Abstract:Litchi and longan are famous southern subtropical fruits in China. Besides closely related to each other taxonomically, litchi and longan share many similarities in origin, cultivation history and environmental requirements; furthermore, the developments of research on litchi and longan germplasm resources are somewhat similar and comparable. This paper reviews the literature on origin and distribution, collection and conservation, classification and identification, selection and breeding of litchi and longan germplasm resources in China. The classification of litchi and longan can be divided into three stages: classification based on morphological traits, classification by isozyme analysis and DNA markers. The expression of morphological traits is often affected by environmental conditions; the number of loci that isozyme analysis can examine is small, cultivars, which are closely related often have identical isozyme patterns. The limitations of classification based on morphological traits and isozyme analysis are then overcome by the utilization of molecular markers. At present, most new litchi and longan cultivars are selected from superior seedlings obtained after open-pollination or sport; germplasm with desirable characteristics such as aborted seeds, rather big-fruit and rather late-bearing have not been fully exploited. In order to achieve the goal of improving fruit quality and agronomic characteristics, it is suggested that additional studies of genetic diversity on litchi and longan germplasm should be carried out with more powerful markers such as SSR and ISSR; marker assisted selection (MAS) should be employed to improve litchi and longan's cross breeding efficiency.

Abstract:The germination and seedling of immature seeds and embryos after anthesis 82～87 d of longan(Dimocarpus longan Lour.'Shixia') were studied.The results showed that the germination and seedling rates of immature embryos sowed directly in clean sand were better than those of in vitro cultured immature seeds.Furthermore,although the germination time of immature embryos stored at 4℃ for 24 hours was slightly delayed in sand,there was no obvious inhibition effect on their germination rate and seedling growth.These results indicated that sowing directly in clean sand was a good way for immature seed and/or embryo rescue in sexual hybridization breeding of longan.

Abstract:The morphological characteristics of pistil and embryogenesis of Chinese plum cultivar Zuili were studied for clari-fying the reason of poor fruit setting with respect to reproductive biologically. Materials were dehydrated, embedded and cut in Paraffin wax. The results were as follows: The style is stuffed; two ovules are contained in the ovary; the ovule was anat-ropous,biteguminous and tenuinucellate. The embryo sac was staying at the 2 to 4 nucleis stage and on the 3rd day after an-thesis reached to the early 8 nucleis stage. The development of the embryo sacs was of polygonum type. The first division of the zygote took place after a dormancy of 2 to 3 days. The development of endosperm conforms to the nuclear type. The pri-mary endosperm nucleus began to divide on the 7th days after anthesis. The globular embryo stage began on 12th day after anthesis. Degeneration of nucellar cells and integument at early stage and abnormal embryo sac structure at mature stage was observed during the development of embryo.
Changes in Specific Proteins in Embryos of Longan at Different Development Stages

Abstract:IEF-SDS-PAGE (isoelectric focusing electrophoresis-sodium dodecyl sulfate-polyacrylamigelelectro- phoresis) technique was applied to comparing and analyzing the changes in protein component during embryo differentiation and development of longan (Dimocarpus longan Lour. cv. Wulongling). The results showed that major components of proteins were generally similar at different development stages, but some changes also took place. The 7 new specific proteins were observed at different stages of the embryo development. They were TE1(27.1 kD, pI 7.3), TE2 (17.5 kD, pI 8.2) on 38 days after anthesis (DAA); TE3 (11.4 kD, pI 7.6), TE4 (13.2 kD, pI 9.9) on 45 DAA; TE5 (22.6 kD, pI 7.2), TE6 (18.6 kD, pI 8.3), TE7 (23.5 kD, pI 3.6) on 52 DAA. These specific proteins showed good repeatability in the experiments. The number of protein points were comparatively higher on 31 DAA gel electrophoretogram, which indicated that rich synthesis and accumulation of proteins appeared at the stage of 31 DAA, and coincided with the results of the protein contents determined. The appearance or disappearance of the specific proteins might play an important role in longan's embryo differentiation and development.

Abstract:Background: The APETALA2/ethylene responsive factor (AP2/ERF) superfamily members are transcription factors that regulate diverse developmental processes and stress responses in plants. They have been identified in many plants. However, little is known about the AP2/ERF superfamily in longan (Dimocarpus longan Lour.), which is an important tropical/subtropical evergreen fruit tree that produces a variety of bioactive compounds with rich nutritional and medicinal value. We conducted a genome-wide analysis of the AP2/ERF superfamily and its roles in somatic embryogenesis (SE) and developmental processes in longan. Results: A genome-wide survey of the AP2/ERF superfamily was carried out to discover its evolution and function in longan. We identified 125 longan AP2/ERF genes and classified them into the ERF (101 members), AP2 (19 members), RAV (four members) families, and one Soloist. The AP2 and Soloist genes contained one to ten introns, whereas 87 genes in the ERF and RAV families had no introns. Hormone signaling molecules such as methyl jasmonate (MeJA), abscisic acid (ABA), gibberellin, auxin, and salicylic acid (SA), and stress response cis-acting element low-temperature (55) and defense (49) boxes also were identified. We detected diverse single nucleotide polymorphisms (SNPs) between the 'Hong He Zi' (HHZ) and 'SI JI MI' (SJM) cultivars. The number of insertions and deletions (InDels) was far fewer than SNPs. The AP2 family members exhibited more alternative splicing (AS) events in different developmental processes of longan than members of the other families. Expression pattern analysis revealed that some AP2/ERF members regulated early SE and developmental processes in longan seed, root, and flower, and responded to exogenous hormones such as MeJA, SA, and ABA, and 2,4-D, a synthetic auxin. Protein interaction predictions indicated that the Baby Boom (BBM) transcription factor, which was up-regulated at the transcriptional level in early SE, may interact with the LALF/AGL15 network. Conclusions: The comprehensive analysis of molecular evolution and expression patterns suggested that the AP2/ERF superfamily may plays an important role in longan, especially in early SE, and in seed, root, flower, and young fruit. This systematic analysis provides a foundation for further functional characterization of the AP2/ERF superfamily with the aim of longan improvement.

Abstract:Abstract Background: CCCH Zinc finger ( Znf ) transcription factors ( TF ), as a novel type Znf genes, regulate genes expression by binding on their mRNA and play important roles in plant abiotic stress, growth and development. However, no overall genome-wide analysis or expression profiling of CCCH ( C3H ) gene family in Dimocarpous longan , especially during the early stages of somatic embryo in longan has been studied. Longan is a tropical/subtropical fruit tree of great economic importance in Southeast Asia,and longan embryogenesis is the main factor affecting fruit quality and yield. Results: In this study, a comprehensive analysis of longan C3H ( DlC3H )gene family was carried out. 49 DlC3H genes were identified from longan genome database,which divided into 3 clades. Besides, genes characteristics, phylogenetic tree, gene structure, motif composition were comprehensively analyzed. The analysis of alternative splicing events (AS) suggested that AS events of DlC3H genes were related to longan non-embryonic and embryonic callus transformation. Promoter analysis indicted that most of DlC3H genes included cis -elements associated with hormones and stress response. Quantitative real-time PCR analysis indicated that 26 DlC3Hs ,which possess MeJA and ABA responsive cis -elements, showed different expression patterns and may involved into ABA and MeJA signaling pathway. The expression profiles of 17 DlC3Hs were performed in four stages of longan, the results showed that only DlC3H01/07/14/16/38 was consistent with the data in the transcriptome. DlC3H 07/14/16/36/49 were highly expressed in EC and only DlC3H 04/38 was in GE , suggesting that they have different functions in embryonic development. Finally, sRNAs were verified involved into regulating 6 DlC3Hs . Conclusion: This study provides the first systematic analysis of CCCH protein in longan somatic embryo. Particularly, CCCH genes may be involved in hormone and stress respond, and somatic embryogenesis. Our results presented here may provide a insight into the characteristics and functions of this family in somatic embryogenesis.

Abstract:ADP-ribosylation modification considered as a model to study histone post-translational modification in chromatin modification. Despite it was reported in many plants, the study of ARFs gene family in longan was still unclear. In this study, 14 longan ARFs genes were identified using the longan genome (the third-generation genome) and further divided into two major groups, including the DlARF in the I-II group and the ARF-like (DlARL) in the III-V group, according to their structure and evolutionary characteristics. Whole-genome duplication (WGD) and segmental duplication events played a major role in the expansion of the DlARFs gene family, the synteny and phylogenetic analyses provided a deeper insight into the evolutionary characteristics of the DlARFs. Protein-protein interactions suggested that some DlARFs proteins may interact to participate in biological processes. Promoter analysis showed more stress response elements in DlARF5, DlGB1, DlARL1, DlARL2, and DlARL8a, suggesting that they may participate in abiotic stress. Expression profiles of DlARFs by quantitative real-time PCR (qRT-PCR) showed that they were abundant accumulation during early somatic embryogenesis (SE). Expression pattern analysis of RNA-seq and qRT-PCR revealed that some ARFs members regulated early SE, and respond to exogenous hormones and abiotic stress such as abscisic acid (ABA), gibberellin A3 (GA3), salicylic acid (SA), methyl jasmonate (MeJA), cold, and heat. Our study provides new insights for further research on the potential function of DlARFs, which may be useful for the improvement of longan.

Abstract:Bamboo is a versatile plant that plays a significant role in the food, raw materials for construction and manufacture, making it one of the most valuable non-timber forestry resources in East Asia. Bamboo propagates through both sexual and asexual reproduction. They often flower synchronously and die collectively after flowering. The time span from seed germination to flowering and fruiting can take several decades or even over a hundred years. Developmental events related to growth occur throughout the entire life cycle of bamboo, and exhibit more diverse developmental patterns than other plants. Till now, a lot of studies focus on growth and development of bamboo organs from aspect of morphology, anatomy, systematics, physiology, cytology, genetics, and molecular biology. Here, we review the current knowledge on the developmental biology of bamboo root, rhizome, culm, leaves and reproductive organs. The inadequate or missing what deserves further investigated in the field of bamboo developmental biology was put forward. Proposal on how to carry out the related research was suggested. Our work provides a reference for bamboo researchers to carry out relevant research.

Abstract:Bupleurum chinense is a commonly used Chinese medicinal material, which has been used medicinally in China for over 2000 years, the development of it is of great value. There have been great advances of studies on its embryology and seed biology in recent years. In this paper, we make a review of the growth of its embryo and fruit, the characteristics of dormancy and storage and the germination of its seed. Besides, we summarize the reasons of its low germination rate and the achievements in improving the situation, for the purpose of providing reference in research on seed physiology of B. chinense.

However, little is known about the AP2/ERF superfamily in longan (Dimocarpus longan Lour.). The release of the whole-genome sequence of longan provided us an opportunity to perform a genome-wide investigation of AP2/ERF superfamily. Results: In the present study, we performed a genome-wide survey of AP2/ERF superfamily longan (DlAP2/ERF), including the gene structure, motif composition, phylogenetic, cis-acting element, single nucleotide polymorphisms (SNPs), insertions and deletions (InDels), alternative splicing (AS) events and the expression patterns. In total, 125 DlAP2/ERF genes were identified and classified into four families, including AP2 (19 members), ERF (101 members), RAV (four members) and Soliost (one member) in the longan genome. The AP2 and soloist genes possessed one to ten introns, while 87 genes within the ERF and RAV family had no introns. A large number of hormone signaling and stress response cis-acting elements were also identified. Besides, a large number of SNPs existed in the DlAP2/ERF superfamily, they may be deverse in the early somatic embryogenesis and stem of longan. However, the numbers of InDels was far less than SNPs. In addition, the AP2 family members existed a large number of AS events in different developmental processes of longan. Expression patterns analysis revealed that the AP2 family may promote the early somatic embryogenesis of longan, and DlAP2/ERF genes were specifically expressed in various organs such as seed, root, flower and young-fruit. The DlAP2/ERF genes can response to exogenous phytohormones, such as 2,4-Dichlorophenoxyacetic acid (2,4-D), Abscisic acid (ABA), Methyl jasmonate (MeJA) and Salicylic acid (SA). Protein interactions prediction indicated that the Baby Boom (BBM) protein may interacted with LALF/AGL15 network and was up-regulated expression at the transcriptional level in the early somatic embryogenesis of longan. Conclusions: A comprehensive analysis of molecular evolution and expression patterns showed that AP2/ERF superfamily played an important role in longan, especially in the early somatic embryogenesis, seed, root, flower and young-fruit. Abstract:The gateway of somatic embryogenesis has been extensively applied in plants' propagation,it is not only an important method for the rapid propagation in a large scale,but also an ideal experimental system for studying the cell totipotency.Great progress has been made with the study of plant somatic embryogenesis in the past few years,but many problems still remain unsolved in this field.Based on the related reports and literature of recent years,the paper summarized the research results of plant somatic embryogenesis,such as enzymes changes,metabolizability of nucleic acid,amino acid and protein,and it also discussed the application prosperity and the existing problems with plant somatic embryogenesis.

Abstract:Plant embryogenic calli (ECs) can undergo somatic embryogenesis to regenerate plants. This process is mediated by regulatory factors, such as transcription factors and specifically expressed genes, but the precise molecular mechanisms underlying somatic embryogenesis at the single-cell level remain unclear. In this study, we performed high-resolution single-cell RNA sequencing analysis to determine the cellular changes in the EC of the woody plant species Dimocarpus longan (longan) and clarify the continuous cell differentiation trajectories at the transcriptome level. The highly heterogeneous cells in the EC were divided into 12 putative clusters (e.g., proliferating, meristematic, vascular, and epidermal cell clusters). We determined cluster-enriched expression marker genes and found that overexpression of the epidermal cell marker gene GDSL ESTERASE/LIPASE-1 inhibited the hydrolysis of triacylglycerol. In addition, the stability of autophagy was critical for the somatic embryogenesis of longan. The pseudo-timeline analysis elucidated the continuous cell differentiation trajectories from early embryonic cell division to vascular and epidermal cell differentiation during the somatic embryogenesis of longan. Moreover, key transcriptional regulators associated with cell fates were revealed. We found that ETHYLENE RESPONSIVE FACTOR 6 was characterized as a heat-sensitive factor that negatively regulates longan somatic embryogenesis under high-temperature stress conditions. The results of this study provide new spatiotemporal insights into cell division and differentiation during longan somatic embryogenesis at single-cell resolution.
