in environmental matrices following an order of sediment > feedstuff > aquatic food > water for most of the target compounds (i.e. 54-376 mu g kg(-1) Pb(II) in sediment, 14-94 mu g kg(-1) Pb(II) in feedstuff and organotin compounds at pg L-1 levels in water), indicating feedstuff as an important exposure source apart from water adsorption. Bioaccumulation factors, biota-sediment accumulation factors and biomagnification factors demonstrated low bioaccumulation probability although the bioaccumulation level followed an order of organotin and organolead > inorganic forms > total metal. In addition, estimated daily intakes, target hazard quotients and cancer risk demonstrated no health risk for all tin compounds and no carcinogenic risks for all lead compounds but moderate non-carcinogenic risks for TML and TEL. Overall, the accumulation level and health risk were in a closer relationship with the chemical forms of Sn and Pb than their total contents, demonstrating a high necessity to risk assessment in terms of chemical forms.

Abstract:Understanding the interactions between different environmental pollutants is necessary in ecotoxicology since environmental contaminants never appear as single components but rather in combination with other substances. Heavy metals and pesticides are commonly detected in the environment, but the characterization of their mixture toxicity has been inadequately explored. This research aimed to elucidate the mixture impacts of the heavy metal lead (Pb) and the pesticide procymidone (PCM) on the hook snout carp ( Opsariichthys bidens ) using an array of biomarkers. The data showed that Pb and PCM possessed almost equivalent acute toxicity to the animals, with 4-days LC 50 values of 120.9 and 85.15 mg L −1 , respectively. Combinations of Pb and PCM generated acute synergistic effects on O. bidens . The contents of malondialdehyde (MDA), antioxidative (SOD), apoptotic (caspase-9), and detoxifying enzymes glutathione S-transferase (GST) and cytochrome P450 (CYP450) significantly changed after most of the mixture exposures compared with the baseline level and the corresponding individual exposures. This suggests the induction of oxidative stress, cell damage, and detoxification dysfunction. The expressions of eight genes ( mn-sod , cu-sod , p53 , cas3 , erβ1 , esr , ap , and klf2α ) associated with oxidative stress, cell apoptosis, immune response, and hormonal functions exhibited pronounced changes when challenged with the mixture compared to the individual treatments. This indicates the occurrence of immune dysregulation and endocrine disorder. These findings provide an overall understanding of fish upon the challenge of sublethal toxicity between Pb and PCM and can be adopted to evaluate the complicated toxic mechanisms in aquatic vertebrates when exposed to heavy metal and pesticide mixtures. Additionally, these results might guide environmental regulation tactics to protect the population of aquatic vertebrates in natural ecosystems.

Abstract:With the increasing occurrence of dietary lead (Pb) contamination in aquaculture, a better understanding of the toxic effects of dietary Pb on aquatic animals is needed. Tilapia (Oreochromis niloticus) were exposed to dietary Pb at concentrations of 0, 100, 400 and 800 mu g g-1 dry weight for 60 days, and Pb accumulation in tissues and blood, oxidative stress in posterior kidney and DNA damage in peripheral blood cells were investigated. The results showed that dietary Pb exposure resulted in significant Pb accumulation in tissues and blood, which increased with the dietary Pb concentrations. Pb accumulated in sampled tissues in the following order: posterior kidney>bone>liver>gill>spleen>testis>muscle>brain. Dietary Pb caused a significant increase in the malondialdehyde level and a significant decrease in the total antioxidant capacity content when compared with the control group (P<0.05), accompanied by concentration-dependent decreases in the glutathione content, glutathione peroxidase and superoxide dismutase activities. Pb dose-dependent DNA damage was observed in peripheral blood cells of tilapia exposed to dietary Pb. The results suggest that dietary Pb exposure can induce significant Pb accumulation in tissues and blood, followed by oxidative stress in posterior kidney and DNA damage in peripheral blood cells of tilapia.

Abstract:The study investigated the bioaccumulation of Cu, Zn, Pb and Cd in Eisenia fetida fed on pig manure in greenhouse, and its relationship to bioavailability of these elements in pig manure. Metals in exchangeable, carbonates-bound and Fe–Mn oxides-bound fractions obtained by sequential extraction were considered to be bioavailable. The bioaccumulation factors (BAFs) of E. fetida to heavy metals were Cd (2.749±0.441), Zn (0.594±0.200), Pb (0.274±0.101) and Cu (0.076±0.030). Variations in the Pb and Cd concentrations of E. fetida were best explained by the concentration of exchangeable fraction, while that of Cu was best explained by the concentrations of exchangeable and Fe–Mn oxides-bound fractions. The concentration of Zn in E. fetida depended strongly on the concentrations of exchangeable, carbonates-bound and Fe–Mn oxides-bound fractions. Further nonlinear regression analysis revealed the positive logarithmic relationship between the BAF and the exchangeable metal concentration of pig manure.

Abstract:Coexistence of heavy metals in aquatic environments exert complex effects on amphibians. Here, the adverse effects of Pb (0.14 μM) combined with Cu at concentrations of 0, 0.25, and 1.0 μM were investigated in Bufo gargarizans tadpoles. Tadpoles were chronically exposed from Gosner stage (Gs) 26 to Gs 38, and morphology of tadpoles as well as intestinal histology and bacterial community were assessed. Our results indicated that Pb+Cu1.0 exposure induced significant retardation of somatic mass, total length, intestine mass, and intestine length as well as intestinal histological alterations. Pb+Cu0.25 and Pb+Cu1.0 exposure were associated with the loss of gut bacterial diversity. Proteobacteria and Bacteroidetes were two dominant phyla in tadpoles independently of heavy metal exposure, but the abundance of Proteobacteria increased significantly in Pb+Cu1.0 group and Bacteroidetes decreased significantly in all treatment groups. Furthermore, functional prediction indicated that metabolic disorders were associated with Pb+Cu0.25 and Pb+Cu1.0 exposure. Overall, relative limited shifts in intestinal bacterial diversity, composition, and functionality caused by Pb+Cu0 exposure, while coexistence of Pb and Cu induced gut dysbiosis and might further cause disturbance of metabolic homeostasis. The findings of this study provide insights into the effects of Pb and Cu coexistence on the health of amphibians.

Abstract:In this work we studied the accumulation of heavy metals in nine species of fish with different life and feeding habitats which are native and major commercial fish in the Baotou Urban Section of the Yellow River. The results showed that the concentration of heavy metals was significantly dependent on fish species; the pollution index of heavy metals in different species were ranked as Hemiculter leucisclus > Carassius auratus auratus > Hemibarbus maculatus > Megalobrama amblycephala > Abbottina rivularis > Cyprinus carpio > Squaliobarbus curriculus > Perccottus glehni > Saurogobio dabryi. Product–moment correlation coefficients among the metal pairs Pb–Zn, Cu–Cd, Cu–Zn, Cu–Pb, Pb–Cd, and Zn–Cd revealed there was no competitions between metals in each tissue. Correlations between heavy metal concentrations and fish length or weight indicated that accumulation of the heavy metals by the different fish species was related to their surrounding environments and their life and feeding habitats. According to the mean bioconcentration factors (BCFs), the heavy metal concentrations in these nine species were ranked Zn ≫ Cu > Cd ≈ Pb. In this work, the bioaccumulation factors (BAFs) were developed by using the sum of exchangeable and bound-to-carbonate heavy metals as Cs values. It was found that BAFs better reveal the accumulation characteristics of the heavy metals in the fish, which might provide an effective method for assessing bioaccumulation of heavy metals.
Copper Uptake Kinetics And Regulation In A Marine Fish After Waterborne Copper Acclimation

Abstract:The uptake kinetics and regulation of copper in a marine predatory fish, the black sea bream Acanthopagrus schlegeli after acclimation to waterborne Cu were examined, using radiotracer techniques. The dissolved Cu uptake followed a linear pattern during the time of exposure, and the calculated uptake rate constant was 6.24L kg(-1) day(-1). The efflux rate constant was 0.091 day(-1) following dietary uptake of Cu, and the dietary assimilation efficiency (AE) of Cu varied between 1.7% and 10.9% after the fish were fed with three types of prey (oysters, clams and brine shrimp). After the fish were acclimated at a nominal concentration of 50 mu g Cu L-1 for 14 days, the Cu uptake rate and efflux rate constant did not change significantly, but the Cu body concentrations and metallothionein (MT) concentrations in fish tissues increased significantly Subcellular Cu distributions were also modified. Significant MT induction was observed in response to increased Cu tissue concentrations, indicating that MT rather than the uptake kinetics may play a primary role in Cu regulation during waterborne Cu acclimation in this marine fish. Moreover, the high Cu efflux may also be important in Cu regulation during long-term exposure. Our modeling calculations indicated that dietary uptake was likely to be the main route for Cu bioaccumulation in the fish, and the relative contribution of waterborne and dietary uptake depended on the bioconcentration factor (BCF) of the prey and ingestion rate of fish. Abstract:A comparison of copper accumulation in Paracheriodon innesi and dialysis membrane in the contaminated fish pond water was conducted both in lab and in situ. The results showed that the amount of copper absorbed by the dialysis membrane was closely correlated to the amount of copper accumulated in fish (gills). It is a feasible way to monitor the accumulation in aquatic creatures in a long term.

Abstract:. Carassius auratus were exposed to mixtures of constant free lead (0.1 mg/L) and particulate lead (adsorbed on gibbsite particles) at varied concentrations (0, 0.4, 0.8, 1.2, 2.0, and 3.0 mg/L). The concentrations of lead and aluminum in the gills and intestines of the fish were determined after exposure. The gills were examined (after the exposure) with a light microscope for any particles adhering to the surface. The examination results revealed that lead accumulations on the gills increased with increased particulate lead concentrations in the ambient water (under conditions of a constant level of free lead), indicating the bioavailability of lead via the gills. Gibbsite particles were also observed on the gill surface after exposure. The ratio of lead and aluminum in the gills was much higher than that in the particles. Our results suggest that there was a two-step process involved in the uptake of particulate lead by fish gills: (1) adherence of the particles on the gill surface where mucus was attached; and (2) desorption of lead from the particles under conditions of the gill microenvironment.

Abstract:Uptake of lead by goldfish ( Carassius auratus L.) and the effect of mucus on the uptake were studied. The mechanisms of lead uptake are discussed. It was demonstrated that the quantity of mucus released from the fish gill increased with the increasing in lead concentration in water and exposure duration. The mucus is capable of binding metals. Lead accumulated in gill was positively proportional to the lead level in water at low concentration (total lead less than 0.6mg/L). The plot of gill lead against water lead leveled off with increased lead concentration. A Langmuir isotherm can fit the observed data well and the saturated sorption capacity was calculated as 0.9424 mmol/kg (dry wt.). The binding cooperativity ( H n ) is very close to unit (1.012) indicating independent binding to the sites with unique apparent affinity. On average, the number of binding sites on the gill of each fish (96.0 mg,wet wt.) was estimated to be 1.62×10 -5 mmol with a conditional binding constant (log K Pb\|gill ) of 4.8. A significant linear relation of lead concentrations between gill and blood was observed during the 10\|day experiment, suggesting that the free lead in water is first bound on fish gill, then migrate across branchial epithelium to blood via passive diffusion or carrier transport.
Single and combined effects of carbamazepine and copper on nervous and antioxidant systems of zebrafish (Danio rerio)

Abstract:Various pollutants co-exist in the aquatic environment such as carbamazepine (CBZ) and copper (Cu), which can cause complex effects on inhabiting organisms. The toxic impacts of the single substance have been studied extensively. However, the studies about their combined adverse impacts are not enough. In the present study, zebrafish were exposed to environmental relevant concentrations of CBZ (1, 10, and 100 μg/L), Cu (0.5, 5, and 10 μg/L) and the mixtures (1 μg/L CBZ + 0.5 μg/L Cu, 10 μg/L CBZ + 5 μg/L Cu, 100 μg/L CBZ + 10 μg/L Cu) for 45 days, the effects on nervous and antioxidant systems of zebrafish were investigated. The results demonstrated that, in comparison with single exposure group, the combined presence of CBZ and Cu exacerbated the effect of antioxidant system (the ability of inhibition of hydroxyl radicals (IHR), superoxide dismutase (SOD), catalase (CAT) and glutathione S-transferase (GST)) but not nervous system (Acetylcholinesterase [AChE]). The qPCR results supported the changes of corresponding enzymes activities. Hepatic histopathological analysis verified the results of biomarkers. Our work illustrated that the toxicity of mixed pollutants is very complicated, which cannot simply be inferred from the toxicity of single pollutant, and calls for more co-exposure experiments to better understanding of the co-effects of pollutants on aquatic organisms.

Abstract:Carp were exposed to copper adsorbed on kaolin particles at various concentrations (0-2.4 mg/L), and net accumulation due to elevated adsorbed copper in the surrounding water was observed. Copper speciation in the water and fish gill microenvironment was modeled in the presence and absence of kaolin using a chemical speciation program (MINT-EQA2). The adsorption affinity constants of kaolin for copper at various pH values used in the speciation calculations were experimentally determined, and the quantitative relationship between the affinity constant and pH was modeled. Copper accumulations in fish gills exposed to kaolin-adsorbed copper was then observed. The results indicate that desorption occurred in the fish gill microenvironment due to both mucus competition of copper and slight increase in water pH. Furthermore, the available copper species increased as a result of desorption, causing a net accumulation of Cu by the gills.
Study on the Effect of Cu (II) and Zn (II) on the Accumulation of Pb (II) from Soil to the Biomass of Vegetable

Abstract:Among soil pollutants, lead (Pb) is one of the toxic metal pollutants widely used in many industrial processes and occurs as a contaminant in environment. In this study, a field experiment was carried out to investigate the accumulation of lead from soil contaminated by this metal ion on the biomass of some vegetables, including spinach, lettuce, carrots, and potatoes. The results showed that lead was cumulative metal. Besides, the level of lead accumulation in soil of the studied vegetables decreased in the order of tubers of carrots, tubers of potato, spinach root, lettuce root, stems and leaves of spinach, stem and leaves of carrot, stems and leaves of potato, and stem and leaves of lettuce, respectively. Our investigations demonstrate the effect of copper and zinc micronutrient elements which play an important role in the growth and development of plants, on the accumulation of lead from contaminated soil of the studied vegetables. The obtained results showed that high concentration of copper and zinc in soil cause competition with lead in the process of absorption and accumulation in the plant. Specifically, copper and zinc showed the inhibition effect on the uptake and accumulation of lead by these plants.
Differential response of heavy metal accumulation in freshwater aquatic organisms to organic matter pathway (δ13C) and trophic level (δ15N)

Abstract:The migration of heavy metals into aquatic ecosystems is a concern for the safety of aquatic organisms and human health. However, the migration of heavy metals from habitats to the food chain in freshwater ecosystems requires extensive exploration. We extensively investigated the levels of heavy metals in multiple media of freshwater ecosystems and explored their migration from freshwater habitats to the food chain. The results showed that the concentrations of Cr, Cu, Cd, Zn, Sb, and Pb in sediments, Cr in mollusks, and Cd in clams exceeded their standard limit values. Feeding habits, species, and body length considerably affected heavy metal levels in fish, whereas regional differences, body length, and weight considerably affected heavy metal levels in mollusks. The bioconcentration capacity (improved biotawater (sediment) accumulation factors) of the muscles was higher than that of the gills and visceral mass. Mollusks were strongly enriched in Cu, Cd, Zn, and Mn in their habitats, whereas fish were more favourable for the accumulation of Cu, Zn, and Cr. Heavy metals in sediments pose a higher risk to aquatic organisms than those in the overlying water. Trophic level (TL) (δ15N) considerably diluted levels of most metals, whereas Cu and Zn levels increased along the organic matter pathway (δ13C) in the entire aquatic food chain. The levels of typical pollutants, such as Cu, Zn, As, Cd, Pb, and Cr changed considerably along the organic matter pathway in mollusks. The levels of heavy metals in fish were not affected by TL or organic matter pathway, except for Cu. The potential risk of consuming aquatic organisms from Poyang Lake was ranked as clams > snails > fish for adults and children and As in aquatic organisms was the primary contributor to health risk.
INDIVIDUAL AND JOINT EFFECTS OF LEAD AND MERCURY ON ACETYLCHOLINESTERASE ACTIVITY IN GOLDFISH BRAIN

Abstract:The combined effects of heavy metals, lead (Pb) and mercury (Hg), in the aquatic environment are not well understood. In this study, the individual and joint effects of Pb and Hg on brain Acetylcholinesterase (AChE EC 3.1.1.7) activity in goldfish (Carassius auratus) were investigated by a 28-day exposure. The results showed that individual Pb or Hg as well as their mixtures had significantly (p<0.05) effects on AChE activities in goldfish brain. However, no significant (p<0.05) inhibition of brain AChE activities in goldfish was found in all the treatments. The brain AChE activities of goldfish in both individual and joint groups were found to be increased initially, followed by a decrease to the normal level over time. It suggested that there may be an adaptation of the fish exposed to either the two individuals or their mixtures during a long-term exposure. In addition, the comparison between the individual and joint groups showed that the effects of mixture were different from those of individuals, suggesting a different action mechanism of mixtures in the joint effects of Pb and Hg. All these results could contribute to a better understanding of the combined effects of Pb and Hg on freshwater fish.

Abstract:Mining effluents are the main source of metals in the surrounding aquatic environment. The mining district of Purple Mountain has a history of copper mining for more than 30 years, but there is limited investigation of metal bioaccumulation in the aquatic creatures from the Tingjiang river catchment affected by the mining activities. In this study, we collected grass carps (Ctenopharyngodon idellus) from four sites, and analyzed the accumulation of chromium (Cr), nickel (Ni), manganese (Mn), copper (Cu), zinc (Zn), arsenic (As), cadmium (Cd), mercury (Hg), and lead (Pb) in ten tissues (scale, skin, muscle, gill, liver, kidney, fish maw, heart, stomach, and intestine) of the fish samples. Among all tissue samples, the highest concentrations (micrograms per gram wet weight) of Ni (0.263), Cu (69.2), Zn (84.0), As (0.259), Cd (0.640), Hg (0.051), and Pb (0.534) were noted in the liver, gill, and kidney tissues, whereas the highest concentrations of Cr (0.356) and Mn (62.7) were detected in the skin and intestine, respectively. These results gave a better understanding of the variability of metals distribution in different fish tissues. In comparison with the sample sites, metals (especially Mn, Cu, Zn, Ni, and Pb) in liver, gill, kidney, stomach, and intestine showed more inter-site differences than other tissues. The inter-site differences also revealed that site 1 and 2 increased fish uptake of Cu, Zn, Ni, and Pb, which may indicate that the copper mine and urban effluents contributed to high levels of these metals in aquatic environments in site 1 and 2. A potential food safety issue may emerge depending on the mining activities in this region because some metals in a few tissue samples exceeded the guideline values for human consumption of fish.
Investigating the Impact of Humic Acid on Copper Accumulation in Sinonovacula constricta Using a Toxicokinetic–Toxicodynamic Model

Abstract:In aquatic ecosystems, the interaction between heavy metals and dissolved organic carbon (DOC) plays a pivotal role in modifying the bioavailability of these metals. This study, employing a toxicokinetic–toxicodynamic model, delves into the interactive effects of humic acid (HA), a significant component of DOC, on the bioaccumulation and toxicity of copper (Cu) in the estuarine economic bivalve Sinonovacula constricta. Utilizing the stable isotope 65Cu as a tracer, we evaluated Cu uptake in S. constricta under varied DOC concentrations in a controlled laboratory setting. Our findings reveal that at DOC concentrations below 3.05 mg L−1, the bioavailability of Cu is reduced due to shifts in the speciation distribution of Cu, resulting in decreased bioaccumulation within S. constricta. Conversely, at DOC levels exceeding 3.05 mg L−1, the formation of colloidal Cu–HA complexes allows its entry into the bivalves' digestive system. Moreover, toxicity assays demonstrate an increase in S. constricta survival rates with higher DOC concentrations, suggesting a protective effect of DOC against Cu toxicity. The integration of accumulation and toxicity data infers that Cu–HA complexes, when ingested via the digestive tract, exhibit lower toxicity compared to Cu directly assimilated from the water phase. These findings emphasize the need to consider environmental DOC levels in assessing Cu pollution risks and provide insights for managing heavy metal toxicity in estuarine aquaculture.

Abstract:Characteristics of accumulation and distribution of Cu and Zn in the freshwater mussel,Cristaria plicata,exposed to individual and mixed metals were investigated under the laboratory conditions.Results showed that the Cristaria plicata mussel could concentrate Cu and Zn in each soft tissue,the contents in which linearly increased with exposure times from 0 to 8 days.The tissues of gill and mantle accumulated Cu and Zn more rapidly than the other tissues(p< 0.05).Significant differences in accumulation rates between Cu and Zn were found at the tissues of gill,viscera and gonad(p <0.05)and the interaction of the two metals during accumulation was antagonistic.Before the metal exposure,metal contents in different tissues were with the sequences of gonad >mantle,viscera >gill,foot >muscle and the contents varied 10-fold range for Cu(p<0.05),and gill>viscera>gonad>foot,mantle>muscle and the contents varied 4-fold range for Zn(p <0.05).The metal exposures caused Cu and Zn contents reached the highest values in the mantle and gill respectively and the differences in Cu contents and Zn contents among each tissue were improved.The ranges of Cu contents varied in different tissues were higher than that of Zn contents varied before and after the exposure.At the different metal exposures,the bioconcentration factor(BCF)of Zn is higher than that of Cu obviously in each tissue of the test mussel.
