Role of Dendritic Cells in Formation of Microchimerism in Host Spleen after Liver Transplantation

Abstract:Objective To investigate the role of dendritic cells(DCs) in the formation of microchimerism in host spleen after liver transplantation. Methods In cross-sex(male Wistar to female SD) rat liver transplantation models(n=4),host rats were sacrificed 7 days postoperatively.Mononuclear cells were collected from host spleen,and spleen-derived DCs culture system was established.DCs surface antigen OX62 was morphologically observed and detected by flow cytometry.PCR was carried out to investigate the sex-determining fragment of Y chromosome in this spleen-derived DCs culture system and illustrate the formation of microchimerism in host spleen. Results Seven days after cross-sex rat liver transplantation,the cells of spleen-derived DCs culture system exhibited morphological characteristics of DCs and expressed OX62.It was revealed by PCR that donor-derived sex-determining region Y gene was obtained in the spleen-derived DCs culture system.Conclusion After allogeneic liver transplantation,donor liver-derived DCs or their precursors migrate into the host spleen and are engaged in the generation of allogeneic microchimerism.
