Effects of orphanin on gastric motility in diabetic gastroparesis rats

Abstract:AIM:To explore the expression level of orphanin (OFQ) and its correlation with diabetic gastroparesis(DGP)and then to evaluate the effect of OFQ expression level on gastric emptying in DGP rats. METHODS: Ninety male SPF Wistar rats were first divided randomly into 2 groups: control (n=15) and experimental group (n=75). The controls were fed ordinary laboratory diet for 20 weeks, but the rats in experimental group were fed fat-and caloric-rich laboratory diet for 6 weeks, injected with small dose of streptozotocin, and then continuously fed fat-and caloric-rich laboratory diet until 20 weeks to develop diabetic models. Radionuclide gastric emptying method was used to screen DGP rats and diabetes mellitus (DM) rats. Gastric emptying (GET1/2) was measured by SPECT. Enzyme-linked immunosorbent assay was employed to examine the level of plasma OFQ. The diabetic gastroparesis rats were then subdivided into 3 groups, treated with OFQ at a dose of 0.6, 6.0 and 60.0 nmol/kg, respectively, once a day for 5 d. GET1/2 was measured again. RESULTS: GET1/2 was significantly delayed [(168.9±10.6) min] in DGP rats compared with that in the control group [(61.9±4.6) min] and DM rats [(66.5±2.3)min, P<0.05]. Similarly, 90 min residual rate was higher in DGP group [(89.5±2.9)%] than that in the control group[ (46.4±3.6)%]and DM group [(50.9±4.1)%] (P<0.05). Plasma level of OFQ in DGP group [(5.30±1.70) ng/L] was lower than that in the control [(9.83±5.70)ng/L] and DM group [(8.90±3.50) ng/L] (P<0.05). A negative correlation was found between plasma OFQ level and GET1/2 in DGP rats (r=-0.6726, P=0.004, <0.01). A significant decrease in GET1/2 in DGP rats was observed pre-and post-treatment with OFQ (P<0.05). A satisfactory treatment effect was achieved at a dose of 6.0 nmol/kg OFQ and further increase in OFQ dose did not result in a better effect. CONCLUSION: OFQ plays an important role in DGP. The plasma OFQ level is negatively correlated with GET1/2 in DGP rats. It elucidates that OFQ can promote the gastric emptying in the DGP rats.
