Abstract:Response surface methodology was employed to optimize the ultrasound-assisted extraction (UAE) conditions for simultaneous optimization of dependent variables, including DPPH radical scavenging activity (RSA), tyrosinase activity inhibition (TAI), and collagenase activity inhibition (CAI) of peanut shell extracts. The effects of the main variables including extraction time (5.0~55.0 min, X1), extraction temperature (26.0~94.0 °C, X2), and ethanol concentration (0.0%~99.5%, X3) were optimized. Based on experimental values from each condition, quadratic regression models were derived for the prediction of optimum conditions. The coefficient of determination (R2) of the independent variable was in the range of 0.89~0.96, which demonstrates that the regression model is suitable for the prediction. In predicting optimal UAE conditions based on the superimposing method, extraction time of 31.2 min, extraction temperature of 36.6 °C, and ethanol concentration of 93.2% were identified. Under these conditions, RSA of 74.9%, TAI of 50.6%, and CAI of 86.8% were predicted, showing good agreement with the experimental values. A reverse transcription polymerase chain reaction showed that peanut shell extract decreased mRNA levels of tyrosinase-related protein-1 and matrix metalloproteinase-3 genes in B16-F0 cell. Therefore, we identified the skin-whitening and anti-wrinkle effects of peanut shell extracts at protein as well as gene expression levels, and the results show that peanut shell is an effective cosmetic material for skin-whitening and anti-wrinkle effects. Based on this study, peanut shell, which was considered a byproduct, can be used for the development of healthy foods, medicines, and cosmetics.
Red fluorescence protein (DsRed2) promotes the screening efficiency in peanut genetic transformation

Abstract:Peanut (Arachis hypogaea L.), one of the leading oilseed crops worldwide, is an important source of vegetable oil, protein, minerals and vitamins. Peanut is widely cultivated in Asia, Africa and America, and China is the largest producer and consumer of peanut. Genetic engineering has shown great potential to alter the DNA makeup of an organism which is largely hindered by the low transformation and screening efficiency including in peanut. DsRed2 is a reporter gene widely utilized in genetic transformation to facilitate the screening of transformants, but never used in peanut genetic transformation. In this study, we have demonstrated the potential of the red fluorescence protein DsRed2 as a visual reporter to improve screening efficiency in peanut. DsRed2 was firstly expressed in protoplasts isolated from peanut cultivar Zhonhua 12 by PEG, and red fluorescence was successfully detected. Then, DsRed2 was expressed in peanut plants Zhonghua 12 driven by 35S promoter via Agrobacterium tumefaciens-mediated transformation. Red fluorescence was visually observed in calli and regenerated shoots, as well as in roots, leaves, flowers, fresh pod shells and mature seeds, suggesting that transgenic screening could be initiated at the early stage of transformation, and continued to the progeny. Upon screening with DsRed2, the positive plant rate was increased from 56.9% to 100%. The transgenic line was then used as the male parent to be crossed with Zhonghua 24, and the hybrid seeds showed red fluorescence as well, indicating that DsRed2 could be applied to hybrid plant identification very efficiently. DsRed2 was also expressed in hairy roots of Huayu 23 via Agrobacterium rhizogenes-mediated transformation, and the transgenic roots were easily selected by red fluorescence. In summary, the DsRed2 is an ideal reporter to achieve maximum screening efficiency and accuracy in peanut genetic transformation.

Abstract:Purple corn pigment is anthocyanins,a kind of natural pigment from the purple corm plant,purple corm cob and purple seed,which is a kind of safety,not poisonous edible.It clearly had antioxidant activity,anti-tumor,anti-aging,reducing blood pressure,regulating blood lipids and blood glucose level and so on.The purple corm pigment was extracted in acid solvent,containing 9 anthocyanins at least and the main active components of red pigment was anthocyanins-3-glucoside.Purple corn pigment is a type of existing natural food additives,and can be used to color for food,medicine,cosmetics,dyes and other industries.Based on the latest research results of home and abroad in recent 10 years,the extraction process,the chemical constitution,the physic-chemical property,the biological active functions,and application of purple corm pigment were summarized.This information may lead to a reference value in depth study,the comprehensive development and application of the purple corm pigment.

Transcriptional networks orchestrating red and pink testa color in peanut

Abstract:Background: Testa color is an important trait of peanut (Arachis hypogaea L.) which is closely related with the nutritional and commercial value. Pink and red are main color of peanut testa. However, the genetic mechanism of testa color regulation in peanut is not fully understood. To elucidate a clear picture of peanut testa regulatory model, samples of pink cultivar (Y9102), red cultivar (ZH12), and two RNA pools (bulk red and bulk pink) constructed from F4 lines of Y9102 x ZH12 were compared through a bulk RNA-seq approach. Results: A total of 2992 differential expressed genes (DEGs) were identified among which 317 and 1334 were up-regulated and 225 and 1116 were down-regulated in the bulk red-vs-bulk pink RNA pools and Y9102-vs-ZH12, respectively. KEGG analysis indicates that these genes were divided into significantly enriched metabolic pathways including phenylpropanoid, flavonoid/anthocyanin, isoflavonoid and lignin biosynthetic pathways. Notably, the expression of the anthocyanin upstream regulatory genes PAL, CHS, and CHI was upregulated in pink and red testa peanuts, indicating that their regulation may occur before to the advent of testa pigmentation. However, the differential expression of down-stream regulatory genes including F3H, DFR, and ANS revealed that deepening of testa color not only depends on their gene expression bias, but also linked with FLS inhibition. In addition, the down-regulation of HCT, IFS, HID, 7-IOMT, and I2'H genes provided an alternative mechanism for promoting anthocyanin accumulation via perturbation of lignin and isoflavone pathways. Furthermore, the co-expression module of MYB, bHLH, and WRKY transcription factors also suggested a fascinating transcriptional activation complex, where MYB-bHLH could utilize WRKY as a co-option during the testa color regulation by augmenting anthocyanin biosynthesis in peanut. Conclusions: These findings reveal candidate functional genes and potential strategies for the manipulation of anthocyanin biosynthesis to improve peanut varieties with desirable testa color.

Abstract:Peanuts are globally cultivated and more than 66.66% of the total production is utilised for oil production, while one-third is either consumed as nut or used in production of peanut butter and other snacks. The peanut processing industries reject large portions of peanut biomass in the form of peanut meal, skins and hulls, which are rich in proteins, phenolics and fibres. Efficient use of peanut skins that are rich in bioactive compounds and antioxidant activity for bio-fortification of foods and dietary supplements is in demand. In the present study, four varieties of peanuts (K-6, K-9, TMV-2 and TAG-24) which are widely cultivated in India were chosen and deduced the proximate analysis. Besides, extraction of total phenolic content (TPC) resulting in seed coat of examined peanut varieties revealed the higher phenolics content in K-9 (60.6 ± 3.0 mg/g) > K-6 (59.4 ± 1.30 mg/g) > TMV-2 (58.4 ± 1.35 mg/g) in the solvent system containing acetone:water:acetic acid at 70:29.8:0.2 ratio, respectively. In addition to K-9, K-6 showed significant antioxidant activity with lower electrical conductivity (an indicator of seed coat intactness) in comparison to the other peanut varieties. Moreover, FTIR analysis confirmed the presence of phenols, amides, esters, amino acids, carboxylic acids, alcohols and flavonoids in the seed coat of different peanut varieties. HPLC analysis further confirmed the presence of wall-bound phenolics, viz., protocatechuic acid, 4-hydroxybenzoic acid, vanillic acid, <i>p</i>-coumaric acid and <i>t</i>-ferulic acid in the skins of various peanut varieties. Owing to the presence of bioactive compounds and antioxidant activity, the peanut skins can be used for bio-fortification of food, dietary supplements, nutraceuticals, cosmetic industries and other value-added food materials.

Abstract:Pigments had been extracted from natural brown cotton fiber(NBCF) and delinted white cottonseed hulls(DWCH) by extensive use of diagnostic reagents,UV-VIS spectrophotometer,infrared absorption spectra and normal phase high-performance liquid chromatography(NP-HPLC) comparative analysis of their chemical compositions.The results of corresponding color reactions of two kinds of pigments showed that they were similar in main chemical constituents and all belonged to condensed tannins(CT).The dominant ingredient of both two pigments were CT with pyrocatechol structure,the main ingredient of DWCH pigments also contained phloroglucinol structure while the content of the component with this structure was very small in NBCF pigments.The NP-HPLC method was developed to separate the two samples,the results showed that there were four compositions with one overwhelming in NBCF pigments,and various kinds of constituents were separated in DWCH pigments,however,constituents with the same molecular structure were not found in two pigments.

Abstract:Natural pigments, including carotenoids, flavonoids and anthocyanidins, determine the attractive color of fruits. These natural pigments are essential secondary metabolites, which play multiple roles in the whole life cycle of plants and are characterized by powerful antioxidant activity. After decades of research and development, multiple benefits of these natural pigments to human health have been explored and recognized and have shown bright application prospects in food, medicine, cosmetics and other industries. In this paper, the research progress of natural fruit pigments in recent years was reviewed, including the structural characteristics and classification, distribution in fruits and analysis methods, biosynthetic process, antioxidant capacity and mechanism, bioaccessibility and bioavailability, and health benefits. Overall, this paper summarizes the recent advances in antioxidant activity and other biological functions of natural fruit pigments, which aims to provide guidance for future research.

Bagging on Pigment Synthesis and Expression of PbcrtZ in 'Chili' Fruit

Abstract:The best bagging time of Chili pear was 60 d after anthesis by determining the appearance quality and the weight loss rate during the storage period. The Chili pear fruit, which was bagged at 60 d after anthesis, was used as the material, and the contents of chlorophyll, flavonoids, carotenoids and the expression of the color-related difference gene. PbcrtZ were determined in fruit peels treated with plastic fabrics and non-woven bags. The results showed that the content of chlorophyll in non-woven fabric and plastic bagged developmental Chili fruits was significantly lower than that in non-bagged fruits, and the contents of flavonoid and carotenoid were significantly higher than those of non-bagged fruits on 180 d after anthesis. In addition, the expression of PbcrtZ in non-woven fabric and plastic bagged fruits were higher than that in non-bagged fruits, and the expression on 180 d after anthesis was 4. 36 and 3. 48 times higher than that of the control group, which was in according with the difference of flavonoid content and carotenoid content. Consequently, the color of the fruits treated with non-woven fabrics and plastic bags was green-yellow and the non-bagged fruit showed a green color, and the fruits treated with non-woven fabrics showed better appearance quality and storage property.

Abstract:Abstract Peanut is one of the important oil and economic crops, among which the variegated testa peanut is a unique member. The molecular mechanisms underlying the pigment synthesis in variegated testa are still unclear. Differentially expressed genes (DEGs) in the flavonoid metabolism pathway in pigmented areas indicated that there were 27 DEGs highly related to the synthesis of variegated testa color among 1,050 DEGs. Of these 27, 13 were up-regulated and 14 were down-regulated, including 3 PALs , 1 C4H , 2 CHSs, 1 F3H, 1 F3'H, 2 DFRs, 2 LARs, 2 IAAs, 4 bHLHs, and 9 MYBs . GO (Gene Ontology) analysis indicated that DEGs were similarly enriched in three branches. KEGG (Kyoto Encyclopedia of Genes and Genomes) analysis suggested flavonoid biosynthesis is the most direct metabolic pathway for the synthesis of testa variegation. The liquid chromatography–tandem mass spectrometry (LC–MS/MS) results showed that cyanidin and delphinidin were the primary metabolites that caused the color differences between the pigmented and the non-pigmented areas. Through the verification of 20 DEGs via qPCR, the results were consistent with transcriptome sequencing in four comparison groups. The results in this study lay the foundation for revealing the molecular regulation mechanisms of flavonoid synthesis in variegated testa peanut.

Abstract:Recently,with extensive cultivation of red Chinese sand pear(Pyrus pyrifolia Nakai) cultivars newly released by hybrid breeding in China,more and more attention has been paid to the germplasm resources and coloring mechanism of red pears(Pyrus L.) by domestic scholars.Red pear germplasm resources with complicated genetic background,abundant genetic diversity and high commercial value nearly cover all cultivated species or types including: P.communis L.,P.pyrifolia Nakai,P.pyrifolia White Pear Group,P.ussuriensis Maxim and P.sinkiangensis Yu.Red coloration on pear skin is known to mainly depend on the accumulation of anthocyanin.Anthocyanins are synthesized via the flavonoid pathway,and the structural genes,e.g.PAL,CHS,CHI,F3H,DFR,ANS and UFGT and a regulatory gene PyMYB 10 related to anthocyanin biosynthesis have been isolated and identified in pears.Coloring patterns in different species are different,and anthocyanin in peel of pear fruit generally distribute in hypodermic cells below the epidermis.Light and temperature are most important external factors in anthocyanin biosynthesis of pear fruit,and some artificial regulatory methods based on the regulatory mechanism of light and on anthocyanin synthesis e.g.bagging,postharvest irradiation and evaporative cooling is being/will be applied in red pear production.Here,we gave a brief review about recent research progresses mentioned above,and hope that this paper will be of help for the future research in the field of red pears.
Extraction and Antioxidative Activities of Pigment from Sweet Cherry

Abstract:The pigment from ripe sweet cherry was obtained by solvent extraction method and purified on a C18 solid-phase column.The main composition of the pigment was characterized by means of HPLC-PAD analysis.At the same time,2,2-diphenyl-1-picrylhydrazyl(DPPH)radical system and hydroxyl radical system were used to investigate its scavenging activities against free radical in vitro.The results indicated that the pigment contained two major components:neochlorogenic acid and p-coumarylquinic acid.It was demonstrated that the pigment exhibits good scavenging effect on DPPH and hydroxyl radical,and the antioxidative activities increase with increasing of the pigment concentration.The results suggested that the sweet cherry pigment may be a natural edible colorant with excellent antioxidative activities.

Abstract:The orthogonal array design method was used to optimize ultrasonic-assisted extraction of resveratrol from peanut sprouts. The results showed that the highest extraction yield of resveratrol using ultrasonic-assisted extraction could be up to 1.1%. The optimal extraction conditions were liquid to solid ratio of 30:1 (mL/g) and ethanol concentration of 80% (v/v) as solvent for 40 min at the temperature of 70 °C. AB-8 macroporous adsorption resin was used to purify the crude extract and the resveratrol content increased to 47.5% after one treatment run. The optimal adsorption parameters were initial concentrations in the sample solution of 2 mg/mL, a pH of 5.0, a flow rate of 2 mL/min, and a temperature of 25 °C. The optimal desorption parameters were 60% ethanol and a flow rate of 1 mL/min. The chemical composition of the peanut sprout's resveratrol sample was investigated via HPLC, and the predominant constituents were found to be protocatechuic acid, catechins, caffeic acid, epicatechuic acid, resveratrol, and rutin. The antioxidant activities of the resveratrol were measured via the following different analytical methods: reducing power, 2,2-diphenyl-1-picrylhdrazyl (DPPH), hydroxyl radical-scavenger activity, superoxide radical-scavenger activity, the β-carotene bleaching test, and the scavenging nitrite test. The results indicated that the resveratrol in peanut sprouts have significant antioxidant activities and can be used as a source of potential antioxidant. And peanut sprout's resveratrol has the potential and valuable application to be used as a new type of resveratrol resource. The finding of this study can provide some theoretical reference for the comprehensive utilization of peanut resources in the development of antioxidant health foods.
