Abstract:An analytical method for determination for tebufenozide,methoxyfenozide and Fuxian residue in cabbage,pakchoi and tomato by high performance liquid chromatography(HPLC) was developed.Samples were extracted by blending with acidic methanol/water.The extract was purified first by n-hexane to remove some fatty impurities,then extracted with dichloromethane to discard water-soluble impurities,and finally cleaned by Florisil column chromatography.The HPLC conditions : Agilent HC-C18,methanol-water=75:25(V/V) as mobile phase,and UV detection at 230nm.The average fortified recoveries for tebufenozide,methoxyfenozide and Fuxian in cabbage,pakchoi and tomato samples were 76.9%～105%.Relative standard deviation was less than 10%,and the limit of detection of three pesticides in cabbage,pakchoi and tomato samples were 0.05mg/kg,respectively.

Abstract:A sensitive and rapid multi‐residue analytical method for plant growth regulators (PGRs) (i.e., chlormequat, mepiquat, paclobutrazol, uniconazole, ethephon and flumetralin) in apples and tomatoes was developed using high‐performance liquid chromatography/tandem mass spectrometry (HPLC/MS/MS). A homogenised sample was extracted with a mixture of methanol/water (90:10, v/v) and adjusted to pH <3 with formic acid. Primary secondary amine (PSA) adsorbent was used to clean up the sample. The determination was performed using electrospray ionisation (ESI) and a triple quadrupole (QqQ) analyser. Under the optimised method, the results showed that, except for ethephon, the recoveries were 81.8–98.1% in apples and tomatoes at the spiked concentrations of 0.005 to 2 mg/kg, with relative standard deviations (RSDs) of less than 11.7%. The limits of quantification (LOQs) were lower than their maximum residue limits (MRLs). The procedure was concluded as a practical method to determine the PGR residues in fruit and vegetables and is also suitable for the simultaneous analysis of the amounts of samples for routine monitoring. The analytical method described herein demonstrates a strong potential for its application in the field of PGR multi‐residue analysis to help assure food safety. Abstract:A new method combining modified QuEChERS (quick, easy, cheap, effective, rugged, and safe) with ultra-high performance liquid chromatography-triple quadrupole linear ion trap mass spectrometry (UHPLC-QTrap-MS/MS) was developed for simultaneous determination of 17 plant growth regulator (PGR) residues in vegetables. Quantitative data acquisition was accomplished in multiple reaction monitoring (MRM) mode, and qualitative analysis and identification of target compounds was achieved by using linear ion trap scan functions. Qualitative and quantitative analysis can be simultaneously achieved by a pair of ions in UHPLC-QTrap-MS/MS. More importantly, QTrap system can minimize the risk of false positive and negative results. To achieve the optimum performance towards the target analytes, several parameters including the content of formic acid in mobile phase and scanning mode were investigated. In the optimized conditions, 17 PGRs showed good linearity over the range of 2~50 μg/L with correlation coefficients of 0.9902~0.9999. The limits of detections (LODs) were in the range of 0.01~1.32 μg/kg. The recoveries of 17 PGRs ranged from 78.5 to 119.2% with the spiked levels of 1, 2, and 5 μg/kg into homogenized radish, and the relative standard deviations (RSDs) ranged from 0.8 to 14.8%. The method illustrated is suitable for routine quantitative and qualitative analyses of PGRs in vegetable samples.

Abstract:Objective To develop a high-throughput method for the determination of 14 plant growth regulators including 4-Chlorophenoxyacetic acid,(2,4-Dichlorphenoxy) acetic acid,1-Naphthyl acetic acid,2-Naphthaleneacetic acid,3-indolyl-acetic acid,4-(3-indolyl)-butyric acid,2,4-D-ethyl ester,2,4-D-1-butyl ester,1-Naphthyl acetamide,chlorflurenol-methy,Paclobutrazol,uniconazole,6-Furfurylaminopurine,6-Benzylaminopurine though the advanced pretreatment methods and gas chromatography tandem mass spectrometry(GC-MS/MS).Methods The sample was separated by DB-5MS column(30 m × 0.25 mm,0.25 μm) using programmed temperature.MS was conducted in multiple reaction monitoring(MRM) mode for detection,and the external standard method was used for quantification.Results A good linearity was obtained in wide range and the correlation coefficients(r) were ≥ 0.997 8.The detection limits were within 0.02 μg/kg-10.00 μg/kg.The recoveries were within 54.8％-131.6％.The relative standard deviations were within 2.39％-6.38％.Conclusion The detection method of 14 plant growth regulators using GC-MS/MS was set up.The method was fast and precise.The detection limit,recovery and the standard deviation can meet the standards of plant growth regulators residue requirement at home and abroad.

Abstract:The present paper describes the development and validation of a new reversed-phase liquid chromatography–electrospray ionization tandem mass spectrometric method for determination of 15 plant growth regulators in bean sprout and tomato. Plant growth regulators were extracted with mixed solvents methanol/acetonitrile/water/acetic acid ( v / v / v / v , 40/40/20/1) from 10 g vegetable and analyzed directly without further cleanup. The identification of each analyte was established by chromatographic retention time, analyte-specific fragmentation patterns, and relative peak area ratios of precursor/product ion pairs. The recoveries of 15 plant growth regulators in bean sprout and tomato were 70.3–110.0 %. The repeatability of this method is excellent, and relative standard deviation value is less than 10 %.

Abstract:In this study,an analytical method was established for the determination of thiamethoxam and its metabolites clothianidin,spirotetramat and its four metabolites such as BYI08330-enol-glucoside,BYI08330-ketohydroxy,BYI08330-enol and BYI08330-mono-hydroxy residues in tomatoes using QuEChERS cleanup with ultra-performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).The residues in samples were extracted with acetonitrile,dehydrated with anhydrous magnesium sulfate and sodium chloride,then purified with primary secondary amine (PSA) sorbent and C18,finally detected by ultra-performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS) under the multi-reactive ion monitoring mode (MRM) and quantified by external standard method.The results showed that good linearities for the seven analytes i.e.thiamethoxam,clothianidin,spirotetramat,BYI08330-enol-glucoside,BYI08330-mono-hydroxy,BYI08330-enol and BYI08330-ketohydroxy were observed in the concentration range of 0.2-2 000 μg/L with the correlation coefficients(r2) not less than 0.999 2.The limits of quantification(LOQs) were 0.002 mg/kg.The average recoveries at the fortified levels of 0.002,0.02,0.20 and 2.O0 mg/kg were in the range of 79.9％-104％ with the relative standard deviations of O.94％-6.4％.With the advantages of rapidness,simplicity,high sensitivity and good reproducibility,the method is suitable for the determination of thiamethoxam,clothianidin,spirotetramat and their four metabolites residues in tomatoes.

Abstract:[Aims]A method was established for determining plant hormones residues in food by ultra performance liquid chromatography-tandem mass spectrometry(UPLC/MS/MS).[Methods] The samples were extracted with methanol-water(90∶10,by vol),and cleaned up by WCX cartridge.The target analytes were separated by Waters C18 column with gradient elution using acetonitrile and 0.1% formate in water as mobile phases.Positive electrospray ionization with multiple reaction monitoring mode was used in the detection.External standard method was used to determine the results.[Results] In the linear range(1-100 μg/L) of each plant hormones,the correlation coefficient was greater than 0.997 for each plant hormones.The limit of detection(LOD) were 0.3-0.5 μg/kg,and the limit of quantitation(LOQ) were 1.0-1.5 μg/kg.In the range of 5-20 μg/kg,the corresponding recoveries were 80.2-119.3%.The relative standard deviation(RSD) of intra-day and inter-day were 0.65-7.28%.[Conclusions] The simplicity,sensitivity and good precision of the method made it be well suitable for determination of plant hormones residues in food.

Abstract:A sensitive and convenient liquid chromatography-tandem mass spectrometric method was developed for the determination of 16 pesticides such as imidacloprid, prochloraz, difenoconazole, azoxystrobin, and thiamethoxam in fruits and vegetables. After compared with methanol and acetone-cyclohexane (1:2, v/v), acetonitrile was chosen as the extraction solvent. The samples were extracted by acetonitrile in high-speed homogenization. The extraction solution was cleaned up by liquid-liquid extraction, and the supernatant was collected. In this work, QuEChERS exhibited much higher efficiency than Carbon-NH2 solid-phase extraction in purification. The pigments and organic acids were removed by purge line (150 mg primary secondary amine (PSA) sorbent and 900 mg absolute magnesium sulfate), leading to the decrease of the background interferences. The average recoveries of the 16 pesticides were almost in the range of 75%-111% at the three spiked levels, and the relative standard deviations were less than 16%. The qualitative analysis and quantitative analysis were investigated by LC-MS/MS and matrix-matched calibration curves. The results showed that the method of QuEChERS combined with LC-MS/MS is rapid, accurate and sensitive for the determination of the 16 pesticide residues in fruits and vegetables.
Determination of 46 Antibiotics Residues in Organic Fertilizer by UPLC-MS/MS with QuEChERS Combined with Column Purification

Abstract:A confirmative method was developed for the simultaneous determination of 46 antibiotics,including sulfonamides,quinolones and macrolides in organic fertilizer by ultrahigh performance liquid chromatography-tandem massspectrometry(UPLC-MS/MS).The samples were extracted with acetonitrile-EDTA buffer solution (pH 10.0),and separated by centrifugation after salting-out.The upper acetonitrile layer was purified by the QuEChERS steps with sorbents.The buffer solution layer was purified with solid-phase extraction cartridge.The separation of target drugs was performed on an ACQUITY UPLC BEH C1scolumn using a mobile phase consisting of 2 mmol/L ammonium acetate with 0.1％ formic acid and methanol by gradient elution.The analysis of target compounds was carried out with positive electrospray ionization (ESI +)source under multiple reaction monitoring (MRM) mode.The matrix-matched external standard calibration was used for quantification.Good linearities for the 46 antibiotics were achieved over the concentration of 1-200 μg/L,with correlation coefficients (r2) of 0.996 6-0.999 9.The recoveries of sulfonamides,quinolones and macrolides at spiked levels of 25,100,400 μg/kg were in the range of 67.8％-95.6％,65.6％-89.4％ and 66.6％-107.8％,respectively,with relative standard deviations(RSDs) of 0.4％-11.9％.The limits of detection (LOD,S/N =3) were 0.6-4.6 μg/kg,and the limits of quantitation(LOQ,S/N=10) were 2.1-15.4 μg/kg.

Abstract:An analytical method was established for the determination of three pyrethroids (bifenthrin, fenpropathrin and flucythrinate) in tomatoes using the quick, easy, cheap, effective, rugged and safe (QuEChERS) cleanup and gas chromatography. The tomato samples were extracted with acetonitrile, cleaned-up by dispersive solid-phase extraction using primary secondary amine as sorbents, concentrated by dispersive liquid-liquid microextraction (DLLME), and analyzed by gas chromatography. Factors affecting the extraction efficiency such as the type and volume of extraction solvent and the volume of dispersive and extraction time were investigated in detail. In the DLLME procedure, 40 microL chloroform was used as the extraction solvent and 1 000 microL acetonitrile was used as the dispersive solvent and the extraction time was 60 s. Under the optimized conditions, the limits of detection for bifenthrin, fenpropathrin and flucythrinate were 0.5, 0.5 and 0.3 microg/kg, and the average recoveries in tomato samples at the spiked levels of 1, 10 and 50 microg/kg were 89% - 109%, 92.5% - 105% and 90% - 108% with the relative standard deviations of 2.5% - 7.6%, 2.8% - 5.7% and 3.8% - 9.1%, respectively. The proposed method is simple, quick, safe, reliable and applicable to analyze pyrethroid residues in tomato samples.

Abstract:Phytohormones are interrelated by synergistic or antagonistic crosstalk and play important roles in the regulation of plant growth and development. In order to understand the interaction between phytohormones in the plant physiological network, it is necessary to determine trace levels of multiple phytohormones simultaneously in a complex matrix. Here, we synthesized ionic liquids containing different functional groups and modified the surface of carbon fibers with them. Based on these carbon fibers-ionic liquid (CFs-IL) materials, a solid phase microextraction method was developed to enable the simultaneous extraction of phytohormones. The adsorption specificity of multiple phytohormones was studied by identifying the hydrophobic, electrostatic, and pi-pi interactions, as well as hydrogen bonds, which favor simultaneous extraction of the relevant acidic, alkaline and neutral phytohormones by improving compatibility. The proposed method, coupled with liquid chromatography-tandem mass spectrometry, was applied to the simultaneous determination of 13 acidic, alkaline and neutral phytohormones in tomato. The limits of quantification were found to be in the range of 0.32-54.05 ng mL(-1) and 4.6-185.8 pg mL(-1), respectively, when measured by QQQ and Q-TOF. All of the relative recoveries were in the range of 94.40-113.37% with RSDs <15.36% (n = 3) for spiked tomato samples. Abstract:A capillary electrophoresis method for the simultaneous determination of seven plant growth regulators including gibberellic acid, abscisic acid, 3-indole acetic acid, 3-indolepropionic acid, 3-indolebutyric acid, 2,4-dichlorophenoxyacetic acid, and 2-methyl-4-chlorophenoxyacetic acid in melons and fruits was established and validated. The samples were ultrasonically extracted with acidified acetonitrile and then dehydrated. The resulting solution was purified with modified QuEChERS method, and then dried-up under weak nitrogen flow, and the residue was redissolved with methanol-water (1:4, v / v ) as sample solution. The electrophoretic separation was performed in an uncoated fused-silica capillary with borax buffer (pH 10.5) containing 29% of ethanol as the running buffer. The running voltage was 25 kV with the column temperature of 30 °C. The linear ranges of gibberellic acid, abscisic acid, and 2-methyl-4-chlorophenoxyacetic acid were from 0.10 to 8.0 μg/mL, while the others were from 0.05 to 4.0 μg/mL with the correlation coefficients greater than 0.997. The limits of detection and the limits of quantification of the method were in the range of 1.54–5.76 and 5.12–19.2 μg/kg, respectively. The recoveries of the method ranged from 69.0 to 109% with the relative standard deviations ranging from 2.01 to 15.4%. The proposed method has been successfully applied to the determination of plant growth regulator residues in 15 melon and fruit samples, and gibberellic acid and abscisic acid were detected in the samples.
[Determination of 50 Pesticide Residues in Fruits by Gas Chromatography-Tandem Mass Spectrometry].

Abstract:A method for the determination of 50 pesticides in fruits by gas chromatography-tandem mass spectrometry (GC-MS/MS) was developed. The three QuEChERS methods (the original one without buffer, the one with acetate buffer and the one with citrate buffer) were compared. The purification effects of primary secondary amine (PSA) and SinChERS-Nano column were also investigated. The results showed that the acetate buffer and the citrate buffer had positive influence on the extraction compared to the original method without buffer, and there was no significant difference between the two methods using buffers. Finally, the QuEChERS method using acetate buffer was chosen as the extraction method. By comparing the purification effect images and the total ion current (TIC) chromatograms, SinChERS-Nano column was revealed to have a better cleaning effect, and was chosen for cleanup. The recoveries of methamidophos, acephate, omethoate, chlorothalonil and dicofol were in ranged of 71.2%-129.2%, the other 45 pesticides were ranged from 79.1% to 122.3%. The limits of detection (LODs) were 0.3-3.0 μ g/kg and the limits of quantification (LOQs) were 1.0-10.0 μ g/kg. The method is rapid and suitable for the screening of the 50 pesticide residues in citrus, grapes and other fruit samples.
Combination of switchable hydrophilic solvent liquid-liquid microextraction with QuEChERS for trace determination of triazole fungicide pesticides by GC-MS

Abstract:This work first proposed a novel green and efficient method based on Quick, Easy, Cheap, Efficient, Rugged, and Safe pretreatment (QuEChERS) combined with switchable hydrophilic solvent homogeneous liquid-liquid microextraction (SHS-HLLME) for trace determination of triazole fungicides (TFs) in agricultural products such as vegetables and fruits by gas chromatography-mass spectrometry (GC-MS). N,N-Dimethyl benzylamine was used for the synthesis of SHS. Box-Behnken design was applied for the optimization of extraction conditions and a mathematical model was obtained. Ultimately, 0.50 mL SHS, 1.0 mL 10 mol L-1 sodium hydroxide, and 45 s ultrasonic time were determined as optimal conditions for the SHS-HLLME method. The limit of detection and limit of quantification determined using the optimal method (SHS-HLLME/GC-MS) were 0.13-0.27 ng mL-1 and 0.43-0.90 ng mL-1, respectively. In addition, the SHS-HLLME method under optimal conditions was combined with the traditional QuEChERS method to realize the advancement of the SHS-HLLME method from simple to complex matrix analysis, and the QuEChERS-SHS-HLLME method was successfully applied to the analysis of TFs in cucumbers, tomatoes, watermelon and grapes in agricultural products. Matrix-matched calibration standards were used to improve the accuracy of TFs in spiked cucumber samples to obtain recovery results close to 100%. It was shown that the new method is green and rapid, enabling fast and inexpensive sample pretreatment with up to 100-fold enrichment factor and low detection limit compared with the original QuEChERS method.

Abstract:An LC-MS/MS method was developed for the identification of 15 compounds including pesticides and plant growth regulators (PGRs) commonly used in cherry cultivation in three main areas (Shanxi, Shandong, Liaoning) in China. Experimental parameters including the column, extraction solvent, eluent, etc. were investigated and optimized. Thirty cherry samples from Shanxi, Liaoning, and Shandong Provinces were analyzed using LC-MS/MS method described herein and demonstrated linearity in the range of 2–2000 ng mL-1 for the 15 compounds. The recovery of analytes were in the range of 70.9 % − 103.7 %. The relative standard deviations varied between 2.1 % and 8.7 % (n = 5). Experimental results highlight the LC-MS/MS method’s convenience, analysis speed, and feasibility for detection and quantification of the 15 compounds used in cherry cultivation.

Abstract:A method was developed for the determination of thiophanate-methyl and carbendazim residues in cucumber and soil by using QuEChERS-high performance liquid chromatography coupled with electrospray ionization tandem mass spectrometry (HPLC-ESI-MS/MS). The residues in the samples were extracted by acetonitrile, cleaned up by developed QuEChERS method, and then analyzed by using LC-MS/MS in multiple reaction monitoring (MRM) mode via positive electrospray ionization with an Agilent ZORBAX SB-C, column (30 mm x 2.1 mm, 5 microm) as the analytical column. The recoveries of thiophanate-methyl spiked at four levels of 0.01, 0.05, 0.1 and 1.0 mg/kg were from 87.3% to 96.0% with the relative standard deviations (RSDs) of 8.0%-9.3% in cucumber, from 88.8% to 93.4% with the RSDs of 5.3%-9.9% in soil; the recoveries of carbendazim spiked at the same levels as those of thiophanate-methyl were from 87.1% to 92.3% with the RSDs of 5.2%-7.5% in cucumber, from 85.8% to 90.9% with the RSDs of 5.3%-13.2% in soil. The method is simple, rapid and characterized with acceptable sensitivity and accuracy to meet the requirements of the pesticide residue analysis. Abstract:A sensitive and simple method for the quantification of metribuzin and its major transformation products in tomato using QuEChERS-based gas chromatography coupled to a triple quadrupole mass analyzer (GC–MS/MS) with multiple reactions monitoring (MRM) was developed and validated. Residues of metribuzin, desamino-metribuzin (DA), diketo-metribuzin (DK), and desamino-diketo-metribuzin (DADK) were extracted from tomato by acetonitrile and purified by graphitized carbon black (GCB) and primary secondary amine (PSA). All analytes gave high correlation coefficients (r2) above 0.995, and the limits of detection (LOD) ranged from 0.002 to 0.008mg/kg. The mean recoveries of the analysis ranged from 72.35% to 95.86% with relative standard deviation (RSD) ranging from 1.98% to 8.70% at three spiking levels (0.05mg/kg, 0.5mg/kg, 1.0mg/kg). The method was also successfully applied to the determination of metribuzin and its transformation products in tomato samples collected from an experimental field.

Abstract:A modified QuEChERS (Quick, Easy, Cheap, Effective, Rugged and Safe) -LC-MS/MS method was developed for the determination of hexachlorophene in fruits and vegetables. Samples were extracted by acetonitrile and then salted with an acetate buffer system. Extractants neutral alumina (Al-N), strong cation exchange silica gel bonded adsorbent (SCX) and graphitized carbon black (GCB) were used for sample purification. The method demonstrates excellent accuracy and reproducibility. Under optimized conditions, the correlation coefficients of hexachlorophene were higher than 0.995 in the range of 0.5-20 ng/mL. The limit of quantification (LOQ) was 2.0 μg/kg. The average recoveries, assessed at three spiked levels (2.0, 4.0, and 20.0μg/kg) across various matrices including cabbage, celery, tomato, eggplant, potato, radish, cowpea, chives, apple, peach, grape, citrus, bitter melon, banana and hami melon ranged from 72.0 to 100.5% with relative standard deviations from 3.2 to 9.8% (n = 6).

Abstract:In this study, a method was developed to determine 45 selected pesticides (of different chemical families) in fruit and vegetable (including apple, spinach and cucumber). Samples were extracted using an improved QuEChERS method with salting out and phase separation in two steps. The target pesticides in concentrated extracts were analyzed by an on-line gel permeation chromatography-gas chromatography/mass spectrometer (online-GPC-GC/MS). Online GPC effectively removed matrix interferences and greatly improved the method sensitivity, recoveries and automation. Method limits of quantification were 10 ng/g for uniconazole and metalaxyl, and 5 ng/g for other 43 target analytes. In three fruit and vegetable matrices each spiked with 45 pesticides (0.01 μg/g), mean recoveries ranged from 80 to 118% for most of the tested pesticides except for profenofos (77% in apple) and chlorpyrifos (68% in apple and 75% in cucumber), with relative standard deviations (RSDs) of less than 14%. The results of the proficiency testing showed that the method is very successful in measuring the certified pesticides with less than 1.3 of the absolute value of Z-score. This method has been applied for routinely monitoring pesticides in fresh fruit and vegetable.

Abstract:In order to cope with the differences in international trade regulations, it is necessary to establish a rapid non-targeted screening method for triazole fungicides in food. Therefore, a non-targeted method based on ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS) was established for the rapid screening of triazole fungicides in tomatoes. Tomato samples were extracted with acetonitrile and then purified by QuEChERS. The purified solution was separated on a BEH C18 column (100 mm×2.1 mm, 1.7 μm) using a gradient mobile phase consisting of acetonitrile and water (both containing 0.1% formic acid). Full information tandem mass spectrometry (MSE) acquisition using electrospray ion source under positive ion mode was performed. The 19 triazole fungicides were divided into eight groups based on their structural differences. The secondary mass fragments of eight typical standards in the eight groups of triazole fungicides were analyzed, and then, the regularities of the characteristic fragment ions and neutral loss fragments produced by collision-induced dissociation of the triazole fungicides were summarized. The fragmentation regularity was further verified by the other 11 triazole fungicides. A total of 16 tomato samples collected from local supermarkets in Beijing were screened using this non-targeted method. Diniconazole was identified and confirmed in one sample, which demonstrated that the method could achieve rapid and accurate non-targeted screening of triazole fungicides in tomatoes. The proposed method is highly efficient, sensitive, and accurate, thus providing a reference for screening triazole fungicide residues in food.
