The Effect of Hydrogen Sulfide on Oleic Acid-Induced Acute Lung Injury in Rats

Abstract:Objective:To explore the role of endogenous cystathionine-γ-lyase(CSE)/hydrogen sulfide(H2S)pathway in oleic acid-induced acute lung injury in rats.Method:Acute lung injury was induced by injecting oleic acid(0.2 mL/kg)intravenously as OA group;NS controls were given normal saline(0.2 mL/kg);NaHS solution(14.0 μmol/kg)was injected intraperitoneally before OA(0.2 mL/kg)injection as NaHS+OA group;and the others were injected NaHS solution(14.0 μmol/kg)intraperitoneally as NaHS group.The concentration of H2S and the contents of MDA,Diene in the lung tissue and the plasma were detected;the activity of CSE and MPST were measured.Result:Compared with NS control group,in OA group the concentration of H2S and the activity of CSE and MPST in lung tissue of rats all decreased(P0.01),The level of plasma H2S increased(P0.01),the contents of MDA and Diene both in lung tissue and in plasma increased significantly(P0.01);In NaHS+OA group,the contents of MDA and Diene decreased significantly(P0.01).Conclusion:Endogenous CSE/H2S pathway was involved in the pathogenesis of acute lung injury;Exogenously applied H2S could attenuate the process of acute lung injury possibly because of reducing oxidative stress.
