Abstract:The biotin-labeled amygdalin probe was successfully synthesized by esterification,acetalization and Sonagarshia coupling,using the hydroxyl group as reactive sites to modify with biotin.The structures were characterized by1 H NMR,13 C NMR,IR and HR-ESI-MS.

Abstract:A radiochemical, propyl 4-[2-(4,6-dimethoxy-2-pyrimidinyloxy)benzylamino]benzoate with all six benzoic ring carbons labeled by 14 C was made from 4-aminobenzoic acid with all six benzoic ring carbons labeled by C-14 through esterification, condensation, reduction and substitution, and purified by preparative HPLC. Its structure was identified by LC-MS and H-1 NMR. HPLC test showed its chemical purity exceeded 98%. HPLC-LSC and TLC-IIA demonstrated that its radiochemical purity exceeded 98% and specific activity was 1.089 +/- 0.011 mCi/mmol. The total chemical yield and radiochemical yield were all 53%.

Abstract:Two analogues to the fluorescent amyloid probe 2,5-bis(4'-hydroxy-3'-carboxy-styryl)benzene (X-34) were synthesized based on the trans-stilbene pyrene scaffold (Py1SA and Py2SA). The compounds show strikingly different emission spectra when bound to preformed Aβ1-42 fibrils. This remarkable emission difference is retained when bound to amyloid fibrils of four distinct proteins, suggesting a common binding configuration for each molecule. Density functional theory calculations show that Py1SA is twisted, while Py2SA is more planar. Still, an analysis of the highest occupied molecular orbitals (HOMOs) and lowest unoccupied molecular orbitals (LUMOs) of the two compounds indicates that the degree of electronic coupling between the pyrene and salicylic acid (SA) moieties is larger in Py1SA than in Py2SA. Excited state intramolecular proton transfer (ESIPT) coupled-charge transfer (ICT) was observed for the anionic form in polar solvents. We conclude that ICT properties of trans-stilbene derivatives can be utilized for amyloid probe design with large changes in emission spectra and decay times from analogous chemical structures depending on the detailed physical nature of the binding site.

Abstract:Activity-based Ubiquitin probes (Ub-ABPs) carrying a reporter group have emerged as effective tools for the investigation of deubiquitinating enzymes (DUBs), such as studying the molecular mechanism of DUBs, profiling new DUBs. But so far, the synthesis of commonly used biotin-bearing Ub-ABPs is a technical challenge. Here, we report a one-pot semi-synthetic strategy for the acquiring of Ub-ABPs carrying a biotin tag through sequential enzymatic ligation, N-S acyl transfer and aminolysis reaction without any purification steps. These probes enable to capture the different family of DUBs for enrichment and immunoblotting using the attached biotin tag.

Abstract:A stable prostacyclin analogue, (15S)-18c, having an azidophenyl group as a photoaffinity labeling functionality has been synthesized. This compound has a sufficiently high affinity to the prostacyclin receptor protein in mastocytoma P-815 cells, exhibiting an IC50 value of 3 nM for the replacement of iloprost bound to the receptor protein. A photoaffinity probe compound, [3H]-(15S)-18c, is obtainable by reduction of the ketone 16c with [3H]NaBH4  CeCl3 followed by alkaline hydrolysis of the methyl ester.

Abstract:We introduce a biotinylated D -amino acid probe capable of metabolically incorporating into bacterial PG. Leveraging the robust affinity between biotin and streptavidin, the probe has demonstrated efficacy in imaging, capture, and targeted inactivation of Gram-positive bacteria through synergistic pairings with commercially available streptavidin-modified fluorescent dyes and nanomaterials. The versatility of the probe is underscored by its compatibility with a variety of commercially available streptavidin-modified reagents. This adaptability allows the probe to be applied across diverse scenarios by integrating with these commercial reagents.

Abstract:Two fluorescent probes SPS1 and SPS2 were designed by connecting biotin to an environment-sensitive coumarin fluorophore. Streptavidin and avidin induced dramatical fluorescence changes in both probes. SPS2 has potential in fluorescent imaging of biotin receptor-enriched tumor cells.

Abstract:Biotinylated polymers with side-chain aldehydes were prepared for use as multifunctional scaffolds. Two different biotin-containing chain transfer agents (CTAs) and an aldehyde-containing monomer, 6-oxohexyl acrylate (6OHA), are synthesized. Poly(ethylene glycol) methyl ether acrylate (PEGA) and 6OHA are copolymerized by reversible addition-fragmentation chain transfer (RAFT) polymerization in the presence of the biotinylated CTAs. The resulting polymers are analyzed by GPC and(1) H NMR spectroscopy. The polymer end groups contained a disulfide bond, which could be readily reduced in solution to remove the biotin. Reactivity of the aldehyde side chains is demonstrated by oxime and hydrazone formation at the polymer side chains, and conjugate formation of fluorescently labeled polymers with streptavidin is investigated by gel electrophoresis.

Abstract:A practical asymmetric total synthesis of (+)-biotin based upon the chiral bifunctional sulfamide 8-promoted enantioselective anhydride desymmetrization has been achieved via the key chiral intermediacy of (3aS,6aR)-1,3-bis(4-methoxybenzyl)tetrahydro-4H-thieno[3,4-d]imidazole-2,4(1H)-dione (4). In addition, the installation of the 4-carboxybutyl side chain at C-4 position of 4 was efficiently introduced by an improved C4+C1 strategy.

Abstract:The simultaneous discrimination of Cys, Hcy, and GSH by a single probe is still an unmet challenge. The design and synthesis of a small molecule probe MeO-BODIPY-Cl (BODIPY=boron dipyrromethene) is presented, which can allow Cys, Hcy, and GSH to be simultaneously discriminated on the basis of three distinct fluorescence turn-on responses. The probe reacts with these thiols to form sulfenyl-substituted BODIPY, which is followed by intramolecular displacement to yield amino-substituted BODIPY. The kinetic rate of the intramolecular displacement reaction determines the observed different sensing behavior. Therefore, the probe responds to Cys, Hcy, and GSH with fluorescence turn-on colors of yellow, yellow and red, and red, respectively. With this promising feature in hand, the probe was successfully used in imaging of Cys, Hcy and GSH in living cells.

Abstract:A novel probe C1 combining benzothiazole with a spiropyran section was developed for the specific detection of human serum albumin (HSA). The molecular docking suggested that the sulphonic acid group modification allowed C1 to form specific hydrogen bonds with lysine (Lys137) at fatty acid site 1 (FA1) of HSA, thus enabling fluorescence differentiation between HSA and BSA.

Abstract:A fluorescent probe (RB–S) containing the N-hydroxysuccinimidyl ester in 2 position was prepared from rhodamine B (RB) by one-step condensation reaction. The reactivity of the fluorescent probe with glycine was studied and the products were identified by LC–MS analysis. The excessive equivalent of the RB–S was hydrolyzed to the corresponding carboxylic acids (RB). UV/vis and fluorescence spectra of the fluorescent probe (RB–S), the labeled derivative (RB–Gly) and the hydrolysate (RB) were also studied. Fluorescence and absorption spectra appear as mirror images. The solvent effect on the spectra of the glycine derivative (RB–Gly) was investigated in methanol/water solution.
An Activated Near-Infrared Fluorescent Probe with Large Stokes Shift for Discrimination of Bio-Thiols

Abstract:Bio-thiols such as cysteine (Cys), homocysteine (Hcy), and glutathione (GSH) play a crucial role in physiological processes. Distinguishing between Cys, Hcy, and GSH remains a challenge because of their similar structure and reactivity. Herein, we conceptualized and synthesized a near-infrared (NIR) fluorescent probe TAI based on acridine-conjugated indole salts. This innovative probe demonstrated effectiveness in sensitively distinguishing Cys, Hcy and GSH by NIR dual-channel and time-dependence fluorescence. Probe displays a significant turn-on NIR fluorescent response toward Cys at 654nm under 488nm excitation with Large Strokes shift (about 166nm) and high selectivity, but also shows turn-on NIR fluorescent response for Hcy/GSH at 695nm under 640nm excitation. More interestingly, the probe also exhibited an NIR fluorescence response at 654nm to Hcy and no fluorescence response to GSH when the response time is over 3h. The sensing mechanism was fully confirmed by NMR, HRMS spectra and DFT theoretical calculations. Moreover, successful intracellular imaging experiments in living cells suggest that the probe is suitable for discrimination imaging endogenous and exogenous bio-thiols. This work offers a promising near-infrared fluorescent imaging tool for the differential detection of bio-thiols.

Abstract:Novel functional AIEgen based on three compact bound aryl skeletons is designed and synthesized. This tri-aryl type luminogen (TA-Catechol) embedded with catechol moiety responds rapidly to series of boronic acids. Real-time visual and quantitative dual-mode detection method is established for the first time with modest precision and low detection limit (8.0 mu M). Detailed mechanistic discussion identifies tetra-coordinated boronic species as the key intermediate within sensing procedure. Wide range of organic boronic acids compatible with this strategy is displayed which is promising in high throughput screening technology. Furthermore, solid-state sensing capability of TA-Catechol is also demonstrated.

Abstract:Glucosyl-novobiocin-based diazirine photoaffinity labelling reagents (PALs) were designed and synthesized to probe the Hsp90 C-terminal domain unknown binding pocket and the structure-activity relationship. Five PALs were successfully synthesized from novobiocin in six consecutive steps employing phase transfer catalytic glycosylation. Reactions were monitored and guided by analytical LC/MS which led to different strategies of adding either a PAL precursor or a sugar moiety first. The structures and bonding linkages of these compounds were characterised by various 2D-NMR spectroscopy and MS techniques. Synthetic techniques provide powerful probes for unknown protein binding pockets.

Abstract:A biotin-tagged analogue of troglitazone was designed, synthesized and applied to affinity chromatography to pulldown EGFR from 293T cell lysates overexpressing EGFR, a membrane protein assumed to be troglitazone's direct binding target by previous work. The results indicate the feasibility of the biotinylated probe as a vigorous tool to clarify the molecular mechanisms for troglitazone's antitumor activities.

Abstract:Here, we report a β-galactosidase (β-Gal)-responsive photochromic fluorescent probe, <b>NpG</b>, that was designed to prebind to human serum albumin (HSA) to form the probe/protein hybrid, . The formation of led to an increase in fluorescence emission (520 nm) corresponding to the binding of the fluorescent naphthalimide unit with HSA. In addition, this enabled visualization of the spiropyran fluorescence emission in aqueous media. Our probe/protein hybrid approach afforded a unique imaging platform with enhanced cell permeability and solubility that was capable of visualizing the cellular uptake of before its activation by β-Gal. The β-Gal-mediated cleavage of the galactose unit within the hybrid resulted in the formation of and an increase in red fluorescence emission (620 nm). The resultant merocyanine unit was then able to undergo photoisomerization (merocyanine ↔ spiropyran) to facilitate STORM (i.e., stochastic optical reconstruction microscopy) imaging with minimal phototoxicity and excellent photostability/reversibility. Using STORM, was able to determine the subcellular distribution of β-Gal activity between cell lines with nanoscale precision.
