Expression and Purification of Antimicrobial Peptide s-Thanatin in Escherichia coli

Abstract:s-thanatin is a cationic 21-amino-acid antimicrobial peptide(AMP) by substitution of thanatin's threonine at position 15 with serine.To make AMP more economically viable,researchers have sought to mass-produce AMP using recombinant means,mostly using Escherichia coli based systems. In this study,s-thanatin gene with optimized codons was synthesized by a PCR-based gene synthesis and cloned into pET-32a plasmid to construct a TrxA fused expression vector,which was transformed into E.coli BL21(DE3) for expression.The fusion protein was concentrated by hollow fiber ultrafiltration and cleaved by thrombin,the mature s-thanatin was separated by DEAE-Sephrose Fast Flow anion ion exchange chromatography and finally purified by prepared reversed-phased HPLC.The antimicrobial activity of Ts against E.coli ATCC25922 and clinical antibiotic-resistant Klebsiella pneumoniae indicated that the recombinant Ts had nearly identical antimicrobial activity of synthetic Ts.This result demonstrates that the expression and purification system provides a simple and reliable strategy for large-scale purification of recombinant antimicrobial peptide.
